Lafora disease in a Chihuahua dog: a case report

AB  - The aging process is still not fully understood, although it has been studied for centuries. One
of the processes in the brain during aging is the accumulation of extracellular and intracellular
deposits of amyloid and lipofuscin. Deposits of various polyglucosan bodies (PGBs) are also
found in brain tissue. The accumulation of the Lafora bodies (LB), a type of PGBs, can cause
the Lafora disease (LD). Initial signs of the disease in humans are tonic-clonic seizures with
blindness and myoclonus seizures. Normally, all haematological and biochemical indices are
within the reference range in dogs with this disease. In this case, a 7-year-old Chihuahua dog with
tonic-clonic seizures was presented. According to history, neurological examination, and blood
test, toxic, metabolic, and infectious causes for the seizures were ruled out. The dog was started
on phenobarbital 2.5 mg/kg twice daily per os. Two years later, the dog died due to complications
caused by a duodenal foreign body. Postmortem examination revealed hyperaemic meninges
and an enlarged, oedematous brain with flattened gyri and narrowed sulci. Histopathological
examination revealed multifocal to diffuse, randomly distributed PGBs that were positive
on periodic acid-Schiff (PAS) staining. The diagnosis of LD in dogs is of great importance because
they are a good experimental model for neurological studies of neurodegenerative diseases
in humans.

Epidemiological and clinical importance of canine dioctophymosis

AB  - Dioctophimosis is an endoparasitism in dogs caused by Dioctophyma
renale, a nematode with an indirect life cycle and high zoonotic potential.
Infection in dogs occurs through ingestion of transitional/paratenic hosts
containing infective larvae. Th e preferred site of parasitism is the right kidney, although the parasite may also be ectopically localized in subcutaneous
tissue, internal organs, peritoneum, and mediastinum. Since dogs are usually infected with a small number of parasites, the disease is asymptomatic,
making the diagnosis of this disease in clinical veterinary practice rather
diffi cult. In addition, as dogs are usually infected with a small number of
parasites, the disease is asymptomatic, which makes the diagnosing of this
disease in clinical veterinary practice diffi cult. Th e long prepatent period,
lasting three to fi ve months, makes it impossible to diagnose the disease in
dogs younger than six months. Clinical suspicion of dioctophimosis in dogs
is established on the basis of anamnestic/epizootiologic data and the clinical picture, while the fi nal diagnosis is made ante mortem or post mortem on
the basis of reliable diagnostic parameters and methods. Th e most practical
application in routine diagnosis of canine dioctophimosis is the detection
of D. renale eggs in urine sediment. When dioctophimosis is suspected in carnivores, the diff erential diagnosis should exclude urinary capillariosis,
feline polycystic nephropathy, leptospirosis, renal fi brosis, and nephritis of
various etiology. Th e outcome of treatment is uncertain and includes surgical removal of the aff ected kidney or drug therapy with avermectin derivatives. Prophylaxis consists of preventing contact of dogs with potential
transitional hosts and controlling the feeding of their heat-untreated meat.
Because of the potential risks that D. renale may have on the health of dog
owners, education on the etiopathogenesis and means of occurrence, maintenance, and spread of this nematode is necessary.
AB  - Dioktofi moza je endoparazitoza pasa koju prouzrokuje Dioctophyma renale, nematoda sa indirektnim životnim ciklusom i visokim zoonoznim potencijalom. Infekcija pasa nastaje ingestijom prelaznih/paratenih domaćina
koji u sebi sadrže infektivne larve. Predilekciono mesto parazitiranja je
desni bubreg, uz mogućnost ektopične lokalizacije parazita u supkutanom
tkivu, unutrašnjim organima, peritoneumu i medijastinumu. Pošto su psi
uglavnom infi cirani malim brojem parazita, oboljenje protiče asimptomatski, što u kliničkoj veterinarskoj praksi otežava dijagnostiku ovog oboljenja.
Dug prepatentni period, koji traje od tri do pet meseci, onemogućava dijagnostiku oboljenja kod pasa mlađih od šest meseci. Klinička sumnja na dioktofi mozu kod pasa postavlja se na osnovu anamnestičkih/ epizootioloških
podataka i kliničke slike, dok se precizna dijagnoza utvrđuje ante mortem
ili post mortem na osnovu pouzdanih dijagnostičkih parametara i metoda. Najveću praktičnu primenu u rutinskoj dijagnostici dioktofi moze pasa
ima nalaz jaja D. renale u sedimentu urina. Kod sumnje na dioktofimozu
mesojeda diferencijalno dijagnostički treba isključiti urinarnu kapilariozu,
policističnu nefropatiju mačaka, leptospirozu, fi brozu bubrega i nefritise
različite etiologije. Lečenje je neizvesno i podrazumeva hirurško uklanjanje
promenjenog bubrega ili medikamentoznu terapiju derivatima avermektina.
Profi laksa je zasnovana na sprečavanju kontakta pasa sa potencijalnim prelaznim domaćinima i kontrolisanje ishrane njihovim termički neobrađenim
mesom. Zbog potencijalnih rizika koje D. renale može imati po zdravlje
vlasnika pasa, neophodna je edukacija o etiopatogenezi i mogućnostima
pojave, održavanja i širenja ove nematode.

