AB  - Homocysteine (Hcy) was investigated as the biomarker of cardiac, renal, and gastrointestinal disorders
in dogs. Data about low Hcy concentrations in the systemic inflammatory response syndrome raised a hypothesis that
Hcy in dogs could be a negative acute-phase reactant. This survey compared Hcy concentrations, serum amyloid A
(SAA), and the routine laboratory parameters between healthy (HD, N=6) and dogs with inflammation of different
extent: mild (dirofilariosis (DIR), N=31), moderate (babesiosis (BAB), N=12), and severe (pyometra (PYO), N=8).
The BAB and PYO groups had lower Hcy er than HD. Also, the levels in the PYO group were below those in the
DIRO group. SAA had the inverse pattern. Across the groups, Hcy and SAA levels correlated negatively (ρ = -0.502,
P<0.001). Hcy and SAA correlated with the erythrocyte count, hematocrit, hemoglobin and mean cellular hemoglobin
concentrations, and neutrophil count, with correlations being positive for Hcy and negative for SAA. Among all dogs,
hemoglobin was the only independent predictor of Hcy concentration. Hcy levels in canine infections, decreased as
acute-phase reaction (APR) intensified. Also, they were related with the hematology changes accompanying the APR.
Further studies will establish the clinical potential of these alterations.

AB  - Prema mehanizmu nastanka, anemije se klasifikuju na
hemoličke i hemoragijske i anemije koje nastaju usled
poremećaja na nivou kostne srži. Hemoličke i hemoragijske
anemije su najčešće praćene ubrzanom eritropoezom i jakim
regenerativnim odgovorom, koji se ogleda u značajnom

kostne srži, koji najčešće uključuju različite oblike
imunosupresije, inhibiciju proliferacije eritroidnih progenitora
i/ili nedostatak nutritivnih faktora, dijagnostikuju se kao

AB  - The highest number of acute Babesia canis cases in dogs is recorded over the February-May (Feb-May) period, which also represents the optimal climate conditions for tick activity in Belgrade, Serbia. A possibility that the acute phase response is more intense in dogs developing the disease in the Feb-May period compared with the response in other time periods of the year was tested. A total of 63 client-owned dogs with acute B. canis infection were enrolled and the routine hematology and biochemistry parameters-serum amyloid A (SAA), IgG against B. canis, level of parasitemia, ceruloplasmin (CER), paraoxonase-1 (PON-1), and fibrinogen-were measured. Acute phase indexes (API) were calculated as (SAA×CER)/(Iron×PON-1) and (SAA×CER)/(Albumin×Iron). Statistics included Kruskal-Wallis test and logistic regression analysis. The results showed that in the Feb-May period, the following parameters were lower: creatinine, albumin, iron, and level of parasitemia. Furthermore, increased API values were more probable in the Feb-May than in the other periods. Together, higher acute phase response intensity and presumptive hemodilution in the Feb-May period indicate a more acute course of B. canis infection than in other time periods of the year.

previously observed that in Belgrade, the capital of the Republic of Serbia, 60% of
cases with the acute-phase response (APR) give a notable maximum between mid-
February and May, the so-called “high season”. The other two peak seasons with
less pronounced maxima are in winter, from January to mid-February, and in
autumn, from October to December. Objectives: Based on clinical observations, we
postulate that dogs infected with B. canis in the “high season” develop a more
intense APR than dogs in the other two seasons. Material and Methods: To test this
hypothesis, we compare the relative parasite load and the level of acute-phase
proteins and reactants, their indexes (calculated as a combination of
concentrations of fast and slow positive, and fast and slow negative acute phase
reactants), as well as the concentration of coagulation factors, complete blood
count, and biochemistry among dogs that developed the disease in the autumn,
winter, and “high season”. Results: The relative parasite load is the lowest, and APR
Indexes are the highest in dogs infected in “high season”. These findings are
accompanied by relatively low creatinine and urea, implying that dogs in “high
season” had hemodilution, thus further suggesting short incubation. Unlike other
acute-phase proteins, fibrinogen showed the lowest concentration in “high
season”. Conclusion: Future research will help unveil the connection between
more intense APR and relatively low parasite load and fibrinogen concentration.

