The influence of zinc and heavy metals in feed and water on the quality of cryopreserved bull semen

AB  - This study aimed to assess the influence of different concentrations of zinc (Zn), lead (Pb), mercury (Hg) and cadmium (Cd) found in the feed and water provided to bulls on the concentrations of these metals in cryopreserved bull semen, and to determine their influence onto semen quality parameters. Correlations between heavy metal concentrations in the semen and the quality parameters of semen as estimated by computer-assisted sperm analysis (CASA) and flow cytometry (FC) methods were determined. A total of 40 cryopreserved semen samples originating from bulls housed in 4 different centers for artificial insemination (A, B, C and D) were examined, making a total of 160 samples. The concentrations of metals and semen quality parameters were determined in cryopreserved semen of 10 bulls from each center, namely 4 samples from each bull. Concentrations of Zn, Pb, Hg and Cd in hay and concentrated feed were within the allowed limits as proposed by the National Research Council (NRC, 2000). A strong negative correlation was detected between curvilinear velocity (VCL) and Zn concentrations in the semen (P < 0.01; r = -0.772) in group D, and a positive correlation of VCL with Pb concentrations (P < 0.05 and r = 0.718) in group B. Mercury concentrations in cryopreserved semen correlated negatively to the percent of live sperm cells with intact acrosomes (V/IA: P < 0.05; r = -0.640) and positively with the percent of dead sperm cells with damaged acrosomes (D/DA: P < 0.01; r = 0.766) in group D. This finding confirms the hypothesis that Hg, even at low concentrations, may cause acrosome damage.
AB  - Cilj rada bio je da se ispita uticaj različitih koncentracija cinka (Zn), olova (Pb), žive
(Hg) i kadmijuma (Cd) iz hrane i vode na koncentracije ovih metala u duboko zamrznutom semenu nakon otapanja i da se ispita njihov uticaj na parametre kvaliteta
semena. Korelacije između koncentracije teških metala u semenu i parametra kvaliteta semena su određivane nakon ispitivanja semena kompjuterski asistiranom analizom
semena (eng. computer-assisted sperm analysis - CASA) i protočnom citometrijom
(eng. flow cytometry - FC).
Iz 4 različita centra za veštačko osemenjavanje (A, B, C i D) ispitivano je po 40 uzoraka duboko zamrznutog semena bikova, što čini ukupno 160 uzoraka. Koncentracije
metala i parametri kvaliteta semena su određivani u duboko zamrznutom semenu od
10 bikova iz svakog centra, odnosno 4 uzorka od svakog bika. Koncentracije Zn, Pb,
Hg i Cd u senu i koncentrovanim hranivima bile su ispod propisanih dozvoljenih
vrednosti Nacionalnog Istraživačkog Veća (eng. National Research Council - NRC,
2000). Jaka negativna korelacija uočena je između krivolinijske brzine spermatozoida
(eng. curvilinear velocity - VCL) i koncentracije Zn u semenu (P < 0.01; r = -0.772) u
grupi D, i pozitivna korelacija između VCL i koncentracije Pb (P < 0.05 and r = 0.718)
u grupi B. Koncentracija žive u duboko zamrznutom semenu negativno je korelirala
sa procentom živih spermatozoida sa neoštećenim akrozomom (eng. live sperm cells
with intact acrosomes - V/IA: P < 0.05; r = -0.640) i pozitivno sa procentom mrtvih
spermatozoida sa oštećenim akrozomom (eng. dead sperm cells with damaged acrosomes - D/DA: P < 0.01; r = 0.766) u grupi D. Ovi rezultati potvrđuju hipotezu da Hg,
čak i u niskim koncenracijama može da prouzrokuje oštećenje akrozoma.

