AB  - This study aimed to measure the total arsenic content deposited in the ovaries of
three consecutive generations of mice. The animals were treated with two different
concentrations to determine whether histological changes were caused in the ovaries.
The control group of mice received tap water, whereas the experimental groups were
given different concentrations of dissolved arsenic (III)-oxide. The arsenic content in
the ovaries in both experimental groups increased with each generation. The highest
content was recorded in the third generation of the second experimental group. Between
the two experimental groups in each generation, significant differences in the average
number of corpora lutea and ovarian follicles were identified. Arsenic caused structural
changes in the ovaries in both experimental groups in all three consecutive generations.

AB  - The supplementation of antimicrobial growth promoters (AGPs) has been banned in many
countries because of the emergence of antimicrobial-resistant pathogens in poultry products and
the environment. Probiotics have been broadly studied and demonstrated as a promising AGP
substitute. Our study is centred on the effects of a multi-strain Bacillus-based probiotic product
on broiler production performance and gut microbial profile in a dexamethasone-induced leaky
gut challenge. Two hundred and fifty-six broiler chicks were hatched and randomly assigned into
four groups (wheat-soybean meal basal diet (BD) = non-supplemented control (C), BD supplemented
with dexamethasone in week 4 (CD), BD containing a probiotic from day one (P), and BD containing
a probiotic from day one and supplemented with dexamethasone during challenge week 4 (PD)).
The production performance and caecal, gizzard, jejunal lumen and jejunal mucosa swab microbiota
were studied by 16S rRNA gene sequencing. The Bacillus probiotic product significantly improved
production performance and altered caecal gut microbiota (p ≤ 0.05), but no significant impact on
microbiota was observed in other gut sections.

Komparativni prikaz animalnih modela u morfološkim analizama placentacije

AB  - Svrha ovog pregleda je da prikaže veliku raznolikost u strukturi i funkciji placente koja se sreće kod sisara, i podobnosti nekih vrsta kao animalnih modela u proučavanju placentacije. Postoje značajne razlike u strukturi i fiziologiji posteljice između čoveka, domaćih i eksperimentalnih životinja. Zbog toga, najčešće upotrebljavani animalni modeli za proučavanje placentacije (miš, kunić, zamorac), nisu adekvatni za sva istraživanja u veterinarskoj i humanoj medicini. Placente se klasifikuju prema svom obliku, strukturi i interhemalnom regionu. Horionske resice placente su obložene prizmatičnim epitelom i preko njega se odvija prenos svih supstanci od majke prema plodu. Ćelije ovog epitela su selektivno propustljive kako za korisne, tako i za potencijalno toksične materije.
	Istraživanja koja se bave analizama i osobenostima placentacije su oskudna, posebno sa aspekta kontakta i razmene materija između majke i ploda. Iz tog razloga, ovaj uvid može da pomogne u sagledavanju različitih problema u kliničkoj praksi i tretmanu koji su bitni za dobijanje zdravog potomstva.

AB  - Epidermis stem cells have a crucial role through the processes of proliferation and
differentiation, to replace cells that are constantly lost during tissue turnover or
following injury. On the other hand, thyroid hormones regulate the proliferation and
differentiation of epidermal cells and thus significantly influence the homeostasis
of the skin. It is well known that maternal hypothyroidism during pregnancy leads
to impaired development of many organ systems in their offspring. However, there
is a lack of data about the influence of maternal subclinical hypothyroidism during
pregnancy and lactation on the development of the skin and its derivatives in the litter.
The aim of this study was to investigate the effects of maternal thyroid dysfunction on
the development of the skin and its derivatives in their offspring in the early postnatal
period. Antithyroid substance 6-n-propyl-2-thiouracil was added into the drinking water
to female Albino Oxfords rats from the beginning of pregnancy and during lactation,
with the aim to induce subclinical and overt form of hypothyroidism. Skin samples
were taken from male pups within twenty-four hours and seven days after birth. The
main findings of this investigation were that both forms of maternal hypothyroidism
lead to serious damage of the epidermis in pups in terms of pronounced hyperkeratosis
and reduction of the germinal layer along with a reduced number of hair follicles and
their delayed morphogenesis. Epidermal impairments were more pronounced in pups
with the overt form of hypothyroidism while offspring with the subclinical form had
impairments that were less pronounced and delayed in occurrence.

