AB  - The work was aimed to research into the vital signs, haematological and biochemical parameters, acute-phase protein concentrations (CRP, albumin), progesterone (PG) and anti-Müllerian hormone (AMH) concentrations in bitches with pyometra and their comparison with those in healthy bitches. In addition, the goal of this work was to assess if some of the tested parameters may be used as a biomarker in the diagnostics of pyometra and the monitoring of the postoperative recovery. Forty 3-6-year-old bitches of various breeds were included in the research and allotted to two groups: the control (C, n = 20) and the diseased – bitches diagnosed with pyometra (P, n = 20). Blood samples for the analyses were taken immediately before surgery (0 h), and 12 h, 24 h, 48 h and 72 h after, excepting for AMH and PG concentrations, which were determined only at 0 h. Clinical examination detected significantly higher respiration rates in bitches with pyometra in comparison to healthy ones at all time points, higher heart rates before and 12 h after surgery, and higher body temperature before and 12 h, 48 h and 72 h post operation (p <.01). Significantly higher concentrations of CRP (p <.01) and lower concentrations of albumin were detected in bitches with pyometra in comparison to their healthy counterparts at all time points. Progesterone and AMH concentration analysis immediately before surgery detected significantly higher levels of the hormone in bitches with pyometra than in the control (p <.01). The inflammatory reaction of the endometrium resulted in a significant AMH concentration increase, whilst increased CRP concentrations and lower albumin concentrations throughout the research may be used as biomarkers of proinflammatory activities, that is of the acute-phase response.

AB  - Background/Aim. Myeloid-derived suppressor cells (MDSCs) suppress immune responses via a series of inhibitory mechanisms, which ultimately could lead to tumor growth. B7-H4 expression is significantly associated with poor outcome and promotion of tumor cell proliferation, invasion and migration in patients with various cancers. Data concerning B7-H4 expression in lung cancers (LC), either on tumor or immunological cells, are still sporadic. The aim was to estimate and correlate the number of CD14+B7-H4+MDSCs in blood and lung tumor microcirculation with clinical stage, histology type of tumor, tumor node metastasis (TNM) stadium, nodal status and disease outspread. Methods. The study included 44 lung cancer patients (III and IV clinical stage) and 30 healthy controls. CD14+B7-H4+ MDSC number was estimated by flow cytometry in blood and tumor microcirculation samples of each patient. Results. CD14+B7-H4+ MDSCs number was significantly higher in patients’ samples compared to controls. CD14+B7-H4+ MDSC number was significantly increased in tumor compared to blood sample of the same patient. Clinical stage III patients had the increased number of the CD14+B7-H4+ MDSC compared to stage IV, in both types of samples. According to histology, small cell lung cancer (SCLC) patients had the highest average CD14+B7-H4+ MDSCs number, significantly increased compared to patients with squamous and large cell LC histology type. Tumor size was directly associated with the number of the CD14+B7-H4+ MDSC, both in blood and tumor samples. Furthermore, nodal involvement was associated with the gradual increase of the CD14+B7-H4+ MDSC number, being the highest in the N3 group, again both in blood and tumor samples. Finally, we detected higher CD14+B7-H4+ MDSCs number in the samples of patients without metastases. Conclusion. CD14+B7-H4+ MDSCs n umber in L C patients is significantly associated with tumor histology type, lymph node involvement, disease extent degree and tumor size. Concerning their large number in LC tumor microenvironment together with immunosuppressive capacities, CD14+B7-H4+ MDSCs could represent important tumor promoting factor in LC pathophysiology.

Moderate radioprotective role of zeolite in rats

AB  - Background/Aim. Exposure of living organisms to γ-radiation results in the overproduction of free radicals. The aim of the study was to test if the subacute administration of micronized zeolite (MZC) accomplishes radioprotective role based on the evaluation of the status of oxidative stress (OS) in the brain and 8-hydroxyguanosine (8-OH-dG) in the plasma of the rats exposed to the single γ-ray irradiation of 2 and/or 10 Gray (Gy). Methods. Wistar rats were on a four-week normal or 5% MZC supplemented diet and afterward exposed to the single γ-ray irradiation of 2 and 10 Gy. Groups of rats were: a) on a normal diet (the control group, and 2Gy and 10Gy groups); b) on 5% MZC supplemented diet (the control group – MZC, MZC + 2Gy, and MZC + 10Gy groups). We measured malondialdehyde (MDA), glutathione (GSH) total, and activity of total super-oxide dismutase (tSOD) and manganese superoxide dismutase (MnSOD) in vulnerable brain regions (cerebellum, hippocampus, and cerebral cortex) and 8-OH-dG in plasma. Results. Lower MDA was found in the MZC+2Gy and MZC+10Gy groups compared to the 2Gy and 10Gy groups. Activity od total SOD was higher in the MZC+10Gy group than in the 10Gy one. GSH was the highest in the 10Gy group. Compared to the control group, 8-OH-dG was extremely higher in groups radiated with 10 Gy regardless of a diet, but slightly lower in the MZC+2Gy and 2Gy groups. Conclusion. Subacute MZC pretreatment accomplished partial radioprotective effect in irradiated rats compared to non-irradiated rats, based on suppressed SOD activity at 2 Gy, and reduced brain MDA when exposed to 2 Gy and 10 Gy.

