AB  - Zarastanje rana je složen proces koji uključuje koordinaciju između različitih tipova ćelija i odgovarajućeg mikrookruženja. Uprkos
visokom regenerativnom potencijalu kože, defekti u vidu dubokih rana ili opekotina, teško spontano zarastaju i zahtevaju terapijski tretman. I pored napretka u bioinžinjeringu kože i intenzivnih istraživanja
u cilju iznalaženja novih biomaterijala, lečenje hroničnih rana i dalje
predstavlja aktuelan klinički problem. Hidrogelni zavoji, karakteristične
porozne i hidratisane molekularne strukture, verno oponašaju mikrookruženje rane, te su se izdvojili kao obećavajuća alternativa i najbolji
kandidati za stimulisanje procesa zarastanja. Hidrogelovi su bazirani
na prirodnim i/ili sintetičkim polimerima, i u vidu trajnih ili privremenih
zavoja podržavaju regeneraciju i zarastanje oštećene kože. Uvođenje
nanotehnologije je omogućilo nove pristupe u sintezi i dobijanje in situ
hibridnih hidrogelova, odličnih svojstava i prilagodljive funkcionalnosti.
Istraživanja u cilju razvoja hidrogelnih zavoja sa mogućnošću procene
mikrookruženja rane i prilagođenog tretmana na zahtev, predstavljaju ključ napretka u polju zbrinjavanja hroničnih rana. Pretklinička ispitivanja in vivo su neophodan uslov za finalnu karakterizaciju svakog
novodizajniranog hidrogela.
Ovaj rad razmatra prednosti upotrebe hidrogelnih zavoja i napretke u razvoju inteligentnih tretmana za rane sa odloženim zarastanjem. Takođe, jedan deo rada je posvećen proceni hidrogelova in vivo, kao i ključnoj ulozi veterinara tokom pretkliničkih ispitivanja na eksperimentalnim animalnim modelima.

Komparativni prikaz animalnih modela u morfološkim analizama placentacije

AB  - Svrha ovog pregleda je da prikaže veliku raznolikost u strukturi i funkciji placente koja se sreće kod sisara, i podobnosti nekih vrsta kao animalnih modela u proučavanju placentacije. Postoje značajne razlike u strukturi i fiziologiji posteljice između čoveka, domaćih i eksperimentalnih životinja. Zbog toga, najčešće upotrebljavani animalni modeli za proučavanje placentacije (miš, kunić, zamorac), nisu adekvatni za sva istraživanja u veterinarskoj i humanoj medicini. Placente se klasifikuju prema svom obliku, strukturi i interhemalnom regionu. Horionske resice placente su obložene prizmatičnim epitelom i preko njega se odvija prenos svih supstanci od majke prema plodu. Ćelije ovog epitela su selektivno propustljive kako za korisne, tako i za potencijalno toksične materije.
	Istraživanja koja se bave analizama i osobenostima placentacije su oskudna, posebno sa aspekta kontakta i razmene materija između majke i ploda. Iz tog razloga, ovaj uvid može da pomogne u sagledavanju različitih problema u kliničkoj praksi i tretmanu koji su bitni za dobijanje zdravog potomstva.

AB  - Epidermis stem cells have a crucial role through the processes of proliferation and
differentiation, to replace cells that are constantly lost during tissue turnover or
following injury. On the other hand, thyroid hormones regulate the proliferation and
differentiation of epidermal cells and thus significantly influence the homeostasis
of the skin. It is well known that maternal hypothyroidism during pregnancy leads
to impaired development of many organ systems in their offspring. However, there
is a lack of data about the influence of maternal subclinical hypothyroidism during
pregnancy and lactation on the development of the skin and its derivatives in the litter.
The aim of this study was to investigate the effects of maternal thyroid dysfunction on
the development of the skin and its derivatives in their offspring in the early postnatal
period. Antithyroid substance 6-n-propyl-2-thiouracil was added into the drinking water
to female Albino Oxfords rats from the beginning of pregnancy and during lactation,
with the aim to induce subclinical and overt form of hypothyroidism. Skin samples
were taken from male pups within twenty-four hours and seven days after birth. The
main findings of this investigation were that both forms of maternal hypothyroidism
lead to serious damage of the epidermis in pups in terms of pronounced hyperkeratosis
and reduction of the germinal layer along with a reduced number of hair follicles and
their delayed morphogenesis. Epidermal impairments were more pronounced in pups
with the overt form of hypothyroidism while offspring with the subclinical form had
impairments that were less pronounced and delayed in occurrence.

AB  - Biocompatibility of materials is one of the most important conditions for their successful application in tissue regeneration
and repair. Cell-surface interactions stimulate adhesion and activation of macrophages whose acquaintance can assist in
designing novel biomaterials that promote favorable macrophage–biomaterial surface interactions for clinical application.
This study is designed to determine the distribution and number of macrophages as a means of biocompatibility evaluation
of two newly synthesized materials [silver/poly(vinyl alcohol) (Ag/PVA) and silver/poly(vinyl alcohol)/graphene (Ag/PVA/
Gr) nanocomposite hydrogels] in vivo, with approval of the Ethics Committee of the Faculty of Veterinary Medicine,
University of Belgrade. Macrophages and giant cells were analyzed in tissue sections stained by routine H&E and im munohistochemical methods (CD68+
). Statistical relevance was determined in the statistical software package SPSS 20
(IBM corp). The results of the study in terms of the number of giant cells localized around the implant showed that their
number was highest on the seventh postoperative day (p.o.d.) in the group implanted with Ag/PVA hydrogels, and on the
30th p.o.d. in the group implanted with Ag/PVA/Gr. Interestingly, the number of macrophages measured in the capsular and
pericapsular space was highest in the group implanted with the commercial Suprasorb© material. The increased mac rophage number, registered around the Ag/PVA/Gr implant on 60th p.o.d. indicates that the addition of graphene can, in a
specific way, modulate different biological responses of tissues in the process of wound healing, regeneration, and
integration.

