AB  - Hepatic stellate cells (HSCs) play a significant role in hepatic fibrogenesis. In the following study we described the distribution of cells that express alpha-smooth muscle actin (α-SMA) and desmin in the cat liver with various degrees of fibrosis, as well as the significance of hepatic stellate cells and portal myofibroblasts in the genesis of fibrosis in cats with polycistic kidney disease. Liver samples from 15 necropsied Persian cats were examined microscopically, using H&E and Masson-trichrom methods and immunohistology for α-SMA and desmin. Liver fibrosis was confirmed in cats with terminal stage of chronic cholangiohepatitis and it was characterized by connective tissue septa which divide the liver parenchyma into irregular lobuli. Inflammation in the cat liver is connected with the activation of periductal myofibroblasts. The intensity of immunopositivity of perisinusoidal HSCs to α-SMA and desmin varied depending on the degree of fibrosis and was the strongest in livers of cats with cirrhosis.
AB  - U ovom radu je opisana distribucija ćelija koje eksprimiraju α-glatkomišićni aktin (α-SMA) i dezmin u jetri mačaka sa različitim stadijumom fibroze, kao i značaj ovih ćelija u razvoju fibroze, kod mačaka sa policističnim bubrezima. Mikroskopski su ispitani uzorci jetre od 15 obdukovanih persijskih mačaka, korišćenjem HE i Mason-trihromnog bojenja i imunohistohemijski na α-SMA i dezmin. Fibroza jetre potvrđena je kod mačaka sa terminalnim stadijumom hroničnog holangiohepatitisa i odlikovala se prisustvom vezivno-tkivnih septi koje dele parenhim na nejednake lobuluse. Inflamacija u jetri mačaka u vezi je sa aktivacijom periduktalnih miofibroblasta. Intenzitet imunohistohemijske reakcije perisinusoidnih HSCs na α-SMA i dezmin razlikovala se od stepena fibroze i bio je najjači u jetri mačaka sa cirozom.

Biological properties of a naturally attenuated infectious bursal disease virus isolated from a backyard chicken flock

AB  - The biological properties of an infectious bursal disease (IBD) virus isolated from bursas collected during an outbreak in a village chicken flock in Macedonia are described. The mortality rate was 50%. Two viruses coexisted in the bursas of infected chickens (IBDVwt and IBDVtc). The virus termed IBDVtc grows on chicken embryo fibroblast (CEF) cells from the first passage. Specific pathogen free chickens inoculated with IBDVtc at passage level 4 did not develop any clinical signs of disease. Some discrete bleeding on the leg muscles was seen and the bursa of Fabricius revealed pathological lesions similar to those caused by classical strains. However, the bursa recovered quickly (bursa lesion score 2) by 14 days post infection (PI). We also found evidence of bursal repopulation by means of perinuclear antigen staining. Strong CD3 influx was evident at 4 days PI, and at 33 days PI the CD3+ cell finding was comparable to the control. The mean antibody titre was 9.2 log(2) at 14 days PI. The amino acid composition of VP2 in IBDVwt (222 Ala, 242 Ile, 253 Gln, 256 Ile, 279 Asp, 284 Ala, 294 Ile and 299 Ser) is described. The same sequence was found in IBDVtc, except for two point mutations, at Gln253 -> His and Ala284 -> Thr. Such amino acid substitution is responsible for partial attenuation and the ability of the strain to replicate in cell culture. None of the commercial vaccine viruses has a similar arrangement of amino acids in the variable domain of IBDV. This strongly suggests that IBDVtc originates from a very virulent strain. To the best of our knowledge, this is the first report of a concomitant infection of chickens with highly pathogenic IBDV and its mutant counterpart.

