AB  - Zarastanje rana je složen proces koji uključuje koordinaciju između različitih tipova ćelija i odgovarajućeg mikrookruženja. Uprkos
visokom regenerativnom potencijalu kože, defekti u vidu dubokih rana ili opekotina, teško spontano zarastaju i zahtevaju terapijski tretman. I pored napretka u bioinžinjeringu kože i intenzivnih istraživanja
u cilju iznalaženja novih biomaterijala, lečenje hroničnih rana i dalje
predstavlja aktuelan klinički problem. Hidrogelni zavoji, karakteristične
porozne i hidratisane molekularne strukture, verno oponašaju mikrookruženje rane, te su se izdvojili kao obećavajuća alternativa i najbolji
kandidati za stimulisanje procesa zarastanja. Hidrogelovi su bazirani
na prirodnim i/ili sintetičkim polimerima, i u vidu trajnih ili privremenih
zavoja podržavaju regeneraciju i zarastanje oštećene kože. Uvođenje
nanotehnologije je omogućilo nove pristupe u sintezi i dobijanje in situ
hibridnih hidrogelova, odličnih svojstava i prilagodljive funkcionalnosti.
Istraživanja u cilju razvoja hidrogelnih zavoja sa mogućnošću procene
mikrookruženja rane i prilagođenog tretmana na zahtev, predstavljaju ključ napretka u polju zbrinjavanja hroničnih rana. Pretklinička ispitivanja in vivo su neophodan uslov za finalnu karakterizaciju svakog
novodizajniranog hidrogela.
Ovaj rad razmatra prednosti upotrebe hidrogelnih zavoja i napretke u razvoju inteligentnih tretmana za rane sa odloženim zarastanjem. Takođe, jedan deo rada je posvećen proceni hidrogelova in vivo, kao i ključnoj ulozi veterinara tokom pretkliničkih ispitivanja na eksperimentalnim animalnim modelima.

Komparativni prikaz animalnih modela u morfološkim analizama placentacije

AB  - Svrha ovog pregleda je da prikaže veliku raznolikost u strukturi i funkciji placente koja se sreće kod sisara, i podobnosti nekih vrsta kao animalnih modela u proučavanju placentacije. Postoje značajne razlike u strukturi i fiziologiji posteljice između čoveka, domaćih i eksperimentalnih životinja. Zbog toga, najčešće upotrebljavani animalni modeli za proučavanje placentacije (miš, kunić, zamorac), nisu adekvatni za sva istraživanja u veterinarskoj i humanoj medicini. Placente se klasifikuju prema svom obliku, strukturi i interhemalnom regionu. Horionske resice placente su obložene prizmatičnim epitelom i preko njega se odvija prenos svih supstanci od majke prema plodu. Ćelije ovog epitela su selektivno propustljive kako za korisne, tako i za potencijalno toksične materije.
	Istraživanja koja se bave analizama i osobenostima placentacije su oskudna, posebno sa aspekta kontakta i razmene materija između majke i ploda. Iz tog razloga, ovaj uvid može da pomogne u sagledavanju različitih problema u kliničkoj praksi i tretmanu koji su bitni za dobijanje zdravog potomstva.

AB  - Epidermis stem cells have a crucial role through the processes of proliferation and
differentiation, to replace cells that are constantly lost during tissue turnover or
following injury. On the other hand, thyroid hormones regulate the proliferation and
differentiation of epidermal cells and thus significantly influence the homeostasis
of the skin. It is well known that maternal hypothyroidism during pregnancy leads
to impaired development of many organ systems in their offspring. However, there
is a lack of data about the influence of maternal subclinical hypothyroidism during
pregnancy and lactation on the development of the skin and its derivatives in the litter.
The aim of this study was to investigate the effects of maternal thyroid dysfunction on
the development of the skin and its derivatives in their offspring in the early postnatal
period. Antithyroid substance 6-n-propyl-2-thiouracil was added into the drinking water
to female Albino Oxfords rats from the beginning of pregnancy and during lactation,
with the aim to induce subclinical and overt form of hypothyroidism. Skin samples
were taken from male pups within twenty-four hours and seven days after birth. The
main findings of this investigation were that both forms of maternal hypothyroidism
lead to serious damage of the epidermis in pups in terms of pronounced hyperkeratosis
and reduction of the germinal layer along with a reduced number of hair follicles and
their delayed morphogenesis. Epidermal impairments were more pronounced in pups
with the overt form of hypothyroidism while offspring with the subclinical form had
impairments that were less pronounced and delayed in occurrence.

AB  - Biocompatibility of materials is one of the most important conditions for their successful application in tissue regeneration
and repair. Cell-surface interactions stimulate adhesion and activation of macrophages whose acquaintance can assist in
designing novel biomaterials that promote favorable macrophage–biomaterial surface interactions for clinical application.
This study is designed to determine the distribution and number of macrophages as a means of biocompatibility evaluation
of two newly synthesized materials [silver/poly(vinyl alcohol) (Ag/PVA) and silver/poly(vinyl alcohol)/graphene (Ag/PVA/
Gr) nanocomposite hydrogels] in vivo, with approval of the Ethics Committee of the Faculty of Veterinary Medicine,
University of Belgrade. Macrophages and giant cells were analyzed in tissue sections stained by routine H&E and im munohistochemical methods (CD68+
). Statistical relevance was determined in the statistical software package SPSS 20
(IBM corp). The results of the study in terms of the number of giant cells localized around the implant showed that their
number was highest on the seventh postoperative day (p.o.d.) in the group implanted with Ag/PVA hydrogels, and on the
30th p.o.d. in the group implanted with Ag/PVA/Gr. Interestingly, the number of macrophages measured in the capsular and
pericapsular space was highest in the group implanted with the commercial Suprasorb© material. The increased mac rophage number, registered around the Ag/PVA/Gr implant on 60th p.o.d. indicates that the addition of graphene can, in a
specific way, modulate different biological responses of tissues in the process of wound healing, regeneration, and
integration.