and repair. Cell-surface interactions stimulate adhesion and activation of macrophages whose acquaintance can assist in
designing novel biomaterials that promote favorable macrophage–biomaterial surface interactions for clinical application.
This study is designed to determine the distribution and number of macrophages as a means of biocompatibility evaluation
of two newly synthesized materials [silver/poly(vinyl alcohol) (Ag/PVA) and silver/poly(vinyl alcohol)/graphene (Ag/PVA/
Gr) nanocomposite hydrogels] in vivo, with approval of the Ethics Committee of the Faculty of Veterinary Medicine,
University of Belgrade. Macrophages and giant cells were analyzed in tissue sections stained by routine H&E and im munohistochemical methods (CD68+
). Statistical relevance was determined in the statistical software package SPSS 20
(IBM corp). The results of the study in terms of the number of giant cells localized around the implant showed that their
number was highest on the seventh postoperative day (p.o.d.) in the group implanted with Ag/PVA hydrogels, and on the
30th p.o.d. in the group implanted with Ag/PVA/Gr. Interestingly, the number of macrophages measured in the capsular and
pericapsular space was highest in the group implanted with the commercial Suprasorb© material. The increased mac rophage number, registered around the Ag/PVA/Gr implant on 60th p.o.d. indicates that the addition of graphene can, in a
specific way, modulate different biological responses of tissues in the process of wound healing, regeneration, and
integration.",

AB  - The application of histological methods in meat composition analysis and identification of prohibited tissues and organs added to meat and meat products is still in the research phase, although there have been some promising results. The aim of this work was to assess the possibility of using histological and histochemical methods for analysis of kulen composition. In this research, six samples of kulen were examined, one of which was produced in domestic conditions, while the rest were commercial products sampled from local markets. The samples were carried through classical histological preparation. The obtained slides were stained with haematoxylin/eosin, Masson-Goldner, toluidine blue and periodic acid-Schiff/alcian blue. The content of muscle, fat and connective tissue was evaluated using histomorphometric analysis. Histological analysis of kulen composition determined the presence of the following structures: muscle, adipose and connective tissues, blood vessels, glandular epithelium, peripheral nerve, cartilage and plant tissue. The histomorphometric analysis showed that the kulen products contained on average 56±2.52% muscle tissue, 7.27±1.38% connective tissue and 19.82±3.24% adipose tissue. The results show that by applying histological methods it is possible to identify different permitted and prohibited animal tissues in kulen, and hence, it is possible to analyse the composition of kulen. It is also possible to confirm the presence of various plant tissues, but for their precise identification, additional histological methods are needed.
AB  - Primena histoloških metoda u analizi sastava mesa i utvrđivanju prisustva nedozvoljenih tkiva u mesu i proizvodima od mesa je još uvek u fazi istraživanja, iako su postojeći rezultati u ovoj oblasti obeć avajuć i. Cilj našeg istraživanja bio je ispitivanje moguć nosti korišć enja histoloških i histohemijskih metoda u analizi sastava fermentisane kobasice u tipu kulena. U ovom istraživanju ispitano je šest uzoraka kulena, od kojih je jedan proizveden u domać im uslovima, dok su ostali komercijalni proizvodi poticali iz lokalnih marketa. Uzorci su sprovedeni klasičnom histološkom procedurom. Dobijeni isečci bojeni su metodama: hematoksilin/ eosin,Masson-Goldner, toluidin plavo i perjodna kiselina/Schiff/alcijan plavo. Sadržaj mišić nog, masnog i vezivnog tkiva procenjen je primenom histomorfometrijskih analiza. Histološkom analizom sastava kulena utvrđeno je prisustvo sledeć ih struktura: mišić no, masno i vezivno tkivo, krvni sudovi, epitel žlezdanog tipa, periferni nerv, hrskavica i biljno tkivo. Histomorfometrijska analiza je pokazala da proizvodi sadrže u proseku 56±2,52% mišić nog tkiva, 7,27±1,38% vezivnog tkiva i 19,82±3,24% masnog tkiva. Rezultati pokazuju da je primenom histoloških metoda moguć e identifikovati različita dozvoljena i nedozvoljena životinjska tkiva u kulenu, pa je samim tim moguće analizirati sastav kulena. Takođe je moguć e potvrditi prisustvo različitih biljnih tkiva, ali je za njihovu preciznu identifikaciju neophodno primeniti dodatne metode.

AB  - Overt and subclinical maternal hypothyroidism affects cartilage and bone extracellular matrix formation during fetal endochondral bone development. Less is known about the
effect maternal hypothyroidism has on cell proliferation and death of growth plate chondrocytes.
This study aimed to evaluate the expression of proliferation (Ki67), autophagy (LC3) and apoptosis (caspase 3) markers in the epiphyseal cartilage of the tibia in pups from 
subclinical and overt hypothyroid does, in their early postnatal development.Hypothyroidism was induced with propylthiouracil using low (1.5 mg/L) for subclinical (H1) and high dose (150 mg/L) for overt (H2) form. Propylthiouracil was
administered through drinking water in pregnant Albino Oxford does from the first day of gravidity and during lactation. Control (C) group was not treated. Six, seven-day-old male
pups from each group were euthanized. Histological examinations were performed on paraffin sections of the proximal tibial growth plate. Immunohistochemistry was used to assess
the expression of Ki67, LC3 and caspase 3. The number of Ki67 positive chondrocytes was evaluated in the resting and proliferating zone using Image J. The expression of LC3 was
performed by counting the number of positive dots in chondrocytes.The number of Ki67 positive chondrocytes was higher in both hypothyroid groups, while the number of LC3 positive dots per cell was lower indicating that the chondrocyte
cell cycle in hypothyroid animals is faster and that the basal level of autophagy is reduced due to frequent mitosis. Compared to controls, caspase 3 expression in the terminal
hypertrophic chondrocytes was reduced in H1 and completely absent in H2 pups which might indicate impaired differentiation.We conclude that both forms of maternal hypothyroidism
in rats lead to accelerated proliferation, slowed autophagy and
compromised apoptosis of terminal hypertrophic chondrocytes in the
early infantile period and a delay in cartilage to bone transition.