Expression of α-SMA, desmin and vimentin in canine liver with fibrosis

AB  - In this work, 36 canine livers with fibrosis were examined. Based on the degree of fibrosis, the livers were classified into three groups: 1- mild portal fibrosis; 2- moderate portal fibrosis with mild periportal and septal fibrosis; 3- severe portal fibrosis with marked periportal and septal fibrosis or cirrhosis. Normal liver sections obtained from five dogs with no evidence of infectious, neoplastic, or cardiovascular diseases were used as controls. The degree of fibrosis was evaluated on liver sections stained according to Masson's trichrome method. It was proved using an immunohistochemical method that HSC and periductal myofibroblasts are immunoreactive to desmin, vimentin and α-SMA. Desmin, vimentin and α-SMA positive cells in the portal triad were significantly more numerous in dogs with fibrosis of the third degree than in the controls or all other examined livers. The density of α- SMA, vimentin and desmin-positive cells in the portal triad was in correlation with the degree of fibrosis, which cannot be said for HSC. These results suggest that portal myofibroblasts have a significant role in the process of liver fibrosis in dogs.
AB  - U ovom radu su prikazani rezultati ispitivanja 36 uzoraka tkiva jetre sa znacima fibroze, poreklom od pasa. Na osnovu stepena fibroze, uzorci su klasifikovani u tri grupe: 1 - blaga portalna fibroza; 2 - umerena portalna fibroza sa blagom periportalnom i septalnom fibrozom; 3 - teška portalna fibroza sa izraženom periportalnom i septalnom fibrozom ili cirozom. Kao kontrola su korišćeni uzorci tkiva jetre dobijeni od pet pasa bez znakova infekcije, neoplazije i kardiovaskularnih bolesti. Evaluacija stepena fibroze je urađena na isečcima jetre bojenim po Masson trihromnoj metodi. Imunohistohemijskom metodom je pokazano da su HSC i periduktalni miofibroblasti imunoreaktivni na dezmin, vimentin i α-SMA. Dezmin, vimentin i α-SMA, pozitivne ćelije u portalnoj trijadi bile su značajno brojnije kod pasa sa fibrozom trećeg stepena u odnosu na kontrolu i na sve druge ispitivane uzorke tkiva jetre. Gustina α-SMA, vimentin i dezmin pozitivnih ćelija u portalnoj trijadi je bila u korelaciji sa stepenom fibroze, što se ne može reći i za HSC. Ovi rezultati sugerišu da portalni miofibroblasti imaju značajnu ulogu u procesu fibroze jetre kod pasa.

Morphological findings in the cranial mesenteric artery of horses with verminous arteritis

AB  - Cranial mesenteric arteries of 18 sacrificed necropsied horses of both sexes and different age groups were described in this paper. After macroscopic examination tissue samples for pathohistological examinations were routinely processed and stained with hematoxylin eosine (HE), Weigert van Gieson and Periodic-Acid-Schiff (PAS) staining. Immunohistochemical staining was performed on selected sections using avidin-biotin-peroxidase complex technique for ásmooth muscle actin (α-SMA). Enlarged, thickened cranial mesenteric arteries with a hardelastic consistency and narrowed lumen, were macroscopically evident in all examined sick horses. Live larvae of Strongylus vulgaris, situated free in the lumen, or attached to the intima of the blood vessel or incorporated in the thrombus were noted macroscopically in 37.5% examined sick horses. Inflammatory and fibrous changes were noticed and were present in the intima, media and adventitia of the blood vessel. The inflammatory infiltrate in the intima of the cranial mesenteric artery consisted of eosinophils, macrophages, plasma cells, lymphocytes and rare multinucleated giant cells and was situated mostly close to the Strongylus vulgaris larvae. Beside inflammatory changes, intimal fibrosis and extension, characterized by increased proliferation of α-SMA positive cells, was notable. Lesions of the internal elastic lamina consequently led to an inflammatory and fibrotic reaction in the tunica media. Fibrosis of the media characterized by the presence of connective tissue cells and fibers, as well as smoothmuscle cells, was present in 93.75% examined sick horses. Inflammation and fibrosis were mildest in the adventitia. Lesions of vasa vasorum were present in 81.25% examined sick animals were characterized by fibrosis, obliteration, perivascular cellular infiltration, mostly with eosinophils and findings of intimal bodies. All described changes are characteristic for cranial mesenteric artery verminous arteritis.
AB  - U ovom radu su ispitane kranijalne mezeneterijalne arterije 18 obdukovanih žrtvovanih konja različitog pola i starosti. Arterije su makroskopski pregledane i uzimani su tkivni isečci za patohistološka ispitivanja. Materijal za histološka ispitivanja fiksiran je u 10% puferizovanom formalinu i posle uobičajene procedure ukalupljen u parafinske blokove. Patohistološki preparati bojeni su hematoksilinom i eozinom (HE), Vajgert van Gison metodom i PAS metodom. Odabrani isečci imunohistohemijski su bojeni avidin-biotin-peroksidaza tehnikom za α-smooth muscle actin (α-SMA). Makroskopski, kranijalne mezenterijalne arterije svih ispitanih konja bile su uvećane, zadebljale, čvrsto-elastične konzistencije i suženog lumena. Žive larve Strongylus vulgaris slobodne u lumenu ili pričvršćene za intimu krvnog suda ili inkorporisane u trombnu masu uočene su makroskopski kod 37,5% obolelih konja. Mikroskopski su uočene promene inflamatornog do fibroznog karaktera i bile su zastupljene u intimi, mediji i adventiciji krvnog suda. Inflamatorni infiltrat u intimi su činili eozinofilni granulociti, makrofage, plazma ćelije, limfociti i višejedarne džinovske ćelije i ovaj infiltrat bio je prisutan uglavnom u blizini larve Strongylus vulgaris. Pored inflamatornih promena uočeno je zadebljavanje sa fibrozom intime koja se karakterisala proliferacijom vezivno-tkivnih ćelija i vlakana, kao i glatko-mišićnih ćelija. Lezije na unutrašnjoj elastičnoj lamini posledično su dovodile do inflamacije i fibroze tunike medije. Fibroza medije se karakterisala prisustvom vezivno-tkivnih ćelija i vlakana, kao i glatko-mišićnih ćelija i bila je prisutna kod 93,75% obolelih konja. Inflamacija i fibroza su najslabije bile izražene u adventiciji, a lezije vasa vasorum su bile prisutne kod 81,25% pregledanih obolelih životinja i karakterisale su se fibrozom, obliteracijom, perivaskularnom ćelijskom infiltracijom, uglavnom sa eozinofilima, kao i nalazom intimalnih tela. Sve opisane promene su karakteristične za verminozni arteritis kranijalne mezenterijalne arterije.