Effects of commercial selenium products on glutathione peroxidase activity and semen quality in stud boars

AB  - The aim of this study was to determine how dietary supplementation of inorganic and organic selenium affects the selenium concentration and glutathione peroxidase activity in blood and sperm of sexually mature stud boars. Twenty-four boars of the Large White, Landrace, Pietrain and Duroc breeds of optimal breeding age (on average 2.5 years old) were used. The study lasted 90 days. The boars were randomly assigned to one of three dietary treatment groups: T1 = control; no added selenium (n = 8 boars), T2 = added 0.3 ppm inorganic selenium (sodium selenite, Microgran (R) Se 1% BMP) (n = 8 boars), and T3 = added 0.3 ppm organic selenium (Se-yeast, Sel-Plex 2000 (R)) (n = 8 boars). The concentration of selenium was determined in whole blood and semen, while the activity of glutathione peroxidase (GPx) was measured in blood plasma and semen. In order to measure GPx activity in semen, reactivation of the enzymatic GPx activity was performed. The determined selenium concentration in blood was lowest in the non-supplemented group of boars. Blood plasma GPx activity was higher in boars fed organic selenium than in boars fed a diet without supplemented selenium. While the supplementation of sodium selenite significantly increased GPx activity in boar semen. The highest-concentration of selenium in semen at the end of the trial was determined in the group of boars supplemented with organic selenium, somewhat lower in boars fed supplemented inorganic selenium, and the lowest in the non-supplemented group of boars. The only significant difference between the selenite and Se-yeast diet supplementation was observed in the Se concentration of the semen. The supplementation of selenium affected semen quality, and organic selenium improved the progressive motility of the spermatozoa and increased their resistance in hypo-osmotic and thermal tests. The storage ability of short term preserved semen was improved by organic selenium supplementation, as well as also increasing the fertility rate in gilts.

The presence of naturally occurring antisperm antibodies in the sera of prepubertal calves

AB  - In this study we investigated the presence of antisperm antibodies in the sera of neonatal and young calves up to the age of 120 days by indirect immunofluorescence assay (IIP) and the sperm-agglutination method. By IIP assay, anil IgG and ants IgM antibodies were detected. In the sera of neonatal calves, before colostrum ingestion, no anti-sperm antibodies were detected due to physiological agammaglobulinemia. Titer obtained values in two day old and older calves were different for native sperm cells and sperm cells previously suspended in TRIS egg yolk extender or Biociphos plus extender (IMV, France) indicating antigenic differences between them. Their liter increased with age. This study confirmed the higher sensitivity of the IIP assay. We have also confirmed the hypothesis that in calves, antisperm antibodies naturally occurring before puberty, are most probably the result of cross reactivity with microbial antigens.
AB  - U ovom radu su izneti rezultati ispitivanja prisustva antitela protiv spermatozoida bika u serumu neonatalne teladi i teladi uzrasta do 120 dana metodama indirektne imunofluorescencije i aglutinacije u želatinu. Za izvođenje testova su korišćeni nativni spermatozoidi iz svežih ejakulata kao i spermatozoidi iz pajeta za veštačko osemenjavanje prethodno suspendovani u TRIS-žumanjčanom razređivaču ili razređivaču Biociphos plus (IMV. France). U serumu teladi pre ingestije kolostruma nisu dokazana antitela što je u skladu sa fiziološkom agamaglobulinemijom kod ove životinjske vrste. Kod teladi stare dva dana dokazano je prisustvo antitela protiv spermatozoida poreklom iz kolostruma i to u različitom titru u odnosu na vrstu korišćenih spermatozoida što ukazuje na njihovu antigensku različitost. Titar antitela se povećavao sa starošću teladi. Ova ispitivanja su potvrdila veću osetljivost indirektne imunofluorescence u odnosu na aglutinaciju. Na osnovu dobijenih rezultata se može pretpostaviti da su kod teladi, prirodna antitela na antigene spermatozoida nastala kao posledica unakrsne reaktivnosti sa mikrobijalnim antigenima.