(P<0.01), and reduction of pathogenic bacteria (Escherichia coli) in guts of broilers than in the group fed only with the basic diet (P<0.05).
On the other hand, the group with the highest inclusion of yeast in diet (0.65 g/kg) achieved similar results for the examined parameters
as did the group with no added yeast. Th erefore, the adequate dose of S. cerevisiae in diet was 0.25 g/kg of yeast, while higher dose of live
yeast in broiler diet (0.65 g/kg) did not achieve better results.

There were significant differences (P<0.05) between all groups for the intestine length, intestine weight, and both height and width of the ileal villus. The highest villus height/crypt depth ratio of jejunum occurred in group II piglets, while the highest villus height/crypt depth ratio of ileum was in piglets from group III. The significant correlations were determined between amount of sodium butyrate and final live weight, intestinal length, intestinal weight, jejunal and ileal crypt depth, and ileal villus widths.

AB  - The concept of designer microbiota in chicken is focused on early exposure of the hatchlings
to pathogen-free microbiota inoculum, limiting the early access to harmful and pathogenic
microorganisms, thus promoting colonisation of the gut with beneficial and natural poultry microbiota.
In this study, we controlled colonisation of the intestine in broiler chickens in a large-scale
industrial setting via at-hatch administration of a commercial product containing a highly diverse
microbiota originating from the chicken caecum. The treatment significantly transformed the microbiota
membership in the crop, proventriculus, jejunum and caecum and significantly altered the
taxa abundance in the jejunum, jejunum mucosa, and caecum estimated using PERMANOVA and
unweighted and weighted UniFrac distances, respectively. The treatment also improved the growth
rate in chickens with no significant alteration in feed conversion ratio. A comparison of inoculum
product microbiota structure revealed that the inoculum had the highest Shannon diversity index
compared to all investigated gut sections, and the number of Observed Species second only to the
caecal community. PCoA plots using weighted or unweighted UniFrac placed the inoculum samples
together with the samples from the caecal origin.

The effects of continual consumption of origanum vulgare on liver transcriptomics

AB  - Pathogen control is re-emerging as a significant challenge to the health of both humans and animals. The livestock industry is in the process of massively replacing in-feed antibiotics with organic production friendly plant-based products. Nutrigenomics as a science of the effects of food constituents on gene expression is shedding more light on both benefits and detrimental side-effects of feed additive prolonged consumption on the host, indicating the need to understand the feed– host interactions and their influence on the host disease profile. In this study, we investigated the effects of 2% oregano powder supplementation on the liver gene expression in healthy male broilers from the hatch to 6 weeks of age. Deep RNAseq was performed on average 113.3 million paired and quality trimmed sequences per sample and four samples for the control and treatment each. The results demonstrate the severity of oregano effect on liver gene expression with substantial modifications in steroid hormone regulation, fat and carbohydrate metabolism alterations and strong influence on the host disease and function profile. Oregano supplementation was able to interfere with the transcriptional effects of a range of registered drugs and to significantly transcriptionally inhibit a range of cancer disease categories including liver cancer, and to modify fat and carbohydrate metabolism.

Histological and histochemical analysis of dry fermented sausage of kulen composition