and repair. Cell-surface interactions stimulate adhesion and activation of macrophages whose acquaintance can assist in
designing novel biomaterials that promote favorable macrophage–biomaterial surface interactions for clinical application.
This study is designed to determine the distribution and number of macrophages as a means of biocompatibility evaluation
of two newly synthesized materials [silver/poly(vinyl alcohol) (Ag/PVA) and silver/poly(vinyl alcohol)/graphene (Ag/PVA/
Gr) nanocomposite hydrogels] in vivo, with approval of the Ethics Committee of the Faculty of Veterinary Medicine,
University of Belgrade. Macrophages and giant cells were analyzed in tissue sections stained by routine H&E and im munohistochemical methods (CD68+
). Statistical relevance was determined in the statistical software package SPSS 20
(IBM corp). The results of the study in terms of the number of giant cells localized around the implant showed that their
number was highest on the seventh postoperative day (p.o.d.) in the group implanted with Ag/PVA hydrogels, and on the
30th p.o.d. in the group implanted with Ag/PVA/Gr. Interestingly, the number of macrophages measured in the capsular and
pericapsular space was highest in the group implanted with the commercial Suprasorb© material. The increased mac rophage number, registered around the Ag/PVA/Gr implant on 60th p.o.d. indicates that the addition of graphene can, in a
specific way, modulate different biological responses of tissues in the process of wound healing, regeneration, and
integration.",

AB  - The ethical dilemmas of using animals as in vivo models in preclinical and clinical examinations have been increasingly present in recent decades. Small laboratory animals (rats, rabbits) will continue to be used because they are cost-effective and permit the formation of statistically testable cohort groups; a task that, for financial, maintenance and care reasons, is almost prohibitive for larger animals. Technological advances in the production of new biomaterials for clinical use are enormous, but screening tests and methods used to assess biocompatibility lag behind these advances. The assessment of biological responses is slow and based on millennial recovery mechanisms in eukaryotic organisms. Therefore, the goal of researchers in this field is to re-evaluate old methods of biocompatibility assessment and introduce new methods of evaluation, especially for in vivo testing. In that sense, a revision of the ISO standards was planned and conducted in 2017, which insisted on cytotoxicity testing in cell lines and produced concrete proposals on how biocompatibility should be quantified. In vivo biocompatibility evaluation of biomaterials used for soft tissue recovery commonly utilises rats. Rabbits are recommended for implants used for hard tissues, because of the rabbit’s size, the possibility of implanting the biomaterials on a larger bone surface, and because of the peculiarities of rabbit bone tissue that favours rapid recovery after bone defects and enables easy reading of the results.

Histological and histochemical analysis of dry fermented sausage of kulen composition

AB  - The application of histological methods in meat composition analysis and identification of prohibited tissues and organs added to meat and meat products is still in the research phase, although there have been some promising results. The aim of this work was to assess the possibility of using histological and histochemical methods for analysis of kulen composition. In this research, six samples of kulen were examined, one of which was produced in domestic conditions, while the rest were commercial products sampled from local markets. The samples were carried through classical histological preparation. The obtained slides were stained with haematoxylin/eosin, Masson-Goldner, toluidine blue and periodic acid-Schiff/alcian blue. The content of muscle, fat and connective tissue was evaluated using histomorphometric analysis. Histological analysis of kulen composition determined the presence of the following structures: muscle, adipose and connective tissues, blood vessels, glandular epithelium, peripheral nerve, cartilage and plant tissue. The histomorphometric analysis showed that the kulen products contained on average 56±2.52% muscle tissue, 7.27±1.38% connective tissue and 19.82±3.24% adipose tissue. The results show that by applying histological methods it is possible to identify different permitted and prohibited animal tissues in kulen, and hence, it is possible to analyse the composition of kulen. It is also possible to confirm the presence of various plant tissues, but for their precise identification, additional histological methods are needed.
AB  - Primena histoloških metoda u analizi sastava mesa i utvrđivanju prisustva nedozvoljenih tkiva u mesu i proizvodima od mesa je još uvek u fazi istraživanja, iako su postojeći rezultati u ovoj oblasti obeć avajuć i. Cilj našeg istraživanja bio je ispitivanje moguć nosti korišć enja histoloških i histohemijskih metoda u analizi sastava fermentisane kobasice u tipu kulena. U ovom istraživanju ispitano je šest uzoraka kulena, od kojih je jedan proizveden u domać im uslovima, dok su ostali komercijalni proizvodi poticali iz lokalnih marketa. Uzorci su sprovedeni klasičnom histološkom procedurom. Dobijeni isečci bojeni su metodama: hematoksilin/ eosin,Masson-Goldner, toluidin plavo i perjodna kiselina/Schiff/alcijan plavo. Sadržaj mišić nog, masnog i vezivnog tkiva procenjen je primenom histomorfometrijskih analiza. Histološkom analizom sastava kulena utvrđeno je prisustvo sledeć ih struktura: mišić no, masno i vezivno tkivo, krvni sudovi, epitel žlezdanog tipa, periferni nerv, hrskavica i biljno tkivo. Histomorfometrijska analiza je pokazala da proizvodi sadrže u proseku 56±2,52% mišić nog tkiva, 7,27±1,38% vezivnog tkiva i 19,82±3,24% masnog tkiva. Rezultati pokazuju da je primenom histoloških metoda moguć e identifikovati različita dozvoljena i nedozvoljena životinjska tkiva u kulenu, pa je samim tim moguće analizirati sastav kulena. Takođe je moguć e potvrditi prisustvo različitih biljnih tkiva, ali je za njihovu preciznu identifikaciju neophodno primeniti dodatne metode.

AB  - Overt and subclinical maternal hypothyroidism affects cartilage and bone extracellular matrix formation during fetal endochondral bone development. Less is known about the
effect maternal hypothyroidism has on cell proliferation and death of growth plate chondrocytes.
This study aimed to evaluate the expression of proliferation (Ki67), autophagy (LC3) and apoptosis (caspase 3) markers in the epiphyseal cartilage of the tibia in pups from 
subclinical and overt hypothyroid does, in their early postnatal development.Hypothyroidism was induced with propylthiouracil using low (1.5 mg/L) for subclinical (H1) and high dose (150 mg/L) for overt (H2) form. Propylthiouracil was
administered through drinking water in pregnant Albino Oxford does from the first day of gravidity and during lactation. Control (C) group was not treated. Six, seven-day-old male
pups from each group were euthanized. Histological examinations were performed on paraffin sections of the proximal tibial growth plate. Immunohistochemistry was used to assess
the expression of Ki67, LC3 and caspase 3. The number of Ki67 positive chondrocytes was evaluated in the resting and proliferating zone using Image J. The expression of LC3 was
performed by counting the number of positive dots in chondrocytes.The number of Ki67 positive chondrocytes was higher in both hypothyroid groups, while the number of LC3 positive dots per cell was lower indicating that the chondrocyte
cell cycle in hypothyroid animals is faster and that the basal level of autophagy is reduced due to frequent mitosis. Compared to controls, caspase 3 expression in the terminal
hypertrophic chondrocytes was reduced in H1 and completely absent in H2 pups which might indicate impaired differentiation.We conclude that both forms of maternal hypothyroidism
in rats lead to accelerated proliferation, slowed autophagy and
compromised apoptosis of terminal hypertrophic chondrocytes in the
early infantile period and a delay in cartilage to bone transition.

