Reticulocyte response in paired blood samples of Babesia canis infected young and adult dogs

AB  - Acute B. canis infection can lead to an acute phase reaction (APR) in dogs. The parasite invades red blood cells causing
anemia through immune-mediated hemolysis and possible erythropoietic suppression. A regenerative response of the
erythroid lineage during the babesiosis has not been described in extension. This research examines hematologic parameters
focusing on the absolute reticulocyte count and apolipoprotein A I (ApoA I) level on the day of admission and 14 days after
treatment with imidocarb-dipropionate in young (n=11) and adult (n=11) dogs naturally infected with B. canis. Metabolic
and inflammatory processes were characterized by analyzing protein and lipid profiles, as well as ApoA I at specified time
points. Automated analyzers were used to determine complete blood count and biochemical parameters, while ApoA I was
assessed using radioimmunoassay. The reticulocyte count was determined using a manual method by means of supravital
staining. Both young and adult dogs with acute B. canis infection showed non-regenerative anemia without difference.
Fourteen days after successful treatment with imidocarb-dipropionate, the anemia was corrected and a high reticulocyte
count was observed (p<0.05). This indicates that the erythroid regenerative response was efficient in young and adult dogs,
although vital signs, leukocyte count and triglyceride concentration suggest a more intense APR in young dogs. A decrease
in ApoA I in both groups 14 days after treatment (p<0.01) confirmed that this lipoprotein acts as a positive acute-phase
protein in acute B. canis infection in dogs, but further studies are needed to connect its role in erythroid lineage regeneration.

AB  - Red blood cell osmotic fragility (RBCOF) refers to susceptibility to hemolysis in
vivo and in vitro. An increase in mean cellular hemoglobin concentration (MCHC)
also reflects in vivo and in vitro hemolysis. Common inflammatory infectious
diseases in dogs are often characterized by an increase in MCHC; however, it is
not known if there is a concomitant increase in RBCOF. The objective of this
study was to determine RBCOF in dogs with babesiosis, dirofilariosis,
parvovirosis, pyometra, and canine distemper and to investigate whether RBCOF
correlates with routine hematologic and biochemical parameters and lipoprotein
fractions. The diagnosis of canine infectious disease was made and complete
blood count, serum biochemical profiles, and lipoprotein electrophoresis were
determined according to current institutional protocols. RBCOF of healthy
control (n=8) and infected dogs (n=73) was measured with a phosphate-buffered
saline dilution test, and mean corpuscular fragility (MCF) was calculated. MCF
was increased in dirofilariosis, pyometra, and distemper; MCHC was increased
in all infected groups; HDL was decreased in babesiosis, pyometra, and
distemper compared with healthy controls. However, no correlation was found
between these parameters. On the other hand, hematological and biochemical
parameters, that are highly predictive of specific infections, correlated with MCF:
ALP for babesiosis (r=0.415, P=0.035), urea (r=-0.488, P=0.034) for
dirofilariosis, and leukocyte count (r=-0.820, P=0.046) for pyometra. Although
elevated MCHC levels in all samples from infected dogs indicate hemolysis,
RBCOF was elevated only in dirofilariosis, pyometra, and canine distemper,
leading to the conclusion that these apparently related phenomena are controlled
by different mechanisms.

Citologija u svakodnevnoj kliničkoj praksi (limfni čvorovi, kutane i subkutane mase)

