AB  - Svrha ovog pregleda je da prikaže veliku raznolikost u strukturi i funkciji placente koja se sreće kod sisara, i podobnosti nekih vrsta kao animalnih modela u proučavanju placentacije. Postoje značajne razlike u strukturi i fiziologiji posteljice između čoveka, domaćih i eksperimentalnih životinja. Zbog toga, najčešće upotrebljavani animalni modeli za proučavanje placentacije (miš, kunić, zamorac), nisu adekvatni za sva istraživanja u veterinarskoj i humanoj medicini. Placente se klasifikuju prema svom obliku, strukturi i interhemalnom regionu. Horionske resice placente su obložene prizmatičnim epitelom i preko njega se odvija prenos svih supstanci od majke prema plodu. Ćelije ovog epitela su selektivno propustljive kako za korisne, tako i za potencijalno toksične materije.
	Istraživanja koja se bave analizama i osobenostima placentacije su oskudna, posebno sa aspekta kontakta i razmene materija između majke i ploda. Iz tog razloga, ovaj uvid može da pomogne u sagledavanju različitih problema u kliničkoj praksi i tretmanu koji su bitni za dobijanje zdravog potomstva.

AB  - Biocompatibility of materials is one of the most important conditions for their successful application in tissue regeneration
and repair. Cell-surface interactions stimulate adhesion and activation of macrophages whose acquaintance can assist in
designing novel biomaterials that promote favorable macrophage–biomaterial surface interactions for clinical application.
This study is designed to determine the distribution and number of macrophages as a means of biocompatibility evaluation
of two newly synthesized materials [silver/poly(vinyl alcohol) (Ag/PVA) and silver/poly(vinyl alcohol)/graphene (Ag/PVA/
Gr) nanocomposite hydrogels] in vivo, with approval of the Ethics Committee of the Faculty of Veterinary Medicine,
University of Belgrade. Macrophages and giant cells were analyzed in tissue sections stained by routine H&E and im munohistochemical methods (CD68+
). Statistical relevance was determined in the statistical software package SPSS 20
(IBM corp). The results of the study in terms of the number of giant cells localized around the implant showed that their
number was highest on the seventh postoperative day (p.o.d.) in the group implanted with Ag/PVA hydrogels, and on the
30th p.o.d. in the group implanted with Ag/PVA/Gr. Interestingly, the number of macrophages measured in the capsular and
pericapsular space was highest in the group implanted with the commercial Suprasorb© material. The increased mac rophage number, registered around the Ag/PVA/Gr implant on 60th p.o.d. indicates that the addition of graphene can, in a
specific way, modulate different biological responses of tissues in the process of wound healing, regeneration, and
integration.

AB  - Babezioza pasa, uzrokovana infekcijom protozoom Babesia canis, sezonska je bolest na području Beograda, glavnog grada Republike Srbije. Sezone pojave oboljenja se poklapaju sa aktivnošću vektora krpelja Dermacentor reticulatus. Period inkubacije varira od četiri do 21 dan, a kliničku sliku karakteriše sistemska akutna inflamacija ili u težim slučajevima sindrom sistemskog inflamatornog odgovora (eng. Systemic Inflammatory Response Syndrome – SIRS). Povezanost klimatskih i epidemioloških faktora koja bi precizno pokazala i okarakterisala sezone u kojima se ovo oboljenje javlja na teritoriji Beograda, do sada nije ispitana. Sa kliničko-epidemiološkog aspekta, nameće se pitanje, da li je pojava SIRS-a vezana za pojedine sezone, odnosno da li postoje razlike u intezitetu odgovora akutne faze kod pasa obolelih u različitim sezonama. Hipoteza ove studije bila je da je tokom prolećne sezone, u kojoj je učestalost pojave pasa akutno inficiranih B. canis najveća, odgovor akutne faze najintenzivniji, odnosno da će reaktanti akutne faze pokazati najveće promene, a da će broj pasa sa SIRS-om, biti najzastupljeniji.Prvi deo studije obuhvatio je ispitivanje učestalosti pojave oboljenja na teritoriji Beograda tokom četiri godine (2013–2016), u različitim sezonama. Ispitan je kratkoročni uticaj (dve nedelje) i dugoročni uticaj meteoroloških parametara (godinu dana) koji su prethodili pojavi 872 zabeležena slučaja babezioze pasa. Drugi deo studije uključio je 66 pasa, sa akutnom infekcijom B. canis tokom 2017–2019. godine, od kojih je prikupljan višak pune krvi i seruma koji su preostali posle dijagnostike oboljenja. Rutinskim laboratorijskim testovima su izvedene hematološke i biohemijske analize, dok je elektroforeza proteina izvedena na agaroznom gelu. Parazitemija je određena komercijalnom metodom lančane reakcije polimeraze, dok je prisustvo IgG antitela protiv B. canis ispitano komercijalnim serološkim testom. Reaktanti/proteini akutne faze određeni su sledećim testovima: koncentracija serum amiloida A komercijalnom ELISA-om (eng. Enzyme Linked Immunosorbent Assay), koncentracija ceruloplazmina i aktivnost paraoksonaze 1 spektrofotometrijski, dok je za koncentraciju fibrinogena primenjen metod zagrevanja uz precipitaciju proteina. Klasifikacija pasa na SIRS pozitivne i negativne izvedena je prema odgovarajućim kliničkim i laboratorijskim promenama, dok su indeksi reaktanata akutne faze (IRAF) dobijeni računskim putem. Razlike između varijabli ispitane su pomoću Kruskal-Wallis, Mann-Whitney U testa i Chi-square testa. Veze između varijabli kvantifikovane su Spirmanovim koeficijentima korelacije odnosno logističkom regresionom analizom.Rezultati analize distribucije pojave babezioze pasa omogućavaju podelu kalendarske godine na četiri sezone: januar–februar, februar–maj, jun–septembar i oktobar–decembar. Broj dijagnostikovanih slučajeva pokazuje bimodalnu distribuciju sa najvećim brojem tokom kasne zime i proleća i nešto manje tokom jeseni. AB  - Canine babesiosis, caused by infection with a protozoan Babesia canis, is a seasonal disease in the area of Belgrade, the capital of the Republic of Serbia. The seasons of the disease coincide with the activity of the vector Dermacentor reticulatus. The disease incubation period varies between 4 and 21 days, and the clinical picture is characterized by systemic acute inflammation or, in severe cases, Systemic Inflammatory Response Syndrome (SIRS). A connection between meteorological parameters and epidemiological factors that could help characterize the seasons in which this disease occurs in the region of Belgrade, has not been investigated previously. From the clinical-epidemiological aspect, there is a question whether the occurrence of SIRS is restricted to some seasons, or, in other words, whether there are differences in the intensity of the acute phase response in dogs infected in different seasons. This study hypothesized that during the spring season, when the frequency of dogs acutely infected with B. canis is the highest, the acute phase response is also the most intense, with acute-phase reactants showing the greatest changes, and the fraction of dogs with SIRS reaching its maximum.The first part of the study investigated the disease frequency in the Belgrade region in different seasons during four years (2013–2016) when a total of 872 were recorded. A short-term influence (two weeks prior) and long-term influence of meteorological parameters (up to one year prior) were analyzed.The second part of the study included 66 dogs with acute B. canis infection diagnosed between 2017 and 2019. Laboratory analyses were conducted using the excess of the whole blood and serum that remained after the diagnosis of the disease. Hematological and biochemical analyses were performed by routine laboratory tests, while agarose gel was used for electrophoretic analysis of proteins. Parasitemia was measured by a polymerase chain reaction method in a commercial laboratory. The presence of IgG antibodies against B. canis was tested by a commercial serological test. The following techniques were used for analyses of acute-phase proteins: commercial Enzyme Linked Immunosorbent Assay for serum amyloid A concentration, spectrophotometry for ceruloplasmin concentration and paraoxonase 1 activity, and heat denaturation method for fibrinogen. The dogs were classified as SIRS positive or negative according to a predefined set of clinical and laboratory changes. The indexes of acute phase reactants (IRAF) were calculated. The significance of the differences between the variables was tested using the Kruskal-Wallis, Mann-Whitney U, and Chi-square tests. The relationships between variables were quantified by Spearman correlation coefficients and logistic regression analyses.The number of diagnosed canine babesiosis cases showed a bimodal distribution wherein four seasons of the year can be distinguished: January–February, February–May, June–September and October–December. The highest number of the cases was recorded during late winter and spring, i.e., in the February–May season, and the other, less prominent maximum, occurred during autumn. AB  - U septembru 2018. godine, štene staro četiri meseca, sa znacima groznice i petehijalnim krvarenjem, je došlo u ambulantu Klinike za male životinje Fakulteta veterinarske medicine Univerziteta u Beogradu. Hematološka analiza je pokazala trombocitopeniju i blagu anemiju. Pregledom krvnog razmaza ustanovljene su inkluzije u trombocitima. Komercijalni serološki test je pokazao prisustvo antitela protiv Anaplasma spp. Sprovedena je terapija doksiciklinom u trajanju od 14 dana, a posle 48 sati od početka terapije znaci bolesti su se povukli. PCR analizom i senkvencioniranjem je potvrđeno prisustvo Anaplasma platys.

