Age, sex and breed effect on laboratory parameters in natural Babesia canis infection

AB  - We tested the hypothesis that age, breed, and sex are related to hematology, biochemistry, acute phase proteins (APPs), seroreactivity and level of parasitemia in dogs with an acute phase response (APR) due to Babesia canis infection. The study enrolled 61 privately owned dogs that naturally acquired B. canis infection. Groups were formed according to the age: young dogs less than one year, and adult dogs more than one year old. Moreover, the group of males was compared to females and purebred to mixed breed dogs. Seroreactivity was tested with immunofluorescence antibody test, level of parasitemia with real-time polymerase chain reaction (real-time PCR), hematology, and biochemistry with automatic analyzers, serum amyloid A with enzyme-linked immunosorbent assay, fibrinogen with heat precipitation and ceruloplasmin and paraoxonase-1 with manual spectrophotometric methods. For protein separation agarose gel electrophoresis was used. The main changes in the whole population of B. canis-infected dogs were fever, pancytopenia, and change in APPs level. One-third of young, and 96% of adult dogs were seropositive (P < 0.001). The level of parasitemia was higher in the young dogs (P < 0.001). Erythroid lineage parameters (P < 0.01), and leukocytes (P < 0.05) were lower in the young, when compared to the adult dogs. Young dogs had lower total globulins (P < 0.001), β- and γ-globulins (P < 0.001), and higher α-globulins (P = 0.022) than adult dogs. Young dogs had higher concentrations of phosphate (P = 0.003) and cholesterol (P < 0.001) and lower amylase (P = 0.014) and lipase activity (P = 0.020) than adult ones. Male dogs had lower neutrophil count than females (P = 0.035), and purebred dogs had more band neutrophils than mixed breed dogs (P = 0.004). In conclusion, dogs with natural Babesia canis infection at a young age have more severe anemia and APR including leukopenia than adults. Male and purebred dogs might also have more severe APR than females and mix-breeds, as they have more pronounced changes related to the myeloid lineage.

AB  - Red blood cell osmotic fragility (RBCOF) refers to susceptibility to hemolysis in
vivo and in vitro. An increase in mean cellular hemoglobin concentration (MCHC)
also reflects in vivo and in vitro hemolysis. Common inflammatory infectious
diseases in dogs are often characterized by an increase in MCHC; however, it is
not known if there is a concomitant increase in RBCOF. The objective of this
study was to determine RBCOF in dogs with babesiosis, dirofilariosis,
parvovirosis, pyometra, and canine distemper and to investigate whether RBCOF
correlates with routine hematologic and biochemical parameters and lipoprotein
fractions. The diagnosis of canine infectious disease was made and complete
blood count, serum biochemical profiles, and lipoprotein electrophoresis were
determined according to current institutional protocols. RBCOF of healthy
control (n=8) and infected dogs (n=73) was measured with a phosphate-buffered
saline dilution test, and mean corpuscular fragility (MCF) was calculated. MCF
was increased in dirofilariosis, pyometra, and distemper; MCHC was increased
in all infected groups; HDL was decreased in babesiosis, pyometra, and
distemper compared with healthy controls. However, no correlation was found
between these parameters. On the other hand, hematological and biochemical
parameters, that are highly predictive of specific infections, correlated with MCF:
ALP for babesiosis (r=0.415, P=0.035), urea (r=-0.488, P=0.034) for
dirofilariosis, and leukocyte count (r=-0.820, P=0.046) for pyometra. Although
elevated MCHC levels in all samples from infected dogs indicate hemolysis,
RBCOF was elevated only in dirofilariosis, pyometra, and canine distemper,
leading to the conclusion that these apparently related phenomena are controlled
by different mechanisms.

AB  - Progressive tissue injury in canine leishmaniosis (CL) is related to the deposition of
immune complexes, which induces vasculitis and leads to endothelial dysfunction.
Homocysteine (Hcy) increase may worsen endothelial dysfunction, but data concerning
its concentration in different CL stages and links to the acute phase response and
oxidative stress are missing. We compared Hcy levels between dogs with mild (N=24)
and moderate CL without treatment (N=17) and treated with anti-Leishmania drugs
and vitamin B supplements (N=9). Dogs with moderate CL, regardless of therapy
administration, had more distinct clinical signs, lower erythron values, and a higher level
of acute-phase proteins (APPs), IgG against Leishmania spp., urea and creatinine, than
dogs with mild CL. Hcy values did not differ between stages, but treated dogs had the
lowest levels of Hcy. An inverse relationship existed between Hcy and the CL stage,
therapy, levels of IgG, and clinical pathology data. The only positive relationship existed
between Hcy and the erythron state. The disease stage and therapeutic intervention
were not related to the oxidative stress level, except in the case of paraoxonase-1/
Hcy ratio, indicating favorable conditions for antioxidative defense in treated dogs. In
conclusion, changes in Hcy levels indicated its possible involvement with endothelial
dysfunction and inverse relationship to tissue injury evaluated by APPs. Finally, Hcy
might be an early marker of favorable conditions for endothelium recovery in CL

AB  - Inflamatorni i degenerativni procesi na zglobovima konja se mogu postepeno razvijati,
pri čemu klinički znaci patoloških promena često nisu vidljivi. Cilj ove studije je
bio da se na osnovu rutinskog pregleda sinovijalne tečnosti (ST) poreklom iz metakarpo/
metatarzofalangealnih (MCP/MTP) zglobova radnih konja, koji su bili upućeni na
klanje i njenih biohemijskih karakteristika utvrdi da li je postojala lokalna inflamacija.
Studija je sprovedena na 6 konja starosti 3 do 12 godina (medijana 8), odnosno 24
MCP/MTP zgloba. Post mortem, iz svakog zgloba je uzorkovana ST. Kliničkim pregledom
pre klanja, nisu uočene promene u izgledu i funkciji mišićno-skeletnog sistema.
Urađen je rutinski pregled ST kojim je opisana boja, prozirnost, viskoznost i broj ćelija.
Određena je koncentracija sledećih biohemijskih parametara: ukupnih proteina,
albumina, aktivnost alkalne fosfataze (ALP), aspartat aminotransferaze (AST), kreatin
kinaze (CK), kao i koncentracije haptoglobina (Hp) i ceruloplazmina (Cp). Kod svih
uzoraka, ST je bila bistra, providna, viskozna i blede do bledo žute boje. Najmlađi konj,
star tri godine, je u sva četiri pregledana zgloba imao ukupan broj ćelija 200-890/μL,
što odgovara fiziološkim vrednostima. Dva konja su imala povećan broj ćelija na jednom,
odnosno dva zgloba (1 400-2 560/μL), dok su tri konja imala promene na sva četiri
zgloba (1 100-2 200/μL). Ispitivani biohemijski parametri se nisu razlikovali u uzorcima
ST iz zglobova sa ukupnim brojem ćelija u okviru fizioloških vrednosti i onih kod
kojih je broj ćelija bio povećan. Pozitivne, umerene korelacije su postojale između koncentracije
proteina i aktivnosti ALP, koncentracije proteina i albumina, aktivnosti AST
i CK. Ispitivanjem ST radnih konja, dokazano je da inflamatorne promene u MCP/MTP
zglobovima nisu prisutne. Nalaz povećanog broja ćelija u ST bez inflamatornih promena
zahteva dodatna ispitivanja.