AB  - A newly produced biomaterial is necessarily subject of standards, which are performed in vivo on animal models. For the evaluation of soft tissue regenerative possibilities after subcutaneous implantation of biomaterials – silver/poly(vinyl alcohol) (Ag/PVA) and novel silver/poly(vinyl alcohol)/graphene (Ag/PVA/Gr) provided for clinical use, sixteen rats were used, according to the instructions of international standards, ISO 10993-6, 2007. Histological sections were observed 7, 15, 30 and 60 days after grafting. These hydrogels were produced by in situ electrochemical synthesis of silver nanoparticles in the polymer matrices, which enabled obtaining completely safe and biocompatible materials, free from any additional toxic chemical reducing agents. Surgical implantation of hydrogels was done according to the permission of the Ethical Committee of the Faculty of Veterinary Medicine, University of Belgrade. Immunohistochemical (IHC) studies included the assessment of  smooth muscle expression actin in blood vessels (α-SMA), the expression of laminin and type I and type III collagen in the skin structures, and, the determination of cell proliferation marker expression (Ki-67) keratinocytes. The results were assessed in a semiquantitative manner. The data were analyzed in the statistical software package IBM SPSS 20. The conclusions indicated that Ag/PVA/Gr might be used as wound dressings to enhance the tissue
AB  - Bilo koji novoproizvedeni biomaterijal nužno podleže standardima, po čijim se protokolima sprovode in vivo istraživanja na životinjskim modelima. Za procenu mogućnosti regeneracije mekog tkiva nakon potkožne implantacije biomaterijala - srebra/poli(vinil alkohola) (Ag/PVA) i novog materijala srebra/poli(vinil alkohola)/grafena (Ag/PVA/ Gr) predviđenog za kliničku upotrebu, šesnaest pacova je korišćeno, po uputstvima međunarodnog standarda, ISO 10993-6 2007. Histološki uzorci su sakupljeni 7, 15, 30 i 60 dana nakon implantacije. Ovi hidrogelovi su proizvedeni in situ elektrohemijskom sintezom nanočestica srebra u polimernim matricama, što je omogućilo dobijanje potpuno sigurnih i biokompatibilnih materijala, bez ikakvih dodatnih toksičnih hemijskih sredstava za redukciju. Hirurška implantacija hidrogela urađena je prema dozvoli Etičkog komiteta Fakulteta veterinarske medicine Univerziteta u Beogradu. Imunohistohemijske (IHC) studije uključivale su procenu ekspresije aktina glatkih mišića u krvnim sudovima (α-SMA), ekspresiju laminina i kolagena tipa I i III u strukturama kože i određivanje ekspresije markera proliferacije (Ki-67) keratinocita. Rezultati su procenjeni semikvantitativnom analizom. Podaci su analizirani u statističkom softverskom paketu IBM SPSS 20. Zaključci su ukazali da se Ag/PVA/Gr može koristiti kao obloga za rane kako bi se povećao potencijal zarastanja tkiva i uspostavila brža integracija i kraće zadržavanje u tkivu.

AB  - Radni konji se u Srbiji koriste kao tovarne životinje koje nose/vuku tovar, često teži od njihove telesne mase. Postavili smo hipotezu da opterećenje koje trpe utiče na pojavu lezija na koštano-zglobnom sistemu. Cilj ovog rada je bio da se uporedno prikaže prisustvo i stepen oštećenja zglobne hrskavice (ZH) i subhondralne kosti (SHK), distalnih kondilusa treće metatarzalne kosti kod starijeg radnog konja pomoću makroskopskog pregleda hrskavice, radioloških i histoloških metoda. Na lokalnoj klanici uzorkovani su kičični zglobovi ekstremiteta radnog pastuva, starog 10 godina. Uz pomoć kompjuterizovane tomografije (CT) utvrđene su promene samo na zglobu prednje leve noge. Nakon disekcije označenog zgloba, izvršena je biopsija ZH i SHK metakarpalne kosti u predelu oštećenja, a bioptati su snimljeni mikro CT-om nakon čega su deklacifikovani i ukalupljeni u parafin, i analizirani su preseci. Makroskopski je bilo vidljivo da je lokalizacija oštećenja ZH bila simetrična na medijalnim i lateralnim epikondilusima matakarpalnih i metatarzalnih kostiju svih ekstremiteta. Linije habanja i palmarne artroze bile su slabo do umereno izražene. Na nativnim rendgenogramima zglobova nisu uočene patološke promene. Analizom snimaka načinjenih CT-om uočena je promena u subhondralnoj zoni distalne epifize treće metakarpalne kosti u vidu pukotine, a oko nje je primetna zona skleroze SHK. Na mikro CT snimku uzorka koštanog tkiva uzetog u predelu lateralnog epikondilusa metakarpane kosti, takođe je uočljiva zona subhondralne skleroze. Najmarkatnije histološke promene na ZH su bile: na površini ZH undulacije i fisure, koje se pružaju i do duboke zone, mnogobrojne izogene grupe sa većim brojem hondrocita u lakuni, fokalni gubitak ćelija i mestimičnio prisustvo hipertrofičnih hondrocita. U SHK su primećene: neznatne lezije koštanog matriksa u vidu svetlijeg prebojavanja, blago remodeliranje kosti koje se karakteriše njenim napredovanjem ka kalcifikovanoj hrskavici. Dugotrajno opterećenje utiče na pojavu oštećenja SHK koje se može uočiti uz pomoć CT-a i histološkom analizom, dok se stepen oštećenja ZH može evaluirati makroskopski i histološki.

