Citoplazmatske kapi spermatozoida nerastova – abnormalnost ili fiziološka pojava?

AB  - Reprodukcija svinja danas predstavlja osetljivo polje delovanja veterinarske službe koje
značajno utiče na profitabilnost svinjarske proizvodnje. Bez obzira na ovu činjenicu, neki
standardni pregledi razređenog semena nerastova kroz duži vremenski period predstavljaju
nezamenljiv način kontrole kvaliteta dobijenog ejakulata. Cito-morfološki pregled izvodi se u
svrhu procene učestalosti pojavljivanja abnormalno građenih spermatozoida. Citoplazmaske kapi
javljaju se na srednjem delu spermatozoida, a po mestu lokacije, dele se na proksimalne i distalne.
Distalne citoplazmatske kapi ukazuju na nezrelost spermatozoida, rano uvođenje mladih
nerastova u reproduktivnu eksploataciju, prekomerno iskorišćavanje priplodnjaka ili pak
posledično pojavljivanje ovakvih spermatozoida kao odgovor na ambijentalne oscilacije ili druge
zdravstvene probleme vezane za nerasta. Ovaj rad predstavlja istraživanje koje je sprovedeno na
nerastovima (n=30), durok i landras rasne pripadnosti. Eksperimentalna grupa životinja dobijala
je peroralno zajedno sa kompletnom smešom za ishranu, dodatak preparata sa antioksidativnim
potencijalom. Za potrebe bojenja spermatozoida korišćena je modifikovana metoda ‚‚u jednom
koraku” (Mortimer, 1994; Jovičin i sar., 2012). Primena dvofaktorske ANOVE za ponovljena
merenja pokazala je da nema statistički značajne razlike između kontrolne i eksperimentalne
grupe nerastova u nalazima citoplazmatskih kapi spermatozoida (%) (F(7, 112)= 1,65, p= 0,13) u
odabranim vremenskim tačkama. Iako nije zabeležena statistički značajna razlika, ovakav
rezultat, odnosno konstanta u broju ovakvih spermatozoida nerastova označava i ujednačenost
uslova pod kojim je ekseriment sproveden.

AB  - Nizak antioksidativni kapacitet spermatozoida nerastova, često može biti i jedan
od glavnih razloga lošeg kvaliteta semena. Iz ove činjenice proizilazi i veoma velika
podložnost spermatozoida oksidativnom stresu i različitim štetnim efektima. Kada
proizvodnja reaktivnih vrsta kiseonika (ROS) nadmaši unutrašnju antioksidativnu
zaštitu, dolazi do probijanja sistema biološke odbrane, što za posledicu ima oštećenje
ćelije. Narušena redoks homeostaza ćelije, usled prekomernog stvaranja, ali i
neadekvatnog uklanjanja slobodnih radikala dovodi do oksidativnog stresa. Napad
reaktivnih vrsta kiseonika dovodi do smanjene pokretljivosti spermatozoida,
smanjene vitalnosti spermatozoida i povećanja morfoloških abnormalnosti
spermatozoida sa štetnim efektima po kapacitaciju spermatozoida i akrozomalnu
reakciju. Sa druge strane, niske koncentracije reaktivnih vrsta kiseonika igraju važnu
ulogu u fiziološkim procesima kao što su kapacitacija spermatozoida, reakcija
akrozoma i fertilizacija. Veća koncentracija antioksidativnih molekula u spermalnoj
plazmi, kompenzuje nizak antioksidativni kapacitet samih spermatozoida nerastova.
Sistem antioksidativne zaštite sastoji se od enzima koji uklanjaju slobodne radikale,
neenzimskih komponenti, specifičnih oksidoreduktaza i proteina. U membrani,
citoplazmi i organelama spermatozoida i spermalnoj plazmi, nalaze se enzimi
supreroksid dismutaza, glutation peroksidaza, glutation reduktaza, katalaza, među
kojima se superoksid dismutaza smatra najvažnijim antioksidativnim enzimom. Cilj
ovog rada je da na osnovu pregleda literature prikaže negativan efekat oksidativnog
stresa, ali i sistem antioksidativne zaštite koju poseduje sprmalna plazma nerastova.

AB  - Svinjarska proizvodnja se u današnjoj praksi ne može zamisliti bez veštačkog
osemenjavanja. Za maksimalnu eksploataciju nerastova uvek mora da se veže dobar kvalitet
ejakulata, kao i dobro zdravstveno stanje nerasta. Spermatozoidi nerastova zbog svog niskog
antioksidativnog kapaciteta kontinuirano su negativno izloženi oksidativnom stresu.
Potencijalna klinička korist za poboljšanje antioksidativne zaštite i smanjivanje
negativnih uticaja na spermatozoide, od strane suplementnih preparata, i dalje izaziva različita i
podeljena mišljenja u naučnim krugovima. Oksidativni stres nastaje kada unutrašnja
antioksidativna odbrana biološkog sistema biva nadvladana od prekomernog nakupljanja/
proizvodnje reaktivnih vrsta kiseonika.
Preliminarna istraživanja ove grupe autora na uzorku od 30 nerastova, gde je
eksperimentalna grupa dobijala kroz ishranu dodatak organskog selena i preparata Oxynat 3D,
govore u prilog da i ova kombinacija može potencijalno poboljšati kvalitet ejakulata. Ispitivani
nerastovi bili su rase durok i landras, prosečno stari godinu dana. Ispitivanje kvaliteta ejakulata
vršeno je ciklusno u vremenskom trajanju jedne spermatogeneze.
Kvalitet razređenog semena je određivan metodom CASA (Computer Assisted Sperm
Analysis), sa integrisanim softverskim sistemom za analizu spermatozoida, u četiri navrata.
Rezultati pokazuju statistički značajne razlike za ukupnu pokretljivost, volumen ejakulata
i broj preporučenih doza, dok je koncentracija spermatozoida bila bez statistički značajne
razlike. Navedene činjenice jasno ukazuju da kombinacija ova dva preparata, koji imaju
antioksidativni potencijal, dovodi do poboljšanja kvaliteta razređene sperme priplodnih
nerastova.

