AB  - Porcine Reproductive and Respiratory Syndrome is a viral disease of swine characterized by reproductive failure of breeding animals and respiratory disorders in all categories. The first PRRS case in Serbia was recorded in 2001 after illegal import of boar semen. PRRS is economically the most important disease due to significant direct and indirect losses. Today, for routine diagnosis of PRRS in infected herds serological methods (ELISA) and molecular methods are used. Although modern diagnostic techniques are very robust, exceptional diversity of the viral strains is often the obstacle for an accurate diagnosis. To estimate the performance of seven different methods for PRRSV genome detection, twenty samples were used. However, none of the methods was able to detect all PRRSV strains. The best sensitivity was obtained by combining two methods. Until today, there is no absolutely accurate test which enables the detection of all circulating strains.

Molecular detection of black queen cell virus and Kashmir bee virus in honey

AB  - Considering the intensive trading nowadays, the honey from the local market was tested for the presence of the six most common bee viruses. To prove the suitability of honey as a sample for the bee viruses detection, the set of different sample types taken directly from the hives we comparatively tested. The study included 30 samples of domestic and 5 samples of imported honey. Additionally, we tested 40 sets of samples including live bees, dead bees, and the honey taken from four apiaries for the evaluation of honey suitability for the virus detection, Two out of the six most common bee viruses were detected in the samples of honey from the market. Black queen cell virus (BQCV) genome was found in 24 domestic honey samples and Kashmir bee virus (KBV) genome was detected in one sample of imported honey. The nucleotide sequences of 24 BQCV isolates showed the highest identity (86.4%) with strains from Europe at the polyprotein gene, whilst the Serbian isolates between each other showed 98.5% similarity. By comparative testing of the different type of samples, in three out of four apiaries BQCV genome was detected in both bees and honey. Evaluating the suitability of honey for the detection of the viral disease by simultaneous testing of live, dead bees, and honey from the same hive, it was shown that the honey can be successfully used for the detection of BQCV. Since, as of yet, there has been no evidence of KBV circulation in Serbia, after its detection in imported honey, there is a substantial risk of its introduction and consequently the need for its surveillance. Therefore, the programs of bee diseases screening should be included in the regular control procedures for the international trade. In addition to this benefit, honey gives an opportunity to beekeepers for continuous monitoring of bees health status.

AB  - The objective of this study was to evaluate the influence of three supplementary diets on the nutritional quality of marketable common carp. Supplementary diets (Carp1-maize, Carp2-extruded and Carp3-pelleted feed) exhibited significantly different (p≤0.05) influences on the protein, lipids, moisture and ash content in market-sized carp. The most abundant saturated FA (SFA) was palmitic acid (C16:0), the most predominant monounsaturated FA (MUFA) was oleic acid (C18:1n-9), and the most abundant polyunsaturated FA (PUFA) was linoleic acid (C18:2n-6). The highest content of SFAs was established in Carp1 (24.57% and 1888.72 mg/100 g wet fillet). MUFAs were presented in the highest quantities (61.77%, I.e. 4854.91 mg/100 g wet fillet) in Carp1, and PUFA accounted the highest quantities (33.48%, I.e. 1763.01 mg/100 g wet fillet) in Carp3. The most nutritionally beneficial n-6/n-3 ratio was obtained in Carp2 (5.83). The results obtained in this study indicate that introducing supplementary diets containing extruded or pelleted feed instead of maize improved carp nutritional quality. PCA (principal component analysis) and LDA (linear discriminant analysis) of the FAs demonstrated that carp could be reliably classified based on their supplementary feed.

Evidence of Aujeszky s disease in wild boar in Serbia

AB  - Background: Aujeszkys disease is a viral disease of suids caused by Suid Herpesvirus 1. The disease has worldwide distribution with significant economic impact. In Serbia, there is neither an Aujeszkys disease eradication nor national vaccination programme of domestic pigs. Since clinical symptoms of Aujeszkys disease are not specific, it is important to establish a link between clinical signs and presence of ADV active infection in wild boars. The aim of this study was to investigate the possibility of active infection within wild boar showing signs of ADV and also to examine relationship between isolates from domestic pigs and wild boar. Having in mind that virus has not been previously isolated from wild boars in Serbia, we report the first isolation of Suid Herpesvirus 1 from this species in Serbia. Results: Tissue and serum samples from 40 wild boars from eastern Serbia were examined for evidence of Aujeszkys disease (AD). Suid Herpesvirus 1 (SHV1), the cause of AD was isolated on PK15 cell line from three tissue samples, inducing cytopathic effect (CPE) with syncytia forming, and viral genome was detected by polymerase chain reaction (PCR) in eight samples. Genetic analysis of us4, us9 and ul49.5 partial sequences showed high homology between ADV isolates from wild boars and between isolates from wild boars and domestic animals. Neutralizing antibodies were not detected by virus neutralisation test (VNT) in sera from four out of eight PCR positive wild boars suggesting recent infection in those animals. Conclusions: This is the first demonstration of Aujeszkys disease virus (ADV) in the wild boar population in Serbia although seroconversion has been detected previously.

