Potential of wormwood and oak bark-based supplement in health improvement of Nosema ceranae-infected honey bees

AB  - Nosema ceranae, a microsporidian parasite, as one of the stressors that contribute to honey bee decline, has a significant negative impact on the longevity, productivity, and reproductive capacity of honey bee colonies. There are several different strategies for Nosema infection control, including natural-based and antibiotic-based products. In this study, we tested wormwood and oak bark-based supplement “Medenko forte” on survival, Nosema infection, oxidative stress, and expression of immune-related genes in artificially N. ceranae-infected bees. The results revealed a positive influence on the survival of Nosema-infected bees, irrespectively of the moment of supplement application (day 1, day 3, or day 6 after bee emergence), as well as reduction of Nosema loads and, consequently, Nosema-induced oxidative stress. Supplementation had no negative effects on bee immunity, but better anti-Nosema than immune-stimulating effects were affirmed based on expression levels of abaecin, defensin, hymenoptaecin, apidaecin, and vitellogenin genes. In conclusion, the tested supplement „Medenko forte” has great potential in the health protection of Nosema-infected bees. However, further investigations need to be performed to elucidate its mechanisms of action.

Uticaj aflatoksina na zdravstvenu bezbednost i kvalitet mleka

AB  - Širom sveta, mogućnost kontaminacije lanca ishrane aflatoksinima je trajno prisutna, sa
ozbiljnim uticajem na zdravlje ljudi i životinja. Činjenica da plesni Aspergillus sp.
proizvode aflatoksine tokom njihovog rasta na supstratima koji su hrana za ljude i
životinje ukazuje na njihov važan uticaj na pojavu različitih mikotoksičnih bolesti.
Ulazak aflatoksina u lanac ishrane predstavlja globalni rizik po javno zdravlje ljudi i
životinja. Akutne aflatoksikoze su veoma retke, ali vremenski duge izloženosti niskim
nivoima aflatoksina su širom sveta rasprostranjene. Postoji više od 20 poznatih
aflatoksina, od kojih su za zdravstvenu bezbednost hrane izrazito značajni aflatoksin B1
(AFB1) i aflatoksin M1 (AFM1). Oba toksina, AFB1 i AFM1 su kancerogeni; međutim,
AFM1 je najotrovniji sekundarni metabolit koji se luči u mleko i svrstan je u grupu 1
kancerogena od strane Međunarodne agencije za istraživanje raka. Trenutno su
potrošači širom sveta neprekidno izloženi niskim dozama AFM1, od kojih najviše deca i
odojčad. Aflatoksini utiču na kvalitet mleka jer krave metabolizuju AFB1 da bi formirale
monohidroksidni derivat AFM1, koji se luči u kravlje mleko. Prisustvo trajne opasnosti
od aflatoksina i njihovog nepoželjnog prisustva u hrani, utiče da su mnoge zemlje
usvojile politiku rigoroznih kontrola. Danas, granica AFM1u Srbiji postavljena je na 0,25
μg/kg mleka, dok standardi Evropske Unije ne dozvoljavaju više od 0,05 μg/kg AFM1.
Dozvoljeni nivo AFM1 u mleku u R.Srbiji, pet puta je veći nego u Evropskoj Uniji i
predviđeno je smanjenje od 1. decembra 2023 godine, prema Pravilniku o izmeni
Pravilnika o maksimalnim koncentracijama određenih kontaminenata u hrani.
Dugoročno posmatrano, u cilju obezbeđenja zdravstveno bezbedne ishrane ljudi i
životinja i ublažavanju nepovoljnih klimatskih promena, potrebno je redovno praćenje
AFB1 od farme za viljuške, kako bi se obezbedio najniži nivo aflatoksina M1 u mleku i
mlečnim proizvodima ili potpuno eliminisalo prisustvo ovih toksina.