AB  - This doctoral dissertation was designed in order to test the biocompatibility of two newly synthesized and previously in vitro characterized hydrogels based on poly(vinyl alcohol) polymers - Ag/PVA and Ag/PVA/Gr, which are intended for making wound dressings or soft tissue implants tissue. Poly(vinyl alcohol) (PVA) is a synthetic polymer commonly used in biomedicine due to its non-toxicity, solubility, biocompatibility, elasticity and the ability to produce reversible hydrogels with high sorption capacity. In the last few years, research has also focused on the incorporation of graphene (Gr) into hydrogel PVAs, in order to improve their mechanical strength and durability. In tissue engineering and regenerative medicine, it is particularly important that implants and bandages contain an antiinflammatory and antibacterial active ingredient to protect the sterility of the wound, or to prevent infection in the implant surrounding tissue. It is known that silver nanoparticles (AgNPs) have excellent antibacterial activity and are therefore often used as an active component of PVA-based hydrogel biomaterials. In order to test the biocompatibility of newly synthesized hydrogels, the soft tissue response to deep subcutaneously implanted Ag/PVA and Ag/PVA/Gr implants was compared to the reaction to the commercial calcium-alginate wound pad (Suprasorb ©, Lohmann & Rauscher GmbH & Co. KG, Neuwied, Germany), which is implanted in an identical manner. The tissue irritation index (IrI) was evaluated as the basic measure of tissue response to implanted hydrogels. In order to determine the degree of biocompatibility, the thickness of the connective tissue capsule around the implanted hydrogels, the number of inflammatory cells, including the Langerhans giant cells, the number of macrophages in the perimplant zone and their eneventual migration to the biomaterial, as well as the number of blood vessels around the implantation zone, were measured. The ratio of proliferating and apoptotic cellular skin and subcutaneous tissue cellular entities was observed, due to their responsibility for regeneration and reparation processes...
AB  - Ova doktorska disertacija dizajnirana je u cilju da se ispita biokompatibilnost dva novosintetisana i prethodno in vitro okarakterisana hidrogela na bazi sintetskog polimera polivinil-alkohola − Ag/PVA i Ag/PVA/Gr, koji su namenjeni za izradu obloga za rane ili za implantate za meka tkiva. Polivinil-alkohol (PVA) je sintetički polimer koji se često koristi u biomedicini zbog svoje netoksičnosti, rastvorljivosti, biokompatibilnosti, elastičnosti i sposobnosti stvaranja reverzibilnih hidrogelova s visokom sposobnošću sorpcije. U poslednjih nekoliko godina, istraživanja se takođe fokusiraju na ugradnju grafena (Gr) u hidrogelove PVA kako bi se poboljšala njihova mehanička čvrstoća i trajnost. U inženjerstvu tkiva i regenerativnoj medicini posebno je važno da implantati i zavoji sadrže antiinflamatornu i antibakterijsku aktivnu komponentu kako bi zaštitili sterilnost rane ili sprečili infekciju u tkivu koje okružuje implantat. Poznato je da nanočestice srebra (eng. silver nanoparticles, AgNPs) poseduju odličnu antibakterijsku aktivnost, pa se zbog toga često koriste kao aktivna komponenta biomaterijala na bazi hidrogela od PVA. U cilju ispitivanja biokompatibilnosti novosintetisanih hidrogelova, praćena je reakcija mekog tkiva na duboko potkožno postavljene implantate od Ag/PVA i Ag/PVA/Gr i poređena je sa reakcijom na komercijalnu oblogu za rane na bazi kalcijum-alginata (Suprasorb©, Lohmann & Rauscher GmbH & Co. KG, Neuwied, Germany), koja je implantirana na identičan način. Procenjivan je tkivni iritacioni indeks (IrI) kao osnovna mera tkivnog odgovora na implantirane hidrogelove. Za određivanje stepena biokompatibilnosti merena je debljina vezivnotkivne kapsule oko implantiranih hidrogelova, broj inflamatornih ćelija, uključujući i Langerhansove džinovske ćelije, broj makrofaga u periimplantnoj zoni i njihova enevntualna migracija u biomaterijal, kao i brojnost krvnih sudova mikrovaskularnog korita oko zone implantacije. Posmatran je odnos proliferacije i apoptoze ćelijskih entiteta kože i potkožnog tkiva odgovornih za procese regeneracije i reparacije...