AB  - A newly produced biomaterial is necessarily subject of standards, which are performed in vivo on animal models. For the evaluation of soft tissue regenerative possibilities after subcutaneous implantation of biomaterials – silver/poly(vinyl alcohol) (Ag/PVA) and novel silver/poly(vinyl alcohol)/graphene (Ag/PVA/Gr) provided for clinical use, sixteen rats were used, according to the instructions of international standards, ISO 10993-6, 2007. Histological sections were observed 7, 15, 30 and 60 days after grafting. These hydrogels were produced by in situ electrochemical synthesis of silver nanoparticles in the polymer matrices, which enabled obtaining completely safe and biocompatible materials, free from any additional toxic chemical reducing agents. Surgical implantation of hydrogels was done according to the permission of the Ethical Committee of the Faculty of Veterinary Medicine, University of Belgrade. Immunohistochemical (IHC) studies included the assessment of  smooth muscle expression actin in blood vessels (α-SMA), the expression of laminin and type I and type III collagen in the skin structures, and, the determination of cell proliferation marker expression (Ki-67) keratinocytes. The results were assessed in a semiquantitative manner. The data were analyzed in the statistical software package IBM SPSS 20. The conclusions indicated that Ag/PVA/Gr might be used as wound dressings to enhance the tissue
AB  - Bilo koji novoproizvedeni biomaterijal nužno podleže standardima, po čijim se protokolima sprovode in vivo istraživanja na životinjskim modelima. Za procenu mogućnosti regeneracije mekog tkiva nakon potkožne implantacije biomaterijala - srebra/poli(vinil alkohola) (Ag/PVA) i novog materijala srebra/poli(vinil alkohola)/grafena (Ag/PVA/ Gr) predviđenog za kliničku upotrebu, šesnaest pacova je korišćeno, po uputstvima međunarodnog standarda, ISO 10993-6 2007. Histološki uzorci su sakupljeni 7, 15, 30 i 60 dana nakon implantacije. Ovi hidrogelovi su proizvedeni in situ elektrohemijskom sintezom nanočestica srebra u polimernim matricama, što je omogućilo dobijanje potpuno sigurnih i biokompatibilnih materijala, bez ikakvih dodatnih toksičnih hemijskih sredstava za redukciju. Hirurška implantacija hidrogela urađena je prema dozvoli Etičkog komiteta Fakulteta veterinarske medicine Univerziteta u Beogradu. Imunohistohemijske (IHC) studije uključivale su procenu ekspresije aktina glatkih mišića u krvnim sudovima (α-SMA), ekspresiju laminina i kolagena tipa I i III u strukturama kože i određivanje ekspresije markera proliferacije (Ki-67) keratinocita. Rezultati su procenjeni semikvantitativnom analizom. Podaci su analizirani u statističkom softverskom paketu IBM SPSS 20. Zaključci su ukazali da se Ag/PVA/Gr može koristiti kao obloga za rane kako bi se povećao potencijal zarastanja tkiva i uspostavila brža integracija i kraće zadržavanje u tkivu.

abstract = "A newly produced biomaterial is necessarily subject of standards, which are performed in vivo on animal models. For the evaluation of soft tissue regenerative possibilities after subcutaneous implantation of biomaterials – silver/poly(vinyl alcohol) (Ag/PVA) and novel silver/poly(vinyl alcohol)/graphene (Ag/PVA/Gr) provided for clinical use, sixteen rats were used, according to the instructions of international standards, ISO 10993-6, 2007. Histological sections were observed 7, 15, 30 and 60 days after grafting. These hydrogels were produced by in situ electrochemical synthesis of silver nanoparticles in the polymer matrices, which enabled obtaining completely safe and biocompatible materials, free from any additional toxic chemical reducing agents. Surgical implantation of hydrogels was done according to the permission of the Ethical Committee of the Faculty of Veterinary Medicine, University of Belgrade. Immunohistochemical (IHC) studies included the assessment of  smooth muscle expression actin in blood vessels (α-SMA), the expression of laminin and type I and type III collagen in the skin structures, and, the determination of cell proliferation marker expression (Ki-67) keratinocytes. The results were assessed in a semiquantitative manner. The data were analyzed in the statistical software package IBM SPSS 20. The conclusions indicated that Ag/PVA/Gr might be used as wound dressings to enhance the tissue, Bilo koji novoproizvedeni biomaterijal nužno podleže standardima, po čijim se protokolima sprovode in vivo istraživanja na životinjskim modelima. Za procenu mogućnosti regeneracije mekog tkiva nakon potkožne implantacije biomaterijala - srebra/poli(vinil alkohola) (Ag/PVA) i novog materijala srebra/poli(vinil alkohola)/grafena (Ag/PVA/ Gr) predviđenog za kliničku upotrebu, šesnaest pacova je korišćeno, po uputstvima međunarodnog standarda, ISO 10993-6 2007. Histološki uzorci su sakupljeni 7, 15, 30 i 60 dana nakon implantacije. Ovi hidrogelovi su proizvedeni in situ elektrohemijskom sintezom nanočestica srebra u polimernim matricama, što je omogućilo dobijanje potpuno sigurnih i biokompatibilnih materijala, bez ikakvih dodatnih toksičnih hemijskih sredstava za redukciju. Hirurška implantacija hidrogela urađena je prema dozvoli Etičkog komiteta Fakulteta veterinarske medicine Univerziteta u Beogradu. Imunohistohemijske (IHC) studije uključivale su procenu ekspresije aktina glatkih mišića u krvnim sudovima (α-SMA), ekspresiju laminina i kolagena tipa I i III u strukturama kože i određivanje ekspresije markera proliferacije (Ki-67) keratinocita. Rezultati su procenjeni semikvantitativnom analizom. Podaci su analizirani u statističkom softverskom paketu IBM SPSS 20. Zaključci su ukazali da se Ag/PVA/Gr može koristiti kao obloga za rane kako bi se povećao potencijal zarastanja tkiva i uspostavila brža integracija i kraće zadržavanje u tkivu.",

