AB  - A new auronolignan, named cotinignan B, was isolated from Cotinus coggygria Scop. Structure elucidation was performed on the basis of 1H, 13C NMR, COSY, NOESY, HSQC, and HMBC experiments, supported with HR-ESI-MS, IR and UV. Additional J-HMBC NMR experiment was essential to resolve the configuration of the trisubstituted double bond. This compound represents the secondly discovered natural compound belonging to the rare auronolignan type.

AB  - Our previous phytochemical studies revealed that the
plant species of the genus Amphoricarpos from
Montenegro are characterized with an appreciable content
(>0.3–2.3% per dry weight of aerial plant material) of γsesquiterpene lactones with guaianolide skeleton, socalled amphoricarpolides that are mostly deposited on the
leaf surface (>1.0–3.4% per dry weight of leaves)
(Djordjević et al., 2021; Jadranin et al., 2013). These
compounds showed in vitro protective effect on
chromosome aberrations in peripheral human
lymphocytes (Cvetković et al., 2021) and in vitro
cytotoxic (Djordjević et al., 2021), antifungal (Jadranin et
al., 2013) and antibacterial (Gavrilović et al., 2016)
activities. The amphoricarpolides are significant as
chemotaxonomic markers as they gave insights about the
taxonomic status of the genus (Djordjević et al., 2021).
This study revealed results from reinvestigation of
sesquiterpene lactones composition in the crude and leaf
surface extracts of Amphoricarpos plants by rapid
resolution liquid chromatography (RRLC) coupled with
quadrupole time-of-flight mass spectrometry (QToF MS)
and the tentative identification of undescribed
sesquiterpene lactones is presented.

AB  - Phytochemistry deals with the study of secondary metabolites produced by plants that synthesize these compounds for many reasons, including their own protection against attack of herbivores and plant diseases. Secondary metabolites are believed to represent plant adaptation to various environmental factors and that they enabled the survival of the species. Secondary metabolites of plants can have curative or toxic effects in humans and animals. Herbal medicine has a long tradition in folk medicine and until the early 20th century, when synthetic organic chemistry began to develop, plants were the main source of medicines. The two basic goals of our phytochemical research are: Isolation and identification of new (biologically active) compounds - potential drugs, and chemotaxonomy (chemosystematics). In the following text through one selected example, the genus Amphoricarpos Vis., our phytochemical research is shown on both aspects.

AB  - Analysis of composition of CH2Cl2 surface extract of leaves of Amphoricarpos neumayeri Vis. revealed 16 sesquiterpene lactones with guaianolide skeleton, so called amphoricarpolides, typical for this genus. Four of them, 13-16, were new derivatives and their structures were elucidated by detailed analyses of IR, NMR and MS data. Amphoricarpolide (9), its 15-O-acetyl derivative (5), and two 9 beta-hydroxyamphoricarpolides, 3,15-di-O-acetyl- and 3-O-acetyl-15-O-isovaleroyl derivatives (3 and 6, respectively) were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (CBMN) assay. The tested compound exerted a beneficial effect by decreasing DNA damage of human lymphocytes.

AB  - New aurone epoxide, 2,10-oxy-10-methoxysulfuretin (14), and new auronolignan (15), named cotinignan A, were isolated by silica gel column and semipreparative HPLC chromatography from the methylene chloride/methanol extract of Cotinus coggygria Scop. heartwood. In addition, thirteen known secondary metabolites namely sulfuretin, 2,3-trans-fustin, fisetin, butin, butein, taxifolin, eriodictyol, 3,5,5,7-tetrahydroxyflavanone, 3,4,7-trihydroxyflavone, 3-O-methyl-2,3-trans-fustin, 3-O-galloyl-2,3-trans-fustin, beta-resorcylic acid and 3-O-beta-sitosterol glucoside were isolated as well. Their structures were elucidated by 1D and 2D NMR, HR-ESI-MS, IR and UV. Ten out of eleven isolated flavonoids possess 7, 3 and 4 hydroxy groups. These structural features could be considered as chemotaxonomic characteristic of flavonoids from C. coggygria. Cotinignan A (15) represents new subclass of secondary metabolites - auronolignans. AB  - The genus Amphoricarpos Vis. is endemic of westpart of Balkan peninsula. In order to get more insight into the ambiguous taxonomy of the genus, the metabolomic study of the Amphoricarpos samples, has been undertaken. The(1)H NMR spectroscopy combined with multivariate data analysis has been applied. OPLS-DA has been shown to be the best method for clear discrimination of these samples based on the metabolites present in the extracts. The main difference between A. autariatus and A. neumayeri has been found to be the presence of characteristic sesquiterpene lactones of guaianolide type named amphoricarpolides. While the main sesquiterpene lactone in A. neumayeri was oxidized in position 2, those present in both subspecies of A. autariatus samples were oxidized in position 9. The use of sesquiterpene lactones as taxonomic characters for differentiating species of the Asteraceae family was reviewed elsewhere. For the differentiation of two subspecies, A. autariatus ssp. autariatus, and A. autariatus ssp. bertisceus, chlorogenic and malic acid have been found to be decisive, but these compounds do not have chemotaxonomic significance. Our results were in accordance with above mentioned classification of Blecic and Mayer but they differentiate from those of the recent genomic study of the genus indicating a single, genetically, morphologically and ecologically variable species, without intraspecific taxa.