The influence of zinc and heavy metals in feed and water on the quality of cryopreserved bull semen

AB  - This study aimed to assess the influence of different concentrations of zinc (Zn), lead (Pb), mercury (Hg) and cadmium (Cd) found in the feed and water provided to bulls on the concentrations of these metals in cryopreserved bull semen, and to determine their influence onto semen quality parameters. Correlations between heavy metal concentrations in the semen and the quality parameters of semen as estimated by computer-assisted sperm analysis (CASA) and flow cytometry (FC) methods were determined. A total of 40 cryopreserved semen samples originating from bulls housed in 4 different centers for artificial insemination (A, B, C and D) were examined, making a total of 160 samples. The concentrations of metals and semen quality parameters were determined in cryopreserved semen of 10 bulls from each center, namely 4 samples from each bull. Concentrations of Zn, Pb, Hg and Cd in hay and concentrated feed were within the allowed limits as proposed by the National Research Council (NRC, 2000). A strong negative correlation was detected between curvilinear velocity (VCL) and Zn concentrations in the semen (P < 0.01; r = -0.772) in group D, and a positive correlation of VCL with Pb concentrations (P < 0.05 and r = 0.718) in group B. Mercury concentrations in cryopreserved semen correlated negatively to the percent of live sperm cells with intact acrosomes (V/IA: P < 0.05; r = -0.640) and positively with the percent of dead sperm cells with damaged acrosomes (D/DA: P < 0.01; r = 0.766) in group D. This finding confirms the hypothesis that Hg, even at low concentrations, may cause acrosome damage.
AB  - Cilj rada bio je da se ispita uticaj različitih koncentracija cinka (Zn), olova (Pb), žive
(Hg) i kadmijuma (Cd) iz hrane i vode na koncentracije ovih metala u duboko zamrznutom semenu nakon otapanja i da se ispita njihov uticaj na parametre kvaliteta
semena. Korelacije između koncentracije teških metala u semenu i parametra kvaliteta semena su određivane nakon ispitivanja semena kompjuterski asistiranom analizom
semena (eng. computer-assisted sperm analysis - CASA) i protočnom citometrijom
(eng. flow cytometry - FC).
Iz 4 različita centra za veštačko osemenjavanje (A, B, C i D) ispitivano je po 40 uzoraka duboko zamrznutog semena bikova, što čini ukupno 160 uzoraka. Koncentracije
metala i parametri kvaliteta semena su određivani u duboko zamrznutom semenu od
10 bikova iz svakog centra, odnosno 4 uzorka od svakog bika. Koncentracije Zn, Pb,
Hg i Cd u senu i koncentrovanim hranivima bile su ispod propisanih dozvoljenih
vrednosti Nacionalnog Istraživačkog Veća (eng. National Research Council - NRC,
2000). Jaka negativna korelacija uočena je između krivolinijske brzine spermatozoida
(eng. curvilinear velocity - VCL) i koncentracije Zn u semenu (P < 0.01; r = -0.772) u

The concentrations of metals and semen quality parameters were determined in cryopreserved semen of 10 bulls from each center, namely 4 samples from each bull. Concentrations of Zn, Pb, Hg and Cd in hay and concentrated feed were within the allowed limits as proposed by the National Research Council (NRC, 2000). A strong negative correlation was detected between curvilinear velocity (VCL) and Zn concentrations in the semen (P < 0.01; r = -0.772) in group D, and a positive correlation of VCL with Pb concentrations (P < 0.05 and r = 0.718) in group B. Mercury concentrations in cryopreserved semen correlated negatively to the percent of live sperm cells with intact acrosomes (V/IA: P < 0.05; r = -0.640) and positively with the percent of dead sperm cells with damaged acrosomes (D/DA: P < 0.01; r = 0.766) in group D. This finding confirms the hypothesis that Hg, even at low concentrations, may cause acrosome damage., Cilj rada bio je da se ispita uticaj različitih koncentracija cinka (Zn), olova (Pb), žive
(Hg) i kadmijuma (Cd) iz hrane i vode na koncentracije ovih metala u duboko zamrznutom semenu nakon otapanja i da se ispita njihov uticaj na parametre kvaliteta
semena. Korelacije između koncentracije teških metala u semenu i parametra kvaliteta semena su određivane nakon ispitivanja semena kompjuterski asistiranom analizom
semena (eng. computer-assisted sperm analysis - CASA) i protočnom citometrijom
(eng. flow cytometry - FC).
Iz 4 različita centra za veštačko osemenjavanje (A, B, C i D) ispitivano je po 40 uzoraka duboko zamrznutog semena bikova, što čini ukupno 160 uzoraka. Koncentracije
metala i parametri kvaliteta semena su određivani u duboko zamrznutom semenu od
10 bikova iz svakog centra, odnosno 4 uzorka od svakog bika. Koncentracije Zn, Pb,
Hg i Cd u senu i koncentrovanim hranivima bile su ispod propisanih dozvoljenih
vrednosti Nacionalnog Istraživačkog Veća (eng. National Research Council - NRC,
2000). Jaka negativna korelacija uočena je između krivolinijske brzine spermatozoida
(eng. curvilinear velocity - VCL) i koncentracije Zn u semenu (P < 0.01; r = -0.772) u
grupi D, i pozitivna korelacija između VCL i koncentracije Pb (P < 0.05 and r = 0.718)