AB  - A total of 703 samples were tested over 1-year period. Listeria monocytogenes was isolated from 12.4% and 2.3% of fish and environmental swabs, respectively. The ratio of n-6/n-3 which is between 1:1 and 4:1, as more desirable parameter of the lipid quality for nutritive benefit e.g. reducing the risk of many diseases, in the fresh salmon, cold and hot smoked salmon was close to 1:1 (fresh salmon, 1.28; cold smoked salmon, 0.98; hot smoked salmon, 1.59). The fatty acid composition of smoked salmon products was also expressed as mg 100 g-1 which is important from the nutritional point of view. The examined salmon products had a high content of eicosapentaenoic acid and docosahexaenoic acid (742 to 1567 mg 100g-1) and fulfill requirements for their sufficient contents of recommendations in the World. On the other hand, cold smoked salmon can be naturally contaminated with low numbers of L. monocytogenes. This could represent a serious hazard for susceptible individuals or 'YOPI' (young, old, pregnant or immuno- compromised individuals).

AB  - The purpose of food packing in modified atmospheric conditions is to extend its sustainability by preventing both biochemical processes and growth of spoilage bacteria. Gases or their mixtures which are mostly used in the modified atmosphere food packing technology are carbon-dioxide (CO2), oxygen (O2) and nitrogen (N2). The aim of our research was to examine the influence of packaging in modified atmosphere and vacuum on the total volatile basic nitrogen (TVB-N) content and pH in muscle of rainbow trout (Oncorhynchus mykiss) and common carp (Cyprinus carpio), as well as to determine the most suitable gas mixtures for packing of these freshwater species. Three sample groups of trout and carp cuts were investigated. The first two groups were packaged in modified atmosphere with different gas ratios: 60%CO2+40%N2 (I group) and 40%CO2+60%N2 (II group), whereas the samples from third, control group, (III group) were vacuum packaged. During trials, samples were stored in refrigerator at +3°C. Determination of TVB-N and pH was performed on 1st, 7th and 14th day of storage. The obtained results indicate that all investigated mixtures of gases as well as vacuum had a significant influence on the values of TVB-N in trout and carp cut samples. The lowest increase in TVB-N was established in trout and carp cut samples from the group I, whereas the highest increase was established in samples from group III. Statistical significant difference (p  lt  0,001) between the average values of TVB-N for trout (I group: 18.17±0.93; II group: 20.90±0.81 and III group: 36.18±2.65 mg N/100 g) and carp cuts (I group: 26.74±1.48; II group: 30.02±0.31 and III group: 35.10±1.75 mg N/100 g) was established on the 14th day. The lowest pH value was measured in samples packaged in modified atmosphere with 60% CO2+40% N2 (I group). On the 14th day of testing obtained value was 6,15 ± 0,09 for trout and 5.94±1.11 for carp samples. Increase in pH value in trout samples packed in vacuum was established during the whole period of investigation (p  lt  0,001), while in carp cut samples packaged in vacuum the increase in pH value (p  lt  0,05) was established up to 7th day of testing. Based on the obtained results it can be concluded that gas mixture consisting of 60% CO2 and 40% N2 was the most suitable for packaging of fresh trout and carp cuts in terms of selected chemical parameters, such as TVB-N and pH.
AB  - Cilj ovih istraživanja bio je da se ispita uticaj pakovanja u modifikovanoj atmosferi i vakuumu na promene vrednosti ukupno isparljivog azota (TVB-N) i pH mesa kalifornijske pastrmke (Oncorhynchus mykiss) i šarana (Cyprinus carpio), kao odabranih hemijskih parametara svežine mesa ribe i da se ustanove najpodesnije smeše gasova za pakovanje ove dve slatkovodne vrste riba. Za potrebe ovog istraživanja formirane su po tri grupe uzoraka očišćene pastrmke i odrezaka šarana. Prve dve grupe su upakovane u modifikovanu atmosferu sa različitim odnosom gasova: 60% CO2 + 40% N2 (I grupa) i 40% CO2 + 60% N2 (II grupa), dok je III, kontrolna, grupa upakovana u vakuum. Svi uzorci su čuvani pri istovetnim uslovima na temperaturi od +3 °C, a zatim su prvog, sedmog i četrnaestog dana čuvanja obavljena ispitivanja. Dobijeni rezultati ukazuju da je na vrednost TVB-N u uzorcima pastrmke i odrezaka šarana bitno uticao sastav upotrebljene gasne smeše. Tokom četrnaest dana čuvanja vrednosti za TVB-N u uzorcima sve tri grupe pastrmke i odrezaka šarana statistički su značajno porasle (p  lt  0,001). Najmanji rast vrednosti za TVB-N ustanovljen je u I grupi uzoraka pastrmke i odrezaka šarana, dok je porast vrednosti za TVB-N bio najveći u uzorcima III grupe. Najniža pH vrednost ustanovljena je u uzorcima pastrmke i odrezaka šarana koji su upakovani u modifikovanu atmosferu sa 60% CO2 + 40% N2 (I grupa). Rast pH vrednosti u uzorcima pastrmke upakovane u vakuum ustanovljen je tokom celog perioda ispitivanja, dok je u uzorcima odrezaka šarana upakovanim u vakuum rast pH vrednosti ustanovljen samo do sedmog dana ispitivanja. Smeša gasova sa 60% CO2 + 40% N2 pokazala se kao najpodesnija za pakovanje sveže pastrmke i odrezaka šarana.