Some of the species investigated, members of the genera Gentiana, Gentianella, and Swertia, are endemic, and the emphasis in this report is on those exhibiting biological activities that could be regarded as a potential source of drugs. This review discusses more than fifty compounds, such as xanthones, secoiridoids, and C-glucoflavonoids.

AB  - The aerial parts of three endemic Centaurea L. species, namely C. tomorosii Micevski, C. soskae Hayek and C. galicicae Micevski, afforded the sesquiterpene lactone cnicin (1) and seven flavonoids: apigenin (2), isokaempferide (3), hispidulin (4), eupatorin (5), cirsimaritin (6), santoflavone (7) and salvigenin (8). The structures of the isolated compounds were determined by UV, H-1-NMR and C-13-NMR spectroscopy and HR-ESI-MS spectrometry. H-1-NMR spectroscopy was used as a method for the quantitative analysis of cnicin.

Nine of them, 9-17, were new compounds, and their structures were elucidated by detailed analyses of IR, NMR, and MS data.

AB  - Dimers tomoroside A (1) and tomoroside B (2) of the co-occuring known chalcone monomer (3), along with the seven known flavonoid glucosides (4-10), were isolated from the aerial parts of Helichrysum zivojinii Cernjavski & Soska. The structures of the isolated compounds were elucidated by spectroscopic techniques. Compound 1 inhibited topo Ha and hif-1 alpha expression and stimulated doxorubicin anticancer effect, while 2 increased the expression of hif-1 alpha a, probably acting as antioxidant and redox status modulator. Notably, 2 synergized with Tipifarnib showing potential to improve the action of this new chemotherapeutic involved in the modulation of mitogene activated protein (MAP) kinase signaling pathway.

AB  - The composition of leaf cuticular neutral lipids of Amphoricarpos autariatus ssp. autariatus collected at canyon of river Tara (North Montenegro) was investigated by GC/MS (nonpolar fraction), LC-ESI TOF MS and H-1 NMR spectroscopy (more polar fraction). The nonpolar fraction (ca. 15% of the whole surface extract) contained C-27 (-) (33) n-alkanes, those with odd-number of carbons predominating. The LC-ESI MS and H-1 NMR of the more polar fraction revealed 13 sesquiterpene lactones, constituting ca. 97.5% of the lactone mixture, identified as the known guaianolides, so-called amphoricarpolides, found previously in the aerial parts of the genus. The lactone fraction exhibited considerable in vitro effect against eight fungi, i.e. Aspergillus ochraceus, A. niger, A. versicolor, Penicillium funiculosum, P. ochrochloron, Trichoderma viride, Fusarium verticillioides and Fulvia fulvum.