AB  - The application of histological methods in meat composition analysis and identification of prohibited tissues and organs added to meat and meat products is still in the research phase, although there have been some promising results. The aim of this work was to assess the possibility of using histological and histochemical methods for analysis of kulen composition. In this research, six samples of kulen were examined, one of which was produced in domestic conditions, while the rest were commercial products sampled from local markets. The samples were carried through classical histological preparation. The obtained slides were stained with haematoxylin/eosin, Masson-Goldner, toluidine blue and periodic acid-Schiff/alcian blue. The content of muscle, fat and connective tissue was evaluated using histomorphometric analysis. Histological analysis of kulen composition determined the presence of the following structures: muscle, adipose and connective tissues, blood vessels, glandular epithelium, peripheral nerve, cartilage and plant tissue. The histomorphometric analysis showed that the kulen products contained on average 56±2.52% muscle tissue, 7.27±1.38% connective tissue and 19.82±3.24% adipose tissue. The results show that by applying histological methods it is possible to identify different permitted and prohibited animal tissues in kulen, and hence, it is possible to analyse the composition of kulen. It is also possible to confirm the presence of various plant tissues, but for their precise identification, additional histological methods are needed.
AB  - Primena histoloških metoda u analizi sastava mesa i utvrđivanju prisustva nedozvoljenih tkiva u mesu i proizvodima od mesa je još uvek u fazi istraživanja, iako su postojeći rezultati u ovoj oblasti obeć avajuć i. Cilj našeg istraživanja bio je ispitivanje moguć nosti korišć enja histoloških i histohemijskih metoda u analizi sastava fermentisane kobasice u tipu kulena. U ovom istraživanju ispitano je šest uzoraka kulena, od kojih je jedan proizveden u domać im uslovima, dok su ostali komercijalni proizvodi poticali iz lokalnih marketa. Uzorci su sprovedeni klasičnom histološkom procedurom. Dobijeni isečci bojeni su metodama: hematoksilin/ eosin,Masson-Goldner, toluidin plavo i perjodna kiselina/Schiff/alcijan plavo. Sadržaj mišić nog, masnog i vezivnog tkiva procenjen je primenom histomorfometrijskih analiza. Histološkom analizom sastava kulena utvrđeno je prisustvo sledeć ih struktura: mišić no, masno i vezivno tkivo, krvni sudovi, epitel žlezdanog tipa, periferni nerv, hrskavica i biljno tkivo. Histomorfometrijska analiza je pokazala da proizvodi sadrže u proseku 56±2,52% mišić nog tkiva, 7,27±1,38% vezivnog tkiva i 19,82±3,24% masnog tkiva. Rezultati pokazuju da je primenom histoloških metoda moguć e identifikovati različita dozvoljena i nedozvoljena životinjska tkiva u kulenu, pa je samim tim moguće analizirati sastav kulena. Takođe je moguć e potvrditi prisustvo različitih biljnih tkiva, ali je za njihovu preciznu identifikaciju neophodno primeniti dodatne metode.

Cilj našeg istraživanja bio je ispitivanje moguć nosti korišć enja histoloških i histohemijskih metoda u analizi sastava fermentisane kobasice u tipu kulena. U ovom istraživanju ispitano je šest uzoraka kulena, od kojih je jedan proizveden u domać im uslovima, dok su ostali komercijalni proizvodi poticali iz lokalnih marketa. Uzorci su sprovedeni klasičnom histološkom procedurom. Dobijeni isečci bojeni su metodama: hematoksilin/ eosin,Masson-Goldner, toluidin plavo i perjodna kiselina/Schiff/alcijan plavo. Sadržaj mišić nog, masnog i vezivnog tkiva procenjen je primenom histomorfometrijskih analiza. Histološkom analizom sastava kulena utvrđeno je prisustvo sledeć ih struktura: mišić no, masno i vezivno tkivo, krvni sudovi, epitel žlezdanog tipa, periferni nerv, hrskavica i biljno tkivo. Histomorfometrijska analiza je pokazala da proizvodi sadrže u proseku 56±2,52% mišić nog tkiva, 7,27±1,38% vezivnog tkiva i 19,82±3,24% masnog tkiva. Rezultati pokazuju da je primenom histoloških metoda moguć e identifikovati različita dozvoljena i nedozvoljena životinjska tkiva u kulenu, pa je samim tim moguće analizirati sastav kulena. Takođe je moguć e potvrditi prisustvo različitih biljnih tkiva, ali je za njihovu preciznu identifikaciju neophodno primeniti dodatne metode.",

cell cycle in hypothyroid animals is faster and that the basal level of autophagy is reduced due to frequent mitosis. Compared to controls, caspase 3 expression in the terminal
hypertrophic chondrocytes was reduced in H1 and completely absent in H2 pups which might indicate impaired differentiation.We conclude that both forms of maternal hypothyroidism
in rats lead to accelerated proliferation, slowed autophagy and
compromised apoptosis of terminal hypertrophic chondrocytes in the
early infantile period and a delay in cartilage to bone transition.