AB  - Background: Canine babesiosis, a tick-borne disease caused by Babesia canis,
shows a seasonality whose relationship with local weather conditions has not been
fully investigated. Objectives: Meteorological conditions can favour the tick-vector
activity, and thus lead to an increased number of cases of canine babesiosis. Hence,
our study looks into the link between the number of recorded cases, on the one
hand, and temperature and relative humidity on the other with an aim to quantify
their correlations. Material and Methods: Over 2013–2016, the data were
collected in Belgrade, the capital of the Republic of Serbia. The meteorological
parameters were obtained from the Republic Hydrometeorological Service of
Serbia. The analysis includes correlations with a time lag, given in number of weeks,
which shifts corresponding correlation pairs and shows a delayed effect of weather
conditions. The time lag ranges between 0 and 52. Results: Canine babesiosis
occurrence shows a pronounced maximum in the spring and a less marked one in
the autumn. For the spring period, statistically significant correlation coefficients
imply that over one year prior to the disease spring peak, temperature is more
strongly linked with the number of cases than relative humidity. Conclusion:
Temperature and relative humidity, through their influence on population of
infected ticks, seem to be important meteorological drivers of the spring maximum
of canine babesios in Belgrade. Further understanding of this interplay can help
better contain the disease, and project its possible spread to other regions
prompted by climate change.

AB  - Background: Lower homocysteine (Hcy) concentraon was noced in dogs with
systemic inflammatory response syndrome of various aeology, in comparison with
healthy dogs. Objecve. The study had two objecves: to compare Hcy
concentraons in serum between dogs naturally infected with Babesia canis and
healthy dogs; to assess the relaonship between Hcy concentraon and the results
of the roune laboratory tests. Materials and methods. The study enrolled twelve
dogs infected with B. canis (BAB) and six healthy dogs (HD). The commercial
immunoassay, spectrophotometry, and haematology kits were used. Stascal
analyses included the Mann-Whitney U test and Spearman's rank correlaon
analysis. Results: BAB had lower concentraons of Hcy than HD (p=0.045). When
analysing the results from all studied dogs together, Hcy had a posive correlaon
with erythrocyte count (r=0.771; p=0.001), haemoglobin concentraon (r=0.696;
p=0.004), haematocrit (r=0.664; p=0.007), leukocyte count (r=0.554; p=0.032), and
total protein concentraon (r=0.689; p=0.004), and a negave with glycaemia (r=-
0.693; p=0.004). Conclusion: The obtained results provide preliminary evidence
about the associaon between acute canine babesiosis and lowered Hcy
concentraon in blood. Further studies will decipher whether this finding has
clinical applicability or represents just an APR epiphenomenon.