AB  - Tissue engineering strategies for a treatment of large bone defects, which is currently implantation of
bone grafts, are based on the development of biomaterials that mimic the bone tissue and improve
regeneration. 3D printed macroporous titanium-based scaffolds, designed specifically for large bone
defect implantation, provide both good mechanical support and architecture of pores suitable for
ingrowth of bone tissue and better osseointegration. However, the bioinert surface of Ti6Al4V alloy
is not the finest substrate for the growth of cells and mineral matrix deposition.
The aim of this study was to modify the surface of the macroporous Ti scaffold, by coating it with
multiple layers of bioceramics: silicate bioactive glasses (BAG) and Ag-doped calcium
hydroxyapatite (AgHAP), in order to provide bioactivity, biocompatibility, and antibacterial
properties. A dip-coating technique was optimized to provide a thin coating that is homogenous and
uniform inside the porous structure of titanium samples, followed by its melting at optimized
temperature to acquire a continuous BAG layer covered with AgHAP particles.
The characterization of the synthesized bioceramic powders and surface morphology of specimen
were conducted using scanning electron microscopy (SEM) and energy-dispersive X-ray
spectroscopy. The bioactivity was evaluated as the growth of hydroxyapatite crystals on the surface
of the samples after 14 days in Simulated body fluid (SBF), which was determined using SEM.
Biocompatibility of the obtained coatings and bare Ti samples was analyzed in vitro on the L929 cell
line, applying the MTT test. Finally, the antibacterial properties of coated scaffolds were evaluated on
E.coli, S. aureus, and C. albicans.
Evaluation of the morphology of the coatings showed that the homogenous deposition was achieved
by a dip-coating method. Improved bioactivity, biocompatibility, and antimicrobial properties of the
coated scaffold showed the great potential of this approach for future clinical application.

AB  - In this paper, we have shown for the first time the soft tissue response of novel silver/poly(vinyl alcohol)/graphene (Ag/PVA/Gr) and silver/poly(vinyl alcohol)/chitosan/graphene (Ag/PVA/CHI/Gr) nanocomposite hydrogels aimed for medical applications. These novel hydrogels were produced by in situ electrochemical synthesis of silver nanoparticles in the polymer matrices as described in our previously published works. Both Ag/PVA/Gr and Ag/PVA/CHI/Gr, as well as controls Ag/PVA, Ag/PVA/CHI and commercial Suprasorb (c) hydrogel discs, were implanted in the subcutaneous tissue of rats. Implants with the surrounding tissue were dissected after post-implantation on days 7, 15, 30 and 60, and then processed for histological examination. The tissue irritation index (TIrI) score, according to ISO 10993-6, 2007, as well as the number of leukocytes in the peri-implant zone and connective tissue capsule thickness were examined. The results show that each TIrI score, the leukocyte number around the implanted materials and capsule thickness gradually decreased during the observation period. At the endpoint of follow-up, the Ag/PVA/CHI/Gr implant was surrounded with a thinner capsule, while both the TIrI score and the number of leukocytes of the peri-implant zone were greater compared to the Ag/PVA/Gr implant. Despite the observed differences, we can conclude that our in vivo experiment suggested that both novel hydrogels were biocompatible and suitable for medical use.

Thyroid hormones affect the proliferation and/or mobilization of bulge stem cell population

AB  - Introduction. Thyroid hormones (TH) play a pivotal role in the development of mammalian skin and are necessary for both, the initiation and maintenance of hair growth. Hypothyroid rats showed  impaired epidermal proliferation, hair growth, and wound healing. Stem cells located at the bulges of the hair follicles are responsible for hair cycling and contribute to the regeneration of the new epidermis after wounding. Therefore a reduction in the num¬ber or function of the bulge stem cells could be responsible for this phenotype. 
We hypothesized that decrease in TH during prenatal development would affect bulge stem cell population.
Material and methods. Subclinical hypothyroidism was induced with propylthiouracil through drinking water in dose 1.5 mg/l in pregnant Albino Oxford rats from the first day of gravidity. Control group was untreated. Seven-day-old pups were euthanized and skin samples were taken from dorsal part of their bodies. For the immunohistochemical study PCNA (Santa Cruz Biotechnology) and NANOG (Thermo scientific) antibodies were used.
Results. . The number of hair follicles was reduced in hypothyroid pups compared to 
controls. They also had a decreased expression of PCNA in cells of inner and outer sheat of hair follicles, but increased number of NANOG positive bulge stem cells. 
Conclusion. This suggests that re¬duced activation of stem cells leading to their accumulation in the bulges which is associated with reduced stem cell prolif¬eration and mobilization.

We hypothesized that decrease in TH during prenatal development would affect bulge stem cell population.
Material and methods. Subclinical hypothyroidism was induced with propylthiouracil through drinking water in dose 1.5 mg/l in pregnant Albino Oxford rats from the first day of gravidity. Control group was untreated. Seven-day-old pups were euthanized and skin samples were taken from dorsal part of their bodies. For the immunohistochemical study PCNA (Santa Cruz Biotechnology) and NANOG (Thermo scientific) antibodies were used.
Results. . The number of hair follicles was reduced in hypothyroid pups compared to 
controls. They also had a decreased expression of PCNA in cells of inner and outer sheat of hair follicles, but increased number of NANOG positive bulge stem cells. 
Conclusion. This suggests that re¬duced activation of stem cells leading to their accumulation in the bulges which is associated with reduced stem cell prolif¬eration and mobilization.",

The effects of maternal hypothyroidism on development and function of different organs of their offspring are well known. It is unclear how its subclinical form affects some of these processes, including endochondral ossification during the prenatal and early postnatal development.
The aim of the study was to assess bone formation by defining the expression of tartarate-resistant acid phosphatase (TRAP), alkaline phosphatase (ALP) and mRNA for dentin matrix acidic phosphoprotein (DMP1) in tibia of pups from subclinical hypothyroid dams in early postnatal development. Subclinical hypothyroidism (SHT) was induced with propylthiouracil through drinking water (1.5 mg/l) in pregnant Albino Oxford dams from the first day of gravidity and during lactation. Control group was untreated. Five seven-day-old pups from each experimental group were euthanized. All histological examinations were performed on paraffin sections of the proximal tibial growth plate. The area of TRAP positive signal per 1µm2 and optical density of ALP immunostaining were evaluated using the Image J. The number of DMP1 positive cells was counted for given area using the same software. Level of thyroxine (T4) and triiodothyronine (T3) of dams and pups were decreased (data published). Expression of TRAP and ALP in the proximal tibial growth plate were lower in hypothyroid pups and the number of cells expressing mRNA for DMP1 was decreased. 
As it is known DMP1 and ALP are expressed in osteoblasts with an important role in endochondral ossification while TRAP represents a marker of osteoclasts function and bone resorption, so we can conclude  that maternal SHT in rats leads to delayed osteogenic differentiation and compromised cartilage to bone transition in early infantile period.