AB  - The objective was to determine optimal thresholds of blood variables that are associated with liver lipid and glycogen content in dairy cows. Liver specimens of 15 Holstein dairy cows were taken 2 weeks postpartum (PP) and used for determination of lipid and glycogen content, respectively. Blood samples were collected weekly, starting from week 2 antepartum (AP) up to week 2 PP. Selected blood parameters and body condition scoring (BCS) were evaluated through receiver operating characteristic curve analysis as indication of fat and glycogen content in the liver. Postpartal low liver glycogen content (mode < 2.0, semiquantitative analysis of glycogen content in the liver tissue samples) can be indicated if values are higher than 0.16 mmol/L, and 3.5 mmol/L for NEFA, and glucose, respectively, and lower than 0.65 mmol/L and 17.94 mu U/mL for BHBA and insulin, respectively, 2 weeks AP. Postaprtal high liver fat content (>20%, stereological quantification of fat content in the liver tissue samples) can be indicated if values are higher than 3.5 for BCS at week 2 AP. Optimal time for indicating both low liver glycogen/high fat content is 1 week PP, with LDL < cut off point set at 0.18 mmol/L and BHBA > cut off point set at 0.88 mmol/L. Established cut off values in this study can be used for indication of fat and glycogen content in the liver of dairy cows.

AB  - The effect of dietary genistein on performance, intestinal morphology, caecal Lactobacillus spp. count, and tibia composition in broiler chickens after 21 and 37 days of feeding was investigated. A total of 360 Cobb 500 broiler chickens (21 days old) were randomly allocated to five treatments with six replicates of 12 birds each. They were fed a basal diet (C) or a basal diet supplemented with 200 (T1), 400 (T2), 600 (T3) and 800 (T4) mg genistein/kg of feed. Genistein supplementation did not affect feed intake, but improved bodyweight, weight gain and feed conversion ratio (FCR) after 21 days, while 600 mg/kg led to a significant increase in FCR after 37 days of feeding. Plasma triglyceride level decreased with dietary genistein after 21 days, while increases were found in T3 and T4 groups after prolonged supplementation. Significantly improved duodenal and jejunal villus length and width, crypt depth and villus/crypt ratio were observed after the first and the second finishing periods, respectively, while adverse effects were found in the ileum for both periods. At 42 days old, greater spleen and heart weights were measured in broilers fed diets with 800 mg/kg than in other broiler groups. The shorter genistein supplementation period (21 days) of 200 and 400 mg/kg had a positive effect on tibia wet weight, ash and calcium (Ca) content, while 37 days of the higher genistein doses administered to the T2, T3 and T4 birds significantly increased caecal lactic acid bacteria (LAB) counts. Thus, recommended doses should not exceed 400 mg/kg.

AB  - Radni konji se u Srbiji koriste kao tovarne životinje koje nose/vuku tovar, često teži od njihove telesne mase. Postavili smo hipotezu da opterećenje koje trpe utiče na pojavu lezija na koštano-zglobnom sistemu. Cilj ovog rada je bio da se uporedno prikaže prisustvo i stepen oštećenja zglobne hrskavice (ZH) i subhondralne kosti (SHK), distalnih kondilusa treće metatarzalne kosti kod starijeg radnog konja pomoću makroskopskog pregleda hrskavice, radioloških i histoloških metoda. Na lokalnoj klanici uzorkovani su kičični zglobovi ekstremiteta radnog pastuva, starog 10 godina. Uz pomoć kompjuterizovane tomografije (CT) utvrđene su promene samo na zglobu prednje leve noge. Nakon disekcije označenog zgloba, izvršena je biopsija ZH i SHK metakarpalne kosti u predelu oštećenja, a bioptati su snimljeni mikro CT-om nakon čega su deklacifikovani i ukalupljeni u parafin, i analizirani su preseci. Makroskopski je bilo vidljivo da je lokalizacija oštećenja ZH bila simetrična na medijalnim i lateralnim epikondilusima matakarpalnih i metatarzalnih kostiju svih ekstremiteta. Linije habanja i palmarne artroze bile su slabo do umereno izražene. Na nativnim rendgenogramima zglobova nisu uočene patološke promene. Analizom snimaka načinjenih CT-om uočena je promena u subhondralnoj zoni distalne epifize treće metakarpalne kosti u vidu pukotine, a oko nje je primetna zona skleroze SHK. Na mikro CT snimku uzorka koštanog tkiva uzetog u predelu lateralnog epikondilusa metakarpane kosti, takođe je uočljiva zona subhondralne skleroze. Najmarkatnije histološke promene na ZH su bile: na površini ZH undulacije i fisure, koje se pružaju i do duboke zone, mnogobrojne izogene grupe sa većim brojem hondrocita u lakuni, fokalni gubitak ćelija i mestimičnio prisustvo hipertrofičnih hondrocita. U SHK su primećene: neznatne lezije koštanog matriksa u vidu svetlijeg prebojavanja, blago remodeliranje kosti koje se karakteriše njenim napredovanjem ka kalcifikovanoj hrskavici. Dugotrajno opterećenje utiče na pojavu oštećenja SHK koje se može uočiti uz pomoć CT-a i histološkom analizom, dok se stepen oštećenja ZH može evaluirati makroskopski i histološki.

