AB  - Examination of cadmium (Cd) toxicity and disulfiram (DSF) effect on liver was focused on oxidative stress (OS), bioelements status, morphological and functional changes. Male Wistar rats were intraperitoneally treated with 1mg CdCl2/kg BW/day; orally with 178.5 mg DSF/kg BW/day for 1, 3, 10 and 21 days; and co-exposed from 22nd to 42nd day. The co-exposure nearly restored previously suppressed total superoxide dismutase (SOD), catalase (CAT) and increased glutathione peroxidase (GPx) activities; increased previously reduced glutathione reductase (GR) and total glutathione-S-transferase (GST) activities; reduced previously increased superoxide anion radical (O-2(center dot-)) and malondialdehyde (MDA) levels; increased zinc (Zn) and iron (Fe), and decreased copper (Cu) (yet above control value), while magnesium (Mg) was not affected; and decreased serum alanine aminotransferases (ALT) levels. Histopathological examination showed signs of inflammation process as previously demonstrated by exposure to Cd. Overall, we ascertained partial liver redox status improvement, compared with the formerly Cd-induced impact.

The effect of carvacrol on inflammatory pain and motor coordination in rats

AB  - Carvacrol is a monoterpenic phenol and an active ingredient of the plant essential oils of the family Lamiaceae. We have investigated the analgesic effect of carvacrol, the possible dependence of the effect in relation to animal sex, and the impact of carvacrol on motor coordination in rats. Hyperalgesia was induced by formalin (1.5%), which was administered SC in the upper lip of rat. Hyperalgesia and effects of carvacrol and indomethacin were measured by using the orofacial formalin test. The influence on motor coordination in animals treated with carvacrol was investigated by using the rota-rod test. Carvacrol administered PO in pre-treatment (45 min. prior to formalin) at a single dose of 50, 75 and 100 mg /kg BW, in the male, 50 and 100 mg /kg BW, in female rats caused a dose-dependent antinociceptive effect. This effect of carvacrol was significantly higher (P lt 0.01, P lt 0.001) in male rats. Compared with indomethacin administered during pre-treatment (2 mg/kg, PO), carvacrol (100 mg/kg) exhibits significantly higher (P  lt 0.05 and P  lt 0.001) antinociceptive effect on formalininduced hyperalgesia in male rats. In the rota-rod test carvacrol did not disturb the motor coordination in male rats, nor the dose of carvacrol with clear antinociceptive properties exhibited depressive effect on the CNS of treated rats. Keeping in mind that the monoterpene carvacrol is of plant origin, with potentially less side effects and without residues, it is realistic to expect the possibility of its therapeutic use in the treatment of inflammatory pain in animals.
AB  - Karvakrol je monoterpenski fenol i sastavni je deo esencijalnih ulja biljaka iz familije Lamiaceae. Cilj ovog istraživanja bio je da se ispita analgetičko dejstvo karvakrola, moguće razlike u farmakološkom odgovoru između polova, i uticaj karvakrola na koordinaciju kretanja kod pacova. Hiperalgezija je izazvana formalinom (1,5%) koji je aplikovan s.c. u gornju usnu pacova. Hiperalgezija i efekti karvakrola i indometacina mereni su orofacijalnim formalinskim testom. Uticaj na motornu koordinaciju kod životinja tretiranih karvakrolom ispitali smo rota-rod testom. Karvakrol primenjen p.o. u pretretmanu (45 min. pre aplikacije formalina) u dozi od 50, 75 i 100 mg/kg t.m. kod mužjaka i dozi od 50 i 100 mg/kg t.m. kod ženki pacova, prouzrokuje dozno zavisan antinociceptivni efekat. Ovaj efekat karvakrola je značajno veći (P lt 0,01, P lt 0,001) kod pacova muškog pola. U poređenju sa indometacinom aplikovanim u pretretmanu (2 mg/kg, p.o.), karvakrol (100 mg/kg) ispoljava značajno veći (P lt 0,05 i P lt 0,001) antinociceptivni efekat na formalinom izazvanu hiperalgeziju kod mužjaka pacova. U rota-rod testu karvakrol nije narušio motornu koordinaciju kod mužjka pacova, niti doza karvakrol sa jasnim antinociceptivnim svojstvima deluje depresivno na CNS tretiranih pacova. Imajući u vidu da je karvakrol monoterpen biljnog porekla sa malo potencijalnih neželjenih dejstava i bez rezidua, realno je očekivati mogućnost njegove primenu u terapiji inflamatornog bola kod životinja.