AB  - Tankoigledna biopsija je sigurna i brza metoda koja je laka za izođenje.
Preporučuje se kao dijagnostička metoda za sve uvećane limfne čvorove, bilo da
se radi o uni- ili multicentričnim promenama kao i za sve kutane i subkutane
mase. Osim tankoiglene biopsije, po ekstirpaciji limfnog čvora, sa njegovog
preseka se može uraditi preparat – otisak.
Citološkim pregledom limfnog čvora se može utvrditi da li postoji
reaktivna hiperplazija, inflamatorne promena u tipu limfadenitisa, primarna
neoplazija limfnog čvora, infiltracija metastatskim tumorskim ćelijama ili
ekstramedularna hematopoeza.
Svaka od navedenih patoloških promena ima određene citološke
karakteristike. Preparat se pregleda na malom uveličanju da bi se uočila
celularnost, raspored ćelija, eventualne metastatske ćelije i optimalno mesto za
pregled. Potrebno je prepoznati tip pozadinskog (engl. background) materijala
primenjujući prvo malo, a potom i veliko uveličanje. To je materijal koji ne
predstavlja patološke ćelije ispitivanog tkiva/mase, već eritrocite koji dospevaju
na preparat oštećenjem krvnih sudova prilikom pravljenja preparata i/ili
ekstracelularni matriks, i/ili specifičan ekstraćelijski materijal koji može biti
tipičan za određeno tkivo i patološku promenu. Na primer, limfoglandularna
telašca predstavljaju delimično razložene delove apoptotičnih limfocita,
karakterističnog su izgleda i javljaju se u većem broju kod limfoma. Potom,
citološki nalaz uključuje prepoznavanje i opis tipova ćelija koje se vide, njihovu
morfologiju i međusobni odnos.
Osnovna pravila prilikom primene tankoiglene biopsije u pravljenju i
tumačenju citoloških preparata limfnih čvorova su sledeća: treba izbegavati da se
analiziraju submandibularni limfni čvorovi koji vrlo često imaju reaktivne promene. Ukoliko postoji generalizovana limfadenopatija treba uzeti uzorke iz
najmanje dva ili svih limfnih čvorova. Igla promera 22G treba da bude plasirana u
nekoliko pravaca i tangencionalno u odnosu na centar limfnog čvora. Kao
problem se može javiti aspiracija tkiva mimo limfnog čvora, a u
submandibularnom regionu su to često pljuvačne žlezde. Takođe, limitacija
citološke dijagnostike postoji kod sitnoćelijskih limfoma mačaka kada su limfociti
dobro diferencirani ili u slučaju fokalne distribucije metastaza. Stepen slaganja u
dijagnostici i prognozi citoloških interpretacija u odnosu na patohistološke zavisi
od tipa limfoma ili metastatske promene.
Kožni ili potkožni noduli su relativno česti problemi kod pasa i mačaka i
mogu imati različitu etiologiju. Dijagnostički pristup se sastoji od uzimanja
detaljne istorije bolesti, pre svega da li je promena nastala naglo ili je prisutna
duži vremenski period. Brzina rasta je takodje od velike važnosti, kao i sam
klinički izgled promene, kao i da li su dobro ograničene. Dubina lezije takođe
može biti od pomoći za izradu diferencijalne liste i dijagnostičkog plana. Ponovno
pojavljanje lezije u oblastima gde je izvršena operacija najčešće ukazuje na
neoplastičnu tvorevinu. Kod starijih pasa česte su potpuno benigne izrasline sa
karakterističnim izgledom. Pozitivan odgovor na antibiotike često ukazuje na
infektivnu etiologiju, ali ne isključuje mogućnost istovremenih bolesti kao što su
dermatomikoze ili tumori.
Prva diferencijacija u proceni nodula je da se utvrdi ili isključi postojanje
neoplastičnih promena za razliku od inflamatornih uzroka ili hiperplazije tkiva. U
te svrhe se koristi tankoiglena biopsija, kako bi se identifikovale prisutne ćelije. U
velikom broju slučajeva kliničar na osnovu mikroskopskog pregleda pripremljenih
pločica, može da proceni da li je u pitanju neoplazija, hiperplazija ili inflamatorni
proces. Na osnovu toga ili preuzima adekvatnu terapiju ili sprovodi dodatne
dijagnostičke postupke.
Sveobuhvatno gledano, sa pojedinim izuzecima, citologija predstavlja
brzu, jednostavnu i pouzdanu metodu za dijagnostiku inflamatornih i
neoplastičnih promena limfnih čvorova, kao i nodula na koži.

AB  - Oxidative stress plays an important role in pathogenesis of idiopathic epilepsy (IE). Although IE is the most
common neurological condition, oxidant–antioxidant status in epileptic dogs is still unknown. The aim of this
study is to evaluate the serum oxidant-antioxidant status in dogs with newly diagnosed IE. The status in 15 dogs
with IE and 15 healthy dogs is estimated through spectrophotometric determination of two oxidant markers:
advanced oxidation protein products-albumin index (AOPP) and thiobarbituric acid reactive substances
(TBARS); and three antioxidant markers: total thiols (R-SH) level, glutathione (GSH) level, and paraoxonase-1
(PON-1) activity. Also, butyrylcholinesterase (BChE) activity is assessed in both groups of dogs. Higher AOPP
is observed in the dogs with newly diagnosed IE, while TBARS level shows no difference when compared to the
healthy dogs. In contrast, lower levels of antioxidants (R-SH, GSH, and PON-1) and BChE activity are found in the
dogs with IE. No significant differences are observed in the oxidant and antioxidant markers and BChE activity
across the investigated IE cases with focal and generalized seizures. Our findings provide evidence that dogs with
IE are characterized by an impaired serum oxidant–antioxidant balance and lower BChE activity, which may
contribute to a better understanding of IE pathogenesis.