Oregano: A potential prophylactic treatment for the intestinal microbiota

AB  - Prophylactic use of antibiotics in poultry diets has been identified as a problematic practice because of its potential to exacerbate the spread of antibiotic resistance to human pathogens. A range of countries have opted to completely ban the use of antibiotics in animal feed. The animal production industries are looking for alternative ways to effectively control pathogens while providing the performance benefits previously secured by antibiotics in feed. Here, we present evidence that oregano (Origanum vulgare) could be a potential alternative for pathogen control in the poultry industry. Broiler diets were supplemented with oregano powder (0%, 0.5%, 1%, and 2%) for six weeks. The capacity for pathogen control was estimated by microbiota profiling of the jejunum, ileum, and caecum content, and in the faeces, by 16S rRNA gene amplicon sequencing. The concentrations of short-chain fatty acids in the caecal content were also measured, as were villus/crypt parameters in the ileum. There were no differences among treatments in weight gain, feed intake, or the concentration of short-chain fatty acids. The height, width, and the surface area of villi in the ileum were not influenced by oregano addition. However, 1% and 2% of oregano produced a significant increase in the villus height to crypt depth ratio. There were no visible histopathological changes in the liver in control and treated groups. Although oregano had no significant effect on overall microbial diversity and gross composition, some specific genera, like Proteus, Klebsiella and Staphylococcus, which include known pathogens, were reduced in relative abundance by oregano treatment. Bifidobacterium, recognized as a beneficial and probiotic genus, was also suppressed by the oregano treatment.

AB  - The stimulative effects of thyroid hormones on mitochondria are realized through 
both non-genomic and genomic mechanisms, affecting respiration, mitochondrial plasticity 
and biogenesis. The subclinical form of maternal hypothyroidism in rats induces significant 
reduction of mitochondria number but also an augmentation of their area in neonatal and 
early infantile offspring dyctiotene oocytes. This study aimed to investigate if this form of 
subclinical hypothyroidism affects mitochondrial morphology and distribution in the early 
prophase of meiosis I oocytes. It was performed on newborn control (C) (n=10) and 
hypothyroid (SCH) (n=10) female rat pups derived from control (n=6) and propylthiouracil
treated pregnant dams (n=6), respectively. Ovaries of all pups were removed and processed 
for transmission electron microscopy. The morphological features of mitochondria in the 
early prophase I oocytes until dyctiotene were assessed. No substantial differences were 
found in leptotene and zygotene oocytes in SCH group comparing to control, except just a 
few mitochondria characterized with shortened cristae, presence of wide pale area centrally 
positioned and membrane disruption. Pachytene mitochondria in treated pup oocytes were 
in great extent with disrupted membrane, shortened cristae and wide pale area centrally 
positioned while these features were rarely observed in control ones. Our results confirm 
altered mitochondria morphology found in primordial and primary follicles in case of 
maternal hypothyroidism, indicating their impaired function and possibly, propensity to 
programmed cell death. Further investigations may indicate to what extent pachytene, as a 
meiotic checkpoint, appears to be a milestone possibly predetermining the future of the 
cell.

AB  - The aim of the present study was to assess the effects of medium-chain fatty acid (MCFA) diet supplementation on growth performance, intestinal histomorphology, serum biochemistry and intestinal microflora of broiler chickens. The study was performed on 180 one-day-old broilers of the same origin (Cobb 500 hybrid), over a 42-day period. They were fed diets supplemented with three treatments: control group (basal diet without supplementation); group with MCFA supplementation; and group with MCFA and coccidiostat supplementation. Broiler chickens fed diets supplemented with MCFAs had a significantly greater final bodyweight. The weights of carcass cuts (breast, drumsticks with thighs and wings) were greater in broilers receiving MCFAs than in control broilers. The addition of MCFAs to broiler diet significantly increased villus length and crypt depth in the duodenum and caecum, and significantly decreased villus width in the duodenum and ileum. Additionally, serum HDL-cholesterol and triglyceride concentrations were increased significantly in broilers with MCFA dietary supplementation. The results indicated that the MCFA diet supplementation had a beneficial effect on the performance of broiler chickens, their intestinal histomorphology and microflora.