Analiza promena na hrskavici i subhondralnoj kosti kiĉiĉnog zgloba radnog konja - radiološki i histološki pristup

AB  - Radni konji se u Srbiji koriste kao tovarne životinje koje nose/vuku tovar, često teži od njihove telesne mase. Postavili smo hipotezu da opterećenje koje trpe utiče na pojavu lezija na koštano-zglobnom sistemu. Cilj ovog rada je bio da se uporedno prikaže prisustvo i stepen oštećenja zglobne hrskavice (ZH) i subhondralne kosti (SHK), distalnih kondilusa treće metatarzalne kosti kod starijeg radnog konja pomoću makroskopskog pregleda hrskavice, radioloških i histoloških metoda. Na lokalnoj klanici uzorkovani su kičični zglobovi ekstremiteta radnog pastuva, starog 10 godina. Uz pomoć kompjuterizovane tomografije (CT) utvrđene su promene samo na zglobu prednje leve noge. Nakon disekcije označenog zgloba, izvršena je biopsija ZH i SHK metakarpalne kosti u predelu oštećenja, a bioptati su snimljeni mikro CT-om nakon čega su deklacifikovani i ukalupljeni u parafin, i analizirani su preseci. Makroskopski je bilo vidljivo da je lokalizacija oštećenja ZH bila simetrična na medijalnim i lateralnim epikondilusima matakarpalnih i metatarzalnih kostiju svih ekstremiteta. Linije habanja i palmarne artroze bile su slabo do umereno izražene. Na nativnim rendgenogramima zglobova nisu uočene patološke promene. Analizom snimaka načinjenih CT-om uočena je promena u subhondralnoj zoni distalne epifize treće metakarpalne kosti u vidu pukotine, a oko nje je primetna zona skleroze SHK. Na mikro CT snimku uzorka koštanog tkiva uzetog u predelu lateralnog epikondilusa metakarpane kosti, takođe je uočljiva zona subhondralne skleroze. Najmarkatnije histološke promene na ZH su bile: na površini ZH undulacije i fisure, koje se pružaju i do duboke zone, mnogobrojne izogene grupe sa većim brojem hondrocita u lakuni, fokalni gubitak ćelija i mestimičnio prisustvo hipertrofičnih hondrocita. U SHK su primećene: neznatne lezije koštanog matriksa u vidu svetlijeg prebojavanja, blago remodeliranje kosti koje se karakteriše njenim napredovanjem ka kalcifikovanoj hrskavici. Dugotrajno opterećenje utiče na pojavu oštećenja SHK koje se može uočiti uz pomoć CT-a i histološkom analizom, dok se stepen oštećenja ZH može evaluirati makroskopski i histološki.

Lung adenocarcinoma as a cause of spontaneous pneumothorax in a cat

AB  - A 15-year-old indoor/outdoor female, neutered, domestic European short haired cat was presented to our clinic with a history of neck-elongation and wheezing during inspiration/expiration. The first signs of respiratory disease occurred two weeks before the clinical presentation. After taking of clinical history, clinical examination and basic laboratory tests (complete blood count, blood smear, snap Feline Immunodeficiency Virus/Feline Leukemia Virus Combo test), a lung x-ray was performed showing the presence of left lung atelectasis and pneumothorax. The primary causes of lung atelectasis were not determined. Given that the general condition of the cat was very bad, she was put in an oxygen cage, thoracocentesis was performed, but the animal’s condition did not improve, and 24 hours after clinical presentation, the cat died. Autopsy revealed the presence of a tumor mass on the caudal lobe of the right lung, tumor formation in the area of trachea bifurcation and changes in the liver in the form of four round structures of different sizes. Histopathologically, the existence of primary undifferentiated lung adenocarcinoma with metastases on the liver and trachea was determined.
AB  - U ovom radu predstavljen je slučaj primarnog adenokarcinoma pluća kod domaće
kratkodlake mačke stare 15 godina. Prvi znaci oboljenja respiratornog sistema pojavili
su se dve nedelje pre dovođenja mačke na pregled, pri čemu su dominirali znaci disanja
sa ispruženim vratom i šištanje tokom inspirijuma i ekspirijuma. Nakon uzimanja istorije
bolesti, kliničkog i laboratorijskog ispitivanja (krvna slika, krvni razmaz, FIV/FeLV
brzi dijagnostički test) urađen je rendgen pluća koji je pokazao postojanje atelektaze
levog plućnog krila i pneumotoraks. Nije utvrđeno koji patofiziološki mehanizam je
odgovoran za atelektazu pluća. S obzirom da je opšte stanje mačke bilo jako loše,
stavljena je u kiseonički kavez, urađena je torakocenteza ali stanje se nije popravilo i
24h od dolaska na kliniku mačka je uginula. Na obdukciji je utvrđeno odstupanje od
normalnog nalaza na levom plućnom krilu, postojanje promena na desnom plućnom
krilu, tumorozna tvorevina u predelu bifurkacije traheje i promene na jetri u vidu četiri
okrugle tvorevine različite veličine. Histopatološki je utvrđeno postojanje primarnog
nediferentovanog adenokarcinoma pluća sa metastazama na već pomenutim organima.