AB  - Babezioza pasa, uzrokovana infekcijom protozoom Babesia canis, sezonska je bolest na području Beograda, glavnog grada Republike Srbije. Sezone pojave oboljenja se poklapaju sa aktivnošću vektora krpelja Dermacentor reticulatus. Period inkubacije varira od četiri do 21 dan, a kliničku sliku karakteriše sistemska akutna inflamacija ili u težim slučajevima sindrom sistemskog inflamatornog odgovora (eng. Systemic Inflammatory Response Syndrome – SIRS). Povezanost klimatskih i epidemioloških faktora koja bi precizno pokazala i okarakterisala sezone u kojima se ovo oboljenje javlja na teritoriji Beograda, do sada nije ispitana. Sa kliničko-epidemiološkog aspekta, nameće se pitanje, da li je pojava SIRS-a vezana za pojedine sezone, odnosno da li postoje razlike u intezitetu odgovora akutne faze kod pasa obolelih u različitim sezonama. Hipoteza ove studije bila je da je tokom prolećne sezone, u kojoj je učestalost pojave pasa akutno inficiranih B. canis najveća, odgovor akutne faze najintenzivniji, odnosno da će reaktanti akutne faze pokazati najveće promene, a da će broj pasa sa SIRS-om, biti najzastupljeniji.Prvi deo studije obuhvatio je ispitivanje učestalosti pojave oboljenja na teritoriji Beograda tokom četiri godine (2013–2016), u različitim sezonama. Ispitan je kratkoročni uticaj (dve nedelje) i dugoročni uticaj meteoroloških parametara (godinu dana) koji su prethodili pojavi 872 zabeležena slučaja babezioze pasa. Drugi deo studije uključio je 66 pasa, sa akutnom infekcijom B. canis tokom 2017–2019. godine, od kojih je prikupljan višak pune krvi i seruma koji su preostali posle dijagnostike oboljenja. Rutinskim laboratorijskim testovima su izvedene hematološke i biohemijske analize, dok je elektroforeza proteina izvedena na agaroznom gelu. Parazitemija je određena komercijalnom metodom lančane reakcije polimeraze, dok je prisustvo IgG antitela protiv B. canis ispitano komercijalnim serološkim testom. Reaktanti/proteini akutne faze određeni su sledećim testovima: koncentracija serum amiloida A komercijalnom ELISA-om (eng. Enzyme Linked Immunosorbent Assay), koncentracija ceruloplazmina i aktivnost paraoksonaze 1 spektrofotometrijski, dok je za koncentraciju fibrinogena primenjen metod zagrevanja uz precipitaciju proteina. Klasifikacija pasa na SIRS pozitivne i negativne izvedena je prema odgovarajućim kliničkim i laboratorijskim promenama, dok su indeksi reaktanata akutne faze (IRAF) dobijeni računskim putem. Razlike između varijabli ispitane su pomoću Kruskal-Wallis, Mann-Whitney U testa i Chi-square testa. Veze između varijabli kvantifikovane su Spirmanovim koeficijentima korelacije odnosno logističkom regresionom analizom.Rezultati analize distribucije pojave babezioze pasa omogućavaju podelu kalendarske godine na četiri sezone: januar–februar, februar–maj, jun–septembar i oktobar–decembar. AB  - Canine babesiosis, caused by infection with a protozoan Babesia canis, is a seasonal disease in the area of Belgrade, the capital of the Republic of Serbia. The seasons of the disease coincide with the activity of the vector Dermacentor reticulatus. The disease incubation period varies between 4 and 21 days, and the clinical picture is characterized by systemic acute inflammation or, in severe cases, Systemic Inflammatory Response Syndrome (SIRS). A connection between meteorological parameters and epidemiological factors that could help characterize the seasons in which this disease occurs in the region of Belgrade, has not been investigated previously. From the clinical-epidemiological aspect, there is a question whether the occurrence of SIRS is restricted to some seasons, or, in other words, whether there are differences in the intensity of the acute phase response in dogs infected in different seasons. This study hypothesized that during the spring season, when the frequency of dogs acutely infected with B. canis is the highest, the acute phase response is also the most intense, with acute-phase reactants showing the greatest changes, and the fraction of dogs with SIRS reaching its maximum.The first part of the study investigated the disease frequency in the Belgrade region in different seasons during four years (2013–2016) when a total of 872 were recorded. Parasitemia was measured by a polymerase chain reaction method in a commercial laboratory. The presence of IgG antibodies against B. canis was tested by a commercial serological test. The following techniques were used for analyses of acute-phase proteins: commercial Enzyme Linked Immunosorbent Assay for serum amyloid A concentration, spectrophotometry for ceruloplasmin concentration and paraoxonase 1 activity, and heat denaturation method for fibrinogen. The dogs were classified as SIRS positive or negative according to a predefined set of clinical and laboratory changes. The indexes of acute phase reactants (IRAF) were calculated. The significance of the differences between the variables was tested using the Kruskal-Wallis, Mann-Whitney U, and Chi-square tests. Hipoteza ove studije bila je da je tokom prolećne sezone, u kojoj je učestalost pojave pasa akutno inficiranih B. canis najveća, odgovor akutne faze najintenzivniji, odnosno da će reaktanti akutne faze pokazati najveće promene, a da će broj pasa sa SIRS-om, biti najzastupljeniji.Prvi deo studije obuhvatio je ispitivanje učestalosti pojave oboljenja na teritoriji Beograda tokom četiri godine (2013–2016), u različitim sezonama. Ispitan je kratkoročni uticaj (dve nedelje) i dugoročni uticaj meteoroloških parametara (godinu dana) koji su prethodili pojavi 872 zabeležena slučaja babezioze pasa. Drugi deo studije uključio je 66 pasa, sa akutnom infekcijom B. canis tokom 2017–2019. godine, od kojih je prikupljan višak pune krvi i seruma koji su preostali posle dijagnostike oboljenja. Rutinskim laboratorijskim testovima su izvedene hematološke i biohemijske analize, dok je elektroforeza proteina izvedena na agaroznom gelu. Parazitemija je određena komercijalnom metodom lančane reakcije polimeraze, dok je prisustvo IgG antitela protiv B. canis ispitano komercijalnim serološkim testom. Reaktanti/proteini akutne faze određeni su sledećim testovima: koncentracija serum amiloida A komercijalnom ELISA-om (eng. Enzyme Linked Immunosorbent Assay), koncentracija ceruloplazmina i aktivnost paraoksonaze 1 spektrofotometrijski, dok je za koncentraciju fibrinogena primenjen metod zagrevanja uz precipitaciju proteina. Klasifikacija pasa na SIRS pozitivne i negativne izvedena je prema odgovarajućim kliničkim i laboratorijskim promenama, dok su indeksi reaktanata akutne faze (IRAF) dobijeni računskim putem. Razlike između varijabli ispitane su pomoću Kruskal-Wallis, Mann-Whitney U testa i Chi-square testa. Veze između varijabli kvantifikovane su Spirmanovim koeficijentima korelacije odnosno logističkom regresionom analizom.Rezultati analize distribucije pojave babezioze pasa omogućavaju podelu kalendarske godine na četiri sezone: januar–februar, februar–maj, jun–septembar i oktobar–decembar. AB  - Hemolysis and systemic acute inflammation characterize canine babesiosis caused by the intraerythrocytic protozoan parasite Babesia canis. Our hypothesis was that blood redox homeostasis of patients that suffered acute B. canis infection might be disturbed even after treatment with imidocarb-dipropionate and successful clinical recovery. Eight owner dogs with acute B. canis infection were used for this study. We analyzed the complete blood count, acute phase proteins (ceruloplasmin, haptoglobin, paraoxonase-1) in the serum, antioxidant enzymes (catalase and glutathione peroxidase) in the erythrocytes, and oxidative stress markers (malondialdehyde in erythrocytes and thiol groups in serum) at presentation and 15 days after treatment. Results were evaluated by corresponding statistical tests. At presentation, anemia, low/normal leukocyte count and severe thrombocytopenia occurred together with increased ceruloplasmin, haptoglobin levels within the reference interval, decreased paraoxonase-1 and compromised antioxidant defense in the red blood cells. After treatment and successful clinical recovery, hematological values generally fitted within the reference intervals, acute phase proteins were within the physiological levels in the majority of cases and the activities of the antioxidant enzymes were increased. However, elevated malondialdehyde levels indicated increased oxidative damage of erythrocytes that remained as a deleterious sequel despite a successful clinical recovery of the dogs.
AB  - Babezioza pasa, koju izaziva protozoa Babesia canis, praćena je hemolizom i sistemskom akutnom infl amacijom. Naša hipoteza je da kod pasa sa akutnom B. canis infekcijom, čak i nakon terapije imidokarb-dipropionatom i uspešnog kliničkog oporavka, postoji poremećaj redoks homeostaze. Studija uključuje osam vlasničkih pasa akutno infi ciranih babezijom. Analizirani su kompletna krvna slika, proteini akutne faze (ceruloplazmin, haptoglobin i paraoksonaza-1) u serumu, antioksidativni enzimi (katalaza i glutation peroksidaza) u eritrocitima i markeri oksidativnog stresa (malondialdehid u eritrocitima, tiolne grupe u serumu) na prezentaciji i 15 dana nakon terapije. Za procenu značajnosti rezultata primenjeni su odgovarajući statistički testovi. Anemija, nizak/normalan broj leukocita i teška trombocitopenija praćeni povišenom koncentracijom ceruloplazmina, haptoglobinom u fi ziološkim intervalima i sniženim nivoom paraoksonaze-1, kao i kompromitovana antioksidativna zaštita, uočeni su na prezentaciji. Nakon terapije i uspešnog kliničkog oporavka, hematološke vrednosti su bile u fi ziološkom opsegu, kao i proteini akutne faze kod većine pacijenata, a aktivnost antioksidativnih enzima je pojačana. Međutim, povišena koncentracija malondialdehida ukazuje na povećano oštećenje eritrocita koje je zaostalo uprkos uspešnom kliničkom oporavku pasa.