Predikcija proizvodnje enetričnog metana u organizmu preživara na osnovu hemijskog sastava hrane

AB  - Procena i ublažavanje emisije metana iz govedarstva, posebno od mlečnih krava,
dobile su globalni značaj za klimatski održivu govedarsku proizvodnju. Metan je gas
koji ima 28 puta veći negativan uticaj na globalno zagrevanje od ugljen dioksida. Udeo
enteričnog metana u globalnoj emisiji metana iznosi 27 procenata. On nastaje kao nusproizvod varenja hrane prvenstveno kod preživara enteričnom fermentacijom u buragu. Preživari (gajeni ili divlji) emituju metan najvećim delom eruktacijom. Proizvedeni
enterični metan predstavlja gubitak energije hrane kod životinja. Količina enteričnog
metana koja se emituje zavisi od tipa digestivnog trakta, starosti i težine životinje, kao
i od kvaliteta i količine unete hrane.
Istraživanjima brojnih autora u proteklom periodu je razvijeno više metoda za
procenu stvarnih emisija ECH4
 i to su: komore za disanje, tehnika praćenja sa sumpor
heksafluoridom (SF6), uzimanje uzoraka izdisaja tokom muže ili ishrane, “Green Feed”
sistem i mnoge druge. Od navedenih, komora za disanje predstavlja zlatni standard i
referentnu metodu za studije metabolizma životinja i emisije gasova. Direktno merenje
ECH4
 je složeno i nepraktično na većem broju grla i zbog toga se za procenu proizvodnje CH4
 koriste predikcioni modeli. Definisanje i primena matematičkih modela predikcije ima za cilj da omogući preciznost i adekvatno prilagođavanje prema uslovima
odgajivanja, kako bi se dobila što realnija vrednost emisije ECH4
. Međutim, definisanje preciznih modela predviđanja zahteva opsežne podatke. Složeni modeli koji koriste
DMI, NDF, EE, MF i BW su imali najbolje performanse za predviđanje proizvodnje ECH4
.
Modeli koji zahtevaju samo podatke o DMI ili DMI sa NDF su imali najbolje prediktivne
sposobnosti i predstavljaju alternativu složenim modelima. Dobra predikciona jednačina ukupne proizvodnje ECH4
 u skupu podataka je usko povezana sa informacijama o
unosu suve materije, a zatim sa sadržajem NDF u obroku. Do danas su razvijeni različiti matematički modeli predikcije emisije eteričkog metana kao što su Holos, NorFor
i drugi.

Contact varroacidal efficacy of lithium citrate and its influence on viral loads, immune parameters and oxidative stress of honey bees in a field experiment

AB  - With an almost global distribution, Varroa destuctor is the leading cause of weakening and loss of honey bee colonies. New substances are constantly being tested in order to find those that will exhibit high anti-Varroa efficacy at low doses/concentrations, without unwanted effects on bees. Lithium (Li) salts stood out as candidates based on previous research. The aims of this study were to evaluate Li citrate hydrate (Li-cit) for its contact efficacy against Varroa, but also the effect of Li-cit on honey bees by estimating loads of honey bee viruses, expression levels of immune-related genes and genes for antioxidative enzymes and oxidative stress parameters on two sampling occasions, before the treatment and after the treatment. Our experiment was performed on four groups, each consisting of seven colonies. Two groups were treated with the test compound, one receiving 5 mM and the other 10 mM of Li-cit; the third received oxalic acid treatment (OA group) and served as positive control, and the fourth was negative control (C group), treated with 50% w/v pure sucrose-water syrup. Single trickling treatment was applied in all groups. Both tested concentrations of Li-cit, 5 and 10 mM, expressed high varroacidal efficacy, 96.85% and 96.80%, respectively. Load of Chronic Bee Paralysis Virus significantly decreased (p < 0.01) after the treatment in group treated with 5 mM of Li-cit. In OA group, loads of Acute Bee Paralysis Virus and Deformed Wing Virus significantly (p < 0.05) increased, and in C group, loads of all viruses significantly (p < 0.01 or p < 0.001) increased. Transcript levels of genes for abaecin, apidaecin, defensin and vitellogenin were significantly higher (p < 0.05—p < 0.001), while all oxidative stress parameters were significantly lower (p < 0.05—p < 0.001) after the treatment in both groups treated with Li-cit. All presented results along with easy application indicate benefits of topical Li-cit treatment and complete the mosaic of evidence on the advantages of this salt in the control of Varroa.