AB  - Asymptomatic outdoor dogs can be carriers of Babesia canis, but data describing the development of an acute phase response (APR) are not available. We hypothesised that these dogs have a moderate APR that could be detected by hematological and biochemical changes. Two groups of Babesia-exposed dogs were represented by nine B. canis PCR-positive and twenty B. canis PCR-negative, seroreactive dogs. The control group consisted of ten Babesia-naïve dogs. Serum amyloid A (SAA), paraoxonase-1 (PON-1), complete blood count, and biochemistry parameters were analysed by standard methodologies. Protein and lipoprotein fractions were separated using agarose gel electrophoresis (GE), and the dominant diameters of lipoproteins were assessed on gradient GE. Results were evaluated using non-parametric tests and the Receiver Operating Characteristic curve. SAA (median 39.0 μg/mL, range 2.2–48.8 μg/mL), total protein (median 74.7 g/L, range 57.1–98.3 g/L) and the dominant diameter of α-lipoproteins (median 13.31 nm, range 12.09–14.17 nm) in B. canis PCR-positive dogs were higher relative to dogs in the control group or dogs that were PCR-negative but seroreactive (p < 0.001 for both groups). Mild to moderate anemia (4/29), thrombocytopenia (7/29), and leukocyte counts that were close to the upper limit of the reference range were encountered in both Babesia-exposed groups. When compared to controls, Babesia-exposed dogs displayed decreased a PON-1 activity and protein GE pattern consistent with low-grade chronic inflammation (p < 0.001 for both groups). Dogs with detectable amounts of B. canis DNA in blood contain increased levels of SAA and total protein along with α-lipoproteins that display an increased diameter relative to those dogs with positive Babesia serology but undetectable levels of B. canis DNA in blood.

AB  - Haematologic abnormalities possibly associated with exposure to vector-borne pathogens are rarely reported in clinically healthy outdoor dogs. Therefore, we analysed changes in the complete blood count (CBC) of clinically healthy outdoor dogs seroreactive to Anaplasma spp. and Babesia spp., with or without microfilariosis. Stray, shelter and hunting dogs, 81 in total, that were polymerase chain reaction negative for Anaplasma spp. and Babesia spp. were divided into groups according to their seroreactive status and results of a modified Knott’s test: seronegative to both Anaplasma spp. and Babesia spp. SN (N=26); seroreactive to A. phagocythophilum SR-A (N=12); seroreactive to B. canis, B. gibsoni and/or B. vogeli SR-B (N=25); and seroreactive to both of the pathogens SR-AB (N=8). These four groups were negative to microfilariosis, unlike the fifth group, seroreactive to either or to both of the pathogens and with microfilariosis SR-M (N=10). The frequencies of CBC alterations among all analysed dogs were: 0.35 – leucocytosis, 0.44 – granulocytosis, 0.28 – anaemia, 0.74 – microcytosis, 0.37 – increased mean cell haemoglobin concentration (MCHC) and 0.33 – thrombocytopenia. The frequency of alterations did not differ across the groups. An exception was the SR-M group wherein increased MCHC peaked with a frequency of 0.80, while in the 
other four groups, the frequency ranged between 0.10 and 0.50. Clinically healthy outdoor dogs have multiple CBC abnormalities, consistent with stress and low-level chronic inflammation, but not associated with a previous exposure to Anaplasma spp. or Babesia spp. The presence of microfilaria increases haemolysis in vitro.
AB  - Klinički zdravi psi koji borave na otvorenom i koji su izloženi patogenima koje prenose vektori mogu imati hematološke premećaje vezane za dejstvo tih patogena. Ipak, hematološke pretrage se retko izvode na toj populaciji pasa. Stoga je cilj našeg rada bio da utvrdimo promene u krvnoj slici zdravih pasa lutalica, pasa smeštenih u azilima i lovačkih pasa, seroreaktivnih na uzročnike bolesti iz rodova Anaplasma i Babesia, sa ili bez mikrofilarioze. U ovom radu je ispitana krvna slika 81 psa. U studiju su uključeni psi kod kojih lančanom reakcijom polimerizacije nije pokazano prisustvo DNK Anaplasma spp. i Babesia spp. Psi su prema serološkom statusu raspoređeni u sledeće grupe: seronegativni na Anaplasma spp. i Babesia spp. SN (N=26), seroreaktivni na A. phagocytophilum SR-A (N=12), seroreaktivni na B. canis, B. gibsoni i/ili B. vogeli SR-B (N=25) i seroreaktivni na oba navedena patogena SR-AB (N=8). Ove četiri grupe su bile negativne na mikrofilariozu, dok je peta grupa, seroreaktivna na neki ili oba patogena imala i mikrofilarije SR-M (N=10). Učestalost promena kompletne krvne slike među svim ispitivanim psima je bila sledeća: 0,35 za leukocitozu, 0,44 za granulocitozu, 0,28 za anemiju, 0,74 za mikrocitozu, 0,37 za povećanu prosečnu koncentraciju hemoglobina u eritrocitima i 0,33 za trombocitopeniju. Grupe se nisu razlikovale u pogledu učestalosti navedenih promena. Izuzetak je grupa SR-M u kojoj je učestalost povećanja vrednosti za prosečnu koncentraciju hemoglobina u eritrocitima iznosila 0,80. U ostale četiri grupe ta učestalost je iznosila između 0,10 i 0,50. Zaključak je da ispitivani psi imaju višestruke poremećaje krvne slike koji najverovatnije odražavaju stres i nizak stepen hronične inflamacije, ali ne i vezu sa prethodnim kontaktom sa Anaplasma spp. i Babesia spp. Prisustvo mikrofilarija izaziva hemolizu in vitro.

