Microbiological characteristics of fish reared in purified wastewater from an abattoir

AB  - Wastewater from abattoirs in some countries is disposed of into water bodies without adequate removal of contaminants. Therefore, the use of wastewater in fish production could pose a serious risk for humans, fish and other aquatic organisms due to possible transfer of pathogenic bacteria in aquatic culture environments. The aims of the present study were to assess the levels of individual microorganisms in different tissues of common carp and to determine any correlation with the season of sampling and the type of analysed sample in common carp reared in an integrated production system that used purified water from an abattoir. A fish pond was filled mostly with purified water from an abattoir, but also partly with well water. Carp fingerlings were stocked in the earthen fishpond in March and reared in ambient conditions. Fish were collected in the spring and autumn of the following year and the microbiological quality was assessed. Carp fillets with skin, gills and digestive tract samples were collected individually under aseptic conditions. All analyses were performed according to standard procedures. The levels of all the examined bacteria in the fish were under prescribed hygiene norms. Also, Listeria spp., sulphite-reducing clostridia and Staphylococcus aureus were not found in the samples. Furthermore, pathogenic bacteria, Salmonella and Listeria monocytogenes were not isolated from the samples. The hygienic quality of the fish produced in purified waste water from an abattoir was acceptable, and the common carp meat was safe for human consumption.

Influence of Anticoccidials on Oxidative Stress, Production Performance and Faecal Oocyst Counts in Broiler Chickens Infected with Eimeria Species

AB  - The influence of certain anticoccidial drugs on oxidative stress in broiler chickens infected with Eimeria species was assessed. There were two untreated (uninfected and infected), and three groups infected and treated with anticoccidials. The first treated group (Ro) was given robenidine, the 2nd a herbal anticoccidial (Herb) and the 3rd the combination of robenidine and the herbal anticoccidial (Ro+Herb). All infected groups were on day 14 challenged with oral inoculation of oocysts. The activities of catalase (CAT), superoxide dismutase (SOD) and glutathione S-transferase (GST), and the concentration of malondialdehyde (MDA) were estimated in blood taken on days 21 and 40. The oocyst numbers were calculated per gram, and chicken body weight and feed conversion ratio (FCR) measured. The activities of CAT, GST and the level of MDA were significantly lower (P<0.05), whilst the activity of SOD was higher in infected chickens treated with anticoccidials (P<0.05) in comparison to those untreated. The most prominent change in the parameters of oxidative stress was recorded in the Ro+Herb group. In chickens treated with anticoccidials body weight was significantly higher (P<0.05), and the FCR and the oocyst counts significantly lower (P<0.05) than in untreated chickens. Oocyst counts were lower in the Ro and Ro+Herb groups than in the Herb group. Our study demonstrated that both anticoccidial substances exerted antioxidant and anticoccidial effects.

Morphometric Parameters of Erythrocytes in Juvenile Mandarin Ducks Aix galerkulata

AB  - The morphometric parameters of erythrocytes in Mandarin ducks Aix galericulata have never been subjected to study even though this species is common in aviculture. The aim of this study is to investigate the morphometric parameters of erythrocytes in juvenile Mandarin ducks. A total of 1500 erythrocytes obtained from thirty Mandarin ducks were analysed. The smears of the peripheral blood were stained using Diff-Quik  stain method, and the length and width of erythrocytes were measured using ImageJ program. The average length of the measured erythrocytes was 11.879 mu m. The maximal measured length of the erythrocytes was 10.O49 mu m, and the minimal measured length was 5.668 pm. The average width of the measured erythrocytes vvas 6.160 pm. The maximal measured width of the erythrocytes was 10.115 pm and the minimal measured width vvas 3.750 mu m. In conclusion, knowing morphometric parameters of Mandarin duck erythrocytes is the first step in evaluation process of erythrocytes functional state and health condition of an individual.

The Influence of Carcass Microlocation on the Speed of Postmortem Changes and Carcass Decomposition

AB  - Determining the post-mortem interval (PMI) is often a very demanding and delicate job which requires a good knowledge of postmortem changes. In this study, 20 domestic pig carcasses (Sus scrofa) whose death occurred within 8 h before the start of the study were simultaneously laid at the same geographical location, but in different environments (on the ground surface - S; buried in the ground - G; placed in a crate and buried in the ground - C; submerged in water - W; and hanging in the air - A). One carcass from each group was sampled on days 14, 28, 120 and 190 from the beginning of the experiment, and on that occasion, a detailed analysis of postmortem changes and an autopsy was carried out. The difference in the rate of decomposition among groups was statistically significant. The fastest decomposition occurred in carcasses placed in a crate and buried, because during the winter period the temperature in the air was below 0 degrees C. At that time, the decomposition process and the insect activity were slowed or stopped on carcasses in groups S, A, W, and G to some extent, while the ground and wooden crate were good thermal insulators for group C carcasses arid provided better conditions for insect activity.