Proteini spermalne plazme nerastova kao genetski markeri kvaliteta semena

AB  - Današnje savremeno svinjarstvo ne može da se zamisli bez primene biotehnološke
metode veštačkog osemenjavanja. Efikasno reproduktivno iskorišćavanje, genetski
superiornih nerastova, zahteva formiranje maksimalnog broja inseminacionih doza po
ejakulatu, odnosno po nerastu godišnje. Rezultati brojnih istraživanja dokazuju da bi
proteini spermalne plazme mogli imati značajan uticaj na variranje stepena progresivne
pokretlјivosti spermatozoida i održavanje fertilizacionog kapaciteta razređene
sperme pojedinih nerastova. U naše istraživanje je bilo uključeno 15 nerastova rase
švedski landras (5) veliki jorkšir (5) i durok (5) približne starosti od 18 do 24 meseca,
u punoj proizvodnoj ekploataciji. Od svakog nerasta su uzeta po tri ejakulata (ukupno
45 ejakulata), u razmaku od mesec dana, u hladnijem delu godine (oktobar-novembar-
decembar). Kod svih nerastova je utvrđivan procenat ukupnih proteina u spermalnoj
plazmi. Na osnovu dobijenih rezulata sadržaja ukupnih proteina u spermalnoj plazmi,
ispitivani nerastovi su podeljeni u dve grupe, grupa (N)-(niski): nerastovi sa niskim
sadržajem proteina (≤ 2,0) i grupa (V)-(visoki): nerastovi sa visokim sadržajem proteina
(≥ 2,1). Poređenjem parametara kvaliteta razređenog semena između ove dve grupe,
utvrđeno je da su prosečne vrednosti svih ispitivanih parametara fertilizacionog potencijala
razređenog semena bile statistički značajno veće u grupi V u odnosu na grupu
N, (p<0,01). Tako su prosečne vrednosti za ukupan broj spermatozoida u dozi iznosile
6,293×109 (V) i 3,595×109 (N), ukupan broj pokretnih spermatozoida u dozi 4,771×109
(V) i 1,325×109(N), procenat ukupno pokretnih spermatozoida 74,20 (V) i 36,26 (N),
ukupan broj progresivno pokretnih spermatozoida 2,982×109 (V) i 731,47×106(N), a
procenat progresivno pokretnih spermatozoida je iznosio 47,90 (V) i 21,42 (N). Nisu
ustanovljene statistički značajne (p>0,05) razlike u prosečom sadržaju proteina u spermalnoj
plazmi, između ispitivanih rasa nerastova. Ova vrednost je iznosila 2,43 procenta
kod rase landras, 2,45 kod rase veliki jorkšir i 1,83 kod rase durok. Prosečna vrednost
sadržaja proteina u spermalnoj plazmi nije značajno (p>0,05) varirala ni u zavisnosti od starosti ispitivanih nerastova. Navedene činjenice, ukazuju da sadržaj proteina u
spermalnoj plazmi, može biti efikasan marker fertilizacionog potencijala nerastova, na
osnovu koga se oni mogu selekcionisati za upotrebu u veštačkom osemenjavanju.

Quality and fertility of extended boar semen after prolonged transport

AB  - The aim of this study was to analyse semen quality parameters of fresh-extended porcine semen imported from Denmark through prolonged importing regulations and transport and to analyse fertility performance in sows inseminated four days post semen collection. Semen was analysed immediately upon the arrival (4 days post collection) using computer-assisted sperm analyses (CASA), cytomorphology and flow cytometry, and sows were inseminated later the same day. The mean sperm concentration was 1718.09 ± 100.11 × 106, with average of 1193.40 ± 90.58 × 106 motile cells (69%) and 756.04 ± 70.57 × 106 progressive motile spermatozoa (43.58%) in a dose, which was almost twice lower in value than producer certified-declared number of 2 billion of motile cells. Acrosome and sperm membrane integrity test indicated high percentage of total damaged acrosome. Mean values of semen samples analysed by sperm chromatin structure assay (SCSA) showed relatively high degree of chromatin damage. Membrane permeability test showed high percentage of sperm with damaged membrane. Prolonged transport negatively affected the sperm quality in terms of both motility and chromatin structure stability. Fertility of sows was affected by semen quality in terms of concentration and sperm motility parameters, as well as ratio of live sperm cells population with damaged acrosome. © 2021 TÜBİTAK.

Uticaj zračenja semena nerastova crvenim delom svetlosnog spektra na veličinu legla krmača

AB  - U ovom izlaganju će biti prikazan uticaj zračenja semena nerastova crvenim delom
svetlosnog spektra neposredno pre osemenjavanja na veličinu legla krmača. Korišćene su LED
lampe ugrađene u klima boks koje emituju spektar crvene boje, talasne dužine od 620 do 630
nm, sa režimom rada od 10 minuta svetlosti, 10 minuta odmora i 10 minuta dodatnog zračenja.
Osemenjavanja su sprovedena u periodu od aprila do maja 2021. godine, na farmi od 1
200 priplodnih krmača. U kontrolnoj grupi su bile 174 krmače, sa prosečnih 4,36 prašenja, a u
oglednoj 64, sa paritetom od 4,14. Nazimice nisu bile uključene u statističku obradu.
U oglednoj grupi je registrovan veći broj živorođene prasadi za 0,87 (14,05 prema
13,17), ali bez statističke značajnosti, dok je u istoj grupi, zabeležen znatno manji broj avitalne
i mrtvorođene prasadi (3,55 prema 4,68; p<0,05). Broj mumificirane prasadi u leglima je bio
identičan (0,45).
Zračenje semena crvenim svetlosnim zracima, neposredno pre osemenjavanja imalo je
pozitivan efekat na broj živorođene prasadi u leglu, ali samo u numeričkoj vrednosti, dok je
jasna statistička razlika uočena u pogledu njihove vitalnosti.
Ovaj ogled je u toku i očekuje se analiza znatno većeg broja prašenja, a time i jasniji
uvid u efekte ovakvog tretmana semena na oprasivost.
AB  - In this presentation, the influence of irradiation of boar semen by red light spectrum
immediately before insemination on the litter size in sows will be presented. LED lamps that
emit a red color spectrum with a wavelength of 620-630 nm built into the air conditioning box
were used. A mode of operation was: 10 minutes of light, 10 minutes of rest and 10 minutes of
additional irradiation.
Inseminations were carried out in the period from April to May 2021, on a farm of 1,200
breeding sows. Control group consisted of 174 sows with an average of 4.36 parities, and
experimental group of 64 sows, with a parity of 4.14. Gillts were excluded from the statistics.
In the experimental group, a higher number of live-born piglets was observed by 0.87
(14.05 vs. 13.17) but with no statistical significance, while in the same group, a significantly
lower number of avital and stillborn piglets was also observed (3.55 vs. 4.68; p <0.05 ). The
number of mummified piglets in the litters was identical (0.45 piglets) for both groups.
Semen irradiation with red light rays immediately before insemination had a positive
effect on the number of live-born piglets in the litter, but only in numerical value, while a clear
statistical difference was observed in terms of vitality of piglets in litters.
This trial is in progress and a significantly larger number of farrowings and a clearer
insight into the efficiency of such semen treatment on the fertility is expected.