“Slow kill” treatment reduces DNA damage in leukocytes of dogs naturally infected with Dirofilaria immitis

AB  - Parasitic diseases are considered to be a cause of oxidative stress which leads to oxidative damage of various
molecules including DNA. This can result in mutations, replication errors, and genome instability. Therefore, aim
of this study was to measure DNA damage induced by Dirofilaria immitis in the single cells such as dogs’ leukocytes using the comet assay. Also, we monitored the effects of antiparasitic treatment on mitigation of
sensitivity to DNA damage in leukocytes treated with H2O2 using the in vivo and ex vivo comet assay. The whole
blood samples from 34 dogs from Serbia were used, both males and females, from one to 13 years old, both pure
and mixed-breeds. A rapid immunochromatographic test (Antigen Rapid Heartworm Ag 2.0 Test Kit, Bionote,
Minnesota, USA) was used for the detection of D. immitis antigens. The modified Knott’s test and PCR were used
in the aim of detecting D. immitis microfilariae in dogs’ blood, and evaluating the number of circulating
microfilariae during the treatment. The genotoxicity evaluation showed that D. immitis infection resulted in DNA
damage in naturally infected dogs, with the highest DNA damage occurring in the group of dogs with severe
clinical signs. Treatment with ivermectin and doxycycline decreased DNA damage in leukocytes of dogs in all
groups, as the intensity of infection decreased due to applied therapy. Ex vivo comet assay results showed that
leukocytes exhibited decreased sensitivity to H2O2-induced DNA damage during treatment. The results of the
modified Knott’s test and PCR in our study showed that treatment with ivermectin and doxycycline was successful in decreasing the average number of microfilariae during the time and at the end eliminating them from
the dogs’ blood.

Istraživanje o mišljenju studenata prve i završne godine Fakulteta veterinarske medicine o pravima i dobrobiti životinja

AB  - У данашње време улога животиња у друштву је промењена. Животиње се
више не посматрају као власништво, већ као жива бића. Када је у питању
ветеринарска медицина, такође постоје промене у интересовањима између
мушкараца и жена, као и професионалним преференцијама. Студије су
показале да пол и брига за добробит животиња такође утичу на ниво
забринутости за здравље животиња. Циљ овог истраживања био је да се
процени разлика у мишљењу између студената прве и завршне године
ветеринарске медицине у односу на етичке дилеме и добробит животиња. У
истраживање је укључено укупно 200 студената ветеринарске медицине прве
и завршне године. Survey Monkey је коришћен за прикупљање информација, а
анкета се састојала од десет питања. Резултати су подељени на демографске
податке, етичке дилеме у области права животиња и мишљење о еутаназији.
Није установљена статистички значајна разлика између односа полова и места
одрастања испитиваних студената. Скоро половина студената прве године
(49%) исказала је жељу да ради са социјалним животињама, а само 11% са
фармским животињама. Када је реч о студентима завршне године, већина
њих такође жели да ради са социјалним животињама (36%), али постоји и
интересовање за рад са фармским животињама (20%). На разлике у мишљењу
о правима животиња и еутаназији више је утицао пол студената у поређењу са
годином студија. Истраживање је показало да у популацији студената постоји
смањена заинтересованост за бављење праксом фармских животиња, и да
је то област којој треба посветити више пажње. Такође, неопходно је више усмеравати студенте за области њиховог интересовања током студирања,
да би када стекну диплому могли одмах знати у којој области ветеринарске
делатности желе да раде.

AB  - Rhinoscopy is an endoscopic visualization of the nasal cavity. It is performed
using special endoscopes and often gives us important information that would be
difficult to obtain using other diagnostic methods. Anatomically, the examination
includes two procedures: anterior rhinoscopy and posterior rhinoscopy. Diseases
of the nasal cavity can be manifested with one or more clinical symptoms such as:
sneezing, nasal discharge, stridor and swelling of the face. This diagnostic method
is indicated when there is a suspicion of chronic inflammation of the nasal
mucosa, aspergillosis in dogs, as well as the presence оf а foreign body or neoplasia
in the nasal cavities. In some cases, in order to confirm the correct diagnosis, a
biopsy of the altered mucous membrane of the nasal cavities is necessary.