The NLR was calculated as the absolute count of neutrophils divided by the absolute
count of lymphocytes, PLR by calculating the absolute platelet divided by the absolute
lymphocyte count, and MPV/PLT by dividing mean platelet volume by absolute platelet
count.
The NLR, MPV, and MPV/PLT ratios were higher in cats with non-septic effusions
suspected to be FIP, but PLR did not differ, when compared to healthy cats. Correlation analysis did not show any association between the selected ratios and hematological and
biochemical parameters.
In the absence of leukocytosis, increased NLR could help us to confirm the presence
of systemic inflammation in cats with non-septic effusions indicative of FIP. However,
a high MPV/PLT ratio should be interpreted with caution, especially in cats.

AB  - The highest number of acute Babesia canis cases in dogs is recorded over the February-May (Feb-May) period, which also represents the optimal climate conditions for tick activity in Belgrade, Serbia. A possibility that the acute phase response is more intense in dogs developing the disease in the Feb-May period compared with the response in other time periods of the year was tested. A total of 63 client-owned dogs with acute B. canis infection were enrolled and the routine hematology and biochemistry parameters-serum amyloid A (SAA), IgG against B. canis, level of parasitemia, ceruloplasmin (CER), paraoxonase-1 (PON-1), and fibrinogen-were measured. Acute phase indexes (API) were calculated as (SAA×CER)/(Iron×PON-1) and (SAA×CER)/(Albumin×Iron). Statistics included Kruskal-Wallis test and logistic regression analysis. The results showed that in the Feb-May period, the following parameters were lower: creatinine, albumin, iron, and level of parasitemia. Furthermore, increased API values were more probable in the Feb-May than in the other periods. Together, higher acute phase response intensity and presumptive hemodilution in the Feb-May period indicate a more acute course of B. canis infection than in other time periods of the year.

Novi laboratorijski parametri dijagnostike poremećaja funkcije bubrega kod pasa i mačaka

AB  - Procenjuje se da prevalenca bubrežnih bolesti u ukupnoj populaciji pasa i mačka iznosi oko 0,5% do 3%, ali da u gerijatrijskoj populaciji prevalenca hronične bubrežne bolesti dostiže čak 80%. Osim
rasne predispozije i starosti, kao faktori rizika navode se hiperkalcemija, bolesti srca, cistitis, urolitijaza, hipertireoidizam, periodontalni i
metabolički poremećaji i hronične ili perzistentne infekcije. Stalni monitoring funkcije bubrega je neophodan da bi se uvela odgovarajuća
terapija, koja može značajno da uspori progresivan tok gubitka funkcije bubrega koji prati bubrežne bolesti. Klasični markeri gubitka funkcije bubrega su koncentracija uree i kreatinina u serumu. Njihova koncentracija raste tek kada je rezervni kapacitet bubrega potrošen, odnosno kada propadne 75% svih nefrona. Na radionici će biti objašnjen
i demonstriran značaj 1) simetričnog dimetilarginina (SDMA) kao novog, osetljivijeg markera premećaja funkcije bubrega, potom 2) značaj
određivanja odnosa proteina i kreatinina u mokraći, kao i 3) potencijal
primene elektroforeze na gelu poliakrilamida za detekciju proteina u
urinu sa ciljem diferencijalne dijagnostike poremećaja funkcije glomerula, odnosno tubula.
AB  - It is estimated that the prevalence of kidney disease in the total population of dogs
and cats is about 0.5% to 3%, but in the geriatric population the prevalence of chronic
kidney disease reaches as much as 80%. In addition to breed predisposition and age,
hypercalcemia, heart disease, periodontal problems, cystitis, urolithiasis, hyperthyroidism,
metabolic disorders, and chronic or persistent infections are listed as risk factors.
Continuous monitoring of the renal function is important for timely and appropriate therapy
managment. Appropriate therapy can significantly slow down the progressive course of
renal disease. The classic markers to test the loss of kidney function are concentration

