Molecular Investigation of Eimeria Species in Broiler Farms in the Province of Vojvodina, Serbia

AB  - Coccidiosis is a significant poultry disease caused by the Eimeria species. This study aims to
determine the prevalence of Eimeria spp. on broiler farms in Vojvodina, along with the identification
of parasite species, and assess the implemented biosecurity measures. The study was conducted on
100 broiler chicken farms (28 small-sized; 34 medium-sized; 38 large-sized farms) from June 2018 to
December 2021. One pooled sample of faeces was collected from three to six-week-old chickens from
each farm, and assessment of biosecurity measures was carried out using a questionnaire. Using the
PCR method, DNA of Eimeria was found in 59 samples (59%), while 41 samples (41%) were negative.
Four species of Eimeria were identified, and their prevalence was the following: E. acervulina (37%),
E. maxima (17%), E. mitis (25%) and E. tenella (48%). A significant difference (p < 0.05) was established
in the number of oocysts in flocks from small-sized farms compared to medium-sized farms. It was
found that regular implementation of disinfection, disinsection and deratisation measures, as well as
all the biosecurity measures, can significantly reduce the occurrence of coccidiosis. These results will
help to develop better strategies for the control and prevention of coccidiosis on farms.

Evaluation of the Effects of New Combined Adsorbent on Occurrence of Pathological Lesions in Aflatoxin B1‐Challenged Broiler Chickens

AB  - This experiment was performed to investigate the protective effect of a new combined adsorbent (bentonite + yeast cellwall extract) previously tested in vitro, on the occurrence of pathological lesions in broilers fed a diet contaminated with aflatoxinB1 until 21 days of age. A total of 96 one-day old Ross 308 hybrid broiler chickens were divided into four groups: group I (control),group II (5 g/kg adsorbent), group III (5 g/kg adsorbent + 2 mg/kg aflatoxin B1), and group IV (2 mg/kg aflatoxin B1). Grossly,AFB1 and AFB1+adsorbent fed birds showed enlargement, friable texture of liver and enlargement and pallorness of kidneys. Spleenrevealed size reduction. No gross-pathological changes were observed in control and group II. Histopathologically, lesions wereobserved in aflatoxicated groups. Liver revealed vacuolar cell degeneration, periportal and perivascular infiltration of mononuclearcells. Kidneys revealed mild to moderate degree of haemorrhages, tubular epithelial necrosis and infiltration of mononuclearcells along with heterophils. In group IV, catarrhal enteritis characterized by desquamation of epithelial cells were found in theduodenum. A milder form of gross and histopathological lesions was seen in group III. In conclusion, the present study revealedthat supplementation of a new combined toxin binder product in the concentration of 5 g/kg could not completely ameliorateaflatoxicity in broilers, although it met the stringent European regulation requirements for the minimum of 90% aflatoxin bindingefficiency in in vitro study.

Influence of Anticoccidials on Oxidative Stress, Production Performance and Faecal Oocyst Counts in Broiler Chickens Infected with Eimeria Species

AB  - The influence of certain anticoccidial drugs on oxidative stress in broiler chickens infected with Eimeria species was assessed. There were two untreated (uninfected and infected), and three groups infected and treated with anticoccidials. The first treated group (Ro) was given robenidine, the 2nd a herbal anticoccidial (Herb) and the 3rd the combination of robenidine and the herbal anticoccidial (Ro+Herb). All infected groups were on day 14 challenged with oral inoculation of oocysts. The activities of catalase (CAT), superoxide dismutase (SOD) and glutathione S-transferase (GST), and the concentration of malondialdehyde (MDA) were estimated in blood taken on days 21 and 40. The oocyst numbers were calculated per gram, and chicken body weight and feed conversion ratio (FCR) measured. The activities of CAT, GST and the level of MDA were significantly lower (P<0.05), whilst the activity of SOD was higher in infected chickens treated with anticoccidials (P<0.05) in comparison to those untreated. The most prominent change in the parameters of oxidative stress was recorded in the Ro+Herb group. In chickens treated with anticoccidials body weight was significantly higher (P<0.05), and the FCR and the oocyst counts significantly lower (P<0.05) than in untreated chickens. Oocyst counts were lower in the Ro and Ro+Herb groups than in the Herb group. Our study demonstrated that both anticoccidial substances exerted antioxidant and anticoccidial effects.