AB  - Although Mycobacterium spp. often cause disease in domestic birds (chickens and companion birds), there are few data on avian tuberculosis in wild populations, especially in birds of prey. We describe here a case of a young adult female, free-living Eurasian griffon vulture (Gyps fulvus) that was found dead. Granulomas were grossly evident in the lungs at autopsy, and tuberculosis was suspected. Ziehl–Neelsen staining revealed large numbers of intracellular acid-fast–positive bacteria within granulomas. Examination on Löwenstein–Jensen medium was negative, but mycobacteria growth indicator tube medium results were positive. For the molecular detection of Mycobacterium spp., the primer set IS901F and IS901R was used. Positive results were observed on gel electrophoresis, indicating the presence of Mycobacterium avium subsp. avium DNA. Although tuberculosis is not considered to be a common cause of death in wild birds, it undoubtedly deserves special attention because vultures are generally considered to be a species resistant to a large number of pathogens. Determination of the cause of death of griffon vultures is important for future conservation measures for this sensitive wild species.

AB  - Prema mehanizmu nastanka, anemije se klasifikuju na
hemoličke i hemoragijske i anemije koje nastaju usled

regenerativnim odgovorom, koji se ogleda u značajnom

i/ili nedostatak nutritivnih faktora, dijagnostikuju se kao

AB  - Ultrazvučni pregled, poznat i kao ultrasonografija, je neinvaziv- na tehnika snimanja koja omogućava uočavanje unutrašnjih telesnih struktura beleženjem odjeka ili refleksije ultrazvučnih talasa. Za razli- ku od rendgenskih zraka koji su potencijalno opasni, ultrazvučni talasi se smatraju sigurnim. Svaki veterinar kliničar treba da bude svestan mogućnosti, ali i ograničenja ove dijagnostičke procedure. On mora da zna na koja pitanja ovaj vid pregleda može dati odgovore, ali i da bude svestan činjenice da negativan ultrazvučni nalaz ne isključuje uvek oboljenje. Najbitniji segment svakog ultrazvučnog pregleda je sistematičnost. Kod svakog pacijenta se treba držati istog redosleda pregleda organa. Ultrazvučni pregled abdomena psa obuhvata pre- gled hepatobilijarnog sistema, želuca, slezine, bubrega, nadbubrež- nih žlezdi, mokraćne bešike, genitalnih organa, creva, limfnih čvorova i krvnih sudova. Standardizovano, precizno i konkretno pisanje izve- štaja, nakon obavljenog ultrazvučnog pregleda, treba da bude velika pomoć veterinaru pri postavljanju dijagnoze ili planiranju sledećih di- jagnostičkih procedura.

Congenital thoracic kyphosis caused by hemivertebra in a Pug puppy

AB  - Congenital spine deformation caused by vertebral malformation is reported in different brachycephalic dog breeds, but also in other large dog breeds. The most severe clinical signs  are  due  to  spinal  cord  compression.  Depending  on  localisation  of   the  lesion,  clinical signs can manifest differently. A five-month-old pug was brought for autopsy to the Department of  Forensic Veterinary Medicine, Faculty of  Veterinary Medicine, University of Belgrade. The first clinical signs, including general weakness and difficulty to jump appeared when he was three and a half  months old. They became more severe until  the  dog  was  four  months  old.  Radiography  and  myelography  showed  severe  kyphosis and spinal cord compression in the T8-T9 region. After euthanasia, an autopsy was  performed.  Pathomorphological  examination  showed  severe  deformation  of   the  spine, kyphosis and a wedge-shaped T8 vertebra. Although, literature data indicates that clinical signs can be latent until five or six months of age, this case shows that even in young puppies, three to four months old, serious clinical signs can be present. This is of  great clinical importance, not just for veterinarians, but also for brachycephalic dog owners and breeders.
AB  - Kongenitalne deformacije kičme uzrokovane malformacijama na vertebrama javljaju se kod različitih brahicefaličnih rasa, ali se mogu javiti i kod velikih rasa pasa. Najteži klinički simptomi se javljaju usled kompresije kičmene moždine. U zavisnosti od lokalizacije lezije, simptomi se mogu različito manifestovati. Leš mopsa starosti pet meseci donešen je zbog obdukcije na Katedru za sudsku veterinarsku medicinu, Fakulteta veterinarske medicine Univerziteta u Beogradu. Prvi klinički simptomi, u vidu opšte slabosti i poteškoća pri skakanju, javili su se kada je štene bilo staro tri i po meseca. Simptomi su u četvrtom mesecu postali mnogo ozbiljniji. Nativno rendgensko i mijelografsko snimanje pokazalo je uznapredovali stepen kifoze i kompresije kičmene moždine u T8-T9 regionu kičme. Nakon eutanazije, pas je obdukovan. Patomorfološki nalaz ukazao je na ozbiljnu deformaciju kičme, izraženu kifozu i klinast oblik T8 pršljena. Iako literaturni podaci ukazuju da klinički simptomi mogu biti nezapaženi sve do pet ili šest meseci starosti, ovaj slučaj pokazuje da čak kod mlade štenadi, starosti tri do četiri meseca, mogu biti prisutni ozbiljni simptomi. Ova činjenica je od velikog značaja, ne samo za veterinare, već i za odgajivače i vlasnike brahicefaličnih rasa pasa.