abstract = "Cranial mesenteric arteries of 18 sacrificed necropsied horses of both sexes and different age groups were described in this paper. After macroscopic examination tissue samples for pathohistological examinations were routinely processed and stained with hematoxylin eosine (HE), Weigert van Gieson and Periodic-Acid-Schiff (PAS) staining. Immunohistochemical staining was performed on selected sections using avidin-biotin-peroxidase complex technique for ásmooth muscle actin (α-SMA). Enlarged, thickened cranial mesenteric arteries with a hardelastic consistency and narrowed lumen, were macroscopically evident in all examined sick horses. Live larvae of Strongylus vulgaris, situated free in the lumen, or attached to the intima of the blood vessel or incorporated in the thrombus were noted macroscopically in 37.5% examined sick horses. Inflammatory and fibrous changes were noticed and were present in the intima, media and adventitia of the blood vessel. The inflammatory infiltrate in the intima of the cranial mesenteric artery consisted of eosinophils, macrophages, plasma cells, lymphocytes and rare multinucleated giant cells and was situated mostly close to the Strongylus vulgaris larvae. Beside inflammatory changes, intimal fibrosis and extension, characterized by increased proliferation of α-SMA positive cells, was notable. Lesions of the internal elastic lamina consequently led to an inflammatory and fibrotic reaction in the tunica media. Fibrosis of the media characterized by the presence of connective tissue cells and fibers, as well as smoothmuscle cells, was present in 93.75% examined sick horses. Inflammation and fibrosis were mildest in the adventitia. Lesions of vasa vasorum were present in 81.25% examined sick animals were characterized by fibrosis, obliteration, perivascular cellular infiltration, mostly with eosinophils and findings of intimal bodies. All described changes are characteristic for cranial mesenteric artery verminous arteritis., U ovom radu su ispitane kranijalne mezeneterijalne arterije 18 obdukovanih žrtvovanih konja različitog pola i starosti. Arterije su makroskopski pregledane i uzimani su tkivni isečci za patohistološka ispitivanja. Materijal za histološka ispitivanja fiksiran je u 10% puferizovanom formalinu i posle uobičajene procedure ukalupljen u parafinske blokove. Patohistološki preparati bojeni su hematoksilinom i eozinom (HE), Vajgert van Gison metodom i PAS metodom. Odabrani isečci imunohistohemijski su bojeni avidin-biotin-peroksidaza tehnikom za α-smooth muscle actin (α-SMA). Histopathological evaluation of most frequent liver diseases in cats