Transcript levels of genes for abaecin, apidaecin, defensin and vitellogenin were significantly higher (p < 0.05—p < 0.001), while all oxidative stress parameters were significantly lower (p < 0.05—p < 0.001) after the treatment in both groups treated with Li-cit. All presented results along with easy application indicate benefits of topical Li-cit treatment and complete the mosaic of evidence on the advantages of this salt in the control of Varroa.",

Detection and characterisation of porcine circoviruses in wild boars in northeastern Serbia

AB  - The objective was to expand and update the knowledge on the presence and genotype diversity of porcine circoviruses 2 and 3 (PCV2 and PCV3) in the wild boar populations from the hunting grounds in northeastern Serbia. The presence of PCV3 was not determined, and PCV2 was confirmed in 40.32% of the organ samples from 124 wild boars hunted from 2018 to 2019, indicating their significance in virus circulation since traditional pig farms with irregular PCV2 vaccination strategies are widespread in this region. The most prevalent genotype was PCV2d, followed by PCV2b and PCV2a in 55.6%, 38.9%, and 5.5% of the examined samples, respectively. Nucleotide sequences of the detected strains were homogenous within the genotype and clustered within the subgroups PCV2d-2, PCV2b-1A/B, and PCV2a-2D with high identity to European, Chinese, and Serbian domestic pig sequences suggesting their origin. Wild boars presented with no clinical or pathological signs of infection, implying that these animals might be less susceptible to disease, particularly since the cofactors present in pig farming systems that support the disease development are absent in the wild. The high PCV2 detection frequency demonstrates the importance of wildlife monitoring to track virus population dynamics, especially in regions with free-range pig farming in order to plan adequate disease control strategies.

AB  - Diabetes is one of the main health concerns, especially in developed countries. During the last few decades, the percentage of diabetic persons is constantly increasing. Although the genetic factors have a strong influence in the development of diabetes, environmental influence (physical inactivity, inadequate nutrition leading to obesity) also have an important impact. The main objective of this investigation was to evaluate the possible influence of organic sprouted whole wheat grain flourless bread Tonus® (product of Trivit, Becej, Serbia) on DNA damage at various stages of progression through type 2 diabetes mellitus (T2DM). In addition to control (non-diabetic) subjects we analyzed obese, pre-diabetic and diabetic patients, for a total of four experimental groups. All subjects used to eat Mediterranean diet for at least two years before being included in our study. In each of four groups we had five persons practicing the Mediterranean diet but instead of bread they consumed flourless bread Tonus®. The DNA damage was evaluated on peripheral blood mononuclear cells by alkaline single cell gel electrophoresis (Comet) assay at the very beginning (before starting the Tonus® bread diet), and after exactly the three months of consumption of Tonus® bread. Statistical analysis revealed that only in patients with type
AB  - Dijabetes predstavlja jedan od glavnih zdravstvenih problema, naročito u razvijenim zemljama. Tokom poslednjih nekoliko decenija neprekidno se povećava procenat osoba obolelih od dijabetesa. Mada genetički faktori imaju snažan uticaj na razvoj dijabetesa, uticaji sredine (fizička neaktivnost, nepravilna ishrana koja dovodi do gojaznosti) takođe imaju veliki uticaj na razvoj ove bolesti. Tokom nekoliko poslednjih decenija procenat osoba sa dijabetesom se konstantno povećava. Glavni cilj ovog istraživanja bio je da se proceni mogući uticaj organskog hleba bez brašna od celih proklijalih zrna pšenice, Tonus® (Trivit, Bečej, Serbia) na oštećenja DNK u različitim stadijumima progresije kroz tip 2 dijabetes melitus. Pored kontrolne grupe (zdrave osobe), analizirali smo gojazne, predijabetičare i dijabetičare, što čini ukupno četiri eksperimentalne grupe. Sve osobe su koristile Mediteransku ishranu najmanje dve godine pre nego su uključene u ovu studiju. U svakoj grupi imali smo pet osoba koje su upražnjavale Mediteransku ishranu, ali su umesto uobičajenog hleba konzumirali hleb bez brašna Tonus®. Oštećenja DNK su evaluirana alkalnom verzijom elektoroforeze DNK pojedinačnih ćelija (komet test) na samom početku (pre uvođenja hleba Tonus® u ishranu) i tačno nakon tri meseca konzumiranja hleba Tonus®. Statistička analiza otkrila je da samo kod pacijenata sa dijabetesom tipa 2 konzumiranje Tonus hleba dovodi do smanjenja oštećenja DNK u poređenju sa stepenom oštećenja DNK kod ovih pacijenata pre nego su otpočeli dijetu sa hlebom Tonus. Pretpostaljamo da je pad hiperinsulinemije usled konzumiranja hleba Tonus mogao da bude jedan od glavnih uzroka smanjenja oštećenja DNK.