Pathomorphological  examination  showed  severe  deformation  of   the  spine, kyphosis and a wedge-shaped T8 vertebra. Although, literature data indicates that clinical signs can be latent until five or six months of age, this case shows that even in young puppies, three to four months old, serious clinical signs can be present. AB  - BackgroundCanine generalized demodicosis is a common parasitic disease caused by the proliferation of Demodex mites. The introduction of isoxazoline class treatments in veterinary dermatology has resulted in apparently effective treatment of generalized demodicosis. The objective of this study was to evaluate the effectiveness of fluralaner for the treatment of canine generalized demodicosis using real-time PCR for the detection and quantification of Demodex DNA.MethodsTwenty privately owned dogs with clinical symptoms of generalized demodicosis and deep skin scrapings positive for Demodex canis mites were enrolled in the study. Following diagnosis (day 0) each dog was treated with fluralaner at the recommended commercial dose for tick and flea treatment (25-56 mg/kg) based on body weight. Clinical and mite count assessments, and hair sampling for molecular analyses were performed on days 0, 28, 56, 84 and 112. Demodex DNA was detected and quantified using real-time PCR.ResultsA single oral dose of fluralaner reduced Demodex mite counts in skin scrapings by an average of 98.9% in all dogs by day 28. No mites were recovered from skin scrapings from any treated dog by day 56, at which time the dog was considered to be clinically cured, with total hair regrowth. There were significant differences among examined dogs in qPCR cycle threshold (Ct) values on days 0, 28, 56, 84 and 112. Demodex DNA levels decreased (increasing Ct values) throughout the study. Mite DNA was present on day 112, possibly from dead mites, at values significantly lower than in samples taken on days 0, 28 and 56. Based on qPCR testing of diluted samples, the Demodex mite population was reduced by approximately 1000-fold on day 112.ConclusionsOral administration of fluralaner at the recommended dose to dogs with generalized demodicosis is highly effective for reducing Demodex mite populations and resolving clinical signs of generalized demodicosis. The presence of mite DNA may indicate that treatment did not kill all Demodex mites.

AB  - BackgroundCulicoides (Diptera: Ceratopogonidae) is a genus of small biting midges (also known as no-see ums) that currently includes 1368 described species. They are proven or suspected vectors for important pathogens affecting animals such as bluetongue virus (BTV) and Schmallenberg virus (SBV). Currently little information is available on the species of Culicoides present in Serbia. Thus, the aim of this study was to examine species diversity, host preference and the presence of BTV and SBV RNA in Culicoides from the Stara Planina Nature Park in south-eastern Serbia.ResultsIn total 19,887 individual Culicoides were collected during three nights of trapping at two farm sites and pooled into six groups (Obsoletus group, Pulicaris group, Others group and further each group according to the blood-feeding status to freshly engorged and non-engorged). Species identification was done on subsamples of 592 individual Culicoides specimens by morphological and molecular methods (MALDI-TOF mass spectrometry and PCR/sequencing). At least 22 Culicoides species were detected. Four animal species (cow, sheep, goat and common blackbird) as well as humans were identified as hosts of Culicoides biting midges. The screening of 8291 Culicoides specimens in 99 pools for the presence of BTV and SBV RNA by reverse-transcription quantitative PCR were negative.ConclusionsThe biodiversity of Culicoides species in the natural reserve Stara Planina was high with at least 22 species present. The presence of C. imicola Kieffer was not recorded in this area. Culicoides showed opportunistic feeding behaviour as determined by host preference. The absence of SBV and BTV viral RNA correlates with the absence of clinical disease in the field during the time of sampling. These data are the direct outcome of a training programme within the Institutional Partnership Project AMSAR: Arbovirus monitoring, research and surveillance-capacity building on mosquitoes and biting midges funded by the programme SCOPES of the Swiss National Science Foundation.

Clinical and epizootiological examination of equine piroplasmosis on the central balkan region and its effect on host DNA damage