AB  - Histological analysis was performed on the liver of 27 cats of different breeds, age and sex, autopsied at the Department of Pathological Morphology of the Faculty of Veterinary Medicine of the University of Belgrade, and findings showed lymphoplasmocytic cholangiohepatitis, different degrees of fibrosis, passive hyperemia, fatty changes, cholestase, and neoplasms. The mentioned entities occurred very rarely as individual morphological manifestations, because the liver has different functions so that one morphological change often causes others to follow. Thus, for example, fibrosis was often followed by intrahepatic cholestase. Histopathological evaluation of liver disease is important not only in the autopsied samples, but also in diagnostics of liver diseases, in samples obtained by biopsy, which is important for making a precise diagnosis. The interpretation of the established histological changes in the liver requires close cooperation between clinicians and pathologists, because the final diagnosis is made on the grounds of morphological, biochemical and clinical findings.
AB  - Histološkom analizom jetre kod 27 mačaka različite rase, pola i starosti, obdukovanih na Katedri za patološku morfologiju Fakulteta veterinarske medicine Univerziteta u Beogradu, ustanovljeni su limfoplazmocitni holangiohepatitis, fibroza različitog stepena, pasivna hiperemija, masna promena, holestaza i neoplazme. Navedeni entiteti su se vrlo retko javljali kao pojedinačne morfološke manifestacije, pošto jetra ima različite funkcije, pa jedna morfološka promena često za sobom povlači i druge. Tako je, na primer, fibroza bila često praćena intrahepatič nom holestazom. Histopatološka procena oboljenja jetre ima značaja ne samo na uzorcima sa obdukcije, već i kod dijagnostike oboljenja jetre, na uzorcima dobijenim biopsijom, što je značajno za postavljanje decidne dijagnoze. Za tumačenje ustanovljenih histoloških promena u jetri potrebna je tesna saradnja između kliničara i patologa, jer se konačna dijagnoza postavlja na osnovu morfoloških, biohemijskih i kliničkih nalaza.

CD 57 expression on lymphocytes present in the caecum and caecal tonsils in broilers infected with Eimeria tenella

AB  - In order to detect and establish the expression of the CD57 lymphocyte population in the caecum and caecal tonsils in broilers, immunohistochemical studies of tissue samples of broilers experimentally infected with E. tenella at 21 days of age were carried out. Immunohistochemical investigations were performed with the aid of the avidin-biotin technique (LSAB), by using mouse monoclonal anti- CD57 antibodies. Increased immunoreactivity of the lamina epithelialis, mucosal lamina propria and caecal submucosa was detected as early as two days after infection, increasing at its highest at the end of the fourth day after infection. On the fifth, sixth and seventh day after infection the degree of expression of CD57 lymphocytes in the caecal mucosa of experimental birds decreases. The immunoreactivity of the tissue of the stroma of the mucosal and submucosal lamina propria of the caecal tonsils was most intensive the fourth, sixth and seventh day after infection. A discrete immunoreactivity decrease was present during the fifth day after infection. The so established CD57 lymphocyte population activity in the caecum and caecal tonsils substantiates the existence of NK cytotoxic activity of intraepithelial lymphocytes, as well as the role of these cells in the intestinal mucosa defense mechanisms against intracellular microorganisms.
AB  - U cilju detekcije i utvrđivanja ekspresije CD57 populacije limfocita u cekumu i cekalnim tonzilama pilića, izvedeno je imunohistohemijsko ispitivanje tkivnih isečaka kod jedinki koje su 21. dana starosti eksperimentalno inficirane protozoom E. tenella. Ova ispitivanja su izvedena avidin-biotin tehnikom (LSAB), korišćenjem monoklonskih mišijih anti-CD57 antitela. Pojačana imunoreaktivnost laminae epithelialis, laminae propriae mukoze i submukoze cekuma zapažena je već drugog dana posle infekcije, sa najvećim intenzitetom krajem četvrtog dana. Petog, šestog i sedmog dana posle infekcije, stepen ekspresije CD57 limfocita u mukozi cekuma eksperimentalnih životinja se smanjivao. Imunoreaktivnost tkiva cekalnih tonzila najintenzivnija je četvrtog, šestog i sedmog dana posle infekcije, u stromi laminae propriae mukoze i submukozi, uz postojanje blagog pada u petom danu posle infekcije. Dokazana imunoreaktivnost CD57 populacije limfocita u cekumima i cekalnim tonzilama, potvrđuje NK ćelijsku citotoksičnu aktivnost populacije intraepitelnih limfocita i ukazuje na važnu ulogu ovih ćelija u organizaciji odbrane crevne mukoze protiv intracelularnih mikroorganizama.