i demonstriran značaj 1) simetričnog dimetilarginina (SDMA) kao novog, osetljivijeg markera premećaja funkcije bubrega, potom 2) značaj
određivanja odnosa proteina i kreatinina u mokraći, kao i 3) potencijal
primene elektroforeze na gelu poliakrilamida za detekciju proteina u
urinu sa ciljem diferencijalne dijagnostike poremećaja funkcije glomerula, odnosno tubula., It is estimated that the prevalence of kidney disease in the total population of dogs
and cats is about 0.5% to 3%, but in the geriatric population the prevalence of chronic
kidney disease reaches as much as 80%. In addition to breed predisposition and age,
hypercalcemia, heart disease, periodontal problems, cystitis, urolithiasis, hyperthyroidism,
metabolic disorders, and chronic or persistent infections are listed as risk factors.
Continuous monitoring of the renal function is important for timely and appropriate therapy
managment. Appropriate therapy can significantly slow down the progressive course of
renal disease. The classic markers to test the loss of kidney function are concentration
of urea and creatinine in the serum. Their concentration increases only when the reserve

Indexes are the highest in dogs infected in “high season”. These findings are
accompanied by relatively low creatinine and urea, implying that dogs in “high
season” had hemodilution, thus further suggesting short incubation. Unlike other
acute-phase proteins, fibrinogen showed the lowest concentration in “high
season”. Conclusion: Future research will help unveil the connection between
more intense APR and relatively low parasite load and fibrinogen concentration.

The epidemiological status concerning many canine tick-borne diseases (TBDs) in Serbia is still insufficiently known.

Objectives
Our study aimed to investigate the presence of tick-borne pathogens of the family Anaplasmataceae and Hepatozoon spp., as a cause of illnesses accompanied by clinical signs that can occur in dogs with anaplasmosis, ehrlichiosis and hepatozoonosis.

Methods
Dogs are included in the study based on the presence of a minimum of three clinical and/or pathological findings that could be associated with anaplasmosis, ehrlichiosis and hepatozoonosis. During the study (April–October 2018), 11 dogs met the conditions to be included in the survey. Identification of the causative agent in the blood of diseased dogs was performed by conventional PCR followed by sequencing.

Results
The presence of the pathogens was confirmed in three animals (3/11, 27.3%). The presence of Ehrlichia canis was confirmed in 3-month-old female Rottweiler puppy, an 8-year old Miniature Schnauzer female was positive for Hepatozoon canis infection, while 4-year-old mixed breed male dog was co-infected with both mentioned pathogens. These are the first cases of autochthonous infection with E. canis and H. canis in dogs from Serbia confirmed by molecular methods.

Conclusions
The results of our study indicate the importance of molecular methods to establish a reliable diagnosis of TBDs. Also, the confirmed presence of causative agents of canine monocytic ehrlichiosis and hepatozoonosis in Serbia appeals to veterinary practitioners that it is necessary to exclude the presence of those diseases in suspicious dogs.

AB  - Inflamatorni i degenerativni procesi na zglobovima konja se mogu postepeno razvijati,
pri čemu klinički znaci patoloških promena često nisu vidljivi. Cilj ove studije je
bio da se na osnovu rutinskog pregleda sinovijalne tečnosti (ST) poreklom iz metakarpo/
metatarzofalangealnih (MCP/MTP) zglobova radnih konja, koji su bili upućeni na
klanje i njenih biohemijskih karakteristika utvrdi da li je postojala lokalna inflamacija.
Studija je sprovedena na 6 konja starosti 3 do 12 godina (medijana 8), odnosno 24
MCP/MTP zgloba. Post mortem, iz svakog zgloba je uzorkovana ST. Kliničkim pregledom
pre klanja, nisu uočene promene u izgledu i funkciji mišićno-skeletnog sistema.
Urađen je rutinski pregled ST kojim je opisana boja, prozirnost, viskoznost i broj ćelija.
Određena je koncentracija sledećih biohemijskih parametara: ukupnih proteina,
albumina, aktivnost alkalne fosfataze (ALP), aspartat aminotransferaze (AST), kreatin
kinaze (CK), kao i koncentracije haptoglobina (Hp) i ceruloplazmina (Cp). Kod svih
uzoraka, ST je bila bistra, providna, viskozna i blede do bledo žute boje. Najmlađi konj,
star tri godine, je u sva četiri pregledana zgloba imao ukupan broj ćelija 200-890/μL,
što odgovara fiziološkim vrednostima. Dva konja su imala povećan broj ćelija na jednom,
odnosno dva zgloba (1 400-2 560/μL), dok su tri konja imala promene na sva četiri
zgloba (1 100-2 200/μL). Ispitivani biohemijski parametri se nisu razlikovali u uzorcima
ST iz zglobova sa ukupnim brojem ćelija u okviru fizioloških vrednosti i onih kod
kojih je broj ćelija bio povećan. Pozitivne, umerene korelacije su postojale između koncentracije
proteina i aktivnosti ALP, koncentracije proteina i albumina, aktivnosti AST
i CK. Ispitivanjem ST radnih konja, dokazano je da inflamatorne promene u MCP/MTP
zglobovima nisu prisutne. Nalaz povećanog broja ćelija u ST bez inflamatornih promena
zahteva dodatna ispitivanja.