AB  - Equine piroplasmosis is a significant vector-prone disease with a high degree of widespread and economic impact on horse industry throughout the world. The causative agents of equine piroplasmosis are protozoas Theileria equi and Babesia caballi. The disease can be present subclinical in horses, but also it can be accompanied by a serious clinical picture. The goal of this doctoral dissertation was the comprehensive clinical and epizootiological examination of the equine piroplasmosis on the central Balkan region, where we would investigate the presence of parasites by molecular detection methods, to follow the haematological and biochemical parameters, the parameters of oxidative stress and at the end, the degree of host cells DNA damage due to parasites presence. In order to gain insight into the prevalence of the disease, we conducted a molecular epidemiological study involving 165 horses, from eight locations from three countries (Serbia, Montenegro, Bosnia and Herzegovina). First, we did a PCR method for the detection of a 450 bp long segment of the 18S rRNA gene which is present in all Theileria and Babesia species. For the definitive species identification, a multiplex PCR was done. In order to confirm obtained results, sequencing of positive samples was made. The overall prevalence of T. equi and B. caballi in the study area was 32.1% and 1.8% respectively. AB  - Piroplazmoza konja predstavlja značajno vektorski prenosivo oboljenje sa velikom raširenošću i ekonomskim uticajem na konjarstvo širom sveta. Uzročnici piroplazmoze konja su protozoe Theileria equi i Babesia caballi. Oboljenje može biti prisutno kod konja i supklinički, ali može biti praćeno i ozbiljnom kliničkom slikom. Cilj ove doktorske disertacije je bio opsežno kliničko- epizootiološko ispitivanje priplazmoze konja na prostoru centralnog Balkana, gde bi ispitivali prisustvo parazita molekularnim metodama detekcije, pratili hematološke i biohemijske parametre, parametre oksidativnog stresa i na kraju stepen oštećenja DNK ćelija domaćina usled prisustva parazita. U cilju sticanja uvida u prevalenciju oboljenja sproveli smo molekularnu epizootiološku studiju koja je uključivala 165 konja, sa osam lokaliteta sa prostora tri države (Srbija, Crna Gora, Bosna i Hercegovina). Prvo smo uradili PCR metodu za detekciju 450 bp dugog segmenta 18S rRNK gena koji je prisutan kod svih Theileria i Babesia vrsta. Za definitivnu detekciju vrste je urađen multipleks PCR. The causative agents of equine piroplasmosis are protozoas Theileria equi and Babesia caballi. The disease can be present subclinical in horses, but also it can be accompanied by a serious clinical picture. The goal of this doctoral dissertation was the comprehensive clinical and epizootiological examination of the equine piroplasmosis on the central Balkan region, where we would investigate the presence of parasites by molecular detection methods, to follow the haematological and biochemical parameters, the parameters of oxidative stress and at the end, the degree of host cells DNA damage due to parasites presence. In order to gain insight into the prevalence of the disease, we conducted a molecular epidemiological study involving 165 horses, from eight locations from three countries (Serbia, Montenegro, Bosnia and Herzegovina). First, we did a PCR method for the detection of a 450 bp long segment of the 18S rRNA gene which is present in all Theileria and Babesia species. For the definitive species identification, a multiplex PCR was done. In order to confirm obtained results, sequencing of positive samples was made. AB  - The main reasons for wildlife forensic research are animal poaching, illegal trade, and falsified game meat products. Small trace amounts, old and degraded materials present the most common samples in revealing criminal activities in this field. This is the reason why it is crucial to use adequate and reliable methods and samples to identify animal species killed outside the hunting season or species protected by law. In this study, different endpoint PCR and real-time PCR protocols were compared in the identification of three Cervidae species (Capreolus capreolus, Cervus elaphus, Dama dama) from old and damaged material found in an enclosed area where the animals were kept. From a total of 129 samples, end point PCR provided results for 119 samples, while real-time PCR was successful in all cases. Also, we created and tested a protocol for simultaneous analyses of different types of samples, which is of great importance as when the amplification is carried out simultaneously it is more cost efficient and speeds up the process.

AB  - Equine piroplasmosis in donkeys has been recognised as a serious problem of major economic importance. The present molecular study is the first investigation of the presence of Theileria equi and Babesia caballi in Balkan donkeys and of the possible haematological alterations related to it. A total of 70 apparently healthy donkeys from Serbia were included in this study. The overall prevalence of T. equi infection in donkeys tested with multiplex PCR was 50%. There was no B. caballi-positive sample. Infections in donkeys included in this study seem to be associated with decreased red blood cell count, haemoglobin concentration, haematocrit and platelet count, and with increased white blood cell count, mean corpuscular haemoglobin and mean corpuscular haemoglobin concentration. Altered haematological parameters in donkeys can lead to a decrease in working capacity and production performance. Further molecular research and long-term monitoring of equine piroplasmosis is needed in Serbia and throughout Europe.

AB  - Intestinal lymphangiectasia is an uncommon disease which can cause severe, chronic protein-losing enteropathy in dogs. Four dogs were presented at the Belgrade Clinic for Small Animals with clinical signs of chronic diarrhea, lethargy, anorexia, vomiting and weight loss. Abnormal physical examination findings included dehydration, signs of pain on abdominal palpation, and ascites. The most important clinicopathological findings were lymphopenia and hypoproteinemia with hypoalbuminemia. Abdominal ultrasound revealed intestinal abnormalities in all dogs. To establish an undoubted diagnosis of intestinal lymphangiectasia, endoscopy and histopathology were conducted.
AB  - Crevna limfangiektazija predstavlja retko oboljenje kod pasa koje može izazvati ozbiljne, hronične enteropatije sa gubitkom proteina. Na Klinici za male životinje u Beogradu su primljena četiri psa sa simptomima hronične dijareje, letargije, anoreksije, povraćanja i gubitka na težini. Opštim pregledom su ustanovljene promene u smislu dehidratacije, bolnosti na palpaciju abdomena i ascitesa. Najznačajnije kliničko-patološke promene su bile limfopenija i hipoproteinemija sa hipoalbuminemijom. Ultrazvučnim pregledom abdomena su ustanovljene promene na crevima kod svih pasa. Da bismo postavili nesumljivu dijagnozu crevne limfangiektazije urađena je endoskopija i histopatologija.