Canine rib osteosynthesis by reinforced acrylate plate: A new surgical method for treating flail chest

AB  - Polymethylmetacrylate (PMMA) is an artificial resin that has a wide application in a number of medical disciplines, surgery included. The use of acrylate is limited due to its modest mechanical features. Hence in this study, as an 'in vivo' experiment on dogs, the usefulness of 'reinforced acrylate' in the form of armored acrylate plates in the surgery of fractured ribs in flail chest has been valued. In earlier studies we have shown that polymethylmetacrylate, when reinforced with non-alkaline glass fibers, has improved mechanical characteristics (bending, stretching and crash resistance) compared with not reinforced, or in any other way strengthened acrylate. In this study the value of armored acrylate plates (AAP) in the sanation of flail chest in dogs has been verified, as well as its good biological tolerability as a new implant material in dogs. An original approach to surgical reposition and osteosynthesis of fractured ribs in dogs has been determined. An innovative approach to rib osteosynthesis with acrylate plates can be an adequate model for its application in human patients with unstable flail chest, which is often the result of injury or tumor excision.
AB  - Polimetilmetakrilat (PMMA) je veštačka smola koja je našla široku primenu u brojnim medicinskim oblastima, uključujući i hirurgiju. Kako je primena akrilata ograničena njegovim relativno slabim mehaničkim osobinama, u ovom radu je u 'in vivo' eksperimentu na psima ispitana upotrebna vrednost 'ojačanog akrilata' odnosno armirane akrilatne pločice u hirurškom lečenju preloma rebara kod nestabilnog zida grudnog koša (torakalnog kapka). Našim ranijim istraživanjima dokazali smo da polimetilmetakrilat armiran bezalkalnim staklenim vlaknima poseduje znatno bolja mehanička svojstva (jačinu na savijanje, istezanje i udar) od nearmiranog ili na neki drugi način ojačanog akrilata. U ovom radu je u ogledima na psima dokazana vrednost armirane akrilatne pločice (AAP) u lečenju torakalnog kapka i potvrđena dobra biološka podnošljivost AAP kao implantnog materijala. Utvrđen je originalni postupak hirurške repozicije i osteosinteze prelomljenih rebara, a usvojena originalna tehnika osteosinteze rebara akrilatnom pločicom kod pasa može da bude adekvatan model postupka za primenu kod ljudi sa nestabilnim zidom grudnog koša, kao posledice teških povreda ili defekta zida toraksa prilikom ekscizije tumora.

abstract = "Polymethylmetacrylate (PMMA) is an artificial resin that has a wide application in a number of medical disciplines, surgery included. The use of acrylate is limited due to its modest mechanical features. Hence in this study, as an 'in vivo' experiment on dogs, the usefulness of 'reinforced acrylate' in the form of armored acrylate plates in the surgery of fractured ribs in flail chest has been valued. In earlier studies we have shown that polymethylmetacrylate, when reinforced with non-alkaline glass fibers, has improved mechanical characteristics (bending, stretching and crash resistance) compared with not reinforced, or in any other way strengthened acrylate. In this study the value of armored acrylate plates (AAP) in the sanation of flail chest in dogs has been verified, as well as its good biological tolerability as a new implant material in dogs. An original approach to surgical reposition and osteosynthesis of fractured ribs in dogs has been determined. An innovative approach to rib osteosynthesis with acrylate plates can be an adequate model for its application in human patients with unstable flail chest, which is often the result of injury or tumor excision., Polimetilmetakrilat (PMMA) je veštačka smola koja je našla široku primenu u brojnim medicinskim oblastima, uključujući i hirurgiju. Kako je primena akrilata ograničena njegovim relativno slabim mehaničkim osobinama, u ovom radu je u 'in vivo' eksperimentu na psima ispitana upotrebna vrednost 'ojačanog akrilata' odnosno armirane akrilatne pločice u hirurškom lečenju preloma rebara kod nestabilnog zida grudnog koša (torakalnog kapka). Našim ranijim istraživanjima dokazali smo da polimetilmetakrilat armiran bezalkalnim staklenim vlaknima poseduje znatno bolja mehanička svojstva (jačinu na savijanje, istezanje i udar) od nearmiranog ili na neki drugi način ojačanog akrilata. U ovom radu je u ogledima na psima dokazana vrednost armirane akrilatne pločice (AAP) u lečenju torakalnog kapka i potvrđena dobra biološka podnošljivost AAP kao implantnog materijala. Utvrđen je originalni postupak hirurške repozicije i osteosinteze prelomljenih rebara, a usvojena originalna tehnika osteosinteze rebara akrilatnom pločicom kod pasa može da bude adekvatan model postupka za primenu kod ljudi sa nestabilnim zidom grudnog koša, kao posledice teških povreda ili defekta zida toraksa prilikom ekscizije tumora.",