Effect of pre-processing of trout by freezing on the characteristics of smoked trout fillets

AB  - In the situation when the capacities of production and further treatment of smoked fish are insufficient, it is necessary to freeze fresh fish during the fishing season and treat it later, when the season is over. That was the reason why the aim of our study was to explore effects of freezing on certain quality parameters of smoked fish. Experimental design included trout separated into two groups: the control group of fresh fish and the experimental group of frozen fish. During production process, total bacterial count was examined, and at the end of the process, chemical composition of the final product (vacuum-packed cold smoked trout) was determined. During the storage of the product at 4°C up to 21 days, microbiological, physicochemical and sensory analyses were performed at regular intervals. Overall, the results of the present study demonstrated that pre-processing freezing of trout is suitable and, in periods of large catches, even recommendable step in smoked trout production, at least for smaller processors.
AB  - U situaciji kada su kapaciteti za proizvodnju i dalji tretman dimljene ribe nedovoljni potrebno je da se sveža riba tokom sezone izlova zamrzne i da se preradi kasnije, kada se sezona završi. To je bio razlog zašto je za cilj naših istraživanja postavljeno da se ispitaju efekti zamrzavanja na određene parametre kvaliteta dimljene ribe. Za eksperiment, pastrmke su bile podeljene u dve grupe: kontrolna grupa, koju je sačinjavala sveža riba i eksperimentalna grupa, koju je sačinjavala smrznuta riba. Tokom procesa proizvodnje ispitan je ukupan broj bakterija, a na kraju procesa, u konačnom proizvodu (vakuum-pakovana hladno dimljena pastrmka), određeni su hemijski parametri. U toku skladištenja proizvoda na +4°C tokom 21 dan, u redovnim intervalima su izvršena mikrobiološka, fizičko-hemijska i senzorna ispitivanja. Generalno, rezultati ovih istraživanja su pokazali da je prethodna obrada pastrmke zamrzavanjem pogodna i, u vreme velikih izlova, čak i preporučljiv korak u proizvodnji dimljene pastrmke, bar što se tiče manjih proizvođača.

Detection and genotyping of classical swine fever virus isolates in Serbia

AB  - Classical swine fever (CSF) is a highly contagious disease of pigs leading to significant economic losses worldwide. Classical swine fever virus can be classified into three genogroups, each consisting of three or four subgroups. However, there is a lack of knowledge on the genotypes of CSFV isolates in Republic of Serbia. This study, based on the sequences analysis of partial E2 gene and 5' non coding region (NCR) of 15 CSFV isolated during 2006-2008 from domestic pigs, revealed that all were clustered into genetic group 2.3. Additionally, we showed that the two most often used real time RT-PCR assays were able to detect all local CSF viruses circulated in Serbia in the last years during intensive vaccination campaign against CSF.
AB  - Klasična kuga svinja (CSF) je visoko kontagiozno oboljenje svinja koje dovodi do značajnih ekonomskih gubitaka širom sveta. Na osnovu genetske strukture, virus klasicne kuge svinja podeljen je utri genogrupe, od kojih svaka ima tri ili četiri podgrupe. Nedostaju podaci o tome koji genotipovi virusa klasične kuge svinja na teritoriji Republike Srbije cirkulišu u prijemčivoj populaciji. Sekvencioniranjem dela E2 gena i 5’nekodirajućeg regiona (NCR) 15 izolata virusa klasične kuge svinja prikupljenih u periodu od 2006-2008 godine, poreklom od domaćih svinja, dokazano je da svi pripadaju genetskoj grupi 2.3. Uz to, dokazano je i da je upotrebom dva najčešće korišćena real time RT-PCR protokola moguće detektovati sve lokalne izolate virusa klasične kuge svinja koji cirkulišu u Srbiji poslednjih godina, u kojoj se uporedo vršila i intenzivna vakcinacija protiv ove bolesti.