abstract = "This doctoral dissertation was designed in order to test the biocompatibility of two newly synthesized and previously in vitro characterized hydrogels based on poly(vinyl alcohol) polymers - Ag/PVA and Ag/PVA/Gr, which are intended for making wound dressings or soft tissue implants tissue. Poly(vinyl alcohol) (PVA) is a synthetic polymer commonly used in biomedicine due to its non-toxicity, solubility, biocompatibility, elasticity and the ability to produce reversible hydrogels with high sorption capacity. In the last few years, research has also focused on the incorporation of graphene (Gr) into hydrogel PVAs, in order to improve their mechanical strength and durability. In tissue engineering and regenerative medicine, it is particularly important that implants and bandages contain an antiinflammatory and antibacterial active ingredient to protect the sterility of the wound, or to prevent infection in the implant surrounding tissue. It is known that silver nanoparticles (AgNPs) have excellent antibacterial activity and are therefore often used as an active component of PVA-based hydrogel biomaterials. In order to test the biocompatibility of newly synthesized hydrogels, the soft tissue response to deep subcutaneously implanted Ag/PVA and Ag/PVA/Gr implants was compared to the reaction to the commercial calcium-alginate wound pad (Suprasorb ©, Lohmann & Rauscher GmbH & Co. KG, Neuwied, Germany), which is implanted in an identical manner. The tissue irritation index (IrI) was evaluated as the basic measure of tissue response to implanted hydrogels. In order to determine the degree of biocompatibility, the thickness of the connective tissue capsule around the implanted hydrogels, the number of inflammatory cells, including the Langerhans giant cells, the number of macrophages in the perimplant zone and their eneventual migration to the biomaterial, as well as the number of blood vessels around the implantation zone, were measured. The ratio of proliferating and apoptotic cellular skin and subcutaneous tissue cellular entities was observed, due to their responsibility for regeneration and reparation processes..., Ova doktorska disertacija dizajnirana je u cilju da se ispita biokompatibilnost dva novosintetisana i prethodno in vitro okarakterisana hidrogela na bazi sintetskog polimera polivinil-alkohola − Ag/PVA i Ag/PVA/Gr, koji su namenjeni za izradu obloga za rane ili za implantate za meka tkiva. Polivinil-alkohol (PVA) je sintetički polimer koji se često koristi u biomedicini zbog svoje netoksičnosti, rastvorljivosti, biokompatibilnosti, elastičnosti i sposobnosti stvaranja reverzibilnih hidrogelova s visokom sposobnošću sorpcije. U poslednjih nekoliko godina, istraživanja se takođe fokusiraju na ugradnju grafena (Gr) u hidrogelove PVA kako bi se poboljšala njihova mehanička čvrstoća i trajnost. U inženjerstvu tkiva i regenerativnoj medicini posebno je važno da implantati i zavoji sadrže antiinflamatornu i antibakterijsku aktivnu komponentu kako bi zaštitili sterilnost rane ili sprečili infekciju u tkivu koje okružuje implantat. Poznato je da nanočestice srebra (eng. silver nanoparticles, AgNPs) poseduju odličnu antibakterijsku aktivnost, pa se zbog toga često koriste kao aktivna komponenta biomaterijala na bazi hidrogela od PVA. U cilju ispitivanja biokompatibilnosti novosintetisanih hidrogelova, praćena je reakcija mekog tkiva na duboko potkožno postavljene implantate od Ag/PVA i Ag/PVA/Gr i poređena je sa reakcijom na komercijalnu oblogu za rane na bazi kalcijum-alginata (Suprasorb©, Lohmann & Rauscher GmbH & Co. KG, Neuwied, Germany), koja je implantirana na identičan način. Procenjivan je tkivni iritacioni indeks (IrI) kao osnovna mera tkivnog odgovora na implantirane hidrogelove. Za određivanje stepena biokompatibilnosti merena je debljina vezivnotkivne kapsule oko implantiranih hidrogelova, broj inflamatornih ćelija, uključujući i Langerhansove džinovske ćelije, broj makrofaga u periimplantnoj zoni i njihova enevntualna migracija u biomaterijal, kao i brojnost krvnih sudova mikrovaskularnog korita oko zone implantacije. Posmatran je odnos proliferacije i apoptoze ćelijskih entiteta kože i potkožnog tkiva odgovornih za procese regeneracije i reparacije...",

Fetlock Joint Cartilage Lesions in Working Horses – A PILOT STUDY

AB  - The horse is still used as a working animal, harnessed in a sled or wheeled chariots 
loaded with cargo several times heavier than the horse itself. There are no data on the extent 
of cartilage lesions in the fetlock joints of front and hind legs of working horses. The aim of 
the study was to determine the size and position of gross cartilage lesions in front and hind 
metacarpo-phalangeal (fetlock) joints of working horses. 
Front and hind legs were collected from a local abattoir after the working horses were 
slaughtered for the reasons not related to this study. The study was conducted on eight horses 
from 5 to 12 years old (mean 8.625 ± 0.91 years), with a body weight of 550 ± 29.46 kg. Thirty two fetlock joints were examined for external gross anatomy changes. After joint dissection, the 
cartilage of metacarpal and metatarsal bones was stained with blue ink and photos were taken. 
The cartilage lesions were stained deep blue, in contrast to light blue surface of the undamaged 
cartilage. The anatomical position of the lesions was recorded. The ImageJ software was used 
to measure the total surface of the cartilage and the surface of the cartilage under the lesions. 
The dimension of the lesions was expressed as a percentage of the total cartilage surface and 
presented as mean ± standard error. 
Gross anatomy changes like skin lesions, wounds, joint oedema or any type of 
deformities were not observed. The position of the cartilage lesions was symmetrical and 
occupied lateral and medial metacarpal and metatarsal condyles next to the sagittal ridge. The 
lesions on the lateral condyles had the same surface area like those on the medial, 12.3 ± 1.03% 
versus 10.7 ± 0.99%, respectively. The cartilage lesions on the front legs (23.62 ± 1.68%) were 
broader than those on the hind legs (22.5 ± 3.35%).
The results of this pilot study indicate no difference between metacarpal and metatarsal 
cartilage surface lesions. Further studies are indicated to determine the severity of cartilage and 
putative subchondral bone lesions by histologic evaluation

Mitochondria in pachytene: the fragile point of maternal subclinical hypothyroidism affection

AB  - The stimulative effects of thyroid hormones on mitochondria are realized through 
both non-genomic and genomic mechanisms, affecting respiration, mitochondrial plasticity 
and biogenesis. The subclinical form of maternal hypothyroidism in rats induces significant 
reduction of mitochondria number but also an augmentation of their area in neonatal and 
early infantile offspring dyctiotene oocytes. This study aimed to investigate if this form of 
subclinical hypothyroidism affects mitochondrial morphology and distribution in the early 
prophase of meiosis I oocytes. It was performed on newborn control (C) (n=10) and 
hypothyroid (SCH) (n=10) female rat pups derived from control (n=6) and propylthiouracil
treated pregnant dams (n=6), respectively. Ovaries of all pups were removed and processed 
for transmission electron microscopy. The morphological features of mitochondria in the 
early prophase I oocytes until dyctiotene were assessed. No substantial differences were 
found in leptotene and zygotene oocytes in SCH group comparing to control, except just a 
few mitochondria characterized with shortened cristae, presence of wide pale area centrally 
positioned and membrane disruption. Pachytene mitochondria in treated pup oocytes were 
in great extent with disrupted membrane, shortened cristae and wide pale area centrally 
positioned while these features were rarely observed in control ones. Our results confirm 
altered mitochondria morphology found in primordial and primary follicles in case of 
maternal hypothyroidism, indicating their impaired function and possibly, propensity to 
programmed cell death. Further investigations may indicate to what extent pachytene, as a 
meiotic checkpoint, appears to be a milestone possibly predetermining the future of the 
cell.