Biocompatibility of silver / poly(vinyl alcohol) / graphen nanocomposite hydrogels in rat subcutaneous tissues for biomedical applications

AB  - This doctoral dissertation was designed in order to test the biocompatibility of two newly synthesized and previously in vitro characterized hydrogels based on poly(vinyl alcohol) polymers - Ag/PVA and Ag/PVA/Gr, which are intended for making wound dressings or soft tissue implants tissue. Poly(vinyl alcohol) (PVA) is a synthetic polymer commonly used in biomedicine due to its non-toxicity, solubility, biocompatibility, elasticity and the ability to produce reversible hydrogels with high sorption capacity. In the last few years, research has also focused on the incorporation of graphene (Gr) into hydrogel PVAs, in order to improve their mechanical strength and durability. In tissue engineering and regenerative medicine, it is particularly important that implants and bandages contain an antiinflammatory and antibacterial active ingredient to protect the sterility of the wound, or to prevent infection in the implant surrounding tissue. It is known that silver nanoparticles (AgNPs) have excellent antibacterial activity and are therefore often used as an active component of PVA-based hydrogel biomaterials. In order to test the biocompatibility of newly synthesized hydrogels, the soft tissue response to deep subcutaneously implanted Ag/PVA and Ag/PVA/Gr implants was compared to the reaction to the commercial calcium-alginate wound pad (Suprasorb ©, Lohmann & Rauscher GmbH & Co. KG, Neuwied, Germany), which is implanted in an identical manner. The tissue irritation index (IrI) was evaluated as the basic measure of tissue response to implanted hydrogels. In order to determine the degree of biocompatibility, the thickness of the connective tissue capsule around the implanted hydrogels, the number of inflammatory cells, including the Langerhans giant cells, the number of macrophages in the perimplant zone and their eneventual migration to the biomaterial, as well as the number of blood vessels around the implantation zone, were measured. The ratio of proliferating and apoptotic cellular skin and subcutaneous tissue cellular entities was observed, due to their responsibility for regeneration and reparation processes...
AB  - Ova doktorska disertacija dizajnirana je u cilju da se ispita biokompatibilnost dva novosintetisana i prethodno in vitro okarakterisana hidrogela na bazi sintetskog polimera polivinil-alkohola − Ag/PVA i Ag/PVA/Gr, koji su namenjeni za izradu obloga za rane ili za implantate za meka tkiva. Polivinil-alkohol (PVA) je sintetički polimer koji se često koristi u biomedicini zbog svoje netoksičnosti, rastvorljivosti, biokompatibilnosti, elastičnosti i sposobnosti stvaranja reverzibilnih hidrogelova s visokom sposobnošću sorpcije. U poslednjih nekoliko godina, istraživanja se takođe fokusiraju na ugradnju grafena (Gr) u hidrogelove PVA kako bi se poboljšala njihova mehanička čvrstoća i trajnost. U inženjerstvu tkiva i regenerativnoj medicini posebno je važno da implantati i zavoji sadrže antiinflamatornu i antibakterijsku aktivnu komponentu kako bi zaštitili sterilnost rane ili sprečili infekciju u tkivu koje okružuje implantat. Poznato je da nanočestice srebra (eng. silver nanoparticles, AgNPs) poseduju odličnu antibakterijsku aktivnost, pa se zbog toga često koriste kao aktivna komponenta biomaterijala na bazi hidrogela od PVA. U cilju ispitivanja biokompatibilnosti novosintetisanih hidrogelova, praćena je reakcija mekog tkiva na duboko potkožno postavljene implantate od Ag/PVA i Ag/PVA/Gr i poređena je sa reakcijom na komercijalnu oblogu za rane na bazi kalcijum-alginata (Suprasorb©, Lohmann & Rauscher GmbH & Co. KG, Neuwied, Germany), koja je implantirana na identičan način. Procenjivan je tkivni iritacioni indeks (IrI) kao osnovna mera tkivnog odgovora na implantirane hidrogelove. Za određivanje stepena biokompatibilnosti merena je debljina vezivnotkivne kapsule oko implantiranih hidrogelova, broj inflamatornih ćelija, uključujući i Langerhansove džinovske ćelije, broj makrofaga u periimplantnoj zoni i njihova enevntualna migracija u biomaterijal, kao i brojnost krvnih sudova mikrovaskularnog korita oko zone implantacije. Posmatran je odnos proliferacije i apoptoze ćelijskih entiteta kože i potkožnog tkiva odgovornih za procese regeneracije i reparacije...

The ratio of proliferating and apoptotic cellular skin and subcutaneous tissue cellular entities was observed, due to their responsibility for regeneration and reparation processes..., Ova doktorska disertacija dizajnirana je u cilju da se ispita biokompatibilnost dva novosintetisana i prethodno in vitro okarakterisana hidrogela na bazi sintetskog polimera polivinil-alkohola − Ag/PVA i Ag/PVA/Gr, koji su namenjeni za izradu obloga za rane ili za implantate za meka tkiva. Polivinil-alkohol (PVA) je sintetički polimer koji se često koristi u biomedicini zbog svoje netoksičnosti, rastvorljivosti, biokompatibilnosti, elastičnosti i sposobnosti stvaranja reverzibilnih hidrogelova s visokom sposobnošću sorpcije. U poslednjih nekoliko godina, istraživanja se takođe fokusiraju na ugradnju grafena (Gr) u hidrogelove PVA kako bi se poboljšala njihova mehanička čvrstoća i trajnost. U inženjerstvu tkiva i regenerativnoj medicini posebno je važno da implantati i zavoji sadrže antiinflamatornu i antibakterijsku aktivnu komponentu kako bi zaštitili sterilnost rane ili sprečili infekciju u tkivu koje okružuje implantat. Poznato je da nanočestice srebra (eng. silver nanoparticles, AgNPs) poseduju odličnu antibakterijsku aktivnost, pa se zbog toga često koriste kao aktivna komponenta biomaterijala na bazi hidrogela od PVA. U cilju ispitivanja biokompatibilnosti novosintetisanih hidrogelova, praćena je reakcija mekog tkiva na duboko potkožno postavljene implantate od Ag/PVA i Ag/PVA/Gr i poređena je sa reakcijom na komercijalnu oblogu za rane na bazi kalcijum-alginata (Suprasorb©, Lohmann & Rauscher GmbH & Co. KG, Neuwied, Germany), koja je implantirana na identičan način. Procenjivan je tkivni iritacioni indeks (IrI) kao osnovna mera tkivnog odgovora na implantirane hidrogelove. Za određivanje stepena biokompatibilnosti merena je debljina vezivnotkivne kapsule oko implantiranih hidrogelova, broj inflamatornih ćelija, uključujući i Langerhansove džinovske ćelije, broj makrofaga u periimplantnoj zoni i njihova enevntualna migracija u biomaterijal, kao i brojnost krvnih sudova mikrovaskularnog korita oko zone implantacije. The influence of the reduced thyroid function in mothers during gravidity and lactation on the endochondral bone development in rat offspring during the early postnatal period