Effect of subclinical and overt form of rat maternal hypothyroidism on offspring endochondral bone formation

AB  - Maternal hypothyroidism in its overt form affects skeletal development of the offspring, but these data are not available for the subclinical form which is becoming very frequent among pregnant women. We hypothesized that the subclinical form of hypothyroidism in rat dams, influences the process of offspring endochondral ossification affecting proliferation and differentiation of chondrocytes, osteoclasts and osteoblasts in pups. Seven-day-old male pups (n=18) derived from control dams and dams treated with a low dose (1.5 mg/L) or high dose (150 mg/L) of propylthiouracil in drinking water during pregnancy and lactation were used. Histomorphometric analysis of pups tibia proximal growth plate, expression of mRNA, immunohistochemical and histochemical visualization of extracellular matrix components was performed. The length of the tibia was reduced in hypothyroid pups. Secretion of type 2 and 10 collagens in the subclinical and overt form were lower while the amount of glycosaminoglycans was higher when compared with controls. Down-regulated tartrate resistant acid phosphatase mRNA indicated altered osteoclasts function while lower expression of dentin matrix acid protein-1 mRNA and reduced synthesis of type 1 collagen accentuated a compromised bone formation in the overt form of hypothyroidism. The subclinical form of maternal hypothyroidism had a negative effect on the differentiation of hypertrophic chondrocytes and calcified cartilage removal in 7-day-old pups. In addition, overt hypothyroidism had a negative effect on the proliferation of chondrocytes and deposition of osteoid. Both forms of hypothyroidism resulted in a decrease of tibia length due to changes in growth plate formation.

Effects of short-term fasting on lipid and lipoprotein concentrations in healthy lean dogs

AB  - Introduction. Analysis of canine lipoprotein fractions after agarose gel electrophoresis (agEF) separation could be an important diagnostic tool in primary and secondary dyslipidemia diagnosis. The aim of this study was to measure concentrations of triglycerides and cholesterol and to analyze lipoprotein fractions in dogs after basal (12 hours) and short-term (24 and 36 hours) fasting, i.e., frequent conditions in clinical practice. Materials and Methods. Blood samples were collected from six lean dogs of both sexes and different breeds, after 12, 24 and 36 hours of fasting. Concentrations of glucose, triglycerides and cholesterol were determined on an automated wet biochemistry analyzer, lipoprotein fractions were separated by agEF and leukocyte numbers were assessed on an automated hematology analyzer. Results and Conclusions. Results showed there was no significant change in glucose, triglyceride and cholesterol concentrations nor in leukocyte numbers during dog fasting. Moreover, there was no change in α1- and α2-fractions, but there was a significant decrease in preβ- and β-fraction of lipoproteins. It is know that high density lipoproteins (HDL) have α-mobility and very low density (VLDL) and low density lipoproteins (LDL) have preβ- and β-mobility. Thus, it is possible that reverse cholesterol transport maintained by HDL is not affected during short-term fasting. On the contrary, synthesis of VLDL and formation of LDL are probably decreased because endogenous synthesis of triglycerides is decreased or their clearance is increased.
AB  - Uvod. Poznato je iz literature da se odnosi lipoproteinskih frakcija seruma pasa koje se dobijaju posle elektroforeze na agaroznom gelu (EFag) menjaju kod oboljenja praćenih dislipidemijama, što može biti jedan od značajnih elemenata dijagnostike pojedinih patoloških stanja. Stoga je cilj ovih ispitivanja bio da se utvrdi koncentracija triglicerida, holesterola i glukoze u serumu pasa kao i odnos lipoproteinskih frakcija dobijenih pomoću EFag, u uslovima koji se često sreću u kliničkoj praksi: posle bazalnog (12 časova) i kratkotrajnog gladovanja od 24 i 36 časova. Materijal i metode. Uzorci krvi su sakupljeni od šest pasa različitih rasa, normalne telesne kondicije, oba pola, posle 12, 24 i 36 časova gladovanja. Koncentracija glukoze, triglicerida i holesterola je određena na automatskom biohemijskom analizatoru. Lipoproteinske frakcije su razdvojene korišćenjem EFag. Broj leukocita je utvrđen na hematološkom analizatoru. Rezultati i zaključak. Rezultati ovog ispitivanja su pokazali da se tokom kratkotrajnog gladovanja kod pasa koncentracija glukoze, triglicerida i holesterola kao i broj leukocita ne menjaju. Osim toga, nije bilo promena vezanih za α1-i α2-elektroforetske zone, ali se javio značajan pad koncentracije lipoproteina preβ-i β-zone. Poznato je da se lipoproteini velike gustine (HDL) nalaze u α-zoni, dok lipoproteini veoma male gustine (VLDL) i lipoproteini male gustine (LDL) pripadaju preβ-i β-zoni. Na osnovu naših nalaza moguće je zaključiti da reverzni transport holesterola koji se odvija posredstvom HDL-a nije poremećen tokom kratkotrajnog gladovanja. Nasuprot tome, kratkotrajno gladovanje verovatno utiče na smanjen obim sinteze triglicerida u jetri, odnosno na smanjenu sintezu VLDL-a i LDL-a ili na njihovo efikasnije uklanjanje iz plazme.