AB  - Niclosamide is applied for the treatment of the disease of common carp hatchlings caused by Bothriocephalus acheilognathi. One of the main characteristics of this drug is a short half-life in water. A series of experiments were performed to determine the distribution of niclosamide residues in the organs and meat of the carp, in controlled conditions. It was shown that 72.42% of niclosamide was deposited in the liver, 18.79% in the kidney and 7.79% in the spleen. However, only about 3% of deposited niclosamide was detected in meat. This drug provides rapid and effective treatment of fish, completely eliminating the tapeworms.

AB  - The aim of the study was to examine the production parameters and chemical composition of fillets in tench, Tinca tinca L., farmed in one of two systems: an extensive system based on only natural food (NF) available in the fish ponds; and a semi-intensive system based on natural food plus the addition of formulated feeds supplemented with various oils: fish (FO), rapeseed (RO), soybean (SO), or linseed oil (LO). Proper pond preparation resulted in a favourable amount and structure of natural food in all ponds. The rearing system had a significant influence on the tench yield, muscle lipid contents and fatty acid composition, and the supplemented feed influenced the fatty acid composition. The percentages of eicosapentaenoic (EPA) and docosahexaenoic (DHA) acid and total highly unsaturated n-3 fatty acids (n-3 HUFA) were higher in the fish oil and the natural food groups than in the others. The n-3/n-6 ratio in the five dietary treatments was notably variable, and in any case higher in fillets than in the diets. In conclusion, the present study suggests that RO, SO and LO represent effective lipid sources for tench fed formulated diets. On the other hand, tench that fed on naturally available pond foods had a high content of n-3 HUFA in their muscle lipids, but poor growth parameters and low yields.

AB  - Telohaneloza je obolenje koje se javlja kod svih kategorija šarana. Uzročnik je protozoa
Thelohanellus nikolskii. Manifestuje se u dva oblika. Prvi se javlja kao telohaneloza
na perajima kod jednogodišnjih mladunaca šarana, a drugi kao telohaneloza
krljušti i kože kod dvogodišnjih, trogodišnjih i višegodišnjih kategorija ribnjačkog šarana
(Cyprinus carpio). Istraživanjima koja su trajala u periodu od 2008-2012. godine
obuhvaćeno je 22 ribnjaka, od čega se 18 nalaze u Srbiji a 4 u BIH, Republika Srpska.
Praćenjem epizootiološke rasprostranjenosti utvrđeno je da su oba oblika oboljenja bila
prisutna na svim ispitivanim ribnjacima. Kod telohaneloze krljušti prevalenca se kretala
od 2-75% a intenzitet infekcije 2-206 cisti po jedinki, dok su iste vrednosti kod telohaneloze
na perajima iznosile 3-30%, odnosno 2-84 ciste. Promene na krljuštima bile
su prisutne tokom aprila i maja, a na perajima tokom jula i avgusta. Tokom epizootiološkog
praćenja i istraživanja promena na krljuštima zapaženo je da na njima dolazi do
formiranja cisti okruglastog oblika koje su dostizale veličinu i do 3 mm. Promene na
krljuštima zabeležene su kod dvogodišnjih mladunaca koji su najčešće bili inficirani,
ali i kod trogodišnjih i četvorogodišnjih konzumnih i matičnih kategorija šarana, dok
kod jednogodišnje mlađi nisu ustanovljene ni u jednom slučaju. Ciste su bile prisutne
kako kod mnogoljuskavih šarana, koji su potpuno prekriveni krljuštima, tako i kod maloljuskavih
gde su se promene nalazile na dorzalnom redu krljušti. Takođe je uočena i
pojava cisti na koži, a ne i na krljušti kod četvorogodišnjih matičnih riba. Tokom čitavog kod mladunaca. Jedinke kojima su otpala repna peraja postajale su plen ribojedih ptica.
Najveći broj cisti nalazio se na repnom peraju, zatim na leđnom, grudnom, trbušnom i
analnom. Merenjem i određivanjem morfoloških karakteristika spora izolovanih iz plazmodijuma
sa krljušti dvogodišnjih mladunaca i plazmodijuma sa peraja jednogodišnje
mlađi dobijene su vrednosti dužine i širine spora, kao i dužine i širine polarnih kapsula.
Prilikom determinacije patohistoloških karakteristika utvrđena je struktura plazmodijuma,
gde se u centralnim delovima nalaze sporame, dok se na periferiji nalaze razvojni
stadijumi i pansporoblasti, te se na osnovu dominacije pojedinih ćelija unutar njega
može utvrditi da li se radi o mladim ili o zrelim cistama. Plazmodijum se razvija unutar
kalcifikovane kolagene kapsule, koja predstavlja tanak sloj kalcifikovanog kolagena
oko kojeg se nalaze izdužene ćelije – skleroblasti, koji su zaduženi za formiranje ciste.
Molekularnim tehnikama i BLAST analizom potvrđeno je da je etiološki uzročnik
Thelohanellus nikolskii isti za oboljenje jednogodišnjih mladunaca na perajima i višegodišnjih
šaranskih riba na krljuštima i koži. Značajan momenat širenja telohaneloze
predstavlja promet mladunaca između ribnjaka gde se ne vodi računa o postojanju ove
bolesti. Drugi bitan faktor je hidrografska povezanost ribnjaka, gde se ispuštanjem i
upuštanjem vode omogućava prenošenje uzročnika bolesti. S obzirom na malu udaljenost
ribnjaka ribojede ptice takođe predstavljaju bitan vektor širenja bolesti. Pošto ne
postoji ni jedno adekvatno terapeutsko sredstvo, kontrola telohaneloze i dalje se bazira
na pridržavanju osnovnih sanitarno–profilaktičkih mera, kao što su isušivanje objekata,
izmrzavanje, mehanička obrada tla i dezinfekcija krečom.