Effect of subclinical and overt form of rat maternal hypothyroidism on offspring endochondral bone formation

AB  - Maternal hypothyroidism in its overt form affects skeletal development of the offspring, but these data are not available for the subclinical form which is becoming very frequent among pregnant women. We hypothesized that the subclinical form of hypothyroidism in rat dams, influences the process of offspring endochondral ossification affecting proliferation and differentiation of chondrocytes, osteoclasts and osteoblasts in pups. Seven-day-old male pups (n=18) derived from control dams and dams treated with a low dose (1.5 mg/L) or high dose (150 mg/L) of propylthiouracil in drinking water during pregnancy and lactation were used. Histomorphometric analysis of pups tibia proximal growth plate, expression of mRNA, immunohistochemical and histochemical visualization of extracellular matrix components was performed. The length of the tibia was reduced in hypothyroid pups. Secretion of type 2 and 10 collagens in the subclinical and overt form were lower while the amount of glycosaminoglycans was higher when compared with controls. Down-regulated tartrate resistant acid phosphatase mRNA indicated altered osteoclasts function while lower expression of dentin matrix acid protein-1 mRNA and reduced synthesis of type 1 collagen accentuated a compromised bone formation in the overt form of hypothyroidism. The subclinical form of maternal hypothyroidism had a negative effect on the differentiation of hypertrophic chondrocytes and calcified cartilage removal in 7-day-old pups. In addition, overt hypothyroidism had a negative effect on the proliferation of chondrocytes and deposition of osteoid. Both forms of hypothyroidism resulted in a decrease of tibia length due to changes in growth plate formation.

Seven-day-old male pups (n=18) derived from control dams and dams treated with a low dose (1.5 mg/L) or high dose (150 mg/L) of propylthiouracil in drinking water during pregnancy and lactation were used. Histomorphometric analysis of pups tibia proximal growth plate, expression of mRNA, immunohistochemical and histochemical visualization of extracellular matrix components was performed. The length of the tibia was reduced in hypothyroid pups. Secretion of type 2 and 10 collagens in the subclinical and overt form were lower while the amount of glycosaminoglycans was higher when compared with controls. Down-regulated tartrate resistant acid phosphatase mRNA indicated altered osteoclasts function while lower expression of dentin matrix acid protein-1 mRNA and reduced synthesis of type 1 collagen accentuated a compromised bone formation in the overt form of hypothyroidism. The subclinical form of maternal hypothyroidism had a negative effect on the differentiation of hypertrophic chondrocytes and calcified cartilage removal in 7-day-old pups. In addition, overt hypothyroidism had a negative effect on the proliferation of chondrocytes and deposition of osteoid. Both forms of hypothyroidism resulted in a decrease of tibia length due to changes in growth plate formation.",

AB  - Introduction. The ovarian surface epithelium (OSE) undergoes intensive regeneration and remodelling after each ovulation during the whole reproductive period. This process increases the risk of one of the most common ovarian tumors in women and the female dog. Considering the fact that maternal hypothyroidism highly impacts cell proliferation and cell death during folliculogenesis in the early neonatal period, we aimed to analyze its effect on OSE morphology and dynamics. Materials and Methods. The study was performed on newborn (24-h-old) and neonatal (4-day-old) female rats, a randomized trial between the control and hypothyroid groups, born under controlled circumstances and hypothyroid mothers, respectively. Their ovaries were analyzed histologically and processed to determine the OSE cell height as an average value of four measurement points. Also, the immunopositivity of the proliferating cell nuclear antigen (PCNA) and caspase-3 were assessed semiquantitatively. Results and Conclusions. No major structural differences of OSE were found between groups within the given ages except for a slight increment of OSE cell height and incompleteness of apical cell membrane with cytoplasmic projections in hypothyroid animals. PCNA immunopositivity of the OSE cells was higher in ovaries of hypothyroid animals of both ages in comparison to the controls. Moreover, only scarce OSE cells were caspase-3 positive in both groups and ages, with no difference in immunopositivity. Our study confirms the impact of hypothyroidism in the early postnatal period on morphology and proliferation rate of OSE cells, with no effect on caspase-3 dependent cell removal, which may serve as a premise for future investigation of potential carcinogenesis, in terms of prevention and treatment of ovarian cancer.
AB  - Uvod. Tokom reproduktivnog perioda, nakon svake ovulacije, površinski epitel (PE) jajnika se intenzivno regeneriše i remodelira. Ovaj proces povećava rizik od nastajanja jednog od najčešćih tumora jajnika kod žena i kuja. Imajući u vidu činjenicu da hipotireoidizam majki u velikoj meri utiče na proliferaciju i ćelijsku smrt u toku folikulogeneze u ranom postnatalnom periodu, cilj rada je bio da analiziramo efekat hipotireoidizma majki na morfologiju i dinamiku PE jajnika potomaka. Materijal i metode. U eksperimentu su korišćeni tek rođeni (24 časa stari) i neonatalni (4 dana stari) mladunci koji potiču od kontrolnih i hipotireoidnih majki. AB  - In this paper, we have shown for the first time the soft tissue response of novel silver/poly(vinyl alcohol)/graphene (Ag/PVA/Gr) and silver/poly(vinyl alcohol)/chitosan/graphene (Ag/PVA/CHI/Gr) nanocomposite hydrogels aimed for medical applications. These novel hydrogels were produced by in situ electrochemical synthesis of silver nanoparticles in the polymer matrices as described in our previously published works. Both Ag/PVA/Gr and Ag/PVA/CHI/Gr, as well as controls Ag/PVA, Ag/PVA/CHI and commercial Suprasorb (c) hydrogel discs, were implanted in the subcutaneous tissue of rats. Implants with the surrounding tissue were dissected after post-implantation on days 7, 15, 30 and 60, and then processed for histological examination. The tissue irritation index (TIrI) score, according to ISO 10993-6, 2007, as well as the number of leukocytes in the peri-implant zone and connective tissue capsule thickness were examined. The results show that each TIrI score, the leukocyte number around the implanted materials and capsule thickness gradually decreased during the observation period. At the endpoint of follow-up, the Ag/PVA/CHI/Gr implant was surrounded with a thinner capsule, while both the TIrI score and the number of leukocytes of the peri-implant zone were greater compared to the Ag/PVA/Gr implant. Despite the observed differences, we can conclude that our in vivo experiment suggested that both novel hydrogels were biocompatible and suitable for medical use.