AB  - Hemolysis and systemic acute inflammation characterize canine babesiosis caused by the intraerythrocytic protozoan parasite Babesia canis. Our hypothesis was that blood redox homeostasis of patients that suffered acute B. canis infection might be disturbed even after treatment with imidocarb-dipropionate and successful clinical recovery. Eight owner dogs with acute B. canis infection were used for this study. We analyzed the complete blood count, acute phase proteins (ceruloplasmin, haptoglobin, paraoxonase-1) in the serum, antioxidant enzymes (catalase and glutathione peroxidase) in the erythrocytes, and oxidative stress markers (malondialdehyde in erythrocytes and thiol groups in serum) at presentation and 15 days after treatment. Results were evaluated by corresponding statistical tests. At presentation, anemia, low/normal leukocyte count and severe thrombocytopenia occurred together with increased ceruloplasmin, haptoglobin levels within the reference interval, decreased paraoxonase-1 and compromised antioxidant defense in the red blood cells. After treatment and successful clinical recovery, hematological values generally fitted within the reference intervals, acute phase proteins were within the physiological levels in the majority of cases and the activities of the antioxidant enzymes were increased. However, elevated malondialdehyde levels indicated increased oxidative damage of erythrocytes that remained as a deleterious sequel despite a successful clinical recovery of the dogs.
AB  - Babezioza pasa, koju izaziva protozoa Babesia canis, praćena je hemolizom i sistemskom akutnom infl amacijom. Naša hipoteza je da kod pasa sa akutnom B. canis infekcijom, čak i nakon terapije imidokarb-dipropionatom i uspešnog kliničkog oporavka, postoji poremećaj redoks homeostaze. Studija uključuje osam vlasničkih pasa akutno infi ciranih babezijom. Analizirani su kompletna krvna slika, proteini akutne faze (ceruloplazmin, haptoglobin i paraoksonaza-1) u serumu, antioksidativni enzimi (katalaza i glutation peroksidaza) u eritrocitima i markeri oksidativnog stresa (malondialdehid u eritrocitima, tiolne grupe u serumu) na prezentaciji i 15 dana nakon terapije. Za procenu značajnosti rezultata primenjeni su odgovarajući statistički testovi. Anemija, nizak/normalan broj leukocita i teška trombocitopenija praćeni povišenom koncentracijom ceruloplazmina, haptoglobinom u fi ziološkim intervalima i sniženim nivoom paraoksonaze-1, kao i kompromitovana antioksidativna zaštita, uočeni su na prezentaciji. Nakon terapije i uspešnog kliničkog oporavka, hematološke vrednosti su bile u fi ziološkom opsegu, kao i proteini akutne faze kod većine pacijenata, a aktivnost antioksidativnih enzima je pojačana. Međutim, povišena koncentracija malondialdehida ukazuje na povećano oštećenje eritrocita koje je zaostalo uprkos uspešnom kliničkom oporavku pasa.

