Fumagillin-induced chromosome aberrations in mouse bone-marrow cells
Hromozomske aberacije prouzrokovane fumagilinom u ćelijama kostne srži miša

The genotoxicity of fumagillin was evaluated assessing the mitotic index and chromosome aberrations in mouse bone-marrow cells. Mice were given fumagillin orally in doses 5, 10 and 20 mg/kg bw. All doses significantly (p lt 0.001) reduced the mitotic index. The medium and maximum doses led to significant (p lt 0.01 and p lt 0.001, respectively) increases in numeric aberrations. The highest dose induced both structural and numeric aberrations (p lt 0.001), and insertions on the first pair of autosomes that were amplified in the 1C and 1E regions. These results pointed to the genotoxic potential of fumagillin in the range of medium and maximum doses applied.
Ispitivana je genotoksičnost fumagilina praćenjem mitotskog indeksa i hromozomskih aberacija u ćelijama kostne srži miša. Miševi su tretirani p.o. dozama 5, 10 i 20 mg/kg t.m. Sve ispitivane doze fumagilina signifikantno smanjuju mitotski indeks (p lt 0,001). Srednja i najveća doza signifikantno (p lt 0,01, odnosno p lt 0,001) povećavaju učestalost numeričkih aberacija. Najviša doza indukuje strukturne i numeričke aberacije visoko signifikantno (p lt 0,001). Fumagilin izaziva pojavu insercija na prvom paru autozoma. Radi se o amplifikaciji u 1C regionu segmenta označenog kao 1C5 traka, odnosno o dva segmenta (1E3 i 1E4 trake) u 1E regionu. Rezultati ukazuju da fumagilin u opsegu srednje i najveće doze ispoljava genotoksičnost.