AB  - The objective of our study was to examine testicular toxicity of cadmium (Cd), focusing on oxidative stress (OS), essential metals and androgenic status and morphological changes. Male Wistar rats [controls and four Cd-subgroups (n = 6) organized according to the exposure (1, 3, 10 and 21 days)] were intraperitoneally (i.p.) treated with 1 mg CdCl2/kg/day. Testicular Cd deposition was noticed from the 1st day. After 10 and 21 days, copper (Cu) and iron (Fe) increased by 60-109% and 43-67%, respectively, while zinc (Zn) decreased by 24-33%. During 121 days of the exposure, decrease in testicular total superoxide dismutase (SOD) and total glutathione-s-transferase (GST) activities occurred gradually by 30-78% and 15-84%, respectively, while superoxide anion radical (O-2(center dot-)) increased gradually by 114-271%. After 10-21 days, decrease in testicular catalase (CAT) activity appeared by 13-31%. After 21 days, malondialdehyde (MDA) decreased by 44% and the ratio of oxidized glutathione/reduced glutathione (GSSG/GSH) increased by 130% in testes of the rats exposed to Cd. Additionally, decreased testicular testosterone level and the relative testes mass, along with induced microscopic and macroscopic changes were occured, what can be explained as the consequence of instantly developed OS, impaired essential metals status and Cd testicular deposition.