AB  - Uvod: Poznato je da su tireoidni hormoni (TH) od esencijalnog značaja za razvoj, funkciju i diferencijaciju svih ćelija u organizmu u toku prenatalnog i postnatalnog života. Prethodna istraživanja dovode u vezu smanjen nivo TH i razvoj reumatoidnog artritisa i drugih oboljenja zglobova, međutim mehanizam nastanka navedenih promena još uvek nije razjašnjen u potpunosti. U sinovijalnim membranama (SM) zglobova zahvaćenih reumatoidnim artritisom, utvrđeno je povećanje broja ćelija koje eksprimiraju PCNA (proliferating cell nuclear antigen). Cilj ovog istraživanja je bio da se definiše uticaj subkliničkog hipotireoidizma majki tokom graviditeta na SM novorođenih jedinki, s obzirom da u dosadašnjim istraživanjima ovaj efekat nije dovoljno proučen.
Materijal i metode: Subklinički hipotireoidizam je kod gravidnih jedinki Albino Oxford pacova prouzrokovan dodavanjem propiltiouracila kroz vodu za piće (1,5 mg/l), počevši od prvog dana graviditeta i tokom laktacije. Kontrolna grupa pacova nije tretirana. Mladunci su žrtvovani sedmog dana nakon rođenja. Histološka ispitivanja sprovedena su na parafinskim presecima debljine 5µm obojenim standardnim hematoksilin/eozin histohemijskim bojenjem, čija je analiza sprovedena na svetlosnom mikroskopu (Olympus BX53). Ekspresija PCNA (Santa Cruz Biotechnology, sc-7907) pokazana je imunohistohemijski, pomoću vizualizacionog kita- Rabbit Specific HRP/DAB (ABC) Detection IHC Kit (ab64261, Abcam, UK). Kvantifikacija ekspresije PCNA izvršena je pomoću softvera Image J. . Prikazan je broj PCNA pozitivnih ćelija u odnosu na 3000 ćelija SM.
Rezultati: Pregledom preseka bojenih hematoksilin/eozinom nisu uočene značajne razlike u građi SM između kontrolne i tretirane grupe. Broj ćelija koje eksprimiraju PCNA u SM tretiranih životinja je značajno veća od broja ćelija u SM kontrolnih životinja. 
Zaključak: Povećana ekspresija PCNA ukazuje na intenziviranu proliferaciju ćelija SM novorođenih pacova starosti 7 dana. Ovaj rezultat može biti osnova za nastavak istraživanja uticaja hipotireoidizma na razviće SM.

Stereological and immunohistochemical study of the spleen in hypothyroid juvenile rats

AB  - The purpose of this work was to investigate the influence of hypothyroidism on spleen tissue morphology and immune cell density in fourteen-day-old juvenile rats. Hypothyroidism in pups (n=10) was induced by administration of propylthiouracil (PTU) in drinking water (1.5 mg/L) to their mothers during pregnancy and period of lactation. Fourteen-day-old pups were sacrificed and the thyroid-stimulating hormone (TSH) serum concentration and thyroid activation index (Ia) were determined. Increased serum level of TSH and increased Ia showed that pups from PTU treated mothers were hypothyroid. White and red spleen pulp, marginal zone and connective tissue volume density has been assessed by using the stereological method. Using immunohistochemistry, the present CD3+ T lymphocytes, CD45RA+ B lymphocytes and CD68+ macrophages were quantified. A significant reduction of volume density of the periarteriolar lymphocyte sheath (Vv(PALS)) and lymphatic follicles (Vvf) due to depletion of T and B lymphocytes respectively, was observed in the spleens of hypothyroid pups compared to controls. The volume density of the red pulp (Vvrp), marginal zone (Vvmz) and connective tissue (Vvct) was increased, as well as the number of CD68+ macrophages in the spleens of hypothyroid pups compared to controls. These results indicate that thyroid hormones might be important for normal development of both, specific and innate immune cells in the spleen during prenatal and early postnatal period.

abstract = "The purpose of this work was to investigate the influence of hypothyroidism on spleen tissue morphology and immune cell density in fourteen-day-old juvenile rats. Hypothyroidism in pups (n=10) was induced by administration of propylthiouracil (PTU) in drinking water (1.5 mg/L) to their mothers during pregnancy and period of lactation. Fourteen-day-old pups were sacrificed and the thyroid-stimulating hormone (TSH) serum concentration and thyroid activation index (Ia) were determined. Increased serum level of TSH and increased Ia showed that pups from PTU treated mothers were hypothyroid. White and red spleen pulp, marginal zone and connective tissue volume density has been assessed by using the stereological method. Using immunohistochemistry, the present CD3+ T lymphocytes, CD45RA+ B lymphocytes and CD68+ macrophages were quantified. A significant reduction of volume density of the periarteriolar lymphocyte sheath (Vv(PALS)) and lymphatic follicles (Vvf) due to depletion of T and B lymphocytes respectively, was observed in the spleens of hypothyroid pups compared to controls. Characterization of deciduous teeth stem cells isolated from crown dental pulp

The last decade has been profoundly marked by persistent attempts to use ex vivo expanded and manipulated mesenchymal stem cells (MSCs), as a tool in different types of regenerative therapy. In the present study we described immunophenotype and the proliferative and differentiation potential of cells isolated from pulp remnants of exfoliated deciduous teeth in the final phase of root resorption. Methods. The initial adherent cell population from five donors was obtained by the outgrowth method. Colony forming unit-fibroblast (CFU-F) assay was performed in passage one. Cell expansion was performed until passage three and all tests were done until passage eight. Cells were labeled for early mesenchymal stem cells markers and analysis have been done using flow cytometry. The proliferative potential was assessed by cell counting in defined time points and population doubling time was calculated. Commercial media were used to induce osteoblastic, chondrogenic and adipogenic differentiation. Cytology and histology methods were used for analysis of differentiated cell morphology and extracellular matrix characteristics. Results. According to immunophenotype analyses all undifferentiated cells were positive for the mesenchymal stem cell markers: CD29 and CD73. Some cells expressed CD146 and CD106. The hematopoietic cell marker, CD34, was not detected. In passage one, incidence of CFU-F was 4.7 ± 0.5/100. Population doubling time did not change significantly during cell subcultivation and was in average 25 h. After induction of differentiation, the multicolony derived cell population had a tri-lineage differentiation potential, since mineralized matrix, cartilage-like tissue and adipocytes were successfully formed after three weeks of incubation. Conclusion. Altogether, these data suggest that remnants of deciduous teeth dental pulp contained cell populations with mesenchymal stem cell-like features, with a high proliferation and tri- lineage differentiation potential and that these cultures are suitable for further in vitro evaluation of cell based therapies.