AB  - In this work we hypothesized that the hypothyroidism in rat dams, influences the process of offspring endochondral ossification affecting proliferation and differentiation of chondrocytes, osteoclasts and osteoblasts in pups. At the day of birth and seven-day-old male pups (n = 36) derived from control dams and dams treated with a low (1.5 mg/L) or high (150 mg/L) dose of propylthiouracil (PTU) in drinking water during pregnancy and lactation (n = 24), were used. Based on the clinical examination, thyroid hormone concentrations and thyroid activation index, it was found that the low dose of PTU caused subclinical, and the high dose of PTU clinical form of hypothyroidism in both mothers and in pups of both age groups. Morphometry, in situ hybridization, immunohistochemistry, histochemistry and electron microscopy were used to analyze the proximal epiphyseal plates of the pups. Chondrocytes in the reserve cartilage zone and proliferation zone contained a lower amount of glycogen, most likely proliferated faster and had a less pronounced autophagy activity in pups from all four treated groups compared to controls. The extracellular matrix contained less collagen type 2 and 10 and more glycosaminoglycans. The most pronounced changes were the shortening of the hypertrophic zone, the reduced height and the increase in the number of hypertrophic chondrocytes that did not express caspase 3. Ossification zone: low amount of tartrate resistant acid phosphatase, and dentin matrix acid protein-1 and collagen type 1 was synthesized in osteoclasts and osteoblasts, respectively. This was associated with the delay of calcified cartilage resorption, decreased deposition and calcification of osteoid, and the formation of shorter and wider trabeculae. All changes were particularly pronounced in pups with a clinical form of hypothyroidism, seven days old. In conclusion, subclinical and clinical hypothyroidism in pups at the day of birth and seven-day-old, most likely stimulates chondrocyte proliferation. It also slows down differentiation and reduces the functional activity of chondrocyte, osteoclasts and osteoblasts, adversely affecting endochondral ossification and tibia length.
AB  - U ovom radu je postavljena hipoteza da hipotireoidizam ženki pacova utiče na proces endohondralnog okoštavanja tako što utiče na proliferaciju i diferencijaciju hondrocita, osteoklasta i osteoblasta kod mladunaca. U ogledu su korišćeni mladunci muškog pola (n=36), eutanazirani odmah na rođenju ili sedmog dana starosti, potomci majki (n=24) iz kontrolne grupe i dve tretirane grupe koje su tokom graviditeta i laktacije dobijale nisku (1.5 mg/L), ili visoku (150 mg/L) dozu propiltiouracila (PTU) u vodi za piće. Na osnovu kliničkog stanja, koncentracije tireoidnih hormona i indeksa aktivacije štitaste žlezde utvrđeno je da je niska doza PTU-a prouzrokovala supklinički, a visoka klinički oblik hipotireoidizma i kod majki i kod mladunaca obe starosne grupe. Za analizu proksimalne epifizne ploče mladunaca korišćena je morfometrija, in situ hibridizacija, imunohistohemija, histohemija i elektronska mikroskopija. Hondrociti u zoni rezervne hrskavice i zoni proliferacije su sadržali manju količinu glikogena, najverovatnije su brže proliferisali i imali su manje izraženu autofagnu aktivnost kod mladunaca iz sve četiri tretirane grupe u odnosu na kontrole. Vanćelijski matriks je sadržala manje kolagena tipa 2 i 10, a više glikozaminoglikana. Najizrazitije promene su skraćenje zone hipertrofije hondrocita, smanjena visina i povećanje broja hipertrofisanih hondrocita koji nisu eksprimirali kaspazu 3. U osteoklastima u zoni osifikacije je smanjenja sinteza tartarat rezistentne kisele fosfataze, dok je u osteoblastima smanjena sinteza kiselog proteina matriksa dentina 1 i kolagena tipa 1. To je povezano sa zaostajanjem kalcifikovane hrskavice, smanjenom depozicijom i kalcifikacijom osteoida te formiranjem kraćih a širih trabekula. Sve promene su naročito izražene kod mladunaca sa kliničkim oblikom hipotireoidizmom starih sedam dana. U zaključku, supklinički i klinički hipotireoidizam kod mladunaca na rođenju i sedmog dana starosti najverovatnije stimuliše proliferaciju hondrocita, a usporava diferencijaciju i smanjuje funkcionalnu aktivnost kako hondrocita, tako i osteoklasta i osteoblasta negativno utičući na endohondralno okoštavanje i dužinu tibije.

Fetlock Joint Cartilage Lesions in Working Horses – A PILOT STUDY

AB  - The horse is still used as a working animal, harnessed in a sled or wheeled chariots 
loaded with cargo several times heavier than the horse itself. There are no data on the extent 
of cartilage lesions in the fetlock joints of front and hind legs of working horses. The aim of 
the study was to determine the size and position of gross cartilage lesions in front and hind 
metacarpo-phalangeal (fetlock) joints of working horses. 
Front and hind legs were collected from a local abattoir after the working horses were 
slaughtered for the reasons not related to this study. The study was conducted on eight horses 
from 5 to 12 years old (mean 8.625 ± 0.91 years), with a body weight of 550 ± 29.46 kg. Thirty two fetlock joints were examined for external gross anatomy changes. After joint dissection, the 
cartilage of metacarpal and metatarsal bones was stained with blue ink and photos were taken. 
The cartilage lesions were stained deep blue, in contrast to light blue surface of the undamaged 
cartilage. The anatomical position of the lesions was recorded. The ImageJ software was used 
to measure the total surface of the cartilage and the surface of the cartilage under the lesions. 
The dimension of the lesions was expressed as a percentage of the total cartilage surface and 
presented as mean ± standard error. 
Gross anatomy changes like skin lesions, wounds, joint oedema or any type of 
deformities were not observed. The position of the cartilage lesions was symmetrical and 
occupied lateral and medial metacarpal and metatarsal condyles next to the sagittal ridge. The 
lesions on the lateral condyles had the same surface area like those on the medial, 12.3 ± 1.03% 
versus 10.7 ± 0.99%, respectively. The cartilage lesions on the front legs (23.62 ± 1.68%) were 
broader than those on the hind legs (22.5 ± 3.35%).
The results of this pilot study indicate no difference between metacarpal and metatarsal 
cartilage surface lesions. Further studies are indicated to determine the severity of cartilage and 
putative subchondral bone lesions by histologic evaluation