Infection with Strongyloides papillosus in sheep: Effect of parasitic infection and treatment with albendazole on basic haematological parameters

AB  - The aim of this study was to determine and evaluate the basic haematological parameters in conditions of natural infection of sheep with Strongyloides papillosus, as well as after the administration of antihelminthic albendazole (ABZ). Based on the intensity of infection with S. papillosus the sheep were divided into three groups: mild, moderate and high, and after that the sheep received a single dose of ABZ of 5mg/kg per body weight, per orally. Sampling of faeces and blood for parasitological and haematological assaying respectively, was performed on the 0 and the 21st day after the treatment with ABZ. The presence of parasitic infection with S. papillosus leads to a decrease of erythrocyte count, while the lowest values were established in the group with the highest intensity of parasitic infection (p ˂ 0.001). After treatment with ABZ the decrease of erythrocyte count was more prominent, which was, based on comparison with control groups C1 and C2, unequivocally established to be the consequence of treatment with ABZ. Detected values of haematocrit and erythrocyte indices indicated the presence of parasitic infection: the lowest values were established in the group with the highest intensity of parasitic infection. After treatment with ABZ haematocrit levels in control group C2 were statistically significantly lower compared to the control group C1 (p ˂ 0.001). In the presence of parasitic infection, the neutrophil and eosinophil counts increased almost linearly up to the value of 44.24±2.50% and 13.29±0.61% respectively, in the group of sheep with the highest intensity of parasitic infection (p ˂ 0.001; compared to control group C1). After treatment with ABZ the decrease of the number of these white blood cells is statistically significant (p ˂ 0.001). Bearing in mind our previous research and the connection of disbalanced redox equilibrium after the treatment with ABZ with changes, it is necessary to include antioxidative substances in the anti-parasitic treatment protocols.
AB  - Cilj ovog istraživanja bio je da se utvrde i procene osnovni hematološki parametri u uslovima prirodne infekcije ovaca sa Strongyloides papillosus, kao i nakon primene antihelmintika albendazola (ABZ). Na osnovu intenziteta infekcije sa S. papillosus ovce su podeljene u tri grupe: niski, srednji i visoki intenzitet infekcije, a zatim su ovce jednokratno dobile peroralno ABZ, u terapijskoj dozi od 5 mg/kg telesne mase. Uzorkovanje fecesa za parazitološka i za hematološka ispitivanja obavljeno je nultog i 21. dana od primene ABZ. Utvrđeno je da prisustvo parazitske infekcije sa S. papillosus dovodi do pada broja eritrocita, pri čemu su najniže vrednosti utvrđene u grupi sa najvećim intenzitetom parazitske infekcije (p ˂ 0,001). Nakon terapije sa ABZ pad broja eritrocita je izraženiji, što je nesumnjivo nastalo kao posledica terapije ABZ (na osnovu poređenja C1 i C2). Utvrđene vrednosti hematokrita i eritrocitnih indeksa su ukazivali na postojanje parazitske infekcije; najniže vrednosti su utvrđene kod grupe sa najvećim intenzitetom parazitske infekcije. AB  - This study examined the activities of aspartate-aminotransferase (AST), gamma-glutamyl-transferase (GGT) and lactate-dehydrogenase (LDH) in the blood serum of 45 dairy Simmental cows divided into three groups according to production period. The first group (n=15) consisted of late pregnant dairy cows, the second group (n=15) cows in the early lactation, and the third group (n=15) cow in mid lactation. The significant higher activity (P lt 0.05) of AST, GGT and LDH were determined in the early lactation period than in dry period and during full lactation. Research results showed possibility of mild degree of hepatic lesions, probably due to fat infiltration in early lactation cows. Serum AST enzyme activities were significant correlated (P lt 0.05) with GGT and LDH activities and may be most sensitive indicator.
AB  - U ovom radu je određivana aktivnost aspartat-aminotransferaze (AST), gama-glutamat-transferaze (GGT) i laktat dehidrogenaze (LDH) u krvnom serumu kod 45 simentalskih mlečnih krava, podeljenih u tri grupe u zavisnosti od produktivnog perioda. Prvu grupu (n=15) su činile visoko gravidne krave, drugu grupu (n=15) krave u ranoj laktaciji, a treću grupu (n=15) krave tokom pune laktacije. Statistički značajno veće aktivnosti AST (P lt 0.05),GGT (P lt 0.05) i LDH (P lt 0.05) u krvnom serumu su utvrđene kod krava u ranoj laktaciji u odnosu na aktivnosti ovih enzima u serumu kod zasušenih krava i krava u punoj laktaciji. Dobijeni rezultati ukazuju na mogućnost blagog stepena oštećenja ćelija jetre, odnosno masnu infiltraciju hepatocita kod krava na početku laktacije. Serumske aktivnosti AST su bile u značajnoj korelaciji (P lt 0.05) sa aktivnostima GGT i LDH u krvnom serumu pa se AST može smatrati pouzdanim indikatorom u tvrđivanju funkcionalnog stanja jetre kod mlečnih krava tokom peripartalnog perioda i pune laktacije.