Ovarian surface epithelium of hypothyroid newborn and neonatal rats: From PCNA and caspase-3 perspective

AB  - Introduction. The ovarian surface epithelium (OSE) undergoes intensive regeneration and remodelling after each ovulation during the whole reproductive period. This process increases the risk of one of the most common ovarian tumors in women and the female dog. Considering the fact that maternal hypothyroidism highly impacts cell proliferation and cell death during folliculogenesis in the early neonatal period, we aimed to analyze its effect on OSE morphology and dynamics. Materials and Methods. The study was performed on newborn (24-h-old) and neonatal (4-day-old) female rats, a randomized trial between the control and hypothyroid groups, born under controlled circumstances and hypothyroid mothers, respectively. Their ovaries were analyzed histologically and processed to determine the OSE cell height as an average value of four measurement points. Also, the immunopositivity of the proliferating cell nuclear antigen (PCNA) and caspase-3 were assessed semiquantitatively. Results and Conclusions. No major structural differences of OSE were found between groups within the given ages except for a slight increment of OSE cell height and incompleteness of apical cell membrane with cytoplasmic projections in hypothyroid animals. PCNA immunopositivity of the OSE cells was higher in ovaries of hypothyroid animals of both ages in comparison to the controls. Moreover, only scarce OSE cells were caspase-3 positive in both groups and ages, with no difference in immunopositivity. Our study confirms the impact of hypothyroidism in the early postnatal period on morphology and proliferation rate of OSE cells, with no effect on caspase-3 dependent cell removal, which may serve as a premise for future investigation of potential carcinogenesis, in terms of prevention and treatment of ovarian cancer.
AB  - Uvod. Tokom reproduktivnog perioda, nakon svake ovulacije, površinski epitel (PE) jajnika se intenzivno regeneriše i remodelira. Ovaj proces povećava rizik od nastajanja jednog od najčešćih tumora jajnika kod žena i kuja. Imajući u vidu činjenicu da hipotireoidizam majki u velikoj meri utiče na proliferaciju i ćelijsku smrt u toku folikulogeneze u ranom postnatalnom periodu, cilj rada je bio da analiziramo efekat hipotireoidizma majki na morfologiju i dinamiku PE jajnika potomaka. Materijal i metode. U eksperimentu su korišćeni tek rođeni (24 časa stari) i neonatalni (4 dana stari) mladunci koji potiču od kontrolnih i hipotireoidnih majki. Morfometrijskom analizom određivana je prosečna visina ćelija PE, izmerena na četiri mesta na svakom ispitivanom preseku jajnika. Takođe, imunopozitivnost proliferativnivnog ćelijskog nuklearnog antigena (PCNA) i kaspaze 3 je određivana semikvantitativno. Rezultati i zaključci. Veće promene u strukturi PE jajnika nisu zapažene između grupa, osim blagog povećanja visine ćelija i nepotpune apikalne membrane sa projekcijama citoplazme kod hipotireoidnih životinja. AB  - The aim of this study was to evaluate the effects of immunocastration on pork meat and carcass quality, compared to meat from surgically castrated males and entire males. Ninety (Duroc x Pietrain) x (Landrace x Yorkshire) crossbred pigs were assigned to three experimental groups: surgically castrated males (barrows; castrated up to the seventh day of age), entire males (males), and vaccinated males (immunocastrates). Carcass and meat quality characteristics such as weight of hot and chilled carcass, meat yield, chilling loss and chemical parameters were examined. Surgically castrated pigs had significantly lower (p lt 0.01; p lt 0.05) weight before slaughter, than males and immunocastrates, and also lower (p lt 0.01) warm carcass weight than males. The average carcass meatiness of castrates was significantly lower (p lt 0.01) than the average meatiness of males and immunocastrates. Chilling loss of barrows was significantly lower (p lt 0.01) than chilling loss of males or immunocastrates. It was also found that the chilling loss of immunocastrates was significantly lower (p lt 0.05) than chilling loss of males. According to the results obtained, it can be concluded that immunocastration could be a good alternative to surgical castration considering meat and carcass quality characteristics.