Nakon terapije i uspešnog kliničkog oporavka, hematološke vrednosti su bile u fi ziološkom opsegu, kao i proteini akutne faze kod većine pacijenata, a aktivnost antioksidativnih enzima je pojačana. AB  - The common signs of canine babesiosis caused by an infection with Babesia canis are fever, anorexia, lethargy, pulse alterations, anemia, and occasionally mild icterus. Dogs with these clinical signs can be divided into two groups: those with acute-phase reaction and those with systemic inflammatory response syndrome (SIRS). Factors associated with the occurrence of SIRS in canine babesiosis have not been thoroughly researched. This article outlines a cross-sectional study of 54 client-owned dogs with an acute B. canis infection, and evaluates the differences in age, gender, laboratory findings, parasite load, and seroreactivity against B. canis between the SIRS and the SIRS-free dogs. We have analyzed a complete blood count, serum biochemistry, serum amyloid A, ceruloplasmin, paraoxonase-1, serology, and PCR testing using standard methodologies. The frequency of SIRS among the investigated dogs reached 0.59. Male dogs and those seronegative against B. canis, were more frequent in the SIRS group, whilst age and parasite load could not be associated with the presence of SIRS. Dogs with SIRS had a lower count of total leukocytes, neutrophils, lymphocytes, and monocytes, and a lower concentration of iron and bilirubin compared with SIRS-free dogs. No significant differences in the concentration of acute-phase proteins have been noticed to exist between the groups of dogs. Further, the seronegative dogs had a lower count of lymphocytes and monocytes and a higher parasite load than the seroreactive dogs. Multivariate logistic regression analysis has identified leukopenia (&lt;6 × 109/L) and monocytopenia (&lt;0.2 × 109/L) as independent associates of SIRS in the investigated dogs, thus implying that these routine tests could be used as reliable markers for SIRS.

Low serum levels of promatrix metalloproteinase-2 and -9 occur during acute Babesia canis infection in dogs

AB  - Inflammation is a hallmark of the acute Babesia canis infection. Promatrix metalloproteinase (proMMP)-2 and -9 are involved in inflammation, but their levels have not been analyzed in canine babesiosis. We hypothesized that in dogs infected with B. canis, serum proMMP-2 and -9 levels change between presentation and recovery. Degree of the change differs if dogs develop systemic inflammatory response syndrome (SIRS). This study included 24 dogs with an acute B. canis infection, at presentation and after two weeks. We used routine hematology and biochemistry methods, spectrophotometry for the acute-phase proteins, microscopy for parasitemia and zymography for (pro)MMPs. In vitro endothelial cells and leukocyte short-term cultures, and platelet lysates were used to detect specific MMP activity. Statistical analyses included Wilcoxon test for paired samples, Mann-Whitney U test and Spearman’s rank correlation. Our results showed that endothelial cells, leukocytes and platelets are the source of proMMP-2 and proMMP-9. Furthermore, both proMMPs were lower at presentation than after recovery (p < 0.001). At presentation, proMMP-9 levels correlated with parasitemia (rho = -0.616, p = 0.009), total leukocyte (rho = 0.704, p < 0.001) and neutrophil counts (rho = 0.741, p < 0.001). Extent of alterations in proMMP-2 levels between presentation and recovery was lower (p = 0.038) in dogs with SIRS than in non-SIRS dogs, while levels of proMMP-9 were comparable between these groups. Our conclusion is that during the acute B. canis infection, low serum levels of proMMP-2 and proMMP-9 at presentation reflect thrombocytopenia and leukopenia. Decreased proMMP-2 level could be associated with SIRS.