AB  - Examination of cadmium (Cd) toxicity and disulfiram (DSF) effect on liver was focused on oxidative stress (OS), bioelements status, morphological and functional changes. Male Wistar rats were intraperitoneally treated with 1mg CdCl2/kg BW/day; orally with 178.5 mg DSF/kg BW/day for 1, 3, 10 and 21 days; and co-exposed from 22nd to 42nd day. The co-exposure nearly restored previously suppressed total superoxide dismutase (SOD), catalase (CAT) and increased glutathione peroxidase (GPx) activities; increased previously reduced glutathione reductase (GR) and total glutathione-S-transferase (GST) activities; reduced previously increased superoxide anion radical (O-2(center dot-)) and malondialdehyde (MDA) levels; increased zinc (Zn) and iron (Fe), and decreased copper (Cu) (yet above control value), while magnesium (Mg) was not affected; and decreased serum alanine aminotransferases (ALT) levels. Histopathological examination showed signs of inflammation process as previously demonstrated by exposure to Cd. Overall, we ascertained partial liver redox status improvement, compared with the formerly Cd-induced impact.

AB  - The objective of our study was to examine testicular toxicity of cadmium (Cd), focusing on oxidative stress (OS), essential metals and androgenic status and morphological changes. Male Wistar rats [controls and four Cd-subgroups (n = 6) organized according to the exposure (1, 3, 10 and 21 days)] were intraperitoneally (i.p.) treated with 1 mg CdCl2/kg/day. Testicular Cd deposition was noticed from the 1st day. After 10 and 21 days, copper (Cu) and iron (Fe) increased by 60-109% and 43-67%, respectively, while zinc (Zn) decreased by 24-33%. During 121 days of the exposure, decrease in testicular total superoxide dismutase (SOD) and total glutathione-s-transferase (GST) activities occurred gradually by 30-78% and 15-84%, respectively, while superoxide anion radical (O-2(center dot-)) increased gradually by 114-271%. After 10-21 days, decrease in testicular catalase (CAT) activity appeared by 13-31%. After 21 days, malondialdehyde (MDA) decreased by 44% and the ratio of oxidized glutathione/reduced glutathione (GSSG/GSH) increased by 130% in testes of the rats exposed to Cd. Additionally, decreased testicular testosterone level and the relative testes mass, along with induced microscopic and macroscopic changes were occured, what can be explained as the consequence of instantly developed OS, impaired essential metals status and Cd testicular deposition.

Effects of different anesthetic agents on GM-CSF, MCP1, IL1α and TNFα levels in rat sepsis model