AB  - The common signs of canine babesiosis caused by an infection with Babesia canis are fever, anorexia, lethargy, pulse alterations, anemia, and occasionally mild icterus. Dogs with these clinical signs can be divided into two groups: those with acute-phase reaction and those with systemic inflammatory response syndrome (SIRS). Factors associated with the occurrence of SIRS in canine babesiosis have not been thoroughly researched. This article outlines a cross-sectional study of 54 client-owned dogs with an acute B. canis infection, and evaluates the differences in age, gender, laboratory findings, parasite load, and seroreactivity against B. canis between the SIRS and the SIRS-free dogs. We have analyzed a complete blood count, serum biochemistry, serum amyloid A, ceruloplasmin, paraoxonase-1, serology, and PCR testing using standard methodologies. The frequency of SIRS among the investigated dogs reached 0.59. Male dogs and those seronegative against B. canis, were more frequent in the SIRS group, whilst age and parasite load could not be associated with the presence of SIRS. Dogs with SIRS had a lower count of total leukocytes, neutrophils, lymphocytes, and monocytes, and a lower concentration of iron and bilirubin compared with SIRS-free dogs. No significant differences in the concentration of acute-phase proteins have been noticed to exist between the groups of dogs. Further, the seronegative dogs had a lower count of lymphocytes and monocytes and a higher parasite load than the seroreactive dogs. Multivariate logistic regression analysis has identified leukopenia (&lt;6 × 109/L) and monocytopenia (&lt;0.2 × 109/L) as independent associates of SIRS in the investigated dogs, thus implying that these routine tests could be used as reliable markers for SIRS.