AB  - Fish is one of the most valuable food products used in human nutrition, due to the content of proteins, fat, minerals, vitamins, essential n -3 polyunsaturated fatty acids (PUFAs) and cholesterol. To meet the growing needs of the population for this kind of food, fish are increasingly grown in aquaculture. Intensive fish production, due to increased stock density, is favoring the occurrence of bacterial diseases. As a consequence, there is increased morbidity and mortality, reduced growth and reduced leasing sockets materials, which pose a serious problem for the aquaculture and lead to massive use of chemotherapeutics. Antibiotics are the common practice for the treatment of bacterial infections in fish ponds, and flurohinolones are used as antibiotics of choice. Fluoroquinolones are a group of antibiotics that have a broad spectrum of activity, low toxicity and only a few side effects in the treated fish. However, residues of antibiotic in fish tissues represent a real risk to human health. Consumption of fish containing residues of antibiotics can cause allergic, toxic, carcinogenic, mutagenic and teratogenic effects. Therefore, it is necessary to establish the residual amounts of antibiotics in fish tissues, that can be achieved by using reliable laboratory methods and techniques. Because of this, the goal of the work was set to investigate the possibility of identification and quantification of fluoroquinolones in the kidneys of carp by microbiological diffusion method, using the test organism E. coli ATCC 11303. The investigations revealed that microbiological diffusion method enables detection of all five fluoroquinolones (oxolinic acid, enrofloxacin, sarafloxacin, difloxacin and flumequine) in kidney of carp at different maximum residue levels, MRL (100 mg/kg, 100 mg/kg, 30 mg/kg, 300 mg/kg and 600mg/kg, respectively). Identification and quantification of fluoroquinolones at MRL levels was only achieved for enrofloxacin, flumequine and difloksacin. These fluoroquinolones can be detected and quantified at the level below the MRL, i.e. at the level of 1/4 MRL. Contrary to these fluoroquinolones, sarafloxacin can be detected only at the level of 2MRL and oxolinic acid can be detected at the level of 4MRL, as well. EU regulations provide that a screening method can be applicable only if a compound can be detected in the amount of at least at the MRL, and the ½ MRL is recommended. This means that the microbiological diffusion method can be used in routine analytical practice for the identification and quantification of enrofloxacin, flumequine and difloxacin in kidney tissues of carp.
AB  - Riba je, zbog sadržaja i količina proteina, masti, minerala, vitamina, esencijalnih n-3 polinezasićenih masnih kiselina (PNMK) i holesterola, jedna od nutritivno najvrednijih namirnica koja se koristi u ishrani ljudi. Da bi se zadovoljile rastuće potrebe stanovništva za ovom vrstom namirnice, riba se sve više gaji u akvakulturi. Intenzivna proizvodnja ribe, zbog povećane gustine nasada, pogoduje nastanku bakterijskih oboljenja. Kao posledica toga, javlja se povećan morbiditet i mortalitet, smanjen prirast i smanjenje nasadnog materjala, što predstavlja ozbiljan problem za akvakulturu i dovodi do masovne upotrebe hemioterapeutika u terapijske svrhe. Za lečenje bakterijskih infekcija riba u ribnjacima koriste se antibiotici. Kao antibiotik izbora u uzgojnom ribarstvu se koriste flurohinoloni. Fluorohinoloni su grupa antibiotika koji imaju širok spektar delovanja, nisku toksičnost i mali broj neželjenih dejstava na tretiranu ribu. Međutim, ostaci antibiotika u tkivima riba predstavljaju realan rizik za zdravlje ljudi. Ishrana ribom koja sadrži ostatke antibiotika može da dovede do pojave alergijskih, toksičnih, karcinogenih, mutagenih i teratogenih efekata. Stoga je neophodno da se ustanove rezidualne količine antibiotika u tkivima riba, što se može postići korišćenjem pouzdanih laboratorijskih metoda i tehnika kojima se ispituju ostaci antibiotika u mesu riba. Zbog navedenog, kao cilj rada definisano je da se ispita mogućnost identifikacije i kvantifikacije fluorohinolona u bubrezima šarana mikrobiološkom difuzionom metodom uz pomoć test mikroorganizma, E. coli ATCC 11303. Ispitivanjima je ustanovljeno da mikrobiološka difuziona metoda omogućava detekciju svih pet fluorohinolona (oksolinska kiselina, enrofloksacin, sarafloksacin, difloksacin i flumekvin) u tkivima bubrega šarana na različitim nivoima maksimalno dozvoljenih količina, MDK (100 μg/kg, 100 μg/kg, 30 μg/kg, 300 μg/kg i 600 μg/kg, respektivno). Identifikacija i kvantifikacija fluorohinolona na nivou MDK moguće je samo za enrofloksacin, difloksacin i flumekvin. Ovi fluorohinoloni mogu da se detektuju i kvantifikuju i na nivou ispod MDK, odnosno na nivou od 1/4 MDK. Za razliku od navedenih fluorohinolona, sarafloksacin se može detektovati samo na nivou od 2 MDK. Oksolinska kiselina se može detektovati na nivou od 4 MDK. Propisi EU predviđaju da trijažna (screening) metoda može biti primenjiva samo ukoliko se neko jedinjenje može detektovati u visini MDK, a preporučljivo je do 1/2 MDK. To znači da se mikrobiološka difuziona metoda može koristiti u rutinskoj analitičkoj praksi za identifikaciju i kvantifikaciju enrofloksacina, difloksacina i flumekvina u tkivima bubrega šarana.