AB  - Psittacine beak and feather disease (PBFD) affects a large number of Psittaciformes species. In this study, five White Cockatoo parrots (Cacatua alba) with clinical signs of PBFD were examined. After euthanasia, a full necropsy of parrots was performed and organs with macroscopic changes were sampled for routine histopathological evaluation. To confirm the presence of psittacine beak and feather disease virus (PBFDv), feather samples were analyzed with the PCR method. Sequence analysis of the obtained PCR products indicated their close relationship (99%) to other PBFDv isolates. Six variable nucleotide sites were discovered, two missense and four silent mutations. This paper presents the evidence of new PBFDv sequence in Cockatoo species.

Molecular detection and investigation of the presence of Dirofilariosis causative agent in dogs on the territory of Belgrade

AB  - Dirofilariosis in dogs is a parasitic disease caused by the strains Dirofilaria immitis and D. Repens. Dirofilaria immitis parasitizes in the right side of the heart and pulmonary artery, causing considerable functional impairment of the heart, while adult forms of D. repens mostly parasitize in subcutaneous tissue causing minor skin damage. In this investigation, during the period from May to November 2015, 60 dogs of different race from the territory of Belgrade were examined. Ater the clinical examination, the blood was sampled for microfilariae detection by Knott test, as well as for molecular detection of the agent. Main clinical signs were: fatigue during exersise, shortness of breath and dry cough, anorexia, rapid heartbeat and skin changes. Microfilariae were detected in 22 dogs (37%) by modified Knott test. By molecular genetic method there was determined the prevalence of D. immitis 42%, and D. repens 22%. In this investigation 7 dogs were negative on Knott test, but by PCR method they showed a positive reaction on the presence of species of the genus Dirofilaria. On the basis of the obtained results, the application of molecular-genetic methods is justified in accurate and reliable species identification of Dirofilaria genus.
AB  - Dirofilarioza pasa je parazitsko oboljenje izazvano vrstama Dirofilaria immitis i D. repens. D. immitis parazitira u desnoj strani srca i plućnoj arteriji, dovodeći do značajnog funkcionalnog oštećenja srca, dok adultni oblici D. repens uglavnom parazitiraju u subkutanom tkivu dovodeći do manjih oštećenja kože. U ovom istraživanju ispitano je 60 pasa, različitih rasa, sa teritorije grada Beograda, u periodu od maja do novembra 2015. godine. Nakon kliničkog pregleda, krv je uzorkovana za detekciju mikrofilarija modifikovanim Knott-ovim testom, kao i za molekularnu detekciju uzročnika. Glavni klinički znaci su bili: brzo zamaranje tokom treninga, otežano disanje i suv kašalj, anoreksija, ubrzan rad srca i promene na koži. Mikrofilarije su detektovane kod 22 psa (37%) modifikovanim Knott-ovim testom. Molekularno genetskom metodom utvrđena je prevalencija D.immitis 42%, a D. repens 22 %. U sprovedenom istraživanju je bilo 7 pasa koji su bili negativni na Knott testu, dok su metodom PCR pokazali pozitivnu reakciju na prisustvo vrsta iz roda Dirofilaria. Na osnovu dobijenih rezultata primena molekularno-genetskih metoda potvrđuje svoju opravdanost u preciznoj i pouzdanoj specijskoj identifikaciji roda Dirofilaria.

Molecular detection of Babesia spp. in ticks sampled from asymptomatic dogs in the area of some Belgrade municipalities