Low serum levels of promatrix metalloproteinase-2 and -9 occur during acute Babesia canis infection in dogs

AB  - Inflammation is a hallmark of the acute Babesia canis infection. Promatrix metalloproteinase (proMMP)-2 and -9 are involved in inflammation, but their levels have not been analyzed in canine babesiosis. We hypothesized that in dogs infected with B. canis, serum proMMP-2 and -9 levels change between presentation and recovery. Degree of the change differs if dogs develop systemic inflammatory response syndrome (SIRS). This study included 24 dogs with an acute B. canis infection, at presentation and after two weeks. We used routine hematology and biochemistry methods, spectrophotometry for the acute-phase proteins, microscopy for parasitemia and zymography for (pro)MMPs. In vitro endothelial cells and leukocyte short-term cultures, and platelet lysates were used to detect specific MMP activity. Statistical analyses included Wilcoxon test for paired samples, Mann-Whitney U test and Spearman’s rank correlation. Our results showed that endothelial cells, leukocytes and platelets are the source of proMMP-2 and proMMP-9. Furthermore, both proMMPs were lower at presentation than after recovery (p < 0.001). At presentation, proMMP-9 levels correlated with parasitemia (rho = -0.616, p = 0.009), total leukocyte (rho = 0.704, p < 0.001) and neutrophil counts (rho = 0.741, p < 0.001). Extent of alterations in proMMP-2 levels between presentation and recovery was lower (p = 0.038) in dogs with SIRS than in non-SIRS dogs, while levels of proMMP-9 were comparable between these groups. Our conclusion is that during the acute B. canis infection, low serum levels of proMMP-2 and proMMP-9 at presentation reflect thrombocytopenia and leukopenia. Decreased proMMP-2 level could be associated with SIRS.