Z-cells and oogonia/oocytes in the advanced process of autophagy are the dominant altered cells in the ovaries of hypothyroid newborn rats

AB  - Induced prenatal hypothyroidism in rat pups leads to accelerated primordial follicle assembly and premature follicular atresia with ovary failure. This work investigates the influence of maternal hypothyroidism induced with 6-n-propyl-2-thyouracil (PTU) on the number and morphology of oogonia/oocytes in newborn rat pups with light and transmission electron microscopy. Expression of apoptosis and autophagy markers in oogonia/oocytes were examined using immunohistochemistry. Hypothyroid newborn pups had a decreased number of mitotic and resting oogonia, while the number of altered oogonia/oocytes was increased. Ultrastructural observations revealed the increased presence of degenerated pachytene oocytes (Z-cells) and oogonia/oocytes undergoing autophagy, apoptosis and combined apoptosis and autophagy, in this group. The most abundant altered oogonia/oocytes in the hypothyroid group were those with morphological features of advanced autophagy and Z-cells. The percentage of TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) positive oogonia/ oocytes was significantly lower in the hypothyroid group. No significant difference was recorded in the expression of caspase-3, ATG7 and LC3 possibly reflecting that these proteins were not involved in the oogonia/ oocyte alteration process during prenatal rat hypothyroidism. The obtained results indicate that developmental hypothyroidism in the offspring enhances the number of Z-cells and oogonia/ oocytes altered with the advanced process of autophagy.

AB  - Introduction. Thyroid hormones (TH) play a pivotal role in the development of mammalian skin and are necessary for both, the initiation and maintenance of hair growth. Hypothyroid rats showed  impaired epidermal proliferation, hair growth, and wound healing. Stem cells located at the bulges of the hair follicles are responsible for hair cycling and contribute to the regeneration of the new epidermis after wounding. Therefore a reduction in the num¬ber or function of the bulge stem cells could be responsible for this phenotype. 
We hypothesized that decrease in TH during prenatal development would affect bulge stem cell population.
Material and methods. Subclinical hypothyroidism was induced with propylthiouracil through drinking water in dose 1.5 mg/l in pregnant Albino Oxford rats from the first day of gravidity. Control group was untreated. Seven-day-old pups were euthanized and skin samples were taken from dorsal part of their bodies. For the immunohistochemical study PCNA (Santa Cruz Biotechnology) and NANOG (Thermo scientific) antibodies were used.
Results. . The number of hair follicles was reduced in hypothyroid pups compared to 
controls. They also had a decreased expression of PCNA in cells of inner and outer sheat of hair follicles, but increased number of NANOG positive bulge stem cells. 
Conclusion. This suggests that re¬duced activation of stem cells leading to their accumulation in the bulges which is associated with reduced stem cell prolif¬eration and mobilization.

AB  - Thyroid hormones (TH) are indispensable for normal development in mammals. The effects of maternal hypothyroidism on development and function of different organs of their offspring are well known. It is unclear how its subclinical form affects some of these processes, including endochondral ossification during the prenatal and early postnatal development.
The aim of the study was to assess bone formation by defining the expression of tartarate-resistant acid phosphatase (TRAP), alkaline phosphatase (ALP) and mRNA for dentin matrix acidic phosphoprotein (DMP1) in tibia of pups from subclinical hypothyroid dams in early postnatal development. Subclinical hypothyroidism (SHT) was induced with propylthiouracil through drinking water (1.5 mg/l) in pregnant Albino Oxford dams from the first day of gravidity and during lactation. Control group was untreated. Five seven-day-old pups from each experimental group were euthanized. All histological examinations were performed on paraffin sections of the proximal tibial growth plate. Histochemistry was used to assess the expression of TRAP, immunohistochemistry for ALP and in situ hybridization for DMP1. The area of TRAP positive signal per 1µm2 and optical density of ALP immunostaining were evaluated using the Image J. The number of DMP1 positive cells was counted for given area using the same software. Level of thyroxine (T4) and triiodothyronine (T3) of dams and pups were decreased (data published). Expression of TRAP and ALP in the proximal tibial growth plate were lower in hypothyroid pups and the number of cells expressing mRNA for DMP1 was decreased. 
As it is known DMP1 and ALP are expressed in osteoblasts with an important role in endochondral ossification while TRAP represents a marker of osteoclasts function and bone resorption, so we can conclude  that maternal SHT in rats leads to delayed osteogenic differentiation and compromised cartilage to bone transition in early infantile period.

AB  - Ovarian surface epithelium (OSE) is a very dynamic part of the ovary with an important role in ovulation during reproductive period. It represents a source of germ cells in fetal period but data concerning that role of OSE in postnatal period are contradictory. Though OSE is a subject of research in past few years, it’s ultrastructure in newborn rats has not been studied yet.
The aim of the study was to define ultrastructure features of OSE in rats in the first 24h hours after birth.
Six newborn rats within first 24 hours after birth were euthanized. Ovaries were fixed in 4% glutaraldehyde, post-fixed in 1% osmium tetroxide, routinely dehydrated and embedded in araldite. Semi-thin sections were stained with toluidine blue and used to select areas of interest. Ultra-thin sections of selected areas were mounted on copper grids and examined on a Philips CM12 transmission electron microscope. Descriptive ultrastrustructure analysis was performed. OSE in newborn rats was mostly single layered and sporadically pseudostratified, consisted from mostly cuboidal and columnar cells. These cells were with centrally positioned nucleus, mostly with irregularly dispersed chromatin. Microvili and cytoplasmic projections extended to peritoneal cavity from the surface of the cells. No continuous tunica albuginea was found, but in places, groups of fibroblast-like cells were present, indicating the onset of its formation. Numerous oogonia and oocytes were identified between OSE cells or up to the basal part of them. This may indicate that extrusion (the release of germ cells into a peritoneal cavity) may be an important way of germ cells elimination. 
Early postnatal period in rat ovaries characterize with massive cell death of oogonia and oocytes, mostly by apoptosis and autophagy, but further investigation might reveal the participation of this way of germ cells elimination realizing the importance of appropriate course of cell death process in terms of reproductive health.