Variability of n-Alkanes and Nonacosan-10-ol in Natural Populations of Picea omorika

AB  - This is the first report of population variability of the contents of n-alkanes and nonacosan-10-ol in the needle epicuticular waxes of Serbian spruce (Picea omorika). The hexane extracts of needle samples originated from three natural populations in Serbia (Vranjak, Zmajevaki potok, and Mileevka Canyon) were investigated by GC and GC/MS analyses. The amount of nonacosan-10-ol varied individually from 50.05 to 74.42% (65.74% in average), but the differences between the three investigated populations were not statistically confirmed. The results exhibited variability of the composition of n-alkanes in the epicuticular waxes with their size ranging from C18 to C35. The most abundant n-alkanes were C29, C31, and C27 (35.22, 13.77, and 12.28% in average, resp.). The carbon preference index of all the n-alkanes (CPItotal) of the P. omorika populations (average of populations IIII) ranged from 3.3 to 11.5 (mean of 5.9), while the average chain length (ACL) ranged from 26.6 to 29.2. The principal component and cluster analyses of the contents of nine n-alkanes showed the greatest difference for the population growing in the Mileevka Canyon. The obtained results were compared with previous literature data given for other Picea species, and this comparison was briefly discussed.

AB  - Four lichen acids, physodalic acid (F1), physodic acid (F2), 3-hydroxyphysodic acid (F3), and isophysodic acid (F4), were isolated from Hypogymnia physodes methanol extract using preparative reversed-phase high performance liquid chromatography and their structures were determined by UV, MS, H-1 NMR and C-13 NMR. This is the first report on the isolation of F4 from H. physodes. Isolated rat thymocytes were cultivated with increasing F1-F4 concentrations (0.1, 1, 10 mu g/well) and proliferative activity, viability, ROS (reactive oxygen species) production and MMP (mitochondrial membrane potential) disturbances were evaluated. Obtained results show significantly decreased thymocytes proliferation was observed when cells were treated with F1 (1 mu g, p < 0.05; 10 mu g; p < 0.001), F2 (10 mu g, p < 0.05) and F3 compound (10 mu g, p < 0.05). Significantly increased cytotoxicity was detected when cells were incubated with F1 (1 mu g, p < 0.05; 10 mu g, p < 0.01), F2 (10 mu g, p < 0.05) and F3 compound (10 mu g, p < 0.001). Increased H2DCF-DA fluorescence intensity, when cells were treated with F1 (1 mu g, p < 0.001; 1 mu g, p < 0.01; 10 mu g, p < 0.001) and F2 (1 mu g, p < 0.05; 10 mu g, p < 0.01) compound, indicating the increase of intracellular ROS production. Simultaneously, increased ROS levels were followed with significantly decreased MMP when thymocytes were cultivated with F1 (0.1 mu g, p < 0.001; 1 mu g, p < 0.001; 10 mu g, p < 0.001) and F2 compound (10 mu g, p < 0.001). Thymocytes exposure to increased (0.1, 1, 10 mu g) concentrations of F3 and F4 compounds did not result with significant alterations in MMP and intracellular ROS production. We have shown that higher F1 and F2 concentrations induce thymocytes toxicity mainly through induction of oxidative stress, while cytotoxicity effect of F3 is followed with altered antioxidant/oxidant balance. The rigid 11H-dibenzo[b,e][1,4]dioxepin-11-one ring in the depsidone structure may play a important role for the examined biological activities.

The carbon preference index of Pinus peuce ranged from 1.0 to 4.3 (1.9 on average). Average chain length ranged from 18.4 to 27.7 (average 25.7). A high level of inidividual quantitative variation in all of these hydrocarbon parameters was also detected. These results were compared with published data on other species from the Pinus genus.