AB  - Promene navika populacije u konzumiranju ribe, kao i sve veća potrošnja ribe iz akvakulture, nameću i određene zahteve koji se odnose na njen kvalitet, a naročito na sadržaj n-3 polinezasićenih masnih kiselina (PNMK), koje su od posebnog značaja za očuvanje zdravlja ljudi. Mnoga istraživanja ukazuju da masnokiselinski sastav hrane utiče na sastav masnih kiselina u mesu ribe. Hrana bogatija n-3 masnim kiselinama, pri istim uslovima uzgoja, značajno utiče na povećanje odnosa n-3/n-6 PNMK u tkivima ribe. Cilj ovoga rada je bio da se utvri uticaj ishrane, odnosno izbora hraniva na sadržaj ukupne masti i masnokiselinski sastav fileta konzumne pastrmke, gajenih u dva različita ribnjaka sa intenzivnom proizvodnjom. Dobijeni rezultati su pokazali razlike u hemijskom (sadržaj masti) i masnokiselinskom sastavu (sadržaj zasićenih (ZMK), mononezasićenih (MNMK), polinezasićenih (PNMK), n-3 i n-6 masnih kiselina) mesa konzumne kalifornijske pastrmke, koje su posledica ishrane različitom hranom i temperature vode ribnjaka. Utvrđeno je da je meso konzumne pastrmke iz Ribnjaka I nutritivno vrednije zbog povoljnijeg n-3/n-6 odnosa.

AB  - For several decades fish welfare has been subject of many researches, and the interest for this subject is connected with the fact that fish can feel pain and suffering. In addition to this stressors' mechanisms of action and their consequences are similar in mammals and fish. Assessment of welfare for farmed fish is based on the same principles as for terrestrial farm animals. However, special methods of collecting data are needed due to morphological characteristics of fish and properties of their environment. In the world and in our country, researches of different ways of fish welfare assessment are being conducted, especially based on non-invasive techniques such as monitoring of behaviour. In this paper a review of basic principles and methods used in assessment of farmed fish welfare is given.
AB  - Dobrobit riba je već nekoliko decenija predmet brojnih istraživanja, a zainteresovanost za ovu temu povezana je sa činjenicom da ribe mogu da osećaju bol i patnju. Osim toga, mehanizmi i posledice delovanja stresora kod riba vrlo su slični onima kod sisara. Procena nivoa dobrobiti kod farmski uzgajanih riba zasniva se na istim principima kao kod kopnenih farmskih životinja. Međutim, voda kao životna sredina riba i njihove morfološke specifičnosti zahtevaju posebne metode prikupljanja podataka. U svetu i kod nas se istražuju različiti načini procene dobrobiti riba, naročito oni koji se zasnivaju na neinvazivnim postupcima, kao što je praćenje ponašanja. U ovom radu dat je prikaz osnovnih principa i metoda koji se koriste u proceni dobrobiti farmski gajenih riba.