AB  - Babesiosis of domestic animals is a vector transmissible and clinically significant disease, caused by protozoa of genus Babesia and Theileria. Possible causative agents for this disease in dogs in Europe are: Babesia canis, B. gibsoni, B. vogeli and B. microti-like. Diagnostics of babesiosis of dogs was for a long time based on the visual inspection of stained blood smear under a microscope, while today there have been increasingly used molecular methods of detection in precise, species diagnostics. The objective of this work was molecular detection of the cause of babesiosis of dogs in the ticks sampled from asymptomatic dogs in the region of some Belgrade municipalities, all for better understanding of epizootiological situation. From three sites in Belgrade, there were collected 49 ticks, sampled from the dogs with no symptoms. There was carried out the determination of the ticks, and after that, DNA was isolated for molecular examination. First, theret was performed Polymerase Chain Reaction (PCR), for determining the species of the genus Babesia, and after that there was also carried out the determining of polymorphism in the length of restriction fragments (RFLP) for the purpose of the causative agent species determination. Out of the total number of the examined ticks, 18,34% were positive on Babesia spp. By RFLP method, in two cases (4,08%) B. Gibsoni was identified, while in 7 cases (14,92%) there were no restriction sites for the used enzymes, what suggests that most likely it was B. canis. The ticks positive on the cause of babesiosis were: Dermacentor reticulatus (4 cases), Rhipicephalus sanguineus (4 cases) i Ixodes ricinus (1 case). This work confirms the presence of Babesia spp. in the ticks sampled from asmptomatic dogs on the territory of Belgrade as well as the significance of PCR-RFLP method in diagnostics and identification of the causative agent of babesiosis in dogs. For the first time in Serbia, there was determined the presence of B. gibsoni in ticks (Species Rhipicephalus sanguineus).
AB  - Babezioza domaćih životinja predstavlja vektorski prenosivo i klinički veoma značajno oboljenje, uzrokovano protozoama rodova Babesia i Theileria. Uzročnici koji mogu da izazovu oboljenje kod pasa u Evropi su: Babesia canis, B. gibsoni, B. vogeli i B. microti-like. Dijagnostika babezioze pasa se dugo vremena bazirala na vizuelizaciji uzročnika pregledom obojenog krvnog razmaza pod mikroskopom, dok se danas sve više koriste molekularne metode detekcije u preciznoj, specijskoj dijagnostici. Cilj ovog rada je bila molekularna detekcija različitih uzročnika babezioze pasa u krpeljima uzorkovanim sa asimptomatskih pasa na prostoru određenih beogradskih opština, radi boljeg razumevanja epizootiološke situacije. Sa tri lokacije u Beogradu je prikupljeno 49 krpelja, uzorkovanih sa pasa bez simptoma bolesti. Izvršena je determinacja krpelja, a nakon toga je izolovana DNK za molekularna ispitivanja. Pozitivni krpelji na uzročnike babezioze su bili Dermacentor reticulatus (4 slučaja), Rhipicephalus sanguineus (4 slučaja) i Ixodes ricinus (1 slučaj). Ovaj rad potvrđuje prisustvo Babesia spp. u krpeljima uzorkovanim sa asimptomatskih pasa na teritoriji grada Beograda, kao i značaj PCR-RFLP metode u dijagnostici i identifikaciji uzročnika babezioze pasa. Molecular detection and prevalence of Theileria equi and Babesia caballi in horses of central Balkan

AB  - Equine piroplasmosis is significant tick-borne disease with wide distribution. The prevalence of equine piroplasmosis in Serbia, Montenegro and Bosnia and Herzegovina is unknown. In aim to obtain a first insight into the prevalence we performed molecular epidemiological study which included 142 horses, on seven locations in these three countries. We first performed PCR for the detection of a 450bp long section of the 18S rRNA of piroplasma-specific region. For all positive samples we have done multiplex PCR for the species detection. Species determination was further confirmed by sequencing PCR products of 10 randomly selected Theileria equi and all Babesia caballi samples. The overall prevalence rates in analysed region for T. equi and B. caballi were 22.5% and 2.1%, respectively. Possible risk factors (such as location, age, sex and activity) associated with PCR positivity were evaluated. Marked differences were found in prevalence between geographic areas. There was no significant association between positivity and age group. T. equi was more prevalent in females and farming horses. This is the first report on the molecular survey of T. equi and B. caballi in central Balkan. Further prevalence studies on definitive host and vectors in this region are necessary.

abstract = "Equine piroplasmosis is significant tick-borne disease with wide distribution. The prevalence of equine piroplasmosis in Serbia, Montenegro and Bosnia and Herzegovina is unknown. In aim to obtain a first insight into the prevalence we performed molecular epidemiological study which included 142 horses, on seven locations in these three countries. We first performed PCR for the detection of a 450bp long section of the 18S rRNA of piroplasma-specific region. For all positive samples we have done multiplex PCR for the species detection. Species determination was further confirmed by sequencing PCR products of 10 randomly selected Theileria equi and all Babesia caballi samples. The overall prevalence rates in analysed region for T. equi and B. caballi were 22.5% and 2.1%, respectively. Possible risk factors (such as location, age, sex and activity) associated with PCR positivity were evaluated. Marked differences were found in prevalence between geographic areas. There was no significant association between positivity and age group. T. equi was more prevalent in females and farming horses. This is the first report on the molecular survey of T. equi and B. caballi in central Balkan. Further prevalence studies on definitive host and vectors in this region are necessary.",