AB  - Background Babesia canis infection induces a marked acute phase response (APR) that might be associated with alteration in lipid and lipoprotein metabolism and disease prognosis. Hypothesis Dogs with B. canis-induced APR develop dyslipidemia with altered lipoprotein concentration and morphology. Animals Twenty-nine client-owned dogs with acute B. canis infection and 10 clinically healthy control dogs. Methods Observational cross-sectional study. Serum amyloid A (SAA) was measured using ELISA. Cholesterol, phospholipids, and triglycerides were determined biochemically. Lipoproteins were separated using agarose gel electrophoresis. Lipoprotein diameter was assessed by polyacrylamide gradient gel electrophoresis; correlation with ApoA-1 (radioimmunoassay) and SAA was determined. Results Dogs with B. canis infection had a marked APR (median SAA, 168.3 mu g/mL; range, 98.1-716.2 mu g/mL) compared with controls (3.2 mu g/mL, 2.0-4.2 mu g/mL) (P < .001). Dogs with B. canis infection had significantly lower median cholesterol (4.79 mmol/L, 1.89-7.64 mmol/L versus 6.15 mmol/L, 4.2-7.4 mmol/L) (P = .02), phospholipid (4.64 mmol/L, 2.6-6.6 mmol/L versus 5.72 mmol/L, 4.68-7.0 mmol/L) (P = .02), and alpha-lipoproteins (77.5%, 27.7%-93.5% versus 89.2%, 75.1%-93.5%) (P = .04), and higher ApoA-1 (1.36 U, 0.8-2.56 U versus 0.95 U, 0.73-1.54 U) concentrations (P = .02). Serum amyloid A correlated with high-density lipoproteins (HDLs) diameter (rho = .43; P = .03) and ApoA-1 (rho = .63, P < .001). Conclusions and Clinical Importance Major changes associated with B. canis-induced APR in dogs are related to concentration, composition, and morphology of HDL particles pointing to an altered reverse cholesterol transport. Parallel ApoA-1 and SAA concentration increase is a unique still unexplained pathophysiological finding.

AB  - Belgrade, the capital of the Republic of Serbia, is an endemic location for canine babesiosis caused by Babesia canis. This tick-borne disease occurs seasonally in regions with moderate continental climate. However, recent clinical data show that 1) some cases of canine babesiosis have been recorded during the winter, and 2) canine babesiosis is spreading to the northern parts of Europe with a colder climate, which is a region previously free of this disease. Our study investigates the occurrence of canine babesiosis in different seasons over 2013-2016, and explores a short-term link between meteorological parameters and number of diagnosed cases of canine babesiosis in Belgrade. It also looks into possible long-term correlations that span one year before the onset of the disease. Based on 872 recorded cases over four years, our results show a bimodal seasonal distribution of canine babesiosis, with a pronounced peak in the spring, and a less conspicuous one in the autumn. Throughout the year, even over the coldest and warmest periods, there is a broad range of temperatures and relative humidifies when the disease is recorded. Over one year prior to the spring and autumn onset of the disease, we found a noticeable impact of temperature and relative humidity, and to a lesser extent, of atmospheric pressure and cloud cover, on the number of diagnosed cases. These findings imply short-term and long-term relationships between occurrence of acute canine babesiosis and certain meteorological parameters, and they open further questions that need to be investigated in order to understand the epidemiology of this disease.

Mitochondria in pachytene: the fragile point of maternal subclinical hypothyroidism affection

AB  - The stimulative effects of thyroid hormones on mitochondria are realized through 
both non-genomic and genomic mechanisms, affecting respiration, mitochondrial plasticity 
and biogenesis. The subclinical form of maternal hypothyroidism in rats induces significant 
reduction of mitochondria number but also an augmentation of their area in neonatal and 
early infantile offspring dyctiotene oocytes. This study aimed to investigate if this form of 
subclinical hypothyroidism affects mitochondrial morphology and distribution in the early 
prophase of meiosis I oocytes. It was performed on newborn control (C) (n=10) and 
hypothyroid (SCH) (n=10) female rat pups derived from control (n=6) and propylthiouracil
treated pregnant dams (n=6), respectively. Ovaries of all pups were removed and processed 
for transmission electron microscopy. The morphological features of mitochondria in the 
early prophase I oocytes until dyctiotene were assessed. No substantial differences were 
found in leptotene and zygotene oocytes in SCH group comparing to control, except just a 
few mitochondria characterized with shortened cristae, presence of wide pale area centrally 
positioned and membrane disruption. Pachytene mitochondria in treated pup oocytes were 
in great extent with disrupted membrane, shortened cristae and wide pale area centrally 
positioned while these features were rarely observed in control ones. Our results confirm 
altered mitochondria morphology found in primordial and primary follicles in case of 
maternal hypothyroidism, indicating their impaired function and possibly, propensity to 
programmed cell death. Further investigations may indicate to what extent pachytene, as a 
meiotic checkpoint, appears to be a milestone possibly predetermining the future of the 
cell.

AB  - Maternal hypothyroidism in its overt form affects skeletal development of the offspring, but these data are not available for the subclinical form which is becoming very frequent among pregnant women. We hypothesized that the subclinical form of hypothyroidism in rat dams, influences the process of offspring endochondral ossification affecting proliferation and differentiation of chondrocytes, osteoclasts and osteoblasts in pups. Seven-day-old male pups (n=18) derived from control dams and dams treated with a low dose (1.5 mg/L) or high dose (150 mg/L) of propylthiouracil in drinking water during pregnancy and lactation were used. Histomorphometric analysis of pups tibia proximal growth plate, expression of mRNA, immunohistochemical and histochemical visualization of extracellular matrix components was performed. The length of the tibia was reduced in hypothyroid pups. Secretion of type 2 and 10 collagens in the subclinical and overt form were lower while the amount of glycosaminoglycans was higher when compared with controls. Down-regulated tartrate resistant acid phosphatase mRNA indicated altered osteoclasts function while lower expression of dentin matrix acid protein-1 mRNA and reduced synthesis of type 1 collagen accentuated a compromised bone formation in the overt form of hypothyroidism. The subclinical form of maternal hypothyroidism had a negative effect on the differentiation of hypertrophic chondrocytes and calcified cartilage removal in 7-day-old pups. In addition, overt hypothyroidism had a negative effect on the proliferation of chondrocytes and deposition of osteoid. Both forms of hypothyroidism resulted in a decrease of tibia length due to changes in growth plate formation.