AB  - Mоноцитна ерлихиоза паса (МЕП) једна је од важнијих крпељима преносивих
болести у јужној Европи. Главни узрочник је грам негативнa бактерије Ehrlichia
canis, која у цитоплазми инфицираних леукоцита формира структуре зване моруле.
У Европи се смеђи псећи крпељ - Rhipicephalus sanguineus сматра главним
вектором. Различите фазе и вишеструке клиничке манифестације МЕП, као и
потенцијалне коинфекције другим патогенима који се преносе крпељима (Babesia
spp., Hepatozoon canis, Anaplasma phagocytophilum, A. platys) компликују
постављање поуздане дијагнозе. Иако су хематолошке, серолошке и цитолошке
дијагностичке технике од велике важности за МЕП, молекуларне методе (PCR,
qPCR) су често неопходне за коначну дијагнозу.
Тромесечно власничко женско штене ротвајлера из околине Београда без историје
путовања у ендемскa подручјa, доведено је на клинику Ветеринарског факултета у
Београду због летаргије, анорексије и генерализоване лифаденопатије. Девет дана
пре довођења на преглед извађен је крпељ. Хематолошке анализе потврдиле су
присуство нормоцитно нормохромне анемије, лимфоцитозе и тромбоцитопеније.
На крвном размазу и размазу издвојених леукоцита (buffy coat) уочено је присуство
гранулираних лимфоцита. Брзи антитело тест (Bionote, Korea) указао на
изложеност Е. canis. Да би се потврдила дијагноза, изолована је укупна ДНК из
ЕДТА крви и изведен је конвенционални PCR праћен секвенцирањем.
Упоређивањем добијене секвенце са секвенцама депонованим у GenBank®,
потврђено је присуство E. canis. Иако серолошке методе могу указивати на
изложеност E. canis (индиректна имунофлуоресценција- ИФА, ЕЛИСА тест), PCR
са великом специфичности и осетљивости је од суштинског значаја за детекцију
ДНК E. canis и овај налаз указује на активну инфекцију. Поред тога, детекција ДНК
је могућа и пре сероконверзије. Недостатак серолошких испитивања такође је и
појава унакрсне реактивности E. canis са другим врстама из породице
Anaplasmacataceae (E. chaffeensis, E. ewingii, A. phagocytophilum). Описани случај
је прва молекуларна потврда МЕП и присуства E.canis у Србији и указује на
важност примене молекуларних метода у циљу успостављања поуздане дијагнозе
код крпељима преносивих болести паса.
AB  - Canine monocytic ehrlichiosis (CME) is one of the major tick-borne diseases in
southern Europe. The main causative agent is gram-negative bacteria Ehrlichia canis,
which in the cytoplasm of infected leukocytes forming structures called morulae. In
Europe, brown dog tick Rhipicephalus sanguineus, is considered as the main vector.
Different phases and multiple clinical manifestations of CME, as well as potential coinfections with other tick-borne pathogens (Babesia spp., Hepatozoon canis,
Anaplasma phagocytophilum, A. platys) complicate reliable diagnosis. Even
haematological, serological and cytological diagnostic techniques are of great
importance for CME, molecular methods (PCR, qPCR) are often necessary for a
definitive diagnosis.
Proprietary, three months old female Rottweiler puppy from the vicinity of Belgrade
city without a history of travel to the endemic area was brought to the clinic of the
Faculty of Veterinary Medicine in Belgrade due to the presence of lethargy, anorexia
and generalized lymphadenopathy. Nine days before presentation tick was removed
from the dog. Haematological analyses showed the presence of normocytic
normochromic anemia, lymphocytosis and thrombocytopenia. On blood and buffy
coat smears granular lymphocytes were noted. Antibody SNAP test (Bionote, Korea)
indicated exposure to E. canis. To confirm the diagnosis, total DNA was extracted
from EDTA blood and conventional PCR followed by sequencing was performed. By
comparing the obtained sequence with the sequences deposited in GenBank®, the
presence of E. canis was confirmed.
Although serological methods may indicate exposure to E. canis (indirect fluorescent
antibody- IFA, enzyme-linked immunosorbent assays- ELISA), PCR with high
specificity and sensitivity is essential to detect DNK of E. canis indicating active
infection. Also, DNA detection is possible before seroconversion. The ambiguity of
serological assays may be also the appearance of cross-reactivity among E. canis and
other species of the Anaplasmacataceae family (E. chaffeensis, E. ewingii, A.
phagocytophilum). The described case was the first molecular confirmation of CME
and E. canis in Serbia, pointing to the importance of molecular methods in reliable
diagnosis.