Prošla dekada je bila posebno obeležena naporima na polju korišćenja ex vivo razvijenih i usmeravanih mezenhimskih matičnih ćelija (MSCs), kao sredstva za različite tipove regenerativne terapije. Cilj ove studije bio je da se utvrdi imunofenotip i potencijal za proliferaciju i diferencijaciju ćelija izolovanih iz zubne pulpe mlečnih zuba dece eksfoliranih u periodu kada je koren zuba bio u poslednjoj fazi resorpcije. Metode. Primarna adherentna populacija ćelija poreklom od pet donora dobijena je metodom eksplanta. Prisustvo progenitorskih ćelija koje obrazuju kolonije fibroblasta (CFU-F) pokazano je u prvoj pasaži. Do treće pasaže ćelije su ekspandirane, a potom korišćene za analiziranje. Imunofenotip je određen korišćenjem protočne citometrije. Proliferativni potencijal i vreme udvajanja ćelija (PDT) u kulturi je definisano na osnovu apsolutnog broja ćelija na početku i na kraju svake pasaže. Posle tronedeljne kultivacije ćelija u komercijalnim medijumima za stimulaciju osteogeneze, hondrogeneze i adipogeneze, citološkim i histološkim metodama je određena morfologija ćelija i karakteristike vanćelijskog matriksa. Rezultati. Antigeni koji karakterišu mezenhimske matične ćelije CD29 i CD73 su bili eksprimirani na svim nediferenciranim ćelijama, dok su antigeni CD146 i CD106 bili eksprimirani na ograničenom broju ćelija. Antigen CD34 (karakterističan za ćelije hematopoetske loze) nije bio eksprimiran. Incidencija CFU-F bila je 4,7 ± 0,5/100 ćelija. PDT se nije menjao tokom osam pasaža i u proseku je iznosio 25 h. Posle tronedeljne stimulacije diferencijacije u kulturama sa adipogenim medijumom došlo je do stvaranja ćelija sa masnim kapljicama, a u kulturama sa osteogenim medijumom došlo je do formiranja vanćelijskog matriksa sa deponovanim kalcijumovim solima. U kulturama sa hondrogenim medijumom došlo je do stvaranja tkiva sličnog hrskavici i vanćelijskog matriksa sa glikozaminoglikanima i kolagenom II. Zaključak. Zubna pulpa mlečnih zuba dece sadrži ćelijsku populaciju koja odgovara mezenhimskim matičnim ćelijama prema svojim karakteristikama, ima visok proliferativni potencijal i potencijal da se diferencira u tri ćelijske linije što je čini pogodnom za dalje in vitro analize i evaluaciju ćelijske terapije.

AB  - Histological evaluations of soft tissue reactions in implanted biomaterials is a long established method of investigating their biocompatibility. It is a very important procedure convenient for various biomaterials, as appropriate for soft as for hard tissue, but at the same time this method is not sufficient for a prediction of their clinic performance. The wide spread practice of this method is in the simplicity of its use in subcutaneous or intramuscular implantations on the animal model, and consequently exploring principal and surrounding tissue is very simple with screening tests for esimating the irritaton index and interaction between tissue and biomaterials. The screening tests (evaluation tests) involve implantation of biomaterials in various locations in the body, different tissue treatment as immunohistochemical tissue analysis, transmissional, scanning and confocal microscopy. Interactions among soft tissue with bioceramics and the evaluation of their cellular and tissue performance in inflammation and immunological response are the first step in the estimation of biocompatibility.
AB  - Histološko ispitivanje reakcije mekih tkiva u odnosu na biomaterijal je davno ustanovljena metoda procenjivanja biokompatibilnosti. To je istovremeno i veoma važna metoda koja je primenjiva na sve oblike biomaterijala, bilo da su namenjeni za kontakt sa mekim tkivom ili ne, ali i metoda koja ima ograničeni značaj za predviđanje kliničkih performansi. Rasprostranjenost korišćenja ove metode proizilazi iz činjenice da je potkožna ili intramuskularna implantacija potencijalnog biomaterijala na životinje jednostavna, a dalja ispitivanja reakcije osnovnog i susednih tkiva su u principu jednostavan skrining test za određivanje iritacije tkiva i interakcije tkiva i biomaterijala. Skrining tehnike (testovi procenjivanja) uključuju implantaciju materijala na različitim lokalizacijama, kao i različite načine obrade tkiva, primenu imunohistohemijskih metoda bojenja, transmisione, skening i konfokalne mikropskopije i primenu drugih sofisticiranih metoda u obradi i interpretaciji podataka. Interactions among soft tissue with bioceramics and the evaluation of their cellular and tissue performance in inflammation and immunological response are the first step in the estimation of biocompatibility., Histološko ispitivanje reakcije mekih tkiva u odnosu na biomaterijal je davno ustanovljena metoda procenjivanja biokompatibilnosti. To je istovremeno i veoma važna metoda koja je primenjiva na sve oblike biomaterijala, bilo da su namenjeni za kontakt sa mekim tkivom ili ne, ali i metoda koja ima ograničeni značaj za predviđanje kliničkih performansi. Rasprostranjenost korišćenja ove metode proizilazi iz činjenice da je potkožna ili intramuskularna implantacija potencijalnog biomaterijala na životinje jednostavna, a dalja ispitivanja reakcije osnovnog i susednih tkiva su u principu jednostavan skrining test za određivanje iritacije tkiva i interakcije tkiva i biomaterijala. Skrining tehnike (testovi procenjivanja) uključuju implantaciju materijala na različitim lokalizacijama, kao i različite načine obrade tkiva, primenu imunohistohemijskih metoda bojenja, transmisione, skening i konfokalne mikropskopije i primenu drugih sofisticiranih metoda u obradi i interpretaciji podataka. Interakcija mekog tkiva sa biokeramikom i procena tkivnog i ćelijskog prikaza u procesu inflamacije i imunološkom odgovoru prvi je korak u određivanju njihove biokompatibilnosti.",