The amount of nonacosan-10-ol varied individually from 27.4 to 73.2% (55.5% in average), but differences between the four investigated populations were not statistically confirmed. The size of the n-alkanes ranged from C18 to C33. The most abundant n-alkanes were C23, C27, and C25 (12.2, 11.2, and 10.8% in average, resp.). The carbon preference index (CPI) of the n-alkanes ranged from 0.8 to 3.1 (1.6 in average), while the average chain length (ACL) ranged from 20.9 to 26.5 (24.4 in average). Long-chain and mid-chain n-alkanes prevailed (49.6 and 37.9% in average, resp.). It was also found that the populations of P. heldreichii var. leucodermis had predominantly a narrower range of n-alkanes (C18?C31) than the trees of the variety pancici (C18?C33). Differences between the varieties were also significant for most of the other characteristics of the n-alkane pattern (e.g., most abundant n-alkanes, CPI, ACL, and relative proportion of short-, mid-, and long-chain n-alkanes). AB  - This is the first report of n-alkanes in needle epicuticular waxes of the variety Bosnian pine, Pinus heldreichii var. pančići. n-Hexane extracts of needle samples, originating from seven isolated localities in Serbia, were analysed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). The results evidenced n-alkanes ranging from C18 to C33 in epicuticular waxes. The most abundant alkanes were C27, C23, C25 and C29 (12.53 %, 12.46 %, 12.00 % and 10.38 % on average, respectively). The carbon preference index (CPItotal) of Pinus heldreichii var. pančići ranged from 1.1 to 2.1 (1.6 on average), while the average chain length (ACLtotal) ranged from 25.0 to 25.8 (25.3 on average). A high level of individual quantitative variation in all of these hydrocarbon parameters was also found. The obtained results were compared with the bibliographic references for Pinus heldreichii var. leucodermis and other species of the Pinus genus.
AB  - Ovo je prvo saopštenje o n-alkanima u voskovima iglica varijeteta munike, Pinus heldreichii var. pančići. n-Heksanski ekstrakti uzoraka iglica koji potiču sa sedam izolovanih lokaliteta u Srbiji analizirani su gasnom hromatografijom (GC) i gasnom hromatografijom-masenom spektrometrijom (GC-MS). Rezultati su pokazali da se n-alkani nalaze u opsegu od C18 do C33. Među njima su najobilniji C27, C23, C25 i C29 (12,53, 12,46, 12,00 i 10,38 % u proseku, redom). Ugljenični preferencijalni indeks (CPItotal) Pinus heldreichii var. pančići je bio u opsegu od 1,1 do 2,1 (prosečno 1,6), a dužina niza ugljenikovih atoma (ACLtotal) u opsegu od 25,0 do 25,8 (prosečno 25,3). Takođe je utvrđen visok nivo individualne kvantitativne varijabilnosti u svim analiziranim parametrima ovih ugljovodonika. AB  - In this study 10 guaianolide-type sesquiterpene gamma-lactones named amphoricarpolides, isolated from the aerial parts of two endemic subspecies of Amphoricarpos neumayeri (ssp. neumayeri and ssp. murbeckii Bosnjak), were investigated by means of reversed-phase thin-layer chromatography. Methanol-water and tetrahydrofuran-water binary mixtures were used as mobile phase in order to determine lipophilicity parameters R-M(0) and C-0. Some of the investigated compounds were screened for their cytotoxic activity against HeLa and B16 cells. Chromatographically obtained lipophilicity parameters were correlated with calculated logP values and IC50 values. AB  - 1H-NMR spectroscopy was applied for the quantitative analysis of cnicin, a bioactive germacranolide type sesquiterpene lactone, in the aerial parts of seven wild-growing Centaurea species collected in Serbia and Montenegro. The analysis was performed by comparison of the integral of the one-proton signal of cnicin (H-13, δ 5.75) with that of the two-proton singlet ( δ 6.98) of 2,6-bis(1,1-dimethylethyl)-4-methylphenol (BHT), used as the internal standard. Cnicin, within the concentration range 1.06-6.12 mg/g, calculated per weight of the fresh plant material was detected in six species, the exception being C. salonitana. This method allows the rapid and simple quantification of cnicin without any pre-purification step.

Analiza je izvršena poređenjem integrala signala jednog protona knicina (H-13, δ 5.75) sa singletom koji potiče od dva protona ( δ 6.98) internog standarda, 2,6-bis(1,1-dimetiletil)-4-metilfenola (BHT). AB  - The aerial parts of Amphoricarpos neumayeri ssp. murbeckii afforded eleven guaianolides with the same relative (1 alpha H,4 beta H,5 alpha H, 7 alpha H) configuration of the basic skeleton. All of them contained a CH2OX (X = H, acetyl or isovaleroyl) group in 4 alpha-position, typical for amphoricarpolides. Four compounds (1-4) were isolated before from the same species, originating from different localities. Guaianolides 5-11 are new compounds. Compounds 7 and 8 were epoxidized at the 10 alpha(14)-position. Instead of the Delta(11(13))-double bond, observed in all previously isolated guaianolides from the same species, the four lactones contained 11 alpha,13-diol (8-10) or 11 alpha-OH, 13-chloro (11) moieties respectively.
