Acute and subchronic exposure of the common carp (Cyprinus carpio) to herbicide S-metolachlor

AB  - S-metolachlor is one of the most frequently used herbicides worldwide. However, toxicity studies of this herbicide to aquatic organisms are scarce. In the present study, two experiments were conducted to test the effects of S-metolachlor on common carp, one of the most economically important fish species, with a distribution throughout the world: (1) 96 h acute semi-static toxicity test, aiming to determine LC50; (2) a subchronic semi-static test that lasted 28 days, in which juvenile carp were exposed to 3%, 8%, and 25% of previously determined LC50—0.5 mg·L−1, 1.4 mg·L−1, and 4.1 mg·L−1, respectively. Several biomarkers were employed to assess fish responses to toxicants. Blood biochemistry analysis and nuclear alterations of erythrocytes did not show any difference among experimental groups. Semi-quantitative histopathological analysis revealed mild alterations in the gills and liver, where oedema of secondary epithelium of gills and leukocyte infiltration in liver were significantly higher in fish exposed to 1.4 mg·L−1 and 4.1 mg·L−1. Histopathological indices in liver, as well as the total histopathological index, also showed significantly higher scores in the same groups. Bioconcentration factors of S-metolachlor ranged from 3.2 to 9.4, depending on the experimental group.

AB  - Toxicological assessment of S-metolachlor, synthetic organic herbicide frequently used for efficient
weeds control, was the subject of the present study. Two exposure experiments (acute - 4 day and subacute - 28 days) were conducted on five months old common carp originated from the same family from the selective breeding program. 96h LCso was calculated to be 16.31 mg L', which is generally high value, since environmental concentrations of S-metolachlor are usually determined to be below 1 mg L'. Concerning this, three concentrations were set for a subacute test, carried out in triplicates: C0 - 0.0 mg L', C1 -0.5 mgl', C2 -1.4 mg L', C3 -4.1 mg L' Biomarkers used for the assessment of fish health status were: histopathology of gills and liver, as well as nuclear abnormalities on erythrocytes. Histological assessment found a number of alterations in both
sampled tissues: hyperaemia, presence of eosinophilic granular cells, epithelial lifting, hyperplasia of epithelial cells and focal necrosis in gills; leukocyte infiltration, hyperaemia, fibrosis of blood vessels and focal necrosis in liver. Only two of all mentioned alterations (epithelial lifting and gills hyperemia) had higher levels in the control, while majority were absent from control group. Different morphological abnormalities were noticed on the erythrocyte nuclei: micronucleus, nuclear buds, fragmented-apoptotic, and bi-nucleated cells. Almost all alterations, either
histopathological or nuclear, were higher compared to control, but due to the high variation in between groups, statistical significance was not established. This study confirms low toxicity of S-metolachlor to common carp,

Principles of fish welfare assessment in farm rearing conditions

AB  - For several decades fish welfare has been subject of many researches, and the interest for this subject is connected with the fact that fish can feel pain and suffering. In addition to this stressors' mechanisms of action and their consequences are similar in mammals and fish. Assessment of welfare for farmed fish is based on the same principles as for terrestrial farm animals. However, special methods of collecting data are needed due to morphological characteristics of fish and properties of their environment. In the world and in our country, researches of different ways of fish welfare assessment are being conducted, especially based on non-invasive techniques such as monitoring of behaviour. In this paper a review of basic principles and methods used in assessment of farmed fish welfare is given.
AB  - Dobrobit riba je već nekoliko decenija predmet brojnih istraživanja, a zainteresovanost za ovu temu povezana je sa činjenicom da ribe mogu da osećaju bol i patnju. Osim toga, mehanizmi i posledice delovanja stresora kod riba vrlo su slični onima kod sisara. Procena nivoa dobrobiti kod farmski uzgajanih riba zasniva se na istim principima kao kod kopnenih farmskih životinja. Međutim, voda kao životna sredina riba i njihove morfološke specifičnosti zahtevaju posebne metode prikupljanja podataka. U svetu i kod nas se istražuju različiti načini procene dobrobiti riba, naročito oni koji se zasnivaju na neinvazivnim postupcima, kao što je praćenje ponašanja. U ovom radu dat je prikaz osnovnih principa i metoda koji se koriste u proceni dobrobiti farmski gajenih riba.