AB  - Haematologic abnormalities possibly associated with exposure to vector-borne pathogens are rarely reported in clinically healthy outdoor dogs. Therefore, we analysed changes in the complete blood count (CBC) of clinically healthy outdoor dogs seroreactive to Anaplasma spp. and Babesia spp., with or without microfilariosis. Stray, shelter and hunting dogs, 81 in total, that were polymerase chain reaction negative for Anaplasma spp. and Babesia spp. were divided into groups according to their seroreactive status and results of a modified Knott’s test: seronegative to both Anaplasma spp. and Babesia spp. SN (N=26); seroreactive to A. phagocythophilum SR-A (N=12); seroreactive to B. canis, B. gibsoni and/or B. vogeli SR-B (N=25); and seroreactive to both of the pathogens SR-AB (N=8). These four groups were negative to microfilariosis, unlike the fifth group, seroreactive to either or to both of the pathogens and with microfilariosis SR-M (N=10). The frequencies of CBC alterations among all analysed dogs were: 0.35 – leucocytosis, 0.44 – granulocytosis, 0.28 – anaemia, 0.74 – microcytosis, 0.37 – increased mean cell haemoglobin concentration (MCHC) and 0.33 – thrombocytopenia. The frequency of alterations did not differ across the groups. An exception was the SR-M group wherein increased MCHC peaked with a frequency of 0.80, while in the 
other four groups, the frequency ranged between 0.10 and 0.50. Clinically healthy outdoor dogs have multiple CBC abnormalities, consistent with stress and low-level chronic inflammation, but not associated with a previous exposure to Anaplasma spp. or Babesia spp. The presence of microfilaria increases haemolysis in vitro.
AB  - Klinički zdravi psi koji borave na otvorenom i koji su izloženi patogenima koje prenose vektori mogu imati hematološke premećaje vezane za dejstvo tih patogena. Ipak, hematološke pretrage se retko izvode na toj populaciji pasa. Stoga je cilj našeg rada bio da utvrdimo promene u krvnoj slici zdravih pasa lutalica, pasa smeštenih u azilima i lovačkih pasa, seroreaktivnih na uzročnike bolesti iz rodova Anaplasma i Babesia, sa ili bez mikrofilarioze. U ovom radu je ispitana krvna slika 81 psa. U studiju su uključeni psi kod kojih lančanom reakcijom polimerizacije nije pokazano prisustvo DNK Anaplasma spp. i Babesia spp. Psi su prema serološkom statusu raspoređeni u sledeće grupe: seronegativni na Anaplasma spp. i Babesia spp. SN (N=26), seroreaktivni na A. phagocytophilum SR-A (N=12), seroreaktivni na B. canis, B. gibsoni i/ili B. vogeli SR-B (N=25) i seroreaktivni na oba navedena patogena SR-AB (N=8). Ove četiri grupe su bile negativne na mikrofilariozu, dok je peta grupa, seroreaktivna na neki ili oba patogena imala i mikrofilarije SR-M (N=10). Grupe se nisu razlikovale u pogledu učestalosti navedenih promena. Izuzetak je grupa SR-M u kojoj je učestalost povećanja vrednosti za prosečnu koncentraciju hemoglobina u eritrocitima iznosila 0,80. U ostale četiri grupe ta učestalost je iznosila između 0,10 i 0,50. Zaključak je da ispitivani psi imaju višestruke poremećaje krvne slike koji najverovatnije odražavaju stres i nizak stepen hronične inflamacije, ali ne i vezu sa prethodnim kontaktom sa Anaplasma spp. i Babesia spp. Prisustvo mikrofilarija izaziva hemolizu in vitro.

A short-term and long-term relationship between occurrence of acute canine babesiosis and meteorological parameters in Belgrade, Serbia

AB  - Belgrade, the capital of the Republic of Serbia, is an endemic location for canine babesiosis caused by Babesia canis. This tick-borne disease occurs seasonally in regions with moderate continental climate. However, recent clinical data show that 1) some cases of canine babesiosis have been recorded during the winter, and 2) canine babesiosis is spreading to the northern parts of Europe with a colder climate, which is a region previously free of this disease. Our study investigates the occurrence of canine babesiosis in different seasons over 2013-2016, and explores a short-term link between meteorological parameters and number of diagnosed cases of canine babesiosis in Belgrade. It also looks into possible long-term correlations that span one year before the onset of the disease. Based on 872 recorded cases over four years, our results show a bimodal seasonal distribution of canine babesiosis, with a pronounced peak in the spring, and a less conspicuous one in the autumn. Throughout the year, even over the coldest and warmest periods, there is a broad range of temperatures and relative humidifies when the disease is recorded. Over one year prior to the spring and autumn onset of the disease, we found a noticeable impact of temperature and relative humidity, and to a lesser extent, of atmospheric pressure and cloud cover, on the number of diagnosed cases. These findings imply short-term and long-term relationships between occurrence of acute canine babesiosis and certain meteorological parameters, and they open further questions that need to be investigated in order to understand the epidemiology of this disease.