AB  - Anesthetic agents could alter the course and outcome of physical trauma, as well as experimentally or naturally occuring severe infections, by regulating several immune response mechanisms. The aim of our study was to investigate the influence of several commercially used anesthetic agents (ketamine, propofole, pentylentetrazole - PTZ) on cytokine concentrations, animal survival and pathohistological changes in the model of rat sepsis. In adult, male Wistar rats after different anesthetic traeatment and induction of sepsis by cecal ligation and punction we estimated serum levels of IL1α, TNFα, GM-CSF and MCP-1 at 12h intervals. After 48h of sepsis induction, the largest number of animals survived in the group treated with PTZ (47%), while the lowest survival rate was in the propofole treatment group (24%). Contrary to survival rate, the most abundant pathohistological changes were seen on preparations from PTZ and than in ketamine/PTZ treated groups, without any significant changes in the CNS of propofole treated animals. In the propofole treated group there was a prominent increament of GM-CSF values at 12h and 24h, followed by a significant decreament at 36h. These changes were negatively correlated to the survival rate in this group. This group had the lowest levels of MCP1 at all evaluated time intervals. After high initial levels, IL1α and TNFα levels fell to undetectable concentrations and at 24h increased to a high level. In PTZ as well as ketamine groups, at 12 h interval, GM-CSF levels were lower than in the propofole treated group. Contrary, MCP-1 levels were higher in these groups comparing to propofole group. After a high initial peak, IL1α levels decreased to low but detectable levels, followed by an intensive rise in ketamine treated, but with further decrement in pentazole treated groups. TNFα levels were low through all evaluated intervals in both these groups. Our results indicate that induction of anaesthesia of animals with sepsis with variuos anesthetic agents is connected to different pathohistological CNS changes, distinct serum cytokine profiles and diverse survival rates.
AB  - Anestetici mogu izmeniti tok i ishod uticaja fizičke traume kao i eksperimentalno ili prirodno nastalih teških infekcija, regulacijom nekoliko mehanizama imunskog odgovora. Cilj naše studije je bio da ispitamo uticaj nekoliko komercijalno korišćenih anestetika (ketamina, propofola i pentzlentetrazola) na koncentraciju citokina, preživljavanje životinja i patohistološke promene u modelu sepse indukovane u pacova. U odraslih pacova, muškog pola, po primeni različitih anestetika i indukcije sepse podvezivanjem i punkcijom cekuma, utvrđivali smo nivoe IL1α, TNFα, IL4, IFNy, GM-CSF i MCp-1 u intervalima od 12 sati. Nakon 48 sati od indukcije sepse, najveći broj životinja preživeo je u grupi tretiranoj pentylenetetrazolom (50%), potom u ketamin/pentylenetetrazol tretiranoj grupi, dok je najmanja stopa preživljavanja bila u grupi tretiranoj propofolom (22%). Nasuprot stopi preživljavanja, najizraženije patohistološke promene su utvrđene na preparatima grupa tratiranih pentazolom i ketamin/pentylenetetrazol, bez značajnih promena u CNS u grupi životinja tretiranih propofolom. U uzorcima grupe tretirane propofolom detektovali smo intezivan porast vrednosti GM-CSF 12h i 24h, praćen značajnim padom posle 36h. Ove promene su negativno korelirale sa stopom preživljavanja u ovoj grupi. Ova grupa je imala i najniže vrednosti MCP-1 u svim praćenim vremenskim intervalima. Posle visokih inicijalnih nivoa, vrednosti IL1a i TNFα pale su na nedetektabilne a u terminu od 24h porasle su na visok nivo. U grupama tretiranim pentazolom i ketaminom, 12. sata, vrednosti GM-CSF bile su niže od grupe tretirane propofolom. Nasuprot tome, vrednosti MCP-1 su bile veće u ovim grupama u odnosu na propofolom tretiranu grupu. Posle visokog inicijalnog skoka, vrednosti IL1α smanjuju se na niske ali detektabilne vrednosti, a zatim sledi intezivan rast u ketaminom tretiranoj grupi i dalji pad vrednosti u pentilenetetrazol tretiranoj grupi. Vrednosti TNFα su u ove dve grupe bile niske u svim ispitivanim intervalima. Vrednosti IL4 i IFNy su bile praktično nedetektabilne u svim ispitivanim grupama. Naši rezultati ukazuju da je indukcija anestezije u životinja sa sepsom različitim anesteticima povezana sa različitim patohistološkim nalazima u CNS, različitim serumskim profilom citokina i različitom stopom preživljavanja.

AB  - Immunomodulation is a phenomenon that is based on the influence of different endogenous and exogenous substances on the function of molecules and cells pertaining to the immune system. In this paper the effects of bovine thymus extract (Thymex - L) on the parameters of humoral immune response (concentration of antibodies to tetanus toxoid - ATTx) and cellular immune response (interleukin 2 - IL-2, interferon γ - IFN-γ and degree of cutaneus basophyl hypersensitivity - CBHR) in rats previously irradiated by X-rays at a dose of 4 or 6 Gy. The studied thymus extract had shown positive effects on IL-2 and IFN-γ serum concentrations in irradiated rats. In the group of rats irradiated with 4 Gy the effects were evident on the 7th day of the trial. Rats irradiated with the higher dose (6 Gy) experienced the positive effects of thymus extract on the 21st day of the experiment. In the skin PHA hypersensitivity test, a positive effect was also recorded, being more pronounced in rats which received the lower irradiation dose. The tested thymus extract had shown a positive effect on the concentration of ATTx as early as the 14th day of the trial.
AB  - Pojedini sastojci ekstrakta goveđeg timusa imaju imunomodulatorna svojstva i mogu, kod ljudi i životinja, sa suprimiranim funkcijama imunskog sistema (humoralnim i/ili celularnim) dovesti do imunostimulacije. U ovom radu su prikazani rezultati ispitivanja efekata preparata Thymex-L dobijenog iz ekstrakta goveđ eg timusa na parametre humoralnog imunskog odgovora pacova kod kojih je prethodno izvršeno jednokratno ozračivanje X zracima u dozi od 4 ili 6 Gy. Određivani su sledeći parametri: koncentracija interleukina 2 (IL-2), interferona γ (IFN-γ), koncentracija antitela posle vakcinacije i revakcinacije tetanusnim toksoidom (ATTx) i stepen kožne preosetljivosti (CBHR) na biljni mitogen (PHA) Ispitivani ekstrakt je ispoljio pozitivan efekat na koncentraciju IL-2 i IFN-γ u serumu ozračenih pacova. Ovaj pozitivan efekat je, kod grupe pacova koja je primila nižu dozu zračenja, vidljiv već nakon 7. dana eksperimenta, dok je kod pacova ozračenih višom dozom zračenja pozitivan efekat registrovan 21. dana eksperimenta. Pri proceni stepena reakcije kožne preosetljivosti na PHA, registrovan je pozitivan efekat primenjenog ekstrakta timusa i on je bio izraženiji kod jedinki kod kojih je bila primenjena niža doza zračenja. Ispitivani ekstrakt goveđeg timusa je ispoljio pozitivan efekat i na koncentraciju ATTx i to već 14. dana eksperimenta.