AB  - Carp (Cyprinus carpio L.) is domesticated in every continent except Antarctica,
owing to its ability in adapting to different values of water quality parameters, including
tolerance to very low dissolved oxygen concentrations. Considering the benefits that
are reflected by the extensive reproductive abilities, breeding and prime selection potentials, and high resistance to various diseases, it is not surprising that carp represents the dominant species in all counties with prerequisites for extensive, semi-intensive and
intensive freshwater fish production. The carp growth in different production systems
depends on many factors, such as varietal, density, welfare and fish feed with proper
nutrients along with geographic location.
Cage production system for carp is a special type of intensive production. It is characterized by lower initial investments in cage construction and minimal labor engagement, ensuring high production yield per volume capacity of water contained in the
constructed units. Modern cages are constructed from metal and synthetic polymers
(Mihailovic et al., 2007). Cage has to be robust enough to endure associated hazards
such as winds and waves, uncomplicated servicing and maintenance. Location choice
is critical as it affects the production and mortality of the produce (Carp) directly impacting the profitability of the production unit.
Below listed are three-selection criterion considered for selection of sites for cage
culture (Bogut et al., 2007).
1. Physical and chemical parameters including temperature, dissolved oxygen
concentration, water flow, pollution and algal blooms.
2. Factors relating to the choice of location taking into account weather conditions, shelter, substrate, water currents and the degree of blur. Additionally the shape and
size of the cage and the depth to which they are installed.
3. Profitability of the farm, and includes legal aspects, accessibility, facilities, security, economic and social conditions.
Superior production practices, quality of water and optimally balanced diet appropriate for the cultured carp category, expected mortality rate is 1-5%. Compared to other
breeding systems, it was found that the cage system morbidity and mortality could vary
significantly in the case of deviation in the values of water quality parameters from the
optimum, the losses can occur quickly and be dramatic. (Orajić et al., 2007)
Shortcoming of cage production system is reflected through allowing of disease
transmission and aquatic ecosystems contamination where the cage system is set up,
this can be prevented by protective systems introduction.
Modern market demands are increasingly directed towards processed fish, especially
fillets (Hough, 1993; Vallod, 1995). By removing the skin, internal organs and intramuscular bones, fillets of carp and other cyprinids grown in polyculture along with the carp,
become high value meals that are easily and quickly prepared (Lin et al., 1989).
Dressing percentage of fish is an economic parameter with significant value attached
to it, in all technological operations related to fish processing. It is expressed as the ratio
of the primary processed carcass (dressed) relative to carcass weight of live fish.
The ratio of edible and non-edible part of the fish carcass can vary significantly depending on the species, weight, size of the fish and season catch (Baltic and Teodorović,
1997). Since the dressing percentage directly influences the economy (profitability) of
production, it is unjustly neglected as the parameter in scientific research/studies (Ljubojević et al, 2012).
The difference in yield of different lines and their hybrids occur due to different
body shape, head length, width and fat of the body. Dressing percentage of fish is significantly higher comparatively than the yield of other animal species (Ćirkovic et al.,
2002). Parts that are rejected may include all or some parts of the head, scales, skin,
intestine, gonads and fins.
According to Dunhamu et al. (1983) fish carcass primary handling involves removal
of the head, scales and internal organs, while Lovell (1981) reference states that primary treatment involves the removal of skin, dorsal and pectoral fins, head and abdominal
organs cavity, with the tail present. Naumovski (1991) and Tumbas (1976) define fish
carcass primary handling as a process which include removal of the tail fin.
In this study, yield was determined as the ratio of the fish weight and the carcass
without the head, scales, fins and internal organs.