200 priplodnih krmača. U kontrolnoj grupi su bile 174 krmače, sa prosečnih 4,36 prašenja, a u
oglednoj 64, sa paritetom od 4,14. Nazimice nisu bile uključene u statističku obradu.
U oglednoj grupi je registrovan veći broj živorođene prasadi za 0,87 (14,05 prema
13,17), ali bez statističke značajnosti, dok je u istoj grupi, zabeležen znatno manji broj avitalne
i mrtvorođene prasadi (3,55 prema 4,68; p<0,05). Broj mumificirane prasadi u leglima je bio
identičan (0,45).
Zračenje semena crvenim svetlosnim zracima, neposredno pre osemenjavanja imalo je
pozitivan efekat na broj živorođene prasadi u leglu, ali samo u numeričkoj vrednosti, dok je
jasna statistička razlika uočena u pogledu njihove vitalnosti.
Ovaj ogled je u toku i očekuje se analiza znatno većeg broja prašenja, a time i jasniji
uvid u efekte ovakvog tretmana semena na oprasivost., In this presentation, the influence of irradiation of boar semen by red light spectrum
immediately before insemination on the litter size in sows will be presented. LED lamps that
emit a red color spectrum with a wavelength of 620-630 nm built into the air conditioning box
were used. A mode of operation was: 10 minutes of light, 10 minutes of rest and 10 minutes of
additional irradiation.
Inseminations were carried out in the period from April to May 2021, on a farm of 1,200
breeding sows. Control group consisted of 174 sows with an average of 4.36 parities, and
experimental group of 64 sows, with a parity of 4.14. Gillts were excluded from the statistics.
In the experimental group, a higher number of live-born piglets was observed by 0.87
(14.05 vs. 13.17) but with no statistical significance, while in the same group, a significantly
lower number of avital and stillborn piglets was also observed (3.55 vs. 4.68; p <0.05 ). The
number of mummified piglets in the litters was identical (0.45 piglets) for both groups.
Semen irradiation with red light rays immediately before insemination had a positive
effect on the number of live-born piglets in the litter, but only in numerical value, while a clear
statistical difference was observed in terms of vitality of piglets in litters.
This trial is in progress and a significantly larger number of farrowings and a clearer
insight into the efficiency of such semen treatment on the fertility is expected.",

AB  - The aim of the study was to determine whether the bacteria from the environment and from the mucous membrane of the boar prepuce have antimicrobial resistance and whether the result obtained is similar/same to the bacteria that can be found in native boar semen. The study addresses the problem of the presence of primarily resistant bacterial strains in the boar sperm, which, due to their reduced sensitivity, cannot be suppressed by antibiotics used in the semen dilution agent, as well as to emphasize the importance of microbiological monitoring of the boar mucous membranes and ambient surfaces before and during their exploitation. Such an examination could contribute to the interchangeable design of the dilution agent for the boar semen relative to the antibiotic content.Resistant strains of bacteria from prepuce swabs and swabs taken from the facility, as well as from native boar semen were isolated. The presence of these bacteria affected the quality of the semen. In conclusion, it should be pointed out that bacterial monitoring of the prepuce and surface of the facility can indicate possible problems related to the quality of semen, and that the design of the dilution agent for boar semen should be adjusted to the established resistance of isolated bacteria.
AB  - Cilj ispitivanja je da se utvrdi da li bakterije iz okoline i sa sluznici prepucijma poseduju antimikrobnu rezistenciju i da li je dobijeni nalaz sličan/isti sa bakterijama koje se mogu naći u nativnom semenu nerasta. Ispitivanje treba da odgovori na problem prisustva, pre svega rezistentnih bakterijskih sojeva u spermi nerasta koji zbog svoje smanjene osteljivosti ne mogu biti suprimirani antibioticima, koji se nalaze u razređivaču za seme, kao i da istakne značaj mikrobiološkog monitoringa sluznice nerasta i ambijentalnih površina pre i tokom njihove eksploatacije. Ovakvo ispitivanje bi moglo da doprinese izmenjivom dizajniranju razređivača za seme nerasta u odnosu na sadržaj antibiotika. Izolovani su rezistentni sojevi bakterija iz briseva prepucijuma i briseva uzetih iz objekta kao i iz nativnog semena nerasta. Prisustvo ovih bakterija uticalo na kvalitet semena. Kao zaključak treba istaći da bakterijski monitoring prepucijuma i površina u objektu može ukazati na moguće probleme vezane za kvalitet semena nerasta kao i da se dizajniranje razređivača za seme nerasta prilagodi utvrđenoj rezistenciji izolovanih bakterija.

