dc.description.abstract | UVOD: Herpesvirusne infekcije su prisutne u populacijama konja širom sveta i najčešće ih izazivaju konjski herpesvirus 1 (EHV-1) i konjski herpesvirus 4 (EHV-4). Konjski herpesvirus 1 predstavlja jedan od najznačajnijih uzročnika pobačaja gravidnih kobila, povećanog mortaliteta novorođene ždrebadi, respiratornog oboljenja mladih jedinki i neuroloških poremećaja. Konjski herpesvirus 4 izaziva respiratornu infekciju konja tzv. rinopneumonitis koji može naneti velike ekonomske gubitke u konjarstvu zbog smanjene radne sposobnosti životinja usled dugog perioda oporavka. CILJ: Identifikacija i molekularna karakterizacija sojeva EHV-1 i EHV-4 u ukupno 40 uzoraka limfnih čvorova, slezine, kičmene moždine i produžene moždine poreklom od konja sa teritorije Republike Srbije. METOD: Identifikacija EHV-1 i EHV-4 primenom metoda lančane reakcije polimeraze (PCR) sa sekvenciranjem delова genoma navedenih virusa koji kodiraju sintezu glikoproteina B (gB) primenom metode po Sanger-u i njihova filogenetska analiza. REZULTAT: Nukleinska kiselina EHV-1 je detektovana u ukupno 36 (90%) uzoraka, a EHV- 4 u ukupno 9 (22,5%) ispitivanih uzoraka, dok je prisustvo mešovite infekcije sa EHV-1 i EHV-4 zabeleženo u ukupno 9 (22,5%) uzoraka. Filogenetskom analizom dobijenih sekvenci navedenih virusa zabeležena je značajna sličnost sa sojevima EHV-1 i EHV-4 izolovanih kod konja u Turskoj i Japanu. ZAKLJUČAK: Dobijeni rezultati potvrđuju opravdanost primene molekularnih metoda virusološke dijagnostike u cilju brze i precizne identifikacije EHV-1 i EHV-4 i značajno doprinose boljem poznavanju epizootiološke situacije na teritoriji Republike Srbije što predstavlja osnovu za preduzimanje preventivnih mera za uspešnu kontrolu oboljenja konja izazvanih navedenim virusima. | sr |
dc.description.abstract | INTRODUCTION: Herpesviral infections are widely distributed in equine populations worldwide and are most often caused by equine herpesvirus 1 (EHV-1) and equine herpesvirus 4 (EHV-4). Equine herpesvirus 1 is one of the most significant causative agents of abortion, neonatal foal mortality, respiratory disease of young horses as well as neurological disease. Equine herpesvirus 4 causes a respiratory infection ie. rhinopneumonitis that leads to major economic losses in the equine industry as a result of prolonged recovery period of diseased horses. AIM: Identification and molecular characterization of EHV-1 and EHV-4 strains in total of 40 samples of lymph nodes, spleen, spinal cord and medulla collected from horses originating from The Republic of Serbia. METHOD: Identification of EHV-1 and EHV-4 by polymerase chain reaction (PCR) with nucleotide sequencing of the parts of the genome coding the synthesis of glycoprotein B (gB) and subsequent phylogenetic analysis. RESULT: The nucleic acid of EHV-1 was detected in 36 samples (90%) and EHV-4 in total of 9 examined samples (22.5%). The presence of mixed infection with both viruses was observed in 9 (22.5%) samples. The phylogenetic analysis of the obtained nucleotide sequences of EHV-1 and EHV-4 identified in this study showed high similarity with Turkish and Japanese EHV-1 and EHV-4 strains. CONCLUSION: The obtained results confirm the applicability of molecular diagnostic methods in rapid and precise identification of EHV-1 and EHV-4 and provide insight to the epizootiological situation in The Republic of Serbia as a basis for undertaking of preventive measures in order to effectively control herpesviral diseases of horses. | sr |