AB  - Introduction. The ovarian surface epithelium (OSE) undergoes intensive regeneration and remodelling after each ovulation during the whole reproductive period. This process increases the risk of one of the most common ovarian tumors in women and the female dog. Considering the fact that maternal hypothyroidism highly impacts cell proliferation and cell death during folliculogenesis in the early neonatal period, we aimed to analyze its effect on OSE morphology and dynamics. Materials and Methods. The study was performed on newborn (24-h-old) and neonatal (4-day-old) female rats, a randomized trial between the control and hypothyroid groups, born under controlled circumstances and hypothyroid mothers, respectively. Their ovaries were analyzed histologically and processed to determine the OSE cell height as an average value of four measurement points. Also, the immunopositivity of the proliferating cell nuclear antigen (PCNA) and caspase-3 were assessed semiquantitatively. Results and Conclusions. No major structural differences of OSE were found between groups within the given ages except for a slight increment of OSE cell height and incompleteness of apical cell membrane with cytoplasmic projections in hypothyroid animals. PCNA immunopositivity of the OSE cells was higher in ovaries of hypothyroid animals of both ages in comparison to the controls. Moreover, only scarce OSE cells were caspase-3 positive in both groups and ages, with no difference in immunopositivity. Our study confirms the impact of hypothyroidism in the early postnatal period on morphology and proliferation rate of OSE cells, with no effect on caspase-3 dependent cell removal, which may serve as a premise for future investigation of potential carcinogenesis, in terms of prevention and treatment of ovarian cancer.
AB  - Uvod. Tokom reproduktivnog perioda, nakon svake ovulacije, površinski epitel (PE) jajnika se intenzivno regeneriše i remodelira. Ovaj proces povećava rizik od nastajanja jednog od najčešćih tumora jajnika kod žena i kuja. Imajući u vidu činjenicu da hipotireoidizam majki u velikoj meri utiče na proliferaciju i ćelijsku smrt u toku folikulogeneze u ranom postnatalnom periodu, cilj rada je bio da analiziramo efekat hipotireoidizma majki na morfologiju i dinamiku PE jajnika potomaka. Materijal i metode. U eksperimentu su korišćeni tek rođeni (24 časa stari) i neonatalni (4 dana stari) mladunci koji potiču od kontrolnih i hipotireoidnih majki. Morfometrijskom analizom određivana je prosečna visina ćelija PE, izmerena na četiri mesta na svakom ispitivanom preseku jajnika. AB  - Objectives The objectives of this study were to estimate the prevalence of feline haemoplasma infections in Northern Serbia, identify potential risk factors and perform molecular subtyping of feline immunodeficiency virus (FIV). Methods PCR analysis for feline haemoplasmas was performed on surplus EDTA blood samples from 373 cats from the Belgrade region, Serbia. An ELISA was used to determine the prevalence of feline leukaemia virus (FeLV) and FIV; PCR was performed on a subpopulation of these cats. FIV subtyping was performed using PCR. Results Within this population, 64/373 cats (17.2%) were infected with one or more haemoplasma species. Mycoplasma haemofelis was detected in 20/373 cats (5.4%), ‘ Candidatus Mycoplasma haemominutum’ in 47/373 cats (12.6%) and ‘ Candidatus Mycoplasma turicensis’ in 23/373 cats (6.2%). Coinfections were observed in 21/373 cats (5.6%). Based on ELISA serological retroviral testing, 4/310 cats (1.3%) were infected with FeLV, whereas 78/331 (23.6%) were infected with FIV. Multivariable analysis identified significant associations between haemoplasma infection and anaemia (anaemic/non-anaemic, odds ratio [OR] 2.7, 95% confidence interval [CI] 1.04–7.1; P = 0.041]), male gender (male/female, OR 4.5, 95% CI 2.22–9.03; P <0.0005), outdoor access (yes/no, OR 5.2, 95% CI 2.28–11.92; P <0.0005), non-pedigree breed (non-pedigree/pedigree, OR 5.5, 95% CI 1.24–24.84; P = 0.025) and FIV seropositive status (positive/negative, OR 2.4, 95% CI 1.21–4.83; P = 0.012). PCR analysis of the FIV ELISA-positive samples revealed clade D as being the most prevalent. Conclusions and relevance All three known species of feline haemoplasma were detected, confirming their presence in Serbia; ‘ Candidatus Mycoplasma haemominutum’ was the most prevalent. We found a high prevalence of FIV-infected cats and FIV clade D was most prevalent.

Influence of absorbed radiation dose following computed tomography on the antioxidative status in rabbit testicles

AB  - In recent years, computed tomography (CT) has become very common in veterinary medicine. It is well known that testicles are organs with high radiosensitivity and their function can be impaired even after exposure to low radiation doses. In this work, we calculated the absorbed radiation doses after CT was performed with different voltage/current levels and correlated it with the activity of antioxidant enzymes in rabbit testicles. Two hours after CT, the activities of catalase (CAT) and glutathione peroxidase (GSH-Px) were increased in the testicles of animals that received an absorbed dose of 29.2 mGy. The same changes, along with elevated glutathione reductase (GR) activity, were observed after 7 days in animals that received the highest absorbed dose (46.3 mGy). It would appear that absorbed doses above 27.8 mGy provoked the antioxidant reaction but the time scale of the reaction was dose-dependent. Examination of the obtained results revealed that the main denominator of CT influence was a higher current. Our results suggest that CT influences the antioxidant status in rabbit testicles. The changes in antioxidant enzyme activities were dose- and time-dependent and influenced by the applied current.

Accordingly, we examined the potential of cell culture supplementation with NAC in prevention of lipid peroxidation, unfavorable changes of total lipids fatty acid composition, and the effects on the activity of antioxidant enzymes. Material and methods We analyzed the extent of oxidative damage in SHED after 48 h treatment with different NAC concentrations. Cellular lipid peroxidation was determined upon reaction with thiobarbituric acid. All enzyme activities were measured spectrophotometrically, based on published methods. Fatty acid methyl esters were analyzed by gas-liquid chromatography. Results Concentration of 0.1 mM NAC showed the most profound effects on SHED, significantly decreasing levels of lipid peroxidation in comparison to control. This dose also diminished the activities of antioxidant enzymes. Furthermore, NAC treatment significantly changed fatty acid composition of cells, reducing levels of oleic acid and monounsaturated fatty acids and increasing linoleic acid, n-6, and total polyunsaturated fatty acid (PUFA) proportions. Conclusion Low dose of NAC significantly decreased lipid peroxidation and altered fatty acid composition towards increasing PUFA. The reduced oxidative damage of cellular lipids could be strongly related to improved SHED survival in vitro.

Z-cells and oogonia/oocytes in the advanced process of autophagy are the dominant altered cells in the ovaries of hypothyroid newborn rats

AB  - Induced prenatal hypothyroidism in rat pups leads to accelerated primordial follicle assembly and premature follicular atresia with ovary failure. This work investigates the influence of maternal hypothyroidism induced with 6-n-propyl-2-thyouracil (PTU) on the number and morphology of oogonia/oocytes in newborn rat pups with light and transmission electron microscopy. Expression of apoptosis and autophagy markers in oogonia/oocytes were examined using immunohistochemistry. Hypothyroid newborn pups had a decreased number of mitotic and resting oogonia, while the number of altered oogonia/oocytes was increased. Ultrastructural observations revealed the increased presence of degenerated pachytene oocytes (Z-cells) and oogonia/oocytes undergoing autophagy, apoptosis and combined apoptosis and autophagy, in this group. The most abundant altered oogonia/oocytes in the hypothyroid group were those with morphological features of advanced autophagy and Z-cells. The percentage of TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) positive oogonia/ oocytes was significantly lower in the hypothyroid group. No significant difference was recorded in the expression of caspase-3, ATG7 and LC3 possibly reflecting that these proteins were not involved in the oogonia/ oocyte alteration process during prenatal rat hypothyroidism. The obtained results indicate that developmental hypothyroidism in the offspring enhances the number of Z-cells and oogonia/ oocytes altered with the advanced process of autophagy.