AB  - The aim of this study was to investigate the effect of different percentage of seminal plasma proteins with different molecular weight on sperm motility and fertility parameters (farrowing rate (FR), number of live-born pigs (PBA) per litter and percentage of unsuccessful insemination). A total of 50 sperm-rich ejaculate fractions were collected (one per boar) using the gloved hand method. The quality parameters of the semen samples were first evaluated at the farm. Further assessment of sperm quality was performed on a CASA - computer assisted semen analysis by two competent operators. Seminal plasma protein fractions were obtained by AOAC -Association of Official Analytical Chemists as a chemical method. The assessment of reproductive performance was carried out based on collected data of three parameters in selected 9696 sows: FR, PBA per litter and percentage of unsuccessful insemination. Protein fractions were divided in to three groups (10 - 20kDa, 21 - 30kDa and 31-40kDa) Proteins with 10 - 20kDa did not have significant effect and correlation with analyzed parameters. Significant differences were recorded in farrowing rate between samples with up to 80 % compared to samples with 10% of proteins with 21 - 30kDa. Significant differences were recorded in unsuccessful insemination between samples with different percentage of proteins with 31 - 40kDa. Results of this study have shown the effect of different percentage of certain fraction of seminal plasma proteins on boar ejaculates fertility potential.

AB  - Introduction. Artificial insemination of sows with cooled semen has become a routine practice in the swine industry and has almost completely ruled out natural mating. The storage time of the cooled semen and preservation of its qualitative parameters are of the upmost importance and depend partly on the semen extender used. The aim of this study was to determine the quality of cooled semen during storage in a thermobox with a commercial extender by assessment of sperm motility, cytomorphology, and assessment of reproductive parameters in sows after intrauterine insemination. Materials and Methods. The semen was preserved with Duragen® (Magapor, Spain) extender and stored at 17±1°C. A total of 110 sows were included in the study and randomly divided into four groups: K1 (n=25) , K2 (n=25), K3 (n=30), and K4 (n=30). The sows were inseminated twice with semen (>1x107/ml spermatozoa) which had been stored for 1 day (S1), 3 days (S3), 5 days (S5), or 7 days (S7). Pregnancy was diagnosed by imaging ultrasound. Results and Conclusions. The best results were obtained when the sows were inseminated with semen stored for one day. However, there were no statistically significant differences in the number and vitality of newborn piglets when semen stored for 3 or 5 days was used. Intrauterine insemination resulted in a satisfactory number of farrowing's and piglets despite the fact that a relatively low number of spermatozoa was determined in each dose used. The use of semen with a higher dilution rate and its intrauterine application ensured a larger number of obtained doses from one ejaculate and indicates this would lead to a more profitable use of boars.
AB  - Uvod. Veštačko osemenjavanje svinja rashlađenim semenom je postala rutinska praksa u industrijskoj proizvodnji svinja, pa je u razvijenim zemljama skoro potpuno zamenilo prirodno parenje. U zavisnosti od vrste upotrebljenog razređivača, dužina čuvanja rashlađenog semena i očuvanje njegovih kvalitativnih parametara je od izuzetne važnosti. Cilj istraživanja je analiza kvaliteta rashlađenog semena tokom njegovog skladištenja u termoboksu pri 17±1°C. Materijal i metode. Seme je konzervirano upotrebom razređivača Duragen® (Magapor, Španija) i skladišteno pri temperaturi od 17±1°C. U ogled je uključeno 110 krmača, podeljenih u četiri grupe (K1, K3, K5, K7). Krmače su osemenjivane dvokratno semenom skladištenim 1 dan (S1), 3 dana (S3), 5 dana (S5) i 7 dana (S7). Ultrazvučnim pregledom je vršena dijagnostika graviditeta. Kvalitet semena je ocenjivan ispitivanjem pokretljivosti spermatozoida i citomorfološkim pregledom i praćenjem reproduktivnih parametara kod intrauterino osemenjenih krmača. Rezultati i zaključak. Najbolji rezultat je postignut osemenjavanjem krmača semenom starim jedan dan. Međutim, nije bilo značajne razlike u broju i vitalnosti oprašene prasadi korišćenjem rashlađenog semena starog 3 i 5 dana. Intrauterinim osemenjavanjem su dobijeni zadovoljavajući rezultati (procenat prašenja i veličina legla) sa manjim brojem spermatozoida u inseminacionoj dozi. Upotrebom semena sa većim razređenjem i njegovim intrauterinim deponovanjem, omogućeno je dobijanje većeg broja inseminacionih doza semena od jednog ejakulata i rentabilnije korišćenje nerastova u reprodukciji.

abstract = "Introduction. Artificial insemination of sows with cooled semen has become a routine practice in the swine industry and has almost completely ruled out natural mating. The storage time of the cooled semen and preservation of its qualitative parameters are of the upmost importance and depend partly on the semen extender used. The aim of this study was to determine the quality of cooled semen during storage in a thermobox with a commercial extender by assessment of sperm motility, cytomorphology, and assessment of reproductive parameters in sows after intrauterine insemination. Materials and Methods. The semen was preserved with Duragen® (Magapor, Spain) extender and stored at 17±1°C. A total of 110 sows were included in the study and randomly divided into four groups: K1 (n=25) , K2 (n=25), K3 (n=30), and K4 (n=30). The sows were inseminated twice with semen (>1x107/ml spermatozoa) which had been stored for 1 day (S1), 3 days (S3), 5 days (S5), or 7 days (S7). Pregnancy was diagnosed by imaging ultrasound. Results and Conclusions. The best results were obtained when the sows were inseminated with semen stored for one day. However, there were no statistically significant differences in the number and vitality of newborn piglets when semen stored for 3 or 5 days was used. Intrauterine insemination resulted in a satisfactory number of farrowing's and piglets despite the fact that a relatively low number of spermatozoa was determined in each dose used. The use of semen with a higher dilution rate and its intrauterine application ensured a larger number of obtained doses from one ejaculate and indicates this would lead to a more profitable use of boars., Uvod. Veštačko osemenjavanje svinja rashlađenim semenom je postala rutinska praksa u industrijskoj proizvodnji svinja, pa je u razvijenim zemljama skoro potpuno zamenilo prirodno parenje. U zavisnosti od vrste upotrebljenog razređivača, dužina čuvanja rashlađenog semena i očuvanje njegovih kvalitativnih parametara je od izuzetne važnosti. Cilj istraživanja je analiza kvaliteta rashlađenog semena tokom njegovog skladištenja u termoboksu pri 17±1°C. Materijal i metode. Seme je konzervirano upotrebom razređivača Duragen® (Magapor, Španija) i skladišteno pri temperaturi od 17±1°C. U ogled je uključeno 110 krmača, podeljenih u četiri grupe (K1, K3, K5, K7). Krmače su osemenjivane dvokratno semenom skladištenim 1 dan (S1), 3 dana (S3), 5 dana (S5) i 7 dana (S7). Ultrazvučnim pregledom je vršena dijagnostika graviditeta. Kvalitet semena je ocenjivan ispitivanjem pokretljivosti spermatozoida i citomorfološkim pregledom i praćenjem reproduktivnih parametara kod intrauterino osemenjenih krmača. Rezultati i zaključak. Najbolji rezultat je postignut osemenjavanjem krmača semenom starim jedan dan. Međutim, nije bilo značajne razlike u broju i vitalnosti oprašene prasadi korišćenjem rashlađenog semena starog 3 i 5 dana. Intrauterinim osemenjavanjem su dobijeni zadovoljavajući rezultati (procenat prašenja i veličina legla) sa manjim brojem spermatozoida u inseminacionoj dozi. Upotrebom semena sa većim razređenjem i njegovim intrauterinim deponovanjem, omogućeno je dobijanje većeg broja inseminacionih doza semena od jednog ejakulata i rentabilnije korišćenje nerastova u reprodukciji.",