AB  - Data concerning the effect of probiotics supplementation on many parameters concurrently at the same cows are lacking. Therefore, the objective of this experiment was to investigate the effects of Saccharomyces cerevisiae feeding on rumen, blood and milk parameters together in high- producing dairy cattle during the transition and early lactation period. Sixteen clinically healthy Holstein cows were divided into 2 groups: a control group of 6 cows and a probiotics-fed group of 10 cows. Rumen fluid and blood samples were collected 21 days before the expected calving as well as 7, 15, 30, 45 and 60 days- in-milk (DIM). Milk yield for each animal was recorded every 2 weeks. Individual milk samples were collected 15, 30, 45 and 60 DIM. Ruminal pH and rumen ammonia nitrogen were significantly lower, whereas total volatile fatty acids were significantly higher in yeast-fed animals compared with controls throughout the study. Serum concentrations of total proteins and globulins were higher, while albumins were lower in the yeast-treated group. Serum glucose levels were significantly higher in yeast-supplemented animals. Serum triglycerides, high density lipoproteins, and low density lipoproteins concentrations were lower, with cholesterol being significantly lower in the treated group. Milk production and milk fat percentage were higher, whereas milk protein percentage and somatic cell count were decreased in yeast-supplemented cows throughout the study. These results suggest that supplementation of S. cerevisiae to dairy cows rations during transition and early lactation period improve their health and milk production parameters.
AB  - Podaci koji bi se odnosili na uticaj suplementacije probiotika na više parametara istovremeno na istim grlima nedostaju u literaturi. Stoga je cilj ovog eksperimenta bio da se ispita uticaj Saccharomices cerevisiae u obroku/hrani na parametre rumena, krvi i mleka zajedno u visoko-proizvodnim mlečnim govedima tokom tranzicije i početkom laktacije. Šesnaest klinički zdravih holštajn krava je podeljeno u 2 grupe: kontrolna grupa od 6 krava i grupa od 10 krava hranjenih probiotikom u obroku. Buražna tečnost i uzorci krvi su sakupljeni 21 dan pre očekivanog teljenja, kao i 7, 15, 30, 45 i 60 dana tokom laktacije (days in milk - DIM). Prinos mleka za svaku životinju zabeležen je svake 2 nedelje. Pojedinačni uzorci mleka su prikupljeni 15, 30, 45 i 60 DIM. pH buraga i buražni amonijačni azot su bili značajno niži, dok su ukupne isparljive masne kiseline bile značajno veće kod životinja hranjenih kvascem u poređenju sa kontrolama kroz celu studiju. Serumske koncentracije ukupnih proteina i globulina bile su više, dok su koncentracije albumina bile niže u grupi sa kvascem. Serumski nivoi glukoze bili su značajno viši kod životinja sa dodatkom kvasca. Trigliceridi u serumu, koncentracije lipoproteina visoke i niske gustine bile su niže, sa holesterolom koji je bio znatno niži u tretiranoj grupi. Proizvodnja mleka i procent mlečne masti bili su viši, dok je sadržaj proteina mleka i somatskih ćelija bio niži u krava hranjenih sa dodatkom kvasca kroz celu studiju. Ovi rezultati ukazuju na to da dodatak S. cerevisiae u obrocima muznih krava tokom tranzicije i rane laktacije poboljšava njihovo zdravlje i parametre proizvodnje mleka.

AB  - Laboratory animals are essential in research, disease diagnostics and
pharmaceutical industry in the production of vaccines and serums, as well as
in potency and safety testing of pharmaceutical products. Like domestic - farm
animals and pets, laboratory animals originate from ancestors that lived in the
wild. For breeding in artifi cial conditions of laboratory and use in experiments
for scientifi c and other purposes rodents are most acceptable. As a rule, these are
animals that are characterized by small size and high fertility, so it is economically
to breed and use.
The most frequent and most important species of laboratory animals is the
mouse. Good knowledge of physiological, medical, genetical, nutritional,
behavioural and other characteristics and needs of laboratory mice is essential
for proper growth and use. Questions and principles of ethics and ensuring the
welfare of laboratory mice in breeding and utilization are important both for
scientifi c and professional as well as for the general public. Principles “3Rs”
obligate scientists and professionals to use their knowledge in the development
of techniques and technologies in breeding and utilization of laboratory mice
in order to be more effective and more humane. In Serbia it is needed to make
technological and ethical improvements in breeding and utilization of laboratory
animals and thus mice, in scientifi c and other purposes, in accordance with
international conventions and scientifi c and professional achievements in the
fi eld of laboratory animal science.