AB  - Thyroid hormones (TH) are one of the key factors for normal prenatal development in mammals. Previously, we showed that subclinical maternal hypothyroidism leads to premature atresia of ovarian follicles in female rat offspring in the pre-pubertal and pubertal periods. The influence of decreased concentration of TH on primordial follicles pool formation during neonatal and early infantile period of rat pups was not investigated previously. Maternal hypothyroidism during pregnancy has irreversible negative influence on primordial follicles pool formation and population of resting oocytes in female rat offspring. The study was done on neonatal and early infantile control (n-10) and hypothyroid (n-10) female rat pups derived from control (n-6) and propylthiouracil (PTU) treated pregnant dams (n-6), respectively. Ovaries of all pups were removed and processed for light and transmission electron microscopy (TEM). Number of nests, oogonia and oocytes per nest, primordial, primary, secondary and preantral follicles were determined. Screening for overall calcium presence in ovarian tissue was done using Alizarin red staining. Morphology and volume density of nucleus, mitochondria and smooth endoplasmic reticulum (sER) in the oocytes in primordial follicles was also assessed. Caspase-3 and terminal deoxynucleotidyl transferase dUTP nick end labelling (TUNEL), both markers for apoptosis, and proliferating cell nuclear antigen (PCNA) for proliferation were determined in oocytes and granulosa cells in different type of follicles. In neonatal period, ovaries of hypothyroid pups had a decreased number of oogonia, oocytes and nests, an increased number of primordial follicles and a decreased number of primary and secondary follicles, while in early infantile period, increased number of primary, secondary and preantral follicles were found. Alizarin red staining was intense in hypothyroid neonatal rats that also had the highest content of dilated sER. Number of mitochondria with altered morphology in both groups of hypothyroid pups was increased. Apoptosis markers have not shown significant difference between groups but PCNA had an increased expression in the oocytes and granulosa cells in primordial follicles of hypothyroid rats. Light and electron microscopy analysis indicate that previously detected premature ovarian follicular atresia in pre-pubertal and pubertal hypothyroid rats is preceded with premature formation of primordial follicles followed by slight changes on sER and mitochondria in examined oocytes, and increased expression of PCNA.

Uticaj subkliničkog hipotireoidizma majki na hipertrofičnu diferencijaciju hondrocita u proksimalnoj epifiznoj ploči tibije novorođenih pacova starih 7 dana

AB  - Thyroid hormones (TH) are indispensable for normal development in mammals. The effects of maternal hypothyroidism on development and function of different organs of their offspring are well known. It is unclear how its subclinical form affects some of these processes, including endochondral ossification during the prenatal and early postnatal development.
The aim of the study was to assess bone formation by defining the expression of tartarate-resistant acid phosphatase (TRAP), alkaline phosphatase (ALP) and mRNA for dentin matrix acidic phosphoprotein (DMP1) in tibia of pups from subclinical hypothyroid dams in early postnatal development. Subclinical hypothyroidism (SHT) was induced with propylthiouracil through drinking water (1.5 mg/l) in pregnant Albino Oxford dams from the first day of gravidity and during lactation. Control group was untreated. Five seven-day-old pups from each experimental group were euthanized. All histological examinations were performed on paraffin sections of the proximal tibial growth plate. Histochemistry was used to assess the expression of TRAP, immunohistochemistry for ALP and in situ hybridization for DMP1. The area of TRAP positive signal per 1µm2 and optical density of ALP immunostaining were evaluated using the Image J. The number of DMP1 positive cells was counted for given area using the same software. Level of thyroxine (T4) and triiodothyronine (T3) of dams and pups were decreased (data published). Expression of TRAP and ALP in the proximal tibial growth plate were lower in hypothyroid pups and the number of cells expressing mRNA for DMP1 was decreased. 
As it is known DMP1 and ALP are expressed in osteoblasts with an important role in endochondral ossification while TRAP represents a marker of osteoclasts function and bone resorption, so we can conclude  that maternal SHT in rats leads to delayed osteogenic differentiation and compromised cartilage to bone transition in early infantile period.

AB  - Uvod: Poznato je da su tireoidni hormoni (TH) od esencijalnog značaja za razvoj, funkciju i diferencijaciju svih ćelija u organizmu u toku prenatalnog i postnatalnog života. Prethodna istraživanja dovode u vezu smanjen nivo TH i razvoj reumatoidnog artritisa i drugih oboljenja zglobova, međutim mehanizam nastanka navedenih promena još uvek nije razjašnjen u potpunosti. U sinovijalnim membranama (SM) zglobova zahvaćenih reumatoidnim artritisom, utvrđeno je povećanje broja ćelija koje eksprimiraju PCNA (proliferating cell nuclear antigen). Cilj ovog istraživanja je bio da se definiše uticaj subkliničkog hipotireoidizma majki tokom graviditeta na SM novorođenih jedinki, s obzirom da u dosadašnjim istraživanjima ovaj efekat nije dovoljno proučen.
Materijal i metode: Subklinički hipotireoidizam je kod gravidnih jedinki Albino Oxford pacova prouzrokovan dodavanjem propiltiouracila kroz vodu za piće (1,5 mg/l), počevši od prvog dana graviditeta i tokom laktacije. Kontrolna grupa pacova nije tretirana. Mladunci su žrtvovani sedmog dana nakon rođenja. Histološka ispitivanja sprovedena su na parafinskim presecima debljine 5µm obojenim standardnim hematoksilin/eozin histohemijskim bojenjem, čija je analiza sprovedena na svetlosnom mikroskopu (Olympus BX53). Ekspresija PCNA (Santa Cruz Biotechnology, sc-7907) pokazana je imunohistohemijski, pomoću vizualizacionog kita- Rabbit Specific HRP/DAB (ABC) Detection IHC Kit (ab64261, Abcam, UK). Kvantifikacija ekspresije PCNA izvršena je pomoću softvera Image J. . Prikazan je broj PCNA pozitivnih ćelija u odnosu na 3000 ćelija SM.
Rezultati: Pregledom preseka bojenih hematoksilin/eozinom nisu uočene značajne razlike u građi SM između kontrolne i tretirane grupe. Broj ćelija koje eksprimiraju PCNA u SM tretiranih životinja je značajno veća od broja ćelija u SM kontrolnih životinja. 
Zaključak: Povećana ekspresija PCNA ukazuje na intenziviranu proliferaciju ćelija SM novorođenih pacova starosti 7 dana. Ovaj rezultat može biti osnova za nastavak istraživanja uticaja hipotireoidizma na razviće SM.