Finding of parasitic nematodes of fishes present in the market

AB  - When placing the fish and fish products on the trade it is necessary to pay attention to the presence of zoonotic parasites that can lead to infection of people especially if the fish is consumed raw or undercooked. Epidemiologically the most important are helminths from the group of Nematoda. The most important are Anisaxis spp. and Eustrongylides excisus. Anisakiasis is a serious zoonotic disease with a dramatic increase in prevalence throughout the world in the last two decades. The larvae are found in marine fish species most frequently in sardines, herring and mackerel. Eustrongylidosis is a disease that occurs primarily in freshwater fish species: catfish, zander and pike, a cause is a parasite of the genus Eustrongylides of which the most important species is Eustrongylides excisus. In this paper, the nematodes that occur in fish that are present in the market in the Republic of Serbia are present. Investigations of Anisaxis spp. were carried out at the Institute of Veterinary Medicine Novi Sad in the period of 2000-2013. Total of 2414 samples of imported marine fish was inspected. In 25 (1.29 % ) samples of herrings (Clupea harengus ) and mackerels ( Scomber scombrus ) was identified Anisakis spp. Investigations of Eustrongylides spp . were carried out in the period 2010-2013 at the Danube-Tisa-Danube Canal. Samples were collected from zander (Sander lucioperca) and European catfish (Silurisglanis) in which the presence of nematodes was found in the abdominal cavity, muscles, the lumen of the stomach and gastric wall where the parasites were encapsulated. Individuals of zander were examined during the 2013 at the Veterinary Institute Novi Sad, where the presence of larvae Eustrongylides spp. u muscle was detected. In order to avoid infecting people with parasites of fish it is necessary to carry out continuous control and monitoring. Fresh fishes and traditional fish products must be inspected for the presence of parasites before they find on the trade. Continuous education is a key factor in combating zoonotic diseases. Avoid consumption of raw or poorly cooked fish is still the best preventive procedure.
AB  - Prilikom stavljanja u promet i pregleda ribljeg mesa i proizvoda od ribe treba obratiti pažnju na prisustvo zoonotskih parazita koji mogu dovesti do inficiranja ljudi naročito ako se riba konzumira sirova ili termički nedovoljno obrađena. Sa epidemiološkog aspekta najveći značaj imaju helminti iz grupe Nematoda. Najznačajnije su Anisaxis spp. i Eustrongylides excisus. Anisakijaza je ozbiljno zoonotsko oboljenje sa dramatičnim porastom prevalence širom sveta u poslednje dve decenije. Larve se mogu naći kod morskih vrsta riba najčešće kod sardine, haringe i skuše. Eustrongilidoza je oboljenje koje se javlja kod slatkovodnih riba prvenstveno grabljivica: soma, smuđa i štuke, a izazivaju je paraziti iz roda Eustrongylides od kojih je najznačajnija vrsta Eustrongylides excisus. U ovom radu prikazane su nematode koje se javljaju kod riba koje su prisutne na tržištu u Republici Srbiji. Istraživanja prisustva Anisaxis spp. su sprovedena na Naučnom institutu za veterinarstvo Novi Sad u periodu 2000-2013. Ukupno je pregledano 2414 uzoraka riba iz uvoza od čega je u 25 (1.29%) utvrđen Anisakis spp., kod haringe (Clupea harengus) i skuše (Scomber scombrus). Istraživanja prisustva Eustrongylides spp. su sprovedena u periodu od 2010-2013 godine na kanalu Dunav-Tisa-Dunav. Prikupljeni su uzorci konzumnog smuđa (Sander lucioperca) i soma (Siluris glanis) kod kojih je utvrđeno prisustvo nematoda u abdomenu, muskulaturi, lumenu želuca i želudačnom zidu gde su paraziti bili inkapsulirani. Jedinke smuda pregledane se tokom 2013. godine i na Naučnom Institutu za veterinarstvo Novi Sad gde je utvrđeno prisustvo larvi Eustrongylides spp. u muskulaturi. Kako ne bi došlo do zaražavanja ljudi zoonotskim parazitima riba neophodno je sprovoditi stalnu kontrolu i monitoring. Sveže meso ribe i tradicionalni riblji proizvodi pre nego što se nadu u prometu moraju biti pregledani na prisustvo parazita. Kontinuirana edukacija je ključni faktor u borbi sa zoonozama a izbegavanje konzumiranja sirovog ili termički slabo obrađenog ribljeg mesa i dalje najbolja preventivna procedura.