AB  - Introduction. Thyroid hormones (TH) play a pivotal role in the development of mammalian skin and are necessary for both, the initiation and maintenance of hair growth. Hypothyroid rats showed  impaired epidermal proliferation, hair growth, and wound healing. Stem cells located at the bulges of the hair follicles are responsible for hair cycling and contribute to the regeneration of the new epidermis after wounding. Therefore a reduction in the num¬ber or function of the bulge stem cells could be responsible for this phenotype. 
We hypothesized that decrease in TH during prenatal development would affect bulge stem cell population.
Material and methods. Subclinical hypothyroidism was induced with propylthiouracil through drinking water in dose 1.5 mg/l in pregnant Albino Oxford rats from the first day of gravidity. Control group was untreated. Seven-day-old pups were euthanized and skin samples were taken from dorsal part of their bodies. For the immunohistochemical study PCNA (Santa Cruz Biotechnology) and NANOG (Thermo scientific) antibodies were used.
Results. . The number of hair follicles was reduced in hypothyroid pups compared to 
controls. They also had a decreased expression of PCNA in cells of inner and outer sheat of hair follicles, but increased number of NANOG positive bulge stem cells. 
Conclusion. This suggests that re¬duced activation of stem cells leading to their accumulation in the bulges which is associated with reduced stem cell prolif¬eration and mobilization.

AB  - Thyroid hormones (TH) are indispensable for normal development in mammals. The effects of maternal hypothyroidism on development and function of different organs of their offspring are well known. It is unclear how its subclinical form affects some of these processes, including endochondral ossification during the prenatal and early postnatal development.
The aim of the study was to assess bone formation by defining the expression of tartarate-resistant acid phosphatase (TRAP), alkaline phosphatase (ALP) and mRNA for dentin matrix acidic phosphoprotein (DMP1) in tibia of pups from subclinical hypothyroid dams in early postnatal development. Subclinical hypothyroidism (SHT) was induced with propylthiouracil through drinking water (1.5 mg/l) in pregnant Albino Oxford dams from the first day of gravidity and during lactation. Control group was untreated. Five seven-day-old pups from each experimental group were euthanized. All histological examinations were performed on paraffin sections of the proximal tibial growth plate. Histochemistry was used to assess the expression of TRAP, immunohistochemistry for ALP and in situ hybridization for DMP1. The area of TRAP positive signal per 1µm2 and optical density of ALP immunostaining were evaluated using the Image J. The number of DMP1 positive cells was counted for given area using the same software. Level of thyroxine (T4) and triiodothyronine (T3) of dams and pups were decreased (data published). Expression of TRAP and ALP in the proximal tibial growth plate were lower in hypothyroid pups and the number of cells expressing mRNA for DMP1 was decreased. 
As it is known DMP1 and ALP are expressed in osteoblasts with an important role in endochondral ossification while TRAP represents a marker of osteoclasts function and bone resorption, so we can conclude  that maternal SHT in rats leads to delayed osteogenic differentiation and compromised cartilage to bone transition in early infantile period.

Ovarian surface epithelium in newborn rats: germ cells warehouse

AB  - Ovarian surface epithelium (OSE) is a very dynamic part of the ovary with an important role in ovulation during reproductive period. It represents a source of germ cells in fetal period but data concerning that role of OSE in postnatal period are contradictory. Though OSE is a subject of research in past few years, it’s ultrastructure in newborn rats has not been studied yet.
The aim of the study was to define ultrastructure features of OSE in rats in the first 24h hours after birth.
Six newborn rats within first 24 hours after birth were euthanized. Ovaries were fixed in 4% glutaraldehyde, post-fixed in 1% osmium tetroxide, routinely dehydrated and embedded in araldite. Semi-thin sections were stained with toluidine blue and used to select areas of interest. Ultra-thin sections of selected areas were mounted on copper grids and examined on a Philips CM12 transmission electron microscope. Descriptive ultrastrustructure analysis was performed. OSE in newborn rats was mostly single layered and sporadically pseudostratified, consisted from mostly cuboidal and columnar cells. These cells were with centrally positioned nucleus, mostly with irregularly dispersed chromatin. Microvili and cytoplasmic projections extended to peritoneal cavity from the surface of the cells. No continuous tunica albuginea was found, but in places, groups of fibroblast-like cells were present, indicating the onset of its formation. Numerous oogonia and oocytes were identified between OSE cells or up to the basal part of them. This may indicate that extrusion (the release of germ cells into a peritoneal cavity) may be an important way of germ cells elimination. 
Early postnatal period in rat ovaries characterize with massive cell death of oogonia and oocytes, mostly by apoptosis and autophagy, but further investigation might reveal the participation of this way of germ cells elimination realizing the importance of appropriate course of cell death process in terms of reproductive health.

abstract = "Ovarian surface epithelium (OSE) is a very dynamic part of the ovary with an important role in ovulation during reproductive period. It represents a source of germ cells in fetal period but data concerning that role of OSE in postnatal period are contradictory. Though OSE is a subject of research in past few years, it’s ultrastructure in newborn rats has not been studied yet.
The aim of the study was to define ultrastructure features of OSE in rats in the first 24h hours after birth.
Six newborn rats within first 24 hours after birth were euthanized. Ovaries were fixed in 4% glutaraldehyde, post-fixed in 1% osmium tetroxide, routinely dehydrated and embedded in araldite. Semi-thin sections were stained with toluidine blue and used to select areas of interest. Ultra-thin sections of selected areas were mounted on copper grids and examined on a Philips CM12 transmission electron microscope. Descriptive ultrastrustructure analysis was performed. OSE in newborn rats was mostly single layered and sporadically pseudostratified, consisted from mostly cuboidal and columnar cells. These cells were with centrally positioned nucleus, mostly with irregularly dispersed chromatin. Microvili and cytoplasmic projections extended to peritoneal cavity from the surface of the cells. No continuous tunica albuginea was found, but in places, groups of fibroblast-like cells were present, indicating the onset of its formation. Numerous oogonia and oocytes were identified between OSE cells or up to the basal part of them. This may indicate that extrusion (the release of germ cells into a peritoneal cavity) may be an important way of germ cells elimination.