Effects of induced maternal hypothyroidism on the ovarian development of offspring rats

AB  - The effects of propylthyouracil (PTU) induced hypothyroidism of rats during pregnancy and lactation on offspring ovarian development and maturation were studied. Thyroid hormones and thyroid stimulating hormone (TSH) concentrations were determined using the radioimmunoassay method in order to verify the hypothyroid status of treated mothers and their two months old pups. The ovaries of the offspring were processed for light microscopy analysis on the day of the first estrus after the 60th day of age. Histological analysis including follicle count was performed on serial sections stained with haematoxyline/eosin and on semithin sections stained with methylene blue. A significant increase of serum TSH and decrease in T3 and T4 levels was observed in treated mothers compared to controls. The levels of measured hormones in the control and PTU-treated two months old rats were not significantly different. Ten percent of 60-dayold treated females did not reach estrus and they were sacrificed in diestrus. The secondary interstitial cells were the dominant structures in the ovaries. The number of healthy growing and early antral follicles was markedly decreased. Ovaries of treated rats contained relatively few antral follicles, significantly more atretic antral follicles and a decreased number of corpora lutea, compared to controls. These results indicate that lack of thyroid hormones during prenatal and early postnatal development impair ovarian development in rats.
AB  - U okviru ovog rada izučavan je uticaj hipotireoidizma majki izazvanog propiltiouracilom (PTU) tokom graviditeta i laktacije na razvoj i sazrevanje jajnika pacova. U cilju određivanja tireoidnog statusa majki i mladunaca, merena je koncentracija tireoidnih hormona i tireostimulirajućeg hormona (TSH) u serumu. Jajnici potomaka hipotireoidnih majki i kontrolne grupe sprovedeni su za analizu svetlosnom mikroskopijom na dan prvog estrusa posle 60. dana starosti. Analizirani su serijski isečci jajnika bojeni hematoksilinom i eozinom, kao i polutanki preseci bojeni metilenskim plavim. Koncentracija T4 i T3 značajno je smanjena u serumu tretiranih majki, dok je nivo TSH značajno povišen u odnosu na kontrole. Kod dva meseca starih kontrolnih i tretiranih ženskih potomaka nije bilo značajne razlike u nivou svih određivanih hormona. Deset procenata tretiranih životinja nije normalno ušlo u estrus do 60. dana, i žrtvovane su u fazi diestrusa. Sekundarne intersticijalne ćelije su dominantne strukture u jajnicima tretiranih pacova. Ukupan broj aktiviranih folikula je značajno smanjen uz izrazitu atreziju folikula, a smanjen je i broj žutih tela. Rezultati pokazuju da hipotireoidizam izazvan u toku graviditeta i laktacije dovodi do promena u razviću jajnika.

AB  - Regenerative medicine is the field of human and veterinary medicine that involves creating living, functional tissue to repair
and replace tissues or organs that have been damaged by injury, disease, aging or birth defects. Regenerative adult cell
therapy with or without supporting biocompatible material, has been currently successfully treated in horses and dogs,
especially in joint, bone and ligament injuries.
The aim of this study was to evaluate the biocompatibility of three different bioceramical materials (BCM) implanted
(paravertebral lumbal region) in rat subcutaneous tissue according to the International standard procedures (ISO 10993-6;
ISO 10993-10) for screening tests by histological examinations. In order for biocompatibility phenomena to be investigated
and to choose of an implantable device we use the routine histological techniques (H&E, TB, AZAN),
immunohistochemical method (for labeling T and B lymphocytes and macrophages) and scanning electron microscopic
investigations on interface of biomaterials. Thirty two rats were assigned to 4 groups, as following: control group; dental
ceramic (DC) implants; alfa tricalcium phosphate (áTCP); hydroxyapatite (HAP). The irritation index (the level of tissue
injuring) was noted microscopically and tabelar system of evaluations were compared in grading scores from 0 to 4, where
results had been described from mild (1) to severe (4). The inflammatory infiltrate consisted of lymphocytes,
polimorphonuclears, macrophages, siderophages and mastocytes. At the same time there were sign of encapsulation and
neovasculogenesis around BCM.
In conclusion, the irritation index among the different BCM were in all cases in the range of biocompatibility. Evaluating
different responses according to BCM, the most intense tissue reaction was noticed at HAP, grading as severe and mild to
moderate for DC and áTCP.

AB  - There are very few data about the effects of endogenous growth factors in vital pulp therapy, and still they are often controversial. This study was undertaken with the aim to evaluate the effects of platelet rich plasma (PRP) in conjugation with hydroxyapatite (HAP), as pulp capping materials, on root and periodontium formation. Eight young monkeys (Cercopithecus Aethiops) with permanent dentition and incomplete root formation were involved in this study. After pulpotomy, the pulp lesion was capped with calcium hydroxide (control), HAP (experimental) or HAP in conjugation with PRP (experimental). Six moths later, the animals were sacrificed, the tissue was removed en block, and prepared for histological analysis in a routine way. The results of histological analysis revealed that the healing process, characterized by dentin bridge formation, maintained morphological and functional integrity of dental pulp and complete formation of dental root and surrounding periodontium. The inflammatory reaction was scored as mild to moderate, in almost all samples in all groups, suggesting the biocompatibility of the used materials. Materials used in this study are convenient as capping agents, contributing in maintaining the integrity of the pulp tissue and facilitating root and periodontium formation. According to histological data it could be suggested that hydroxyapatite in conjugation with endogenous growth factors, represent a superior alternative to other materials used in this study.
AB  - Podaci iz literature o primeni faktora rasta u vitalnoj terapiji pulpe su oskudni i često kontraverzni. Cilj našeg rada je bio da se ispita uticaj plazme bogate trombocitima u kombinaciji sa hidroksiapatitom, kao materijala za direktno prekrivanje pulpe, na završetak rasta korena i formiranje okolnog parodoncijuma. Istraživanje je sprovedeno na osam mladih majmuna (Cercopithecus aethiops), sa stalnom denticijom i nezavršenim rastom korena. Nakon pulpotomije, pulpna lezija je prekrivena kalcijum hidroksidom (kontolna grupa), hidroksiapatitom (eksperimentalna grupa I) ili hidroksiapatitom u kombinaciji sa plazmom bogatom trombocitima (eksperimentalna grupa II). Šest meseci kasnije životinje su žrtvovane, a uzeto tkivo pripremljeno je za histološku analizu. Rezultati histološke analize ukazali su da je zarastanje bilo karakterisano stvaranjem mineralizovanog dentinskog mostića, uz očuvanje morfološkog i funkcionalnog integriteta pulpe i završetak rasta korena i okolnog parodoncijuma u uzorcima svih ispitivanih grupa. Zapaljenska reakcija je gotovo u svim ispitivnim uzorcima bila ocenjena kao blaga ili umerena, što govori u prilog biokompatibilnosti primenjenih materijala. Na osnovu dobijenih rezultata može se reći da su svi primenjeni materijali pogodni za direktno prekrivanje pulpe, da doprinose očuvanju integriteta pulpe i ubrzavaju završetak rasta korena i formiranje apikalnog parodoncijuma, s tim što hidroksipatit u kombinaciji sa endogenim faktorima rasta predstavlja superiorniju alternativu u odnosu na druge materijale korišćene u ovoj studiji.