са великом специфичности и осетљивости је од суштинског значаја за детекцију
ДНК E. canis и овај налаз указује на активну инфекцију. Поред тога, детекција ДНК
је могућа и пре сероконверзије. Недостатак серолошких испитивања такође је и
појава унакрсне реактивности E. canis са другим врстама из породице
Anaplasmacataceae (E. chaffeensis, E. ewingii, A. phagocytophilum). Описани случај
је прва молекуларна потврда МЕП и присуства E.canis у Србији и указује на
важност примене молекуларних метода у циљу успостављања поуздане дијагнозе
код крпељима преносивих болести паса., Canine monocytic ehrlichiosis (CME) is one of the major tick-borne diseases in
southern Europe. The main causative agent is gram-negative bacteria Ehrlichia canis,
which in the cytoplasm of infected leukocytes forming structures called morulae. In
Europe, brown dog tick Rhipicephalus sanguineus, is considered as the main vector.
Different phases and multiple clinical manifestations of CME, as well as potential coinfections with other tick-borne pathogens (Babesia spp., Hepatozoon canis,
Anaplasma phagocytophilum, A. platys) complicate reliable diagnosis. Even
haematological, serological and cytological diagnostic techniques are of great
importance for CME, molecular methods (PCR, qPCR) are often necessary for a
definitive diagnosis.
Proprietary, three months old female Rottweiler puppy from the vicinity of Belgrade
city without a history of travel to the endemic area was brought to the clinic of the
Faculty of Veterinary Medicine in Belgrade due to the presence of lethargy, anorexia
and generalized lymphadenopathy. Nine days before presentation tick was removed
from the dog. Haematological analyses showed the presence of normocytic
normochromic anemia, lymphocytosis and thrombocytopenia. On blood and buffy
coat smears granular lymphocytes were noted. Antibody SNAP test (Bionote, Korea)
indicated exposure to E. canis. To confirm the diagnosis, total DNA was extracted
from EDTA blood and conventional PCR followed by sequencing was performed. By
comparing the obtained sequence with the sequences deposited in GenBank®, the
presence of E. canis was confirmed.
Although serological methods may indicate exposure to E. canis (indirect fluorescent
antibody- IFA, enzyme-linked immunosorbent assays- ELISA), PCR with high
specificity and sensitivity is essential to detect DNK of E. canis indicating active
infection. Also, DNA detection is possible before seroconversion. The ambiguity of
serological assays may be also the appearance of cross-reactivity among E. canis and
other species of the Anaplasmacataceae family (E. chaffeensis, E. ewingii, A.
phagocytophilum). The described case was the first molecular confirmation of CME
and E. canis in Serbia, pointing to the importance of molecular methods in reliable
diagnosis.",

Lung adenocarcinoma as a cause of spontaneous pneumothorax in a cat

AB  - A 15-year-old indoor/outdoor female, neutered, domestic European short haired cat was presented to our clinic with a history of neck-elongation and wheezing during inspiration/expiration. The first signs of respiratory disease occurred two weeks before the clinical presentation. After taking of clinical history, clinical examination and basic laboratory tests (complete blood count, blood smear, snap Feline Immunodeficiency Virus/Feline Leukemia Virus Combo test), a lung x-ray was performed showing the presence of left lung atelectasis and pneumothorax. The primary causes of lung atelectasis were not determined. Given that the general condition of the cat was very bad, she was put in an oxygen cage, thoracocentesis was performed, but the animal’s condition did not improve, and 24 hours after clinical presentation, the cat died. Autopsy revealed the presence of a tumor mass on the caudal lobe of the right lung, tumor formation in the area of trachea bifurcation and changes in the liver in the form of four round structures of different sizes. Histopathologically, the existence of primary undifferentiated lung adenocarcinoma with metastases on the liver and trachea was determined.
AB  - U ovom radu predstavljen je slučaj primarnog adenokarcinoma pluća kod domaće
kratkodlake mačke stare 15 godina. Prvi znaci oboljenja respiratornog sistema pojavili
su se dve nedelje pre dovođenja mačke na pregled, pri čemu su dominirali znaci disanja
sa ispruženim vratom i šištanje tokom inspirijuma i ekspirijuma. Nakon uzimanja istorije
bolesti, kliničkog i laboratorijskog ispitivanja (krvna slika, krvni razmaz, FIV/FeLV
brzi dijagnostički test) urađen je rendgen pluća koji je pokazao postojanje atelektaze
levog plućnog krila i pneumotoraks. Nije utvrđeno koji patofiziološki mehanizam je
odgovoran za atelektazu pluća. S obzirom da je opšte stanje mačke bilo jako loše,
stavljena je u kiseonički kavez, urađena je torakocenteza ali stanje se nije popravilo i
24h od dolaska na kliniku mačka je uginula. Na obdukciji je utvrđeno odstupanje od
normalnog nalaza na levom plućnom krilu, postojanje promena na desnom plućnom
krilu, tumorozna tvorevina u predelu bifurkacije traheje i promene na jetri u vidu četiri
okrugle tvorevine različite veličine. Histopatološki je utvrđeno postojanje primarnog
nediferentovanog adenokarcinoma pluća sa metastazama na već pomenutim organima.