AB  - Twenty-one specimens of pike-perch (Sander lucioperca) were caught in the Danube-Tisa-Danube Canal in the city area of Novi Sad for parasitological examination. The presence of nematodes in the muscles was revealed in three fish. The parasites were identified to belong to the species Eustrongylides excisus, for which the pike-perch is a paratenic host. This finding represents the first determination of the larvae in the pike-perch in Serbia. The pike-perch is infected by ingestion of benthos- or plankton-eating fishes, the second intermediate hosts harbouring the fourth-stage nematode larvae. E. excisus is pathogenic to humans, who may be infected by consuming raw or undercooked fish.

Different manifestations of Thelohanellus nikolskii infection in carp (Cyprinus carpio)

AB  - Thelohanellosis is a disease that occurs in all carp categories. The cause is protozoa Thelohanellus nikolskii Achmerov, 1955. It manifests itself in two forms. The first form occurs on the fins in oneyear- old carp fry and the second one occurs on scales in two-yearold, three-year-old and older categories of common carp (Cyprinus carpio). The researches conducted between 2008 and 2012 covered 22 fish ponds, 18 of which are located in Serbia, and 4 in Bosnia and Herzegovina, Republic of Srpska. It has been concluded that both forms of the disease were present in all observed fish ponds. Prevalence of telohanellosis located on the fins ranged between 3% and 30%, and the infection intensity was between 2 and 86 cysts per individual, while in the case of thelohanellosis on scales the values were higher. Changes on the scales were present during April and May, and on the fins during July and August. After determination of morphological characteristics of spores and histopathological characteristics of plasmodia located on the fins and scales, no significant differences were noticed between them. Also, artificial infestation of carp fingerlings for species differentiation was done. Since there are no adequate therapeutic measures, control of thelohanellosis is still based on compliance with basic sanitaryprophylactic measures such as drying of equipment, freezing, mechanical cleaning and disinfection with lime.
AB  - Telohaneloza je obolenje koje se javlja kod svih kategorija šarana. Uzročnik je protozoa Thelohanellus nikolskii Achmerov, 1955. Manifestuje se u dva oblika. Prvi se javlja kao telohaneloza na perajima kod jednogodišnjih mladunaca šarana a drugi kao telohaneloza krljušti kod dvogodišnjih, trogodišnjih i višegodišnjih kategorija ribnjačkog šarana (Cyprinus carpio). Istraživanjima koja su trajala u periodu od 2008.-2012. godine obuhvaćeno je 22 ribnjaka od čega se 18 nalaze u Srbiji a 4 u BIH, Republika Srpska. Utvrđeno je da su oba oblika obolenja bila prisutna na svim ispitivanim ribnjacima. Kod telohaneloze na perajima, prevalenca se kretala od 3-30% a intenzitet infekcije od 2-84 cisti po jedinki, dok su te vrednosti kod telohaneloze na krljištima bile veće. Promene na krljuštima bile su prisutne tokom aprila i maja a na perajima tokom jula i avgusta. Utvrđivanjem morfoloških karakteristika spora iz cisti sa peraja i krljušti, kao i prilikom determinacije patohistoloških karakteristika plazmodijuma nisu uočene značajne razlike između njih. Takođe je urađen i ogled veštačkog zaražavanja mladunaca šarana radi diferencijacije vrste. Pošto ne postoji ni jedno adekvatno terapeutsko sredstvo kontrola telohaneloze i dalje se bazira na pridržavanju osnovnih sanitarno profilaktičkih mera kao što su isušivanje objekata, izmrzavanje, mehanička obrada tla i dezinfekcija krečom.