Indirect immunofluoroscence in evaluation of bulls sperm antigenic characteristics

AB  - Our investigations were conducted in order to evaluate variations in anti-sperm antibodies in rabbits sera. Rabbits were previously immunized with TRIS - egg yolk extender, Biociphos plus extender (IMV, France) and with content of insemination straws prepared with same extenders. We investigated sperm cells from ejaculates of 12 black and white spotted bulls by indirect immunofluorescence method. Both extenders and content of insemination straws were capable in raising antibody production. Titer of antibodies had the highest values when sperm cells from insemination straws were used for the test. .
AB  - U ovom radu su izneti rezultati ispitivanja varijacija u titru antitela protiv spermatozoida bika i razređivača za spermu bikova u serumu kunića imunizovanih TRIS žumanjčanim razređivačem, razređivačem Biociphos plus (IMV, France) i sadržajem pajeta za VO krava pripremljenim pomoću njih. Ispitivanja su vršena metodom indirektne imuno-fluorescence a ukupno je testirano 12 bikova crno-bele šarene rase. Postignuti rezultati ukazuju da oba razređivača dovode do nastanka antitela kod kunića a da je titar antitela na antigene spermatozoida značajno viši kada se za izvođenje testa koriste spermatozoidi iz pajeta za VO.

abstract = "Our investigations were conducted in order to evaluate variations in anti-sperm antibodies in rabbits sera. Rabbits were previously immunized with TRIS - egg yolk extender, Biociphos plus extender (IMV, France) and with content of insemination straws prepared with same extenders. We investigated sperm cells from ejaculates of 12 black and white spotted bulls by indirect immunofluorescence method. Both extenders and content of insemination straws were capable in raising antibody production. AB  - Samples of native bulls sperm of Holstein - Friesian breed from Center for Artificial Insemination, were investigated. In bulls sperm numerous types of microorganisms are represented. Of conditionally pathogenic microorganisms Corynebacterium rénale and Staphylococcus auteur were isolated. By microbiological investigation it was established the important representation of saprophytic micro flora: Bacillus spp., Proteus mirabilis, coagulase of negative Staphylococcus, micrococcus, Aspergilus spp., etc. In technological process of seed treatment combinations of antibiotics of a large spectrum of influence are added. Taking into consideration that antimicrobial effect depends, first of all, on present microorganisms, it is clear why microbiological examination of semen as a part of healthy control of breeding bulls has a great importance for the success in cattle reproduction. .
AB  - Ispitivani su uzorci nativne i zamrznute sperme bikova Holštajn Frizijske rase Centra za veslačko osemenjavanje. U nativnoj spermi bikova zastupljene su brojne vrste mikroorganizama. Od uslovno patogenih mikroorganizama izolovani su Corynebacterium rénale i Staphylococcus aureus. Mikrobiološkom pretragom nativne sperme ustanovljena je značajna zastupljenost saprofitne mikroflore: Bacillus spp., Proteus mirabilis, koagulaza negativne stafilokoke, mikrokoke, Aspergilus spp., i dr. U tehnološkom procesu obrade semena dodaju se kombinacije antibiotika širokog spektra delovanja. Mikrobiološkom analizom uzoraka zamrznute sperme utvrđena je isključivo saprofitna mikroflora, dok je značajno veći broj uzoraka bio mikrobiološki negativan. Obzirom da antimikrobni efekat dodatih antibiotika putem razređivača, zavisi prevashodno od prisutnih mikroorganizama, jasno je zašto mikrobiološki pregled semena kao deo zdravstvene kontrole priplodnih bikova ima veliki značaj za uspeh u reprodukciji goveda. .

AB  - The aim of this investigation was to estimate the possible influence of bovine seminal plasma (BSP) on the replication in vitro of some sexually transmitted viruses: BHV - 1 (bovine herpes virus - 1) and BVD (bovine viral diarrhea) significant for bovine reproduction. In addition, we tested the influence of the on EHV-1 (equine herpes virus - 1) and Morbus Aujeszcky virus replication in the same model. Our results showed that BSP has an inhibitory effect on the replication in vitro of all tested viruses and their strains. This activity was lost following dialysis suggesting that the active inhibitory factor is of small molecular veight (less than 12 kDa).Furthermore, inhibition was preserved after repeated freezing and thawing and after inactivation for 30 min at 56 °C indicating that the inhibitory factor is heat stable.