AB  - Induced prenatal hypothyroidism in rat pups leads to accelerated primordial follicle assembly and premature follicular atresia with ovary failure. This work investigates the influence of maternal hypothyroidism induced with 6-n-propyl-2-thyouracil (PTU) on the number and morphology of oogonia/oocytes in newborn rat pups with light and transmission electron microscopy. Expression of apoptosis and autophagy markers in oogonia/oocytes were examined using immunohistochemistry. Hypothyroid newborn pups had a decreased number of mitotic and resting oogonia, while the number of altered oogonia/oocytes was increased. Ultrastructural observations revealed the increased presence of degenerated pachytene oocytes (Z-cells) and oogonia/oocytes undergoing autophagy, apoptosis and combined apoptosis and autophagy, in this group. The most abundant altered oogonia/oocytes in the hypothyroid group were those with morphological features of advanced autophagy and Z-cells. The percentage of TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) positive oogonia/ oocytes was significantly lower in the hypothyroid group. No significant difference was recorded in the expression of caspase-3, ATG7 and LC3 possibly reflecting that these proteins were not involved in the oogonia/ oocyte alteration process during prenatal rat hypothyroidism. The obtained results indicate that developmental hypothyroidism in the offspring enhances the number of Z-cells and oogonia/ oocytes altered with the advanced process of autophagy.

AB  - Rabbit is the third most commonly used animal model in different fields of scientific research, such as reproductive biology, fertility and embryo transfer studies, and immunology. This animal species, often used in antibodies production, has minority of scientific records about the immunological status of its reproductive organs. The aim of this study was to find histological and immunological changes in rabbit female reproductive tract during different reproductive stages. The study was carried out on female rabbits, divided in three groups, according to the following stages of reproductive cycle: Estrous, ovulation and pregnancy. Histological and immunohistochemical stains for T- and B-cells were performed on tissue samples of cornu uteri and cervix. T lymphocytes were predominant in all anatomical parts of the uterus, in all stages of the cycle. The highest number of those cells was recorded at estrous, while the lowest was recorded at pregnancy. Cervix expressed more immunological activity than cornu uteri. The distribution and the number of immune positive cells in the rabbit female reproductive tract depend on its hormonal status.

AB  - Introduction. Thyroid hormones (TH) play a pivotal role in the development of mammalian skin and are necessary for both, the initiation and maintenance of hair growth. Hypothyroid rats showed  impaired epidermal proliferation, hair growth, and wound healing. Stem cells located at the bulges of the hair follicles are responsible for hair cycling and contribute to the regeneration of the new epidermis after wounding. Therefore a reduction in the num¬ber or function of the bulge stem cells could be responsible for this phenotype. 
We hypothesized that decrease in TH during prenatal development would affect bulge stem cell population.
Material and methods. Subclinical hypothyroidism was induced with propylthiouracil through drinking water in dose 1.5 mg/l in pregnant Albino Oxford rats from the first day of gravidity. Control group was untreated. Seven-day-old pups were euthanized and skin samples were taken from dorsal part of their bodies. For the immunohistochemical study PCNA (Santa Cruz Biotechnology) and NANOG (Thermo scientific) antibodies were used.
Results. . The number of hair follicles was reduced in hypothyroid pups compared to 
controls. They also had a decreased expression of PCNA in cells of inner and outer sheat of hair follicles, but increased number of NANOG positive bulge stem cells. 
Conclusion. This suggests that re¬duced activation of stem cells leading to their accumulation in the bulges which is associated with reduced stem cell prolif¬eration and mobilization.