AB  - The aim of the present study was to investigate the influence of the protein content of seminal plasma on the motility, viability and acrosome integrity of spermatozoa in extended semen stored for 3 days. A total of 32 semen samples (from four boars) with high (4 mg/ml) and 32 semen samples (from four boars) with low (2 mg/ml) protein content were investigated. The semen samples were diluted by BTS at a ratio of 1:4, and stored for 72 h at 17oC. The percentages of live sperm (LS), live sperm with damaged acrosome (LDA) and total sperm with damaged acrosome (TDA) were detected by flow cytometry. Sperm progressive motility (PM) was detected using CASA. After 72 h of storage, the percentage of LS and PM was significantly (P  lt  0.01) higher, and the LDA and TDA were significantly (P  lt  0.01) lower in samples with high protein content than in the samples with low protein content (LS = 66 vs. 44%, PM = 64 vs. 48%, LDA = 15 vs. 21% and TDA = 29 vs. 45%, respectively). When comparing the difference between 0 and 72 h of storage, the percentage decrease in LS and PM, while increase in LDA and TDA were significantly higher in the samples with low (LS: 75 to 44%; PM: 68 to 48%; LDA: 11 to 21% and TDA: 23 to 45%) than in the samples with high protein content (LS: 78 to 66%; PM: 70 to 64%; LDA: 9 to 15% and TDA: 17 to 29%). We concluded that protein content in seminal plasma has a significant influence on progressive motility, viability and acrosome integrity in diluted semen stored for 3 days.

Boar and season effects on some parameters of semen fertilizing potential

AB  - In order to determine the more accurate fertile potential of sperm, it seems that the conventional parameters of boar semen quality (the ejaculate volume, sperm concentration, progressive motility, percentage of live sperm and of those with intact acrosomal morphology) are insufficient. Since recently, there have been numerous studies proving that protein concentration in sperm plasma has high positive correlation with boar fertile potential. The research objective was to determine the effect of boars as well as the season on the variation of protein content in the sperm plasma. For the research there were used spermal fractions of 2 boars with high (V-boar) and 2 boars with low (N-boar) protein content in spermal plasma. The ejaculates of boars were taken once a week, for a month, during one year (4 *12 = 48 ejaculates per boar). For protein analysis in the spermal plasma, the samples were prepared by centrifugation. The ejaculate volume, protein concentration and progressive motility varied considerably (p  lt  0.05 or p  lt  0.01) among the boars as well as in one and the same boar. The variations of the same parameters were determined also during both warm and cold season. On the other hand, protein concentration was rather constant, and in V-boars (ranged from 4 to 4.5%) while in N-boars (ranged from 2.3 to 2.6%). The season did not significantly affect (p > 0.01) protein content in sperm plasma (V-boars: 4 to 4.5% in warm and cold season; N-boars: 2.3 do 2.6% in warm and 2.3 to 2.5% in cold season). The obtained results showed that measurement of protein content in boar sperm plasma could be a useful method for their ranking, based on fertile potential of fresh semen.
AB  - U cilju određivanja što tačnijeg fertilizacionog potencijala spermatozoida, čini se da konvencionalni parametri kvaliteta semena nerastova (volumen ejakulata, koncentracja spermatozoida, progresivna pokretljivost, procenat živih spermatozoida, kao i onih sa intaktnom akrozomalnom morfologijom) nisu dovoljni. Od nedavno, postoje brojna istraživanja koja dokazuju da koncentracja proteina u spermalnoj plazmi ima visoku pozitivnu korelaciju sa fertilizacionim potencijalom nerastova. Cilj ovog istraživanja bio je da se utvrdi efekt nerasta kao i uticaj godišnje sezone na varijaciju sadržaja proteina u spermalnoj plazmi nerastova. Za ispitivanja su korištene spermalne frakcje 2 nerasta sa visokim (V-nerast) i 2 narasta sa niskim (N-nerast) sadržajem proteina u spermalnoj plazmi. Ejakulati su od nerastova uzimani jednom nedeljno, u okviru jednog meseca, tokom godinu dana (4*12=48 ejakulata po nerastu). Za analizu proteina u spermalnoj plazmi, uzorci su pripremljeni centrifugiranjem. Volumen ejakulata, koncentacja spermatozoida kao i progresivna pokretljivost su značajno varirali (p  lt  0,05 ili p  lt  0,01) između nerastova kao i kod jednog te istog nerasta. Varjacje istih parametara utvrđene su i tokom tople odnosno hladne sezone godine. Dobijeni rezultati pokazuju da bi merenje sadržaja proteina u spermalnoj plazmi nerastova mogao biti koristan metod za njihovo rangiranje, a na osnovu fertilizacionog potencijala svežeg semena.