Oxidative stress and DNA damage in horses naturally infected with Theileria equi

AB  - The aim of this study was to determine the concentrations of oxidative stress parameters and DNA damage in horses infected by Theileria equi. Initial screening of 110 horses with duplex PCR enabled the selection of 30 infected horses with T. equi and 30 free of infection (control). Specimens from the 60 horses were further analysed by determining the following oxidative stress parameters: extent of haemolysis (EH), plasma free haemoglobin (PHb), catalase (CAT), Cu,Zn superoxide dismutase (SOD1), paraoxonase (PON1), nitrite (NO2-), total nitrate and nitrite (NOx), malondialdehyde (MDA) and free thiol groups (-SH). In addition, relative distribution of lactate dehydrogenase (LDH1-LDH5) activity and the DNA-damaging effects of T. equi infection were evaluated. Compared to control horses, horses infected with T. equi had significantly higher SOD1 activities (P < 0.05) and PHb (P < 0.01), NO2- (P < 0.001), NOx (P < 0.05) and MDA concentrations (P < 0.001), and significantly-lower EH (P < 0.001), CAT (P < 0.01) and PON1 (P < 0.001) activities, and thiol group concentrations (P < 0.05). The comet assay demonstrated significantly increased DNA damage in T. equi infected cells compared to non-infected cells (P < 0.001). Infected horses had significantly increased LDH5 isoenzyme activities (P < 0.05). There was higher production of ROS/RNS in T. equi-infected horses, which resulted in changes in osmotic fragility, damage to lipids, proteins and DNA, haemolysis and hepatocellular damage. Oxidative stress in horses naturally infected with T equi could contribute to the pathogenesis of the infection.

AB  - Polycystic kidney disease (PKD) is an inherited autosomal disorder in cats, mostly diagnosed in Persian cats. Renal cysts can be diagnosed by ultrasound, but cats must be at least 16 weeks old. The goals of this study were to assess the occurrence of PKD in Serbia using a randomly selected group of Persian cats, to compare the diagnostic efficacy of ultrasound and genetic tests, and to measure haematological and selected biochemical parameters. We examined 70 cats of Persian breed, between 4 months and 8 years of age. Complete blood count and selected biochemical parameters were measured, renal ultrasound was performed. Swabs of the oral cavity were obtained for genetic testing. Percentage of PKD positive cats identified by genetic testing was 48.6%, whilst only 18.6% were detected through ultrasound. Animals that were polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) positive and ultrasound negative ranged from 4 months to 3.5 years. All haematological and biochemical parameters were within the the normal range values in all examined cats. Genetic methods proved to be the most effective for reliable and early diagnosis of PKD in Persian cats. DNA analysis can be used right after birth, and excludes the need for other diagnostic procedures, such as ultrasound.

Clinical babesiosis and molecular identification of babesia canis and babesia gibsoni infections in dogs from Serbia

AB  - Canine babesiosis is a frequent and clinically significant tick-borne disease. Sixty symptomatic dogs with clinical findings compatible with babesiosis were included in this study conducted in Serbia. After clinical examination, blood samples were taken for microscopic examination, complete blood count (CBC), Canine SNAP 4Dx Test, DNA analyses and sequencing. The main clinical signs included apathy, anorexia, fever, brown/red discoloration of urine, pale mucous membranes, icterus, splenomegaly, and vomiting. The main clinicopathological findings in Babesia infections were a slight to severe thrombocytopenia and a mild to very severe normocytic normochromic anaemia. Microscopic evaluation revealed 58 positive samples with the presence of large and small intraerythrocytic piroplasms in 57 and 1 sample(s), respectively. No co-infections were found using SNAP test. Two Babesia species, B. canis (58/60) and B. gibsoni (2/60), were differentiated by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Species identification was further confirmed by sequencing PCR products of B. gibsoni samples and six randomly selected B. canis samples. All dogs were treated with imidocarb dipropionate (6.6 mg/kg of body weight), given intramuscularly twice at an interval of 14 days. This report presents the first molecular evidence of the occurrence of B. gibsoni and B. canis, confirmed by DNA sequencing, in sick dogs from Serbia.

AB  - The aim of this study was to evaluate the parameters of intraoperative and postoperative pain in bitches undergoing laparoscopic or conventional ovariectomy. The study was conducted on 20 healthy, adult bitches of medium and large breeds. All the bitches were subjected to the same anesthetic protocol. Carprofen was applied preoperatively, and general anaesthesia was carried out with a combination of medetomidine, propofol and sevoflurane. The bitches were divided into two groups of 10 individuals each. The first group underwent conventional ovariectomy while the second one was subjected to the laparoscopic procedure. The intensity of intraoperative pain was estimated at different time points by measuring changes in heart and respiratory rate, arterial blood pressure, and body temperature. A multifactor pain scale was used to assess the intensity of postoperative pain. Dogs in the group which was scheduled for conventional ovariectomy (COV group) responded to painful intra-operative stimuli with increased heart rate and respiratory rate. Arterial blood pressure and body temperature decrease was detected in COV group. Laparoscopic ovariectomy induced less intra- and postoperative pain when compared to conventional ovariectomy. Beside that, the total anaesthesia time and postoperative recovery were shorter in dogs that underwent laparascopic ovariectomy.
