AB - Proliferative enteropathy, also known as proliferative ileitis, is considered to be one of the most economically important diseases in pig production worldwide. The estimated losses per affected growing pig usually range from US $1 to $5. The disease is caused by Lawsonia intrawellularis, a Gram-negative, obligately intracellular bacterium. Control of the disease can be achieved with the use of vaccines or antibiotics. Recently there has been an increase in the efforts in the control of certain pathologies of the digestive system with phytogenic additives. The aim of this work was to assess the effects of a phytogenic additive on the histomorphometric characteristics of the intestines in weaned pigs with a subclinical infection with L. intracellularis acquired spontaneously. Histomorphometry analysis showed that crypt depth was significantly shorter (P <0.05), and the villus-height-to-crypt-depth ratio (P<0.05) significantly greater in the treatment group than the control. This improvement in the histological parameters of the intestine, considered to be indicators of its health, proved the positive effect of the tested additive on the digestive system in pigs.
značajno (P<0,05) pliće, a količnik visine vilusa i dubine kripti značajno veći (P<0,05)
u tretiranoj grupi prasadi nego u kontroli. Ovo poboljšanje histoloških parametara
creva, koje se smatraju indikatorima njegovog zdravlja, dokaz je pozitivnog efekta ispitivanog aditiva na digestivni sistem svinja.

AB - Lawsonia intracellularis is known to cause proliferative enteropathy (PE), one of the economically most important swine diseases with global distribution. Not unlike other enteric diseases, PE is a frequent indication for antibiotic therapy. However, their unjustified use leads to an emerging problem antimicrobial resistance. Thus, the aim of this research was to assess if a phytogenic additive may replace antibiotics in the control of PE in 144 weaned piglets (72 treated and 72 controls) naturally infected with L. intracellularis. The quantity of L intracellularis faecal shedding was monitored by real-time polymerase chain reaction (PCR) assay in faecal samples on day 0, 14 and 28, whilst the level of the ileum damage was determined by immunohistochemistry (IHC) assay performed on gut sections. Real-time PCR assay revealed that cycle-threshold (Ct) values in the treatment group increased significantly over time and were higher than in the control. These results indicate that the use of the phytogenic additive decreases the faecal excretion of L. intracellularis both throughout the experiment and in comparison to the control. The expression of the L. intracellularis antigen in IHC assay was lower in treated animals, implying that the additive leads to the decrease in the pathogen quantity in the ileum. Significantly higher feed conversion ratio was recorded in the treatment group. The results indicate that the phytogenic additive may be beneficial in the control of PE, but additional research is necessary to assess its use in various pig categories and define the optimum concentrations.

AB - Micromorphological and anatomical analyses of the indumentum of Nepeta rtanjensis, a Serbian/local endemic species, were performed by means of light and electron microscopy and described in detail. The leaves and stems bear numerous glandular trichomes, both peltate and capitate, as well as non-glandular unbranched trichomes. A basal epidermal cell, a short stalk cell and a large round head of four secretory cells constitute one peltate trichome. Capitate trichomes are short-and long-stalked and can be divided into two types, based on the number of glandular head cells, and further into a few subtypes according to the length and number of cells in stalk. Capitate trichomes of smaller length with one-cellular or with a bicellular head have been previously reported in different Nepeta species; however, according to our best knowledge, no existing study has reported the presence of long capitate trichomes on vegetative organs in any other species of this genus.

AB - Control of swine dysentery with antibiotics is often ineffective due to the resistance of Brachyspira hyodysenteriae. The potential of some herbal-based components against B. hyodysenteriae was previously studied in vitro. This study aims at the evaluation of in vivo efficacy of phytogemc feed additives in the control of swine dysentery. The study involved 64 seven-week old weaned pigs allotted to 4 groups: two were fed on feed supplemented with either Patente Herba(R)or Patente Herba(R) Plus, the third received tiamulin (positive control), while the negative control was not given antibiotics or additives. Fecal consistency was recorded daily. The presence of B. hyodysenteriae in the feces was investigated weekly using microbiological assays and the PCR test. Weight gain and feed conversion ratio were calculated for each week, and for the whole experiment. B. hyodysenteriae was detected in all samples by both methods. The additives showed efficacy in the prevention and control of swine dysentery as only normal and soft stool was observed in the treated groups. By contrast, in the negative control all feces categories were detected. Frequencies of feces categories significantly differed (p<0.001) between feed-supplemented groups and the negative control. Efficacy of both additives in the prevention of SD is comparable to tiamulin, based on insignificant differences in the frequency of the various feces categories. Beneficial effects of both additives resulted in significantly (p <= 0.05) higher weight gain and lower feed conversion ratio in comparison to the negative control. The average weight gains between additive-fed groups and tiamulin-treated group did not differ significantly.

The average weight gains between additive-fed groups and tiamulin-treated group did not differ significantly.",

AB - The success of antioxidant therapy in hyperthyroidism implies that disease is mediated by oxidative stress, which is known as one of the causing agents of ageing, degenerative diseases, and cancer. The main objective of our study was to determine possible protective effects of methanolic extract of N. rtanjensis in triiodothyronine (T3)-induced DNA breaks of human lymphocytes under in vitro conditions, based upon plant antioxidant capacity related to its phytochemical profile, mainly its polyphenolic complex. The total phenolic and flavonoid content and the antioxidant activity using in vitro 1,1-dyphenyl-2- picrylhydrazyl reagent (DPPH) was determined in methanolic extracts of plant leaves and flowers. The phenolic compound content of 62.73±1.80mg of GaA/g, exhibited solid antioxidant activity (IC50= 112.59±0.95μg/ml). The antigenotoxic activity of 0.2, 0.5 and 1.0mg/ml N. rtanjensis methanol extracts mixture with 100µM of T3 was studied in human lymphocytes in vitro using the Comet assay. It is supposed that the antigenotoxicity of N. rtanjensis methanol extracts was caused by high presence of chlorogenic acid, rosmarinic acid and rutin, all known as efficient antioxidant bioactive compounds, which were determined by ultrahigh-pressure liquid chromatograph with MS/MS Mass Spectroscopy (UHPLC/-HESI-MS / MS).

AB - There is increasing evidence that substances which are normally present in human or animal bodies may, under the certain circumstances, exhibit deleterious effects on genetic material, therefore acting as endogenous mutagenic agents. Since hormones represent one of the best studied endogenous mutagens, some research focused on the possible role of thyroid hormone in mutagenesis and carcinogenesis. Indeed, thyroid hormones accelerate aerobic metabolism and production of reactive oxygen species (ROS) and, therefore, may exhibit mutagenic effects in various test systems on mammalian cells. However, possible mutagenic effects on prokaryotic DNA has not been investigated so far. Hence, the aim of this research was to compare the sensitivity of TA 100 Salmonella typhimurium with and without metabolic activation with S9 fraction, and human lymphocytes to possible genotoxic effects of triiodothyronine (T-3). Therefore, we used the reverse mutation assay on S. typhimurium (Ames test) and in vitro Comet assay in isolated peripheral blood human lymphocytes. In both tests-systems a broad spectrum of T-3 concentrations was applied. The obtained results showed absence of genotoxic effects of T-3 in bacterial reverse mutation assay and very profound genotoxic effects in human lymphocytes at concentrations higher than 15 mu M. We only observed cytotoxic effects in bacterial system at very high T-3 concentrations (300 and 500 mu M). In conclusion, T-3 was unable to increase the level of reverse mutations in Ames test both with and without S9 mix. Therefore, it seems that ROS production in mitochondria may be the primary cause of DNA damage caused by T-3 in mammalian cells.