AB  - Recently, it was frequently demonstrated that fertility of sows after artificially inseminated is lower than after mating. This is associated with a reduced fertilization capacity of overdiluted insemination doses. The aim of this study was to investigate the sperm motility in the semen samples, forming from the ejaculates with high or low protein content, stored in vitro on 17 degrees C for 3 days. Progressive motility was significantly higher (p<0.01) in the ejaculates with high, compared to the ejaculates with low protein content (82% vs. 76%). After 3 days of storage, in the1:4 dilution proportion, the average progressive motility was significantly (p<0.01) decreased in relation to this value in native semen from the boars with high (82% to 64%), as well from the boars with low protein content in seminal plasma (76% to 48%). However, the average diluted semen progressive motility was significantly greater (p<0.01) in the boars with high (64%), compared to the boars with low protein content in seminal plasma (48%). The number of good diluted semen samples (>= 65% progressive motility), was also significantly (p<0.01) greater in the boars with high (41%), compared to the boars with low protein content in seminal plasma (12%). These results show that seminal plasma proteins play an important role in maintaining the sperm progressive motility of diluted semen in vitro stored for 3 days.

AB  - The aim of this study was to investigate: (a) seminal plasma protein content in the Land-race (n = 38), Large White (n = 44) and Duroc (n = 24) boars breed, as well as boars distribution according to protein content in seminal plasma, and (b) the sperm motility in the semen samples, forming from the ejaculates with high or low protein content, stored in vitro on 17°C for 3 days. Not significant (p > 0.05) differences was found in seminal plasma protein content between three boar breeds. Only 31% of the 106 investigated boars had > 3.6% protein in seminal plasma. Progressive motility was significantly higher (p  lt  0.01) in the ejaculates with high, compared to the ejaculates with low protein content (82% vs. 76%). After 3 days of storage, in thel:4 dilution proportion, the average progressive motility was significantly (p  lt  0.01) decreased in relation to this value in native semen from the boars with high (82% to 64%), as well from the boars with low protein content in seminal plasma (76% to 48%). However, the average diluted semen progressive motility was significantly greater (p  lt  0.01) in the boars with high (64%), compared to the boars with low protein content in seminal plasma (48%). The number of good diluted semen samples (>65% progressive motility), was also significantly (p  lt  0.01) greater in the boars with high (41%), compared to the boars with low protein content in seminal plasma (12%).

Effect of season on boast semen quality

AB  - It was demonstrated that boar sperm production and characteristics significantly decrease in the wormer period of the year. As a result, the boars reproductive exploitation efficiency decrease in the wormer season. Therefore, the aim of the present paper was to investigate the influence of cold and warm season on the boars semen quality, in the our intensive pig production conditions. The obtained results clearly show that the main semen quality parameters were significantly (p lt 0.05 or p lt 0.01) higher in the cold, compared with the warm season (ejaculate volume=274ml, sperm concentration=229'106/ml, total sperm number=60'109, progressive motility=79%, and good ejaculates=96%, vs. 218ml, 208'106/ml, 45'109, 69% and 78%, resp.). In conclusion, keeping boars cool during summer and frequent observation of boars to determine if they are being heat- stressed, can significant reduce the negative influence of elevated ambient temperature on sperm production and, consequently, improve boars reproductive exploitation efficiency.
AB  - Ustanovljeno je da su produkcija i osobine sperme nerasta značajno niže tokom toplijeg perioda godine. Posledično, u ovom periodu godine značajno se smanjuje efikasnost reproduktivnog iskorištavanja nerastova. Zbog toga je cilj ovog rada bio da se ustanovi uticaj hladne i tople godišnje sezone na parametre kvaliteta sperme u našim uslovima intenzivne proizvodnje svinja. Dobijeni rezultati jasno pokazuju da su osnovni parametri kvaliteta ejakulata značajno veći (p lt 0.05 or p lt 0.01) u hladnoj, nego u toploj godišnjoj sezoni (volumen ejakulata=274ml, koncentracija spermatozoida=229'106/ml, ukupan broj spermatozoida=60'109, progresivna pokretljivost=79% i dobrih ejakulata=96%, prema 218ml, 208'106/ml, 45'109, 69% i 78%). Zaključak je da rashlađivanje nerastova tokom leta i češća observacija prisustva toplotnog stresa, može značajno smanjiti negativan uticaj povišene ambijentalne temperature na produkciju sperme i, posledično, povećati efikasnost reproduktivnog iskorištavanja nerastova.

AB  - The aim of the present study was to investigate the possibility of increasing boars reproductive exploitation by using AI doses of doubly reduced volume and sperm count in the intrauterine AI procedure. The experiment was conducted at a commercial pig farm in Serbia in 2014. Classic intracervical insemination (ICI) was performed by using 50 mL or 100 mL volume doses containing 4 x 10(9) or 2 x 10(9) progressively motile spermatozoa. The same volumes and sperm numbers per dose were used with intrauterine insemination (IUI). Each dose combination was used to inseminate 30 sows. Intrauterine insemination with AI doses of reduced volume (50 mL) and sperm count (2 x 10(9)) did not produce a statistically significant difference (P < 0.05) in the farrowing rate (76.7%) as compared with 4 x 10(9) spermatozoa in the same volume (83.3%) or with insemination by doses of 100 mL with a 2 x 10(9) (83.3%) or a 4 x 10(9) sperm count (86.7%). The number of live-born piglets (10.82) was larger following IUI using a 50 mL volume dose with a 2 x 10(9) sperm count as compared with ICI with the same AI dose volume and sperm count (9.85). The results show that the use of reduced AI dosages provides an opportunity for the swine industry to considerably exploit the reproductive potential of genetically superior boars.