Effect of eugenol on hematological parameters in rats

AB  - Investigations covered the possible hematotoxic effect of eugenol in rats following two-week and four-week continuous p.o. application. An experiment was conducted on 72 maleWistar rats divided into six groups. Four groups were treated with different doses of eugenol (10 mg/kg bm/day, 50 mg/kg/day, 200 mg/kg/day and 400 mg/kg bm/day), the control group was administered a vehiculum (0,5 % methylcellulose, 20 % propylene glycol and water), and the sixth group was the absolute untreated control. Eugenol and the vehiculum were administered using a gastric probe in a volume of 1 ml/100 g body mass of rat. Blood was sampled using cardiac puncture on days 14 and 28 of the experiment in order to determine hematological parameters (hematocrit, number of erythrocytes, MCV, haemoglobin concentration, MCH, number of leukocytes, leukocyte formula, and number of thrombocytes). The results have shown that eugenol administered over 14 and 28 days in doses of 10, 50, 200 and 400 mg/kg bm/day has no hemolytic activity. AB  - Ispitivan je mogući hematotoksični efekat eugenola kod pacova posle dvonedeljne i četvoronedeljne kontinuirane p.o. primene. Ogled je izveden na 72 mužjaka pacova soja vistar podeljenih u šest grupa. Četiri grupe tretirane su različitim dozama eugenola (10 mg/kg tm/dan, 50 mg/kg/dan, 200 mg/kg/dan i 400 mg/kg tm/dan), peta kontrolna grupa dobijala je vehikulum (0,5 % metil-celuloza, 20 % propilen-glikol i voda), a šesta je bila netretirana, apsolutno kontrolna. Eugenol i vehikulum aplikovani su gastričnom sondom u volumenu od 1 ml/100 g telesne mase pacova. AB  - Дирофилариоза је паразитско обољење, пре свега − паса, изазвано нематодом Dirofilaria immitis. Током последњих година на просторима Србије код паса и мачака све чешће се документује налаз паразита Dirofilaria immitis, било као проузроковача изненадног угинућа, било као споредан налаз на обдукцији. Пас је једини сисар код којег се Dirofilaria immitis јавља уобичајено и уједно представља најзначајнији резервоар овог паразита, мада и друге животиње могу бити инфициране овим паразитом, a у ретким ситуацијама може бити инфициран и човек. Патоморфолошким прегледом, код 36 паса и две мачке које су обдуковане на катедрама за патолошку морфологију и судску ветеринарску медицину Факултета ветеринарске медицине у Београду пронађени су адултни облици Dirofilaria immitis, али су они у само четири случаја били узрок изненадног угинућа. Након макроскопског прегледа узети су узорци ткива различитих органа за патохистолошка испитивања. Материјал за патохистолошку анализу је фиксиран у 10% пуферисаном формалину, и после уобичајене процедуре калупљен у парафин. Парафински исечци дебљине око 5 μm су бојени хематоксилином и еозином (HE) и Periodic-Acid-Schiff техником (PAS), толуидин плавим (TB) и Weigert van Gieson методом. Услед присуства паразита у десној комори и плућној артерији последично долази до парцијалне васкуларне блокаде, развоја интензивне плућне хипертензије и интензивног оптерећења десне срчане коморе, као и њене последичне хипертрофије. Временом, у хроничном току болести може доћи до развоја слабости − дилатације десног срца. На плућној артерији и њеним огранцима развија се артеритис и периартеритис, а микрофиларије изазивају имунопосредован гломерулонефритис. Код младих паса и мачака може доћи до развијања „синдрома вене каве“. Аберантно, адултни облици се могу наћи и у кожи, оку, мозгу, кичменом каналу, перитонеалној дупљи и др. Третман дирофилариозе може бити хируршки или медикаментозни. Медикаментозни третман може бити: а) адултицидни, б) микрофиларицидни и в) превентивни.
AB  - Dirofilariosis is parasitic disease mostly of dogs, caused by nematode Dirofilaria immitis. During past
few years on the teritory of Republic of Serbia, this parasite is diagnosed in dogs and cats either as a direct cause of death or as accidental finding on the post mortem exam.Dog is the only mammal in which Dirofilaria immitis is commonly found, and at the same time it is the most important reservoir of this parasite, although the other animals, in rare cases even a human, can also be infected with this parasite. Adult forms of Dirofilaria immitis were found during pathomorphological examination of 36 dogs and 2 cats, but these parasites were direct cause of sudden death in only 4 cases. These post mortem examinations were performed on the Department of pathology and Department of forensic medicine, on Faculty of Veterinary Medicine of Belgrade. After macroscopic examination tissue samples of different organs were taken. Tissue samples for pathohistologic analysis after fixation in 10% buffered formalin, and routine procedure, were embeded in paraffin blocks. Pathohistological preparations were stained with hematoxylin eosine (HE) and Periodic-Acid-Schiff staining (PAS), toluidine blue (TB), and Weigert van Gieson (WvG). The presence of parasites in right cardiac ventricle causes partial vascular blockade, development of intensive pulmonary hypertension, and intensive load of right cardiac ventricle, as well as its consenquentional hypertrophy. In time, during the chronic course of disease the dilatation of right heart can develop. Arteritis and periarteritis
develop on the pulmonary artery and its branches. Microfilaria also causes immuno-mediated glomerulonephritis. In young dogs and cats “caval syndrome” can develop. Adult forms can aberrantly be found beneath the skin, eye, brain, spinal canal, peritoneal cavity, etc. Treatment of dirofilariosis can be surgical and medicamentous. Medicamentous treatment can be: a) adulticide, b) microfilaricide, and c) preventional. Adulticide treatment consists of melarsomin hydrochloride, after symptomatic therapy. Macrocyclic lactons, milbecins and avermectins: moxidectin, ivermectin, selamectin etc. are used for microfilaricidal and preventive treatment in form of topical and injectible drugs according to different protocols.