Cilj našeg rada je bio da se ispita uticaj plazme bogate trombocitima u kombinaciji sa hidroksiapatitom, kao materijala za direktno prekrivanje pulpe, na završetak rasta korena i formiranje okolnog parodoncijuma. Istraživanje je sprovedeno na osam mladih majmuna (Cercopithecus aethiops), sa stalnom denticijom i nezavršenim rastom korena. Nakon pulpotomije, pulpna lezija je prekrivena kalcijum hidroksidom (kontolna grupa), hidroksiapatitom (eksperimentalna grupa I) ili hidroksiapatitom u kombinaciji sa plazmom bogatom trombocitima (eksperimentalna grupa II). Šest meseci kasnije životinje su žrtvovane, a uzeto tkivo pripremljeno je za histološku analizu. Rezultati histološke analize ukazali su da je zarastanje bilo karakterisano stvaranjem mineralizovanog dentinskog mostića, uz očuvanje morfološkog i funkcionalnog integriteta pulpe i završetak rasta korena i okolnog parodoncijuma u uzorcima svih ispitivanih grupa. Zapaljenska reakcija je gotovo u svim ispitivnim uzorcima bila ocenjena kao blaga ili umerena, što govori u prilog biokompatibilnosti primenjenih materijala. Na osnovu dobijenih rezultata može se reći da su svi primenjeni materijali pogodni za direktno prekrivanje pulpe, da doprinose očuvanju integriteta pulpe i ubrzavaju završetak rasta korena i formiranje apikalnog parodoncijuma, s tim što hidroksipatit u kombinaciji sa endogenim faktorima rasta predstavlja superiorniju alternativu u odnosu na druge materijale korišćene u ovoj studiji.",

Histological and immunohistochemical evaluations of rat soft tissue response to bioceramical implants

AB  - The purpose of this study was to evaluate the tissue response induced by bioceramical materials (BCM) implanted in rat subcutaneous tissue. Thirty two rats were assigned to 6 groups, as following: control group i.e. not treated; dental ceramic (DC) implants; alfa tricalcium phosphate synthetised at 22 MPa (αTCP22) and at 45 MPa (αTCP45); hydroxyapatite synthetised at 22 MPa (HAP22) and at 45 MPa (HAP45). Contra lateral sides of all test animals represented the blank control surgically incisied at the paravertebral sockets. The experimental groups received subcutaneous implants of the test BCM materials. Samples were collected after 14 days and processed histologically. Inflammatory reactions were noted microscopically. The inflammatory infiltrate consisted of lymphocytes, polimorphonuclears, macrophages and mastocytes. At same time there were signs of increased fibrosis around the graft places. In conclusion, the implant sides compared with surgical incision wounds induced a foreign body-type granuloma with subsequent fibrosis around the grafts. The presence of a capsule at the interface with BCM in rat subcutis did not elicit any remarkable immune responses, thus being considered biocompatible. By evaluating different responses according to BCM, the most intense tissue reaction was noticed at HAP45, graded as severe, and mild to moderate for DC, HAP22 and alfa tricaltium phosphate.
AB  - Svrha ove studije je bila procena tkivnog odgovora indukovanog biokeramič kim materijalima (BCM) inplantiranim u subkutano tkivo pacova. Trideset dva pacova je bilo podeljeno u sledećih 6 grupa: kontrolna grupa-neoperisana; inplantirane sa: dentalnom keramikom (DC); alfa trikalcijum fosfatima sintetisanim na 22 MPa (αTCP22) i na 45 MPa (αTCP45); hidroksiapatitom sintetisanim na 22 MPa (HAP22) i na 45 MPa (HAP45). Kontralateralne strane svih životinja bile su slepa kontrola i urađene su kao džepovi hirurške incizije u subkutanom tkivu. Eksperimentalne grupe su imale subkutano inplantirane biokeramičke materijale. Uzorci su sakupljeni posle 14 dana i sprovedeni u histološkoj proceduri, a inflamatorna reakcija je praćena mikroskopski. Inflamatorni infiltrat se sastojao od limfocita, polimorfonuklearnih leukocita, makrofaga i mastocita. Istovremeno su uočeni znaci rastuće fibroze oko mesta graftova. Inplantirana mesta u poređenju sa hirurškim ranama indukuju granulom karakterističnog tipa na strano telo sa posledičnom fibrozom oko graftova, kapsulom na dodirnoj površini sa BCM u subkutisu pacova, ne izazivajući uočljivu imunološ ku reakciju, tako da odgovor tkiva ukazuje na biokompatibilnost. Procenjivanjem odgovora sa različitim BCM, zabeležena je najintenzivnija (snažna) tkivna reakcija kod HAP45 inplanta, a blaga do umerena kod DC, HAP22 i alfa trikalcijumfosfata.