Pararectal treatment of boars with primary semen contamination and astenospermia

AB  - Importing boars of high genetic value is an important issue in Serbia.
Therefore, frequent replacement of boars is not a primary approach
and there is a need for their prolonged exploitation. Ejaculates from
6 boars were selected for bacterial control and for treatment based
on lower CASA (Computer Assisted Semen Analyses) results, flow
cytometry and cytological sperm quality parameters in extended
semen. According to the sensitivity of bacterial isolates from fresh
semen, boars were treated with Enroxil  10% (1 ml/40 kg BW;
Krka, Slovenia), during 8 consecutive days. An antibiotic was
injected between the tail and rectum (pararectal therapy). On the 8th
day, boars received 8 ml of Baytril  10% (Bayer, Germany). On the
first and last day of treatment boars received 20 ml of AD3E
vitamins with selenium i/m and C vitamin (20 ml, 100 mg/ml).
Semen quality was controlled 45 days following treatment. Total and
progressive sperm motility increased from 39.66% and 12.87% to
83.98% and 61.98%, respectively (p < 0.001). The percentage of live
spermatozoa with intact acrosomes also improved (48.3–63.4%,
PNA-FITC, n.s.). The percentage of agglutinated cells decreased
from 66.00 to 6.67% and the average bacterial contamination in
fresh semen decreased from 18.450 to 1.233 CFU/ml. It is evident
that boars with primary semen contamination are able to renew
sperm production after prolonged antibiotic and supportive treatment. This could be a strategy to decrease the culling rate in elite
stud boars.

Finding of parasitic nematodes of fishes present in the market

AB  - When placing the fish and fish products on the trade it is necessary to pay attention to the presence of zoonotic parasites that can lead to infection of people especially if the fish is consumed raw or undercooked. Epidemiologically the most important are helminths from the group of Nematoda. The most important are Anisaxis spp. and Eustrongylides excisus. Anisakiasis is a serious zoonotic disease with a dramatic increase in prevalence throughout the world in the last two decades. The larvae are found in marine fish species most frequently in sardines, herring and mackerel. Eustrongylidosis is a disease that occurs primarily in freshwater fish species: catfish, zander and pike, a cause is a parasite of the genus Eustrongylides of which the most important species is Eustrongylides excisus. In this paper, the nematodes that occur in fish that are present in the market in the Republic of Serbia are present. Investigations of Anisaxis spp. were carried out at the Institute of Veterinary Medicine Novi Sad in the period of 2000-2013. Total of 2414 samples of imported marine fish was inspected. In 25 (1.29 % ) samples of herrings (Clupea harengus ) and mackerels ( Scomber scombrus ) was identified Anisakis spp. Investigations of Eustrongylides spp . were carried out in the period 2010-2013 at the Danube-Tisa-Danube Canal. Samples were collected from zander (Sander lucioperca) and European catfish (Silurisglanis) in which the presence of nematodes was found in the abdominal cavity, muscles, the lumen of the stomach and gastric wall where the parasites were encapsulated. Individuals of zander were examined during the 2013 at the Veterinary Institute Novi Sad, where the presence of larvae Eustrongylides spp. u muscle was detected. In order to avoid infecting people with parasites of fish it is necessary to carry out continuous control and monitoring. Fresh fishes and traditional fish products must be inspected for the presence of parasites before they find on the trade. Continuous education is a key factor in combating zoonotic diseases. Avoid consumption of raw or poorly cooked fish is still the best preventive procedure.
AB  - Prilikom stavljanja u promet i pregleda ribljeg mesa i proizvoda od ribe treba obratiti pažnju na prisustvo zoonotskih parazita koji mogu dovesti do inficiranja ljudi naročito ako se riba konzumira sirova ili termički nedovoljno obrađena. Sa epidemiološkog aspekta najveći značaj imaju helminti iz grupe Nematoda. Najznačajnije su Anisaxis spp. i Eustrongylides excisus. Anisakijaza je ozbiljno zoonotsko oboljenje sa dramatičnim porastom prevalence širom sveta u poslednje dve decenije. Larve se mogu naći kod morskih vrsta riba najčešće kod sardine, haringe i skuše. Eustrongilidoza je oboljenje koje se javlja kod slatkovodnih riba prvenstveno grabljivica: soma, smuđa i štuke, a izazivaju je paraziti iz roda Eustrongylides od kojih je najznačajnija vrsta Eustrongylides excisus. U ovom radu prikazane su nematode koje se javljaju kod riba koje su prisutne na tržištu u Republici Srbiji. Istraživanja prisustva Anisaxis spp. su sprovedena na Naučnom institutu za veterinarstvo Novi Sad u periodu 2000-2013. Ukupno je pregledano 2414 uzoraka riba iz uvoza od čega je u 25 (1.29%) utvrđen Anisakis spp., kod haringe (Clupea harengus) i skuše (Scomber scombrus). Istraživanja prisustva Eustrongylides spp. su sprovedena u periodu od 2010-2013 godine na kanalu Dunav-Tisa-Dunav. Prikupljeni su uzorci konzumnog smuđa (Sander lucioperca) i soma (Siluris glanis) kod kojih je utvrđeno prisustvo nematoda u abdomenu, muskulaturi, lumenu želuca i želudačnom zidu gde su paraziti bili inkapsulirani. Jedinke smuda pregledane se tokom 2013. godine i na Naučnom Institutu za veterinarstvo Novi Sad gde je utvrđeno prisustvo larvi Eustrongylides spp. u muskulaturi. Kako ne bi došlo do zaražavanja ljudi zoonotskim parazitima riba neophodno je sprovoditi stalnu kontrolu i monitoring. Sveže meso ribe i tradicionalni riblji proizvodi pre nego što se nadu u prometu moraju biti pregledani na prisustvo parazita. Kontinuirana edukacija je ključni faktor u borbi sa zoonozama a izbegavanje konzumiranja sirovog ili termički slabo obrađenog ribljeg mesa i dalje najbolja preventivna procedura.
